Description
本願は、2012年2月3日に出願の米国仮特許出願第61/594867号“EXTERNAL FILES FOR DISTRIBUTION OF MOLECULAR DIAGNOSTIC TESTS AND DETERMINATION OF COMPATIBILITY BETWEEN TESTS”の優先権を主張するものであって、この文献の全開示は参照により全て本明細書に組み込まれる。
本明細書で開示の実施形態は、自動化されたアッセイを実行するための方法及びシステム、特には複数の試料について自動化装置で複数のアッセイを実行することに関する。
本明細書で提供する方法及びシステムの実施形態において、異なる各自動化アッセイワークフローは、関連する固有アッセイ定義又はユーザ定義プロトコルファイルを有する。本明細書で使用の用語「アッセイ定義ファイル(ADF)」は、そのワークフローに関するアッセイ特有パラメータの少なくとも一部、典型的には全てを提供するファイルを意味する。加えて、ADFは特定のワークフローに関するコンパチビリティインデックス値を提供し得る。典型的な実施形態において、ADFは、自動化装置でアッセイを実行するのに必要な情報の全てを格納し得る。ADFの1つの機能は、装置とアッセイとの間に独立性をもたらすことである。この独立性により、装置製造業者又はアッセイ試薬製造業者は、装置についての新しいアッセイプロトコルを、装置ソフトウェアを大きく改訂することなくリリースすることができる。
特定の実施形態において、アッセイパラメータは、製造業者ではなくユーザによって決定される。これらのユーザ定義プロトコル(UDP)にも第1レベル及び第2レベルコンパチビリティインデックス値を割り当てて他の商業的に開発されたアッセイプロトコルとのコンパチビリティを確保することができる。インデックス及びアッセイ定義ファイルのメリットの1つは、ユーザ定義アッセイプロトコルと、同じようにコンパチビリティをカバーする商業的に開発されたアッセイプロトコルとの間でファイアウォールが得られることである。インデックス値を使用することで、商業的に開発されたアッセイプロトコルのインデックス値とは異なるユーザ定義プロトコルについて固有の制御手段を設定することができる。図1に示す実施形態において、ユーザ定義プロトコルは、UDP及び「抽出のみ」とラベリングされたボックスで表される。
幾つかの実施形態において、アッセイ定義又はユーザ定義プロトコルファイルは、第1レベルコンパチビリティインデックス値を含み得る。典型的には、第1レベルコンパチビリティインデックス値は、ラックコンパチビリティに言及している。しかしながら、特定の他の実施形態において、第1レベルコンパチビリティインデックス値は、ランコンパチビリティに言及しており、第2レベルインデックス値は、ラックコンパチビリティに言及している。このため、この方法は、(a)利用可能なアッセイプロトコルの第1リストから第1アッセイプロトコルを選択し;(b)利用可能な他の複数のアッセイプロトコルのどれが第1アッセイと同じ第1レベルコンパチビリティインデックス値を含むアッセイ定義ファイルを有するかを評価することを含み得る。典型的な実施形態において、同じ第1レベルコンパチビリティインデックス値を有する2つのアッセイプロトコルは、第1レベルコンパチビリティを示す。しかしながら、当然のことながら、コンパチビリティ値を個々のアッセイに割り当て且つ特定することができるいずれの適切な手段も、本明細書で提供する方法及びシステムにおいて役立ち得る。したがって、幾つかの実施形態において、ラックコンパチブルである2つのアッセイプロトコルは異なる第1レベルインデックス値を有し得る。しかしながら、本開示における便宜上、第1レベルコンパチビリティを有する2つのアッセイプロトコルは、同じ第1レベルコンパチビリティインデックス値を有するとみなされる。

