Description
該反応容器11は、開口部13を有する円筒状の貯留室12と、該貯留室12と連通し該貯留室12よりも薄く薄層化された反応室15を挟んで層状に形成され、その全体が透光性の反応部14とを有するものである。前記開口部13は、後述するキャップ20を介して回転体としてのノズル22に装着可能である。該反応室15は、前記貯留室12の下方で、かつ、前記開口部13または貯留室12の軸線に対して、貯留室12よりも位置座標として遠い位置に設けられている。前記開口部13または貯留室12の軸線は、後述する回転体としてのノズル22に該反応容器11が装着された際には、該回転体の回転軸線と一致する。
なお、該液回転処理装置50は、上側から吊り下げられるように設けられ、前記反応測定処理システム10の全域を覆うように、図示しない直動機構を利用したX軸Y軸移動機構によって移動可能に設けられている。
ここでは、所定の検体に含まれるDNAの量を測定するために、リアルタイムPCR法を用いて測定する場合について説明する。リアルタイムPCRは、核酸プローブを用いた核酸の濃度を測定する方法である。その方法は、例えば、蛍光色素で標識化された核酸プローブが、目的核酸にハイブリダイズした際に、蛍光色素が結合した部分の塩基の種類及び塩基配列に依存した程度で、蛍光色素の発光が減少するという現象、または、核酸プローブを目的核酸から除去することによって発光強度が増大するという現象(発光消光現象)を利用したり、または二本鎖DNAに挿入されることで蛍光を発する試薬を反応系に加え、増幅に伴う蛍光を検出する方法を利用し、その蛍光強度を検出することで定量を行う方法(インターカレーター法)を用いる。

Claims (16)
- 開口部を有し液を貯留可能な貯留室と、該貯留室と連通し該貯留室よりも薄くまたは細く形成された反応室とを有する容器であって、該容器は外部に設けた回転可能な回転体に装着可能であり、該容器が前記回転体に装着された際には、該回転体の回転軸線は前記容器を貫き前記反応室は前記貯留室よりも前記回転軸線から遠くに位置するように形成された反応容器。
- 前記回転体は気体の吸引吐出が可能で自転可能なノズルであって、該ノズルはその軸方向に沿った回転軸線を有する請求項1に記載の反応容器。
- 前記回転体は、その回転軸線が前記貯留室の開口部を貫くように、該開口部に装着可能である請求項1または請求項2のいずれかに記載の反応容器。
- 前記反応室の少なくとも一部が透光性または半透光性である請求項1乃至請求項3のいずれかに記載の反応容器。
- 前記反応室の一部または全部は軟質材で形成され、該軟質材を変形することによって、該反応室を密封可能とする請求項1乃至請求項4のいずれかに記載の反応容器。
- 前記回転体の下端部に着脱自在に装着されて該下端部を覆うキャップを、前記開口部を覆うために該開口部に着脱自在に設けた請求項3に記載の反応容器。
- 開口部を有し液の貯留が可能な貯留室と、該貯留室と連通し該貯留室よりも薄くまたは細く形成された反応室と、前記反応室と連通した流体の吸引吐出口とを有する容器であって、前記貯留室の開口部は流体の吸引吐出を行うノズルの下端部によって閉塞可能となるように形成された反応容器。
- 前記反応室の少なくとも一部が透光性または半透光性である請求項7に記載の反応容器。
- 前記反応室の一部または全部は軟質材で形成され、該軟質材を変形することによって、該反応室を密封可能とする請求項7または請求項8のいずれかに記載の反応容器。
- 請求項1乃至請求項9のいずれかに記載の1または2以上の反応容器と、1または2以上の前記反応室と接触しまたは近接して設けられて該反応室を加熱または冷却する加熱冷却部と、1または2以上の前記反応室内の光情報を得る光情報測定部とを有する反応測定装置。
- 前記光情報測定部は、2以上の前記反応容器の前記反応室の各照射位置に設けた2以上の照射端部と、複数種類の波長をもつ光を各々発生する複数種類の光源と、前記光源からの光の内の1種類の光を時間的に切り換えて選択して、前記各照射端部に一斉に導光する光源選択部と、2以上の前記反応容器の前記反応室の各受光位置に設けた2以上の受光端部と、該各受光端部からの光を時間的に切り換えて選択する受光位置選択部と、選択された受光位置からの光が通過すべき複数種類のフィルタを時間的に切り換えて選択するフィルタ選択部と、選択された受光位置からの光であって選択されたフィルタを通過した光を順次入力する光電素子とを有する請求項10に記載の反応測定装置。
- 前記光情報測定部は、2以上の前記反応容器の前記反応室の各照射位置に設けた2以上の照射端部と、複数種類の波長をもつ光を各々発生する複数種類の光源と、前記光源からの光の内の1種類の光を時間的に切り換えて選択し、選択された光を時間的に切り換えて各受光端部に導光する光源照射位置選択部と、2以上の前記反応容器の前記反応室の各受光位置に設けた2以上の受光端部と、前記受光位置からの光が通過すべき複数種類のフィルタを時間的に切り換えて選択するフィルタ選択部と、選択されたフィルタを通過した光を順次入力する光電素子とを有する請求項10に記載の反応測定装置。
- 回転可能な1または2以上の回転体と、該回転体に着脱自在に装着可能に形成された1または2以上の反応容器と、該回転体を回転駆動する回転駆動部とを有し、前記反応容器は、開口部を有し液を貯留可能な貯留室、および該貯留室と連通し該貯留室よりも薄くまたは細く形成された反応室を有し、前記回転体の回転軸線は前記容器を貫き前記反応室は前記貯留室よりも前記回転軸線から遠くに位置するように形成されて、前記反応容器の前記貯留室に収容された液体を前記反応室に導入する液回転処理装置。
- 前記回転体は気体の吸引吐出が可能で自転可能なノズルであって、該ノズルはその軸方向に沿った回転軸線を有する請求項13に記載の液回転処理装置。
- 前記回転体は、その回転軸線が前記貯留室の開口部を貫くように、該開口部に装着可能である請求項13または請求項14のいずれかに記載の液回転処理装置。
- 前記回転体の下端部には着脱自在に装着されて前記下端部を覆うキャップを有し、該キャップは、前記反応容器の開口部を覆うために該開口部に着脱自在に装着された請求項15に記載の液回転処理装置。
