- ターゲット分子と結合しうるアプタマーを同定するための方法であり、アプタマーはそのターゲット分子からの遅い解離速度をもち、またターゲット分子はタンパク質またはペプチドであって:
(a)ターゲット分子と候補混合物を接触させ、試料混合物を形成して、核酸−ターゲット複合体を形成させ、ここで、核酸がターゲット分子に対してアフィニティを有するときに核酸−ターゲット分子複合体が形成され、また前記候補混合物は、核酸の少なくとも1つもしくはそれぞれにおいて1つ、複数もしくは全てのピリミジンが5位で化学的に修飾された修飾核酸を含み、修飾ピリミジンが5−(N−ベンジルカルボキシアミド)−2’−デオキシウリジン、5−(N−イソブチルカルボキシアミド)−2’−デオキシウリジンおよび5−(N−[2−(1H−インドール−3イル)エチル]カルボキシアミド)−2’−デオキシウリジンからなる群より選択され;
(b)少なくとも1つの競合剤分子を試料混合物に添加し、ここで競合剤分子は硫酸デキストランであり;
(c)残りの候補混合物から核酸−ターゲット分子複合体を分配し;
(d)核酸−ターゲット分子複合体を解離させて、未結合核酸を生成し、ここで未結合核酸からの少なくとも1つの核酸がターゲット分子と結合可能である
工程を含み、これによってターゲット分子に対するアプタマーが同定される、前記方法。 - (a)から(d)の工程を1回以上繰り返すことをさらに含む請求項1〜4または17のいずれかに記載の方法。
- (a)から(d)の工程を1回以上繰り返すことをさらに含む請求項34記載の方法。