本明細書では自動化されたアッセイを実行するためのシステムも提示し、このシステムは、第1ワークステーションと第2ワークステーションとを含む自動化装置を含み、第1ワークステーション及び第2ワークステーションのそれぞれは複数の異なる自動化アッセイワークフローに従って複数の試料を受け取って処理するように構成され、また単一のサービスリソースによってサポートされている。異なる各自動化アッセイワークフローは典型的には、関連した固有のアッセイ定義ファイル又はユーザ定義プロトコルファイルを含む。システムは、プロセッサ、ストレージ容量及び自動化されたアッセイを実行するためのプログラムも含み、プログラムは、2つの別個のアッセイワークフローが自動化装置での同時処理に関して互いにコンパチブルであるかインコンパチブルであるかを判断し、これらのアッセイがコンパチブルである場合に、これら別個のアッセイワークフローを装置で同時に実行するための命令を含む。
本明細書で提示する方法及びシステムには、既存のアプローチに勝る多数の利点がある。例えば、アッセイプロトコルを配布するために製造業者がADFを使用することで、装置のソフトウェアを合わせてアップデートする必要なく、装置プロットフォーム上で新しい又は修正したアッセイプロトコルをリリースする方法が提供される。装置ソフトウェア改訂の必要性を排除することで、このアプローチはアッセイのリリースをより直接的なものにしている。加えて、必要に応じて、製造業者は、アッセイプロトコル間のコンパチビリティを、装置ソフトウェアを改訂することなく、ビジネス又は他のニーズに合うように修正することができる。
ユーザ定義プロトコル及び市販のアッセイプロトコルに関して第1レベル及び第2レベルコンパチビリティインデックス値を割り当てる
本実施例では、第1及び第2レベルコンパチビリティインデックス値を特定のアッセイワークフロー又はユーザ定義プロトコル(UDP)に割り当てる処理について説明する。自動化試料処理/分析装置は、2つの別個の試料処理ワークフロー又はラックを同時に実行する能力を有する(ランコンパチブル)。しかしながら、試料処理ワークフローには、修正されても依然としてコンパチビリティを維持する特定のアクション(ラックコンパチブル)と、装置の同じランではワークフローがインコンパチブルとなる特定のアクション(インコンパチブル)とがある。物理的な制限に加えて、ある装置で複数のアッセイプロトコルが一緒に行われないようにするビジネス上の要件がある場合もある。
アッセイコンパチビリティの特定
本実施例では、一実施形態による、2つのアッセイプロトコル間での第1レベルコンパチビリティ及び第2レベルコンパチビリティの特定について説明する。図2に示す例示的な方法において、第1アッセイと第2アッセイとの間のコンパチビリティは、2つのレベルのコンパチビリティインデックス値を比較することで判断される。コンパチブルではないアッセイを同時に実行しないようにするための、ユーザは、同じラックで同時に実行できるか否か、あるいは同じ装置の異なるラックであっても実行できるか否かを判断するためのチャート又は表でアッセイプロトコルを手作業で比較しなくてはならなかった。そのようなルックアップテーブルの例を図5に示す。このような手作業のアプローチは誤りが起きやすく、また効率が悪く、労働集約的である。本実施例では、コンパチブルなアッセイプロトコルを特定し且つコンパチブルではないアッセイプロトコルが同時に実行されないようにする自動化された方法の例を挙げる。
図2の概略図に記載されるように、第1アッセイはユーザによって、全ての利用可能なアッセイプロトコルのリストから選択される。ユーザの入力に基づいて、選択された第1アッセイについての第1レベル(ラック)コンパチビリティインデックス値が第1アッセイについてのアッセイ定義ファイル(ADF)から、又は選択されたアッセイがユーザ定義されたものの場合、UDPから得られる。次に、そのコンパチビリティインデックス値を、利用可能な残りのアッセイプロトコルのそれぞれの第1レベルコンパチビリティインデックス値(各アッセイについてADF又はUDPから得られる)と比較する。選択されたアッセイと第1レベルコンパチビリティインデックス値を共有する全てのアッセイプロトコルが特定され、コンパチブルではないアッセイプロトコルは、更なる検討から除外される。
