Abstract
Description
Claims (28)
- 1.流体、細胞を含有する試料を分析するための装置であって、試料流入ポート と; 該流入ポートから伸びる試料流動チャンネル;および該流動チャンネルと流体連 絡して設けられた細胞を処理するための細胞取扱い領域からなるメンスケール流 動システム;とを形成するように微細加工された固体基材よりなる該装置。
- 2.さらに、該メンスケール流動チャンネルおよび該細胞取扱い領域を通しての 該試料中の細胞の流動を誘起させるための手段よりなる請求項1記載の装置。
- 3.該細胞取扱い領域か細胞溶解手段よりなり;ここに、流動を誘起させるため の該手段を用いて、該試料中の細胞を該細胞溶解手段と細胞膜破壊的接触させ、 それにより、該試料中の細胞を溶解する請求項2記載の装置。
- 4.該細胞溶解手段が、その壁から伸びる細胞膜貫通突起物を有する流動チャン ネルの部分よりなる請求項3記載の装置。
- 5.該細胞溶解手段が、該細胞取扱い領域の中に捕捉された鋭いエッジの粒子よ りなる請求項3記載の装置。
- 6.該細胞溶解手段が、細胞の通過を阻害するが、細胞内分子を通過させるのに 十分な制限された断面寸法の領域よりなる請求項3記載の装置。
- 7.さらに、該試料中の細胞の細胞内分子成分の存在を検出するための、該細胞 溶解領域の下流にある手段よりなる請求項3記載の装置。
- 8.さらに、不溶性の細胞夾雑物を収集するための、該細胞溶解手段の下流に設 けられた手段よりなる請求項3記載の装置。
- 9.さらに、該細胞溶解手段の下流に設けられた濾過手段よりなる請求項3記載 の装置。
- 10.該基材が、さらに、少なくとも2つのさらなるポートと連絡した分岐メン スケール流動チャンネルを形成し、該装置が、さらに、該流動チャンネルのうち 選択された1つの内に流体流動を向けるバルブ手段よりなる請求項2記載の装置 。
- 11.該細胞取扱い領域が: 該細胞試料中の該細胞集団の予め選択された細胞表面分子に可逆的に結合する固 定化された結合部位よりなる細胞捕捉領域よりなり;ここに、該細胞含有試料の 流動を誘起するための該手段を用いて;該結合部位によって該細胞集団中の細胞 の捕捉を可能とし、それにより、該試料から該細胞集団を分離するのに十分遅い 第1の流速;および該分離された細胞を該捕捉領域から放出するのに十分な第2 の速い流速にての流動を誘起する請求項2記載の装置。
- 12.該流動システムが、さらに、該試料の細胞外成分の存在を検出するための 、該細胞捕捉領域の下流にある手段よりなる請求項11記載の装置。
- 13.該流動システムが、さらに、該細胞捕捉領域から下流にある細胞溶解手段 よりなり、ここに、該流動誘起手段が、細胞を該細胞溶解手段へ押し込むための 手段を含有し、それにより、該試料中の細胞を溶解し;該装置が、さらに、該捕 捉された細胞中の細胞内成分の存在を検出するための手段よりなる請求項11記 載の装置。
- 14.さらに、該試料から細胞夾雑物を濾過するための、該細胞溶解手段および 該検出手段の間に設けられたフィルター手段よりなる請求項13記載の装置。
- 15.該基材が、さらに、十分に小さな直径の細胞のみを通過させる制限された 大きさの複数の流動経路を形成する手段からなる細胞ふるいを形成する請求項2 記載の装置。
- 16.該基材が、微細加工されたシリコンよりなる請求項1記載の装置。
- 17.さらに、該基材と組み合わせて用いるための器具よりなり、該器具が;該 基材を保持するための手段;および 該基材上の流入ポートと適合する流体投入手段よりなり; ここに、流動を誘起するための該手段が、流体が該保持手段に保持される場合に 、該流体を該基材の流動システムに通すための、該器具に設けられている、ポン プ手段よりなる請求項2記載の装置。
- 18.さらに、該基材と組み合わせて用いるための器具よりなり、該器具が;該 基材を保持するための手段;および 該基材中の該メンスケール流動システムの内容物を観察するための光学的手段よ りなる請求項2記載の装置。
- 19.該光学的手段が、拡大する光学機器およびビデオカメラよりなり、ここに 、該器具が、さらに; 該装置の角度および位置を手動で調整するための傾斜機構;および該流動システ ムの内容物を観察するためのビデオスクリーンよりなる請求項18記載の装置。
- 20.流体試料を分抗するための装置であって;試料流入ポート; 該流入ポートから伸びた試料流動チャンネル;該流動チャンネルと流体連絡した 分岐チャンネル;および各々が、該流動チャンネルおよび該分岐チャンネルの間 、ならびに該流動システムの外部に連絡する少なくとも2つのさらなるポート; および該流動システム内に含有されている試料中の分析物の存在または濃度を示 すデータを光学的あるいは電気的に収集するための検出領域よりなるメソケール 流動システムを形成するように微細加工された固形基材;および該流動システム を通る流動を、該さらなるポートのうち選択された1つに向けるためのパルプ手 段よりなる該装置。
- 21.さらに、該基材と組み合わせて用いるための器具よりなり、該器具が;該 基材を保持するための手段; 該基材が該保持手段中に保持されている場合に、該ポートのうち少なくとも2つ に連絡している流体流動チャンネル;および該流動システム内で流動を誘起する ためのポンプ手段よりなる請求項20記載の装置。
- 22.該バルブ手段が該器具内に設けられている請求項21記載の装置。
- 23.流体試料を分析するための装置であって;その各々か流動チャンネルおよ び分析物検出領域よりなり、該流動システムの1つが試料を分析するために適合 したものであり、他のものが対照として適合するものである、少なくとも2つの メンスケール流動システムを形成するように微細加工された固体基材;および 該流動システムを両方を通る試料の流動を実質的に同時に誘起し、それにより、 該システムの検出領域からのデータと比較することができる手段よりなる該装置 。
- 24.流体試料を分析するための装置において、試料流入ポートと、該ポートお よび分析物検出領域の間で連結する試料流動チャンネルよりなるメンスケール流 動システムとを形成するように微細加工された固体基材よりなる装置であって、 改良点が; 分析物を検出する前に該試料から不溶性物質を除去するために、該検出領域の上 流に、該流動チャンネル中に設けられたフィルターよりなる該装置。
- 25.さらに、該フィルターに隣接して設けられた、不溶性の夾雑物を収集する ための排水溜よりなる請求項24記載の装置。
- 26.(A)標的の細胞集団に特徴的な細胞膜結合クンバク質に特異的な結合蛋 白がその上に固定化された固体壁よりなるメンスケール試料流動経路を供し;( B)可逆的な細胞表面タンパク質−固定化タンパク質結合により、細胞標的亜集 団の膜は捕捉でき、一方他の細胞はそれを通過させる条件下において細胞含有液 体試料を該流動経路に通し;次いで、(C)該標的の細胞亜集団を放出させるた めに該流物経路における条件を変化させる工程よりなることを特徴とする細胞含 有流体試料中の標的細胞亜集団を分離する方法。
- 27.該流動経路中の流体の該流動速度を工程Cで増大させて、該固体壁から細 胞を剪断により除く請求項26記載の方法。
- 28.工程Cを、該細胞を該固体壁から脱着させる溶媒を、該流動チャンネルに 導入することによって行う請求項26記載の方法。
