Description
Claims (16)
- 細胞を含む体液における標的分子の存在を決定する方法であって、
溶解物を生成するために前記体液における細胞を溶解させるステップ、
前記溶解物を、第1の検出分子を有する粒子と混ぜ合わせるステップ、
第1の検出分子を介して標的分子を前記粒子に結合させるステップ、
第2の検出分子を有した基板を前記溶解物に曝露するステップ、
第2の検出分子を介して標的分子を前記基板に結合させるステップ、
前記基板に付着した粒子を検出するステップ、
を含む方法。 - 前記細胞は、1つの界面活性剤又は複数の界面活性剤の混合物によって溶解される、請求項1に記載の方法。
- 前記界面活性剤は、Nonidet−P40、Nonidet−P40代用品、スクロースモノラウラート、Chemal LA−9及びTritonX100のうち少なくとも1つを含む、請求項2に記載の方法。
- タンパク質及び炭水化物の非存在下で、少なくとも塩が前記界面活性剤に添加される、請求項2又は3に記載の方法。
- 前記細胞が、物理的溶解方法によって溶解される、請求項1乃至4のいずれか一項に記載の方法。
- 前記体液の細胞の溶解の開始から特定の時間の後、前記溶解物は前記粒子と混ぜ合わされ、且つ/或いは、前記基板は前記溶解物に曝露される、請求項1乃至5のいずれか一項に記載の方法。
- 第1の位置にて前記体液の細胞は溶解され、さらに、第2の位置にて前記溶解物は前記粒子と混ぜ合わされ、且つ/或いは、前記基板は前記溶解物に曝露される、請求項1乃至6のいずれか一項に記載の方法。
- 前記溶解された細胞から放出される物質を結合させるために、前記溶解物に結合剤を添加するステップ、
をさらに含む、請求項1乃至7のいずれか一項に記載の方法。 - 前記標的分子は、特定のタイプのタンパク質である、請求項1乃至8のいずれか一項に記載の方法。
- 前記体液は血液を含む、請求項1乃至9のいずれか一項に記載の方法。
- 前記粒子は磁気粒子であり、
当該方法は、
磁力によって前記基板の方向に前記磁気粒子を動かすステップ、
反対の磁力によって、前記基板に付着していない磁気粒子を取り除くステップ、
前記粒子により反射された放射物を検出することによって、前記基板に付着した前記磁気粒子を検出するステップ、
を含む、請求項1乃至10のいずれか一項に記載の方法。 - 分析装置内に挿入するためのカートリッジであって、
体液における細胞を溶解させるための界面活性剤、
第1の検出分子を有した粒子、
第2の検出分子を有した基板、
を含むカートリッジ。 - チャネルによって接続される第1の空洞及び第2の空洞、
をさらに含み、
前記界面活性剤は、前記第1の空洞内に配置され、
前記粒子及び前記基板は、前記第2の空洞内に配置される、請求項12に記載のカートリッジ。 - 前記チャネル内に弁、
をさらに含み、
前記弁は、前記体液が前記第1の空洞に入ってから特定の時間の後で開くように適応される、請求項13に記載のカートリッジ。 - 請求項1乃至11のいずれか一項に記載の方法を行うために適応される分析装置。
- 請求項11乃至13のいずれか一項に記載のカートリッジを受けるためのスロット、
を含む、請求項15に記載の分析装置。