あるいは、システムは、同じラックで一緒に実行するにはコンパチブルではない場合に、アッセイプロトコルを特定し、別々のラックでアッセイプロトコルを実行し得る。図2の概略図に示すように、第1アッセイはユーザによって、全ての利用可能なアッセイプロトコルのリストから選択される。ユーザの入力に基づいて、選択された第1アッセイについての第1レベル(ラック)コンパチビリティインデックス値が第1アッセイについてのアッセイ定義ファイル(ADF)から又は、選択されたアッセイがユーザ定義されたものの場合、UDPから得られる。次に、そのコンパチビリティインデックス値を、利用可能な残りのアッセイプロトコルのそれぞれの第1レベルコンパチビリティインデックス値(各アッセイについてADF又はUDPから得られる)と比較する。選択されたアッセイと第1レベルコンパチビリティインデックス値を共有するアッセイプロトコルが特定されないならば、次にシステムは選択された第1アッセイについて第2レベル(ラン)コンパチビリティインデックス値を得て、その値を他の利用可能なアッセイプロトコル全ての第2レベルコンパチビリティインデックス値と比較する。選択されたアッセイと第2レベルコンパチビリティインデックス値を共有する全てのアッセイプロトコルが特定され、コンパチブルではないアッセイプロトコルは更なる検討から除外される。次に、別々のラックでの同時の実行に関してコンパチブルなアッセイプロトコルだけが載ったリストが表示される。ユーザはそのリストから第2アッセイを選択し、選択が完了したら、システムはこれら2つのアッセイプロトコルを別々のラック上で同時に実行し始める。
システムがラックコンパチブル又はランコンパチブルな他のアッセイプロトコルを特定しない場合、ユーザは、1つまたは複数の試料を使用して、同じ又は別々のラック上で単一のアッセイプロトコルを実行することを選択することができる。
ランワークリストへの試験の追加

Claims (26)
- 複数の試料について自動化されたアッセイを実行する方法であって、前記方法が、
第1ワークステーションと第2ワークステーションとを含む自動化装置を用意する工程であって、前記第1ワークステーション及び前記第2ワークステーションのそれぞれが複数の異なる自動化アッセイワークフローに従って複数の試料を受け取って処理するように構成され、異なる各自動化アッセイワークフローが、関連した固有のアッセイ定義ファイル又はユーザ定義プロトコルファイルを有し、前記アッセイ定義ファイル又は前記ユーザ定義プロトコルファイルは、それぞれ、第1レベルコンパチビリティインデックス値、第2レベルコンパチビリティインデックス値を含み、前記自動化アッセイワークフローを実行するための1つ以上のパラメータを提供する工程と、
2つの別個のアッセイワークフローが自動化装置での同時処理に関して互いにコンパチブルであるかインコンパチブルであるかを判断する工程と、

前記利用可能な他の複数のアッセイワークフローのいずれも、前記第1アッセイワークフローと同じ第1レベルコンパチビリティインデックス値を含むアッセイ定義ファイル又はユーザ定義プロトコルファイルを有するものはないと判断する工程とを含む、請求項1に記載の方法。 - 同じ第1レベルコンパチビリティインデックス値を有するアッセイワークフローが、自動化装置の同じワークステーション又は異なるワークステーションにおいて同時に処理される、請求項1に記載の方法。
- 前記第1レベルコンパチビリティインデックス値は、インキュベーション時間、溶解時間、試薬体積、試薬タイプ、インキュベーション温度、溶解温度、ワークステーション時間要求、規制区分、ビジネス上考慮すべき事柄及びこれらの組み合わせから成る群から選択されるパラメータに基づいて決定される、請求項8に記載の方法。
