Claims (17)
- 염기성 물질을 생산하는 능력을 갖는 미생물을 액체 배지를 함유하는 발효조 중에서 배양하여 당해 배지 중에서 염기성 물질을 생성 및 축적시킴을 포함하고, 전체 배양공정 내의 적어도 일부의 기간 동안, 배지중 총 암모니아 농도를 일정한 농도 범위로 조정함으로써, 염기성 물질의 카운터 이온으로서 사용되는 황산 이온 및/또는 염화물 이온의 사용양을 감소시킴을 특징으로 하는, 발효에 의한 염기성 물질의 제조법.
- 제1항에 있어서, 총 암모니아 농도의 일정 범위가,(A) 배지 중의 암모늄 이온 농도는, 배지 중에 용존된 중탄산 이온 및/또는 탄산 이온 및 다른 음이온의 이온 당량의 합계가 배지 중에 축적된 염기성 물질 중의 이온화된 염기성 물질의 이온 당량보다 커지도록 하는 농도이고,(B) 배지 중의 총 암모니아 농도는, 미생물을 배지의 pH 및 총 암모니아 농도를 변경하여 배양하고, 염기성 물질의 생산성을 측정하고, 최적 조건에서의 염기성 물질의 생산성의 50% 이상의 생산성을 나타내는 총 암모니아 농도를 각각의 pH에서 측정함으로써 미리 결정된 상기 미생물의 염기성 물질의 생산을 억제하지 않는 농도인 조건을 만족시키는 범위인, 발효에 의한 염기성 물질의 제조법.
- 제1항에 있어서, 총 암모니아 농도의 일정 범위가,(A') 목적하는 염기성 물질의 카운터 이온원으로서 pH 7.2 이하에서 배양하기에 충분한 양의 황산 이온 및/또는 염화물 이온을 함유하고 pH가 암모니아 가스, 암모니아수 및 요소 중 하나 이상을 첨가함으로써 pH 6.5 내지 pH 7.2의 범위로 유지되는 배지 중에서 배양하고, 염기성 물질의 생산성을 측정하고,(B') 상기 단계(A')에서 사용되는 배지 성분에서 황산 이온 및/또는 염화물 이온을 목적하는 양만큼 저하시킨 배지를 사용하여 배양을 실시하고, 목적하는 염기성 물질의 축적에 의해 카운터 이온인 황산 이온 및/또는 염화물 이온이 부족됨으로 인해 배지의 pH를 7.2 이하로 유지할 수 없는 기간 동안에는 총 암모니아 농도를 변화시켜 배양을 실시하여, (A')에서 측정된 생산성의 50% 이상의 생산성을 제공하는 총 암모니아 농도의 범위를 측정함으로써 미리 측정되는, 발효에 의한 염기성 물질의 제조법.
- 제1항에 있어서, 적어도 일부의 기간이 목적하는 염기성 물질의 축적에 의해 카운터 이온이 부족됨으로써 배지의 pH가 상승하는 기간 및 배지에 양이온을 첨가함으로써 pH가 상승하는 기간 중 하나 이상을 포함하는, 발효에 의한 염기성 물질의 제조법.
- 제1항에 있어서, 배지 중의 총 암모니아 농도가, 배지 중의 용존 산소 농도, 배지 중의 탄소원의 소비속도, 배지의 탁도, 염기성 물질의 생산성 및 배지의 pH 변화를 지표로 하여 측정되는 바에 따라 미생물의 활성이 저하되거나 정지할 때에 배지에 암모니아 또는 요소를 첨가함으로써 조정됨을 특징으로 하는, 발효에 의한 염기성 물질의 제조법.
- 제1항에 있어서, 배지로서, pH 7.2 이하에서 배양하기에 충분한 양의 황산 이온 및/또는 염화물 이온을 함유하는 배지의 조성으로부터 황산 이온 및/또는 염화물 이온을 목적하는 양만큼 감소시킨 배지를 사용하며,적어도 일부의 기간이 배지에 축적되는 염기성 물질에 대한 카운터 이온이 부족됨으로써 pH를 7.2이하로 유지할 수 없는 기간임을 특징으로 하는, 발효에 의한 염기성 물질의 제조법.
- 제2항에 있어서, 다른 음이온이 황산 이온, 염화물 이온, 인산 이온 및 이온화된 유기산으로부터 선택되는, 발효에 의한 염기성 물질의 제조법.
- 제2항 또는 제7항에 있어서, 다른 음이온의 합계가 900meq/l 이하임을 특징으로 하는, 발효에 의한 염기성 물질의 제조법.
- 제1항에 있어서, 배양액 중의 총 암모니아 농도가 200mM 이하로 조정됨을 특징으로 하는, 발효에 의한 염기성 물질의 제조법.
- 제1항에 있어서, 미생물을 증식시키는 공정을 포함하는, 발효에 의한 염기성 물질의 제조법.
- 제10항에 있어서, 미생물을 증식시키는 공정 동안에는 총 암모니아 농도를 조정하지 않음을 특징으로 하는, 발효에 의한 염기성 물질의 제조법.
- 제1항에 있어서, 염기성 물질이 L-라이신, L-아르기닌 및 L-히스티딘으로부터 선택되는, 발효에 의한 염기성 물질의 제조법.
- 제12항에 있어서, 염기성 물질이 L-라이신인, 발효에 의한 염기성 물질의 제조법.
- 제12항에 있어서, 염기성 물질이 L-아르기닌인, 발효에 의한 염기성 물질의 제조법.
- 제1항에 있어서, 발효후에 배지 또는 이의 처리물을 가열하여, 중탄산 이온 및 탄산 이온을 제거함을 특징으로 하는, 발효에 의한 염기성 물질의 제조법.
- 제1항에 있어서, 미생물이 코리네형 세균 또는 에세리키아(Escherichia) 속 세균인, 발효에 의한 염기성 물질의 제조법.
- 제15항의 방법에 따라 수득되는, 염기성 물질을 함유하는 발효액 또는 발효 생산물.