- 前記自動化装置は、前記第1レベルコンパチビリティインデックス値が異なる場合に、インコンパチブルなアッセイを同じワークステーション内で同時に実行することを妨げる、請求項1に記載の方法。
- 前記2つの別個のアッセイワークフローが第2レベルコンパチブルである場合、前記方法が、以下の工程:
アッセイ特有試料作製スクリプトを前記自動化装置で開始する工程、
消耗品パッケージ上の識別表示を前記自動化装置に保存された一連のアッセイ特有識別データと比較する工程、

装填されたカートリッジにおける蛍光比を前記自動化装置に保存された一連のアッセイ特有蛍光比データと比較して、前記カートリッジがうまく装填されたか否かを判断する工程、
アッセイ特有反応スクリプトを前記自動化装置で開始する工程、
アッセイ特有データ分析アルゴリズムを前記自動化装置で開始する工程、
1つ以上のアッセイ特有リザルトアルゴリズム又はスクリプトに基づいてアッセイワークフローに対する最終判断を導き出す工程、
の1つ以上を更に含む、請求項1に記載の方法。 - 前記アッセイワークフローの少なくとも1つが、ポリメラーゼ連鎖反応(PCR)、転写増幅(TMA)、オリゴヌクレオチドライゲーション法(OLA)、リガーゼ連鎖反応(LCR)、ローリングサークル増幅(RCA)、鎖置換増幅(SDA)及びハイブリダイゼーション反応から成る群から選択される反応を含む、請求項1に記載の方法。
- 第1ワークステーションと第2ワークステーションとを含む自動化装置と、
プロセッサと、
ストレージ容量と、
自動化されたアッセイを実行するためのプログラムとを含む自動化されたアッセイを実行するためのシステムであって、
前記第1ワークステーション及び前記第2ワークステーションのそれぞれは複数の異なる自動化アッセイワークフローに従って複数の試料を受け取って処理するように構成され、異なる各自動化アッセイワークフローは、関連した固有のアッセイ定義ファイル又はユーザ定義プロトコルファイルを有し、
各アッセイ定義ファイル又はユーザ定義プロトコルファイルは、第1レベルコンパチビリティインデックス値、第2レベルコンパチビリティインデックス値を含み、前記自動化アッセイワークフローを実行するための1つ以上のパラメータを提供し、
前記プログラムが、
2つの別個のアッセイワークフローが自動化装置での同時処理に関して互いにコンパチブルであるかインコンパチブルであるかを判断する工程を自動化アッセイに行わせ、
前記判断工程が、
利用可能なアッセイワークフローの第1リストから第1アッセイワークフローを選択する工程と、
利用可能な他の複数のアッセイワークフローのどれが前記第1アッセイワークフローと同じ第2レベルコンパチビリティインデックス値を含むアッセイ定義ファイル又はユーザ定義プロトコルファイルを有するかを評価し、
前記同じ第2レベルコンパチビリティインデックス値を含むアッセイ定義ファイル又はユーザ定義プロトコルファイルを有するアッセイワークフローが第2レベルコンパチブルである、工程と、
前記2つの別個のアッセイワークフローが第2レベルコンパチブルである場合に、自動化装置の異なるワークステーションにおいて同時に、それぞれがそれ自身の1以上のパラメータに従って、前記2つの別個のアッセイワークフローを実行する工程とを含む、システム。 - 同じ第2レベルコンパチビリティインデックス値を有するアッセイワークフローは、自動化装置の異なるワークステーション上でのみ同時に処理される、請求項13に記載のシステム。
- 前記自動化装置は、少なくとも1つの共有リソースを利用して、複数の異なる自動化アッセイワークフローに従って前記複数の試料を受け取り処理するように構成される、請求項13に記載のシステム。
- 前記2つの別個のアッセイワークフローは、前記2つの別個のアッセイワークフローに関連するパラメータが前記少なくとも1つの共有リソースの第2レベルコンパチビリティを与える範囲外にはない場合に、前記2つの別個のアッセイワークフローは第2レベルコンパチビリティである、請求項15に記載のシステム。
- 前記利用可能な他の複数のアッセイワークフローのどれが前記第1アッセイワークフローと同じ第2レベルコンパチビリティインデックス値を含むアッセイ定義ファイル又はユーザ定義プロトコルファイルを有するかを評価する工程は、
前記第1アッセイワークフローの第2コンパチビリティインデックス値とは異なる第2レベルコンパチビリティインデックス値を有するアッセイワークフローを特定する工程と、
第2アッセイワークフローの第2リストを用意する工程とを含み、
前記第2リストでは前記第1アッセイワークフローの第2レベルコンパチビリティインデックス値とは異なる第2レベルコンパチビリティインデックス値を有するアッセイワークフローが除外される、請求項13に記載のシステム。 - 前記第2レベルコンパチビリティインデックス値は、規制区分、ワークフローのインコンパチビリティ、ビジネス上考慮すべき事柄及びこれらの組み合わせから成る群から選択されるパラメータに基づいて決定される、請求項13に記載のシステム。
- 前記判断工程は更に、
前記利用可能な他の複数のアッセイワークフローのどれが前記第1アッセイワークフローと同じ第2レベルコンパチビリティインデックス値を含むアッセイ定義ファイル又はユーザ定義プロトコルファイルを有するかを評価する工程の前に、
利用可能な他の複数のアッセイワークフローのどれが、前記第1アッセイワークフローと同じ第1レベルコンパチビリティインデックス値を含むアッセイ定義ファイル又はユーザ定義プロトコルファイルを有するかを評価する工程と、
前記利用可能な他の複数のアッセイワークフローのいずれも、前記第1アッセイワークフローと同じ第1レベルコンパチビリティインデックス値を含むアッセイ定義ファイル又はユーザ定義プロトコルファイルを有するものはないと判断する工程とを含む、請求項13に記載のシステム。 - 同じ第1レベルコンパチビリティインデックス値を有するアッセイワークフローが、自動化装置の同じワークステーション又は異なるワークステーションにおいて同時に処理される、請求項13に記載のシステム。
- 前記第1レベルコンパチビリティインデックス値は、インキュベーション時間、溶解時間、試薬体積、試薬タイプ、インキュベーション温度、溶解温度、ワークステーション時間要求、規制区分、ビジネス上考慮すべき事柄及びこれらの組み合わせから成る群から選択されるパラメータに基づいて決定される、請求項13に記載のシステム。
- 前記自動化装置は、前記第1レベルコンパチビリティインデックス値が異なる場合に、インコンパチブルなアッセイを同じワークステーション内で同時に実行することを妨げる、請求項13に記載のシステム。
- 前記2つの別個のアッセイワークフローがコンパチブルである場合、前記プログラムが、以下の工程:
アッセイ特有試料作製スクリプトを前記自動化装置で開始する工程、
消耗品パッケージ上の識別表示を前記自動化装置に保存された一連のアッセイ特有識別データと比較する工程、

アッセイ特有データ分析アルゴリズムを前記自動化装置で開始する工程、
1つ以上のアッセイ特有リザルトアルゴリズム又はスクリプトに基づいてアッセイワークフローに対する最終判断を導き出す工程、
の1つ以上を更に実行するように構成された、請求項13に記載のシステム。 - 前記識別表示が、バーコードを含む、請求項23に記載のシステム。
- 前記システムが、バーコードリーダーを更に含む、請求項24に記載のシステム。
- 前記アッセイワークフローの少なくとも1つが、ポリメラーゼ連鎖反応(PCR)、転写増幅(TMA)、オリゴヌクレオチドライゲーション法(OLA)、リガーゼ連鎖反応(LCR)、ローリングサークル増幅(RCA)、鎖置換増幅(SDA)及びハイブリダイゼーション反応から成る群から選択される反応を含む、請求項13に記載のシステム。
