第1のサブユニット及び第2のサブユニットを含み、
前記第1のサブユニットが前記認識配列の第1の認識した半分の部位に結合し、第1の超可変(HVR1)領域を含み、

をコードする、請求項30又は31に記載のポリヌクレオチド。 - 前記ポリシストロニックmRNAが、配列番号22のアミノ酸配列を含む遺伝子操作されたメガヌクレアーゼをコードし、且つ配列番号33のアミノ酸配列を含む遺伝子操作されたメガヌクレアーゼをコードする、請求項30〜32のいずれか1項に記載のポリヌクレオチド。
- 請求項1〜27のいずれか1項に記載の遺伝子操作されたメガヌクレアーゼをコードする核酸配列を含む、組換えDNA構築物。
- プロモータと請求項1〜27のいずれか1項に記載の遺伝子操作されたメガヌクレアーゼをコードする核酸配列とを含むカセットを含む、請求項34に記載の組換えDNA構築物。
- 2以上のカセットを含み、前記カセットの各々が、プロモータと、請求項1〜27のいずれか1項に記載の遺伝子操作されたメガヌクレアーゼをコードする核酸配列とを含み、前記遺伝子操作されたメガヌクレアーゼの各々が、異なるHBV認識配列に対して特異性を有する、請求項34に記載の組換えDNA構築物。
- プロモータと、請求項1〜27のいずれか1項に記載の遺伝子操作されたメガヌクレアーゼの1以上をコードするポリシストロニック核酸配列と、を含むカセットを含み、前記プロモータが、前記ポリシストロニック核酸配列の発現を駆動して、標的細胞においてポリシストロニックmRNAを生成する、請求項34に記載の組換えDNA構築物。
- 前記ポリシストロニックmRNAが、請求項30〜33のいずれか1項に記載のポリシストロニックmRNAである、請求項36に記載の組換えDNA構築物。
- 請求項1〜27のいずれか1項に記載の遺伝子操作されたメガヌクレアーゼをコードする前記核酸配列を含むウイルスベクターをコードする、請求項34に記載の組換えDNA構築物。

- 2以上のカセットを含み、前記カセットの各々が、プロモータと、請求項1〜27のいずれか1項に記載の遺伝子操作されたメガヌクレアーゼをコードする核酸配列と、を含み、前記遺伝子操作されたメガヌクレアーゼの各々が、異なるHBV認識配列に対して特異性を有する、請求項41に記載のウイルスベクター。
- プロモータと、請求項1〜27のいずれか1項に記載の遺伝子操作されたメガヌクレアーゼの1以上をコードするポリシストロニック核酸配列と、を含むカセットを含み、前記プロモータが、前記ポリシストロニック核酸配列の発現を駆動して、標的細胞においてポリシストロニックmRNAを生成する、請求項41に記載のウイルスベクター。
- 前記ポリシストロニックmRNAが、請求項30〜33のいずれか1項に記載の前記ポリシストロニックmRNAである、請求項45に記載のウイルスベクター。
- B型肝炎ウイルス(HBV)又はHBVに起因する肝細胞がんを有する対象を治療するための医薬組成物であって、医薬的に許容される担体、及び
(a)請求項1〜27のいずれか1項に記載の遺伝子操作されたメガヌクレアーゼをコードする核酸;又は
(b)請求項1〜27のいずれか1項に記載の遺伝子操作されたメガヌクレアーゼ;
を含む、医薬組成物。 - 前記遺伝子操作されたメガヌクレアーゼをコードする前記核酸が、請求項29〜33のいずれか1項に記載の前記mRNAである、請求項47に記載の医薬組成物。
- 請求項34〜40のいずれか1項に記載の組換えDNA構築物を含む、請求項47に記載の医薬組成物。
- 請求項41〜46のいずれか1項に記載のウイルスベクターを含む、請求項47に記載の医薬組成物。
- 請求項1〜27のいずれか1項に記載の遺伝子操作されたメガヌクレアーゼを含む、請求項47に記載の医薬組成物。
- 請求項1〜27のいずれか1項に記載の遺伝子操作されたメガヌクレアーゼの2以上を含み、前記遺伝子操作されたメガヌクレアーゼが、異なるHBV認識配列に対して特異性を有する、請求項47に記載の医薬組成物。
- 請求項1〜27のいずれか1項に記載の遺伝子操作されたメガヌクレアーゼの2以上をコードする2以上の核酸を含み、前記遺伝子操作されたメガヌクレアーゼが、異なるHBV認識配列に対して特異性を有する、請求項47に記載の医薬組成物。
- 脂質ナノ粒子内に封入されている請求項29〜33のいずれか1項に記載の前記mRNAの1以上を含む、請求項47に記載の医薬組成物。
- HBVを有する対象を治療する方法であって、前記対象の標的細胞に、
(a)遺伝子操作されたメガヌクレアーゼをコードし、前記標的細胞においてインビボで発現される核酸;又は
(b)遺伝子操作されたメガヌクレアーゼ;
を送達することを含み、
前記遺伝子操作されたメガヌクレアーゼは、B型肝炎ウイルスの少なくとも2つの遺伝子型のゲノムのORF中の認識配列に対する特異性を有し、前記遺伝子操作されたメガヌクレアーゼは、前記標的細胞中の前記認識配列を認識し切断し、前記対象におけるHBVの感染及び/又は増殖が減少又は排除される方法。 - 前記認識配列が、遺伝子型A、並びに遺伝子型B(配列番号4)、遺伝子型C(配列番号5)、遺伝子型D(配列番号6)、遺伝子型E(配列番号7)、遺伝子型F(配列番号8)、及び遺伝子型G(配列番号9)の1以上のORF内にある、請求項55に記載の方法。
- 前記認識配列が、B型肝炎ウイルスの少なくとも3つ、少なくとも4つ、少なくとも5つ、又は少なくとも6つの遺伝子型のORF内に見出される、請求項55又は56に記載の方法。
- 前記認識配列が、ポリメラーゼ(P)タンパク質、大表面(preS1/preS2/S)タンパク質、中表面(preS2/S)タンパク質、及び小表面(S)タンパク質からなる群から選択されるタンパク質をコードする、少なくとも1つのORF内にある、請求項55〜57のいずれか1項に記載の方法。
- 前記認識配列が配列番号12、配列番号16、配列番号10、又は配列番号14を含む、請求項55〜58のいずれか1項に記載の方法。
- 前記認識配列が配列番号12を含む、請求項55〜59のいずれか1項に記載の方法。
- 前記遺伝子操作されたメガヌクレアーゼが、請求項4〜15のいずれか1項に記載の遺伝子操作されたメガヌクレアーゼである、請求項55〜59のいずれか1項に記載の方法。
- 前記認識配列が配列番号16を含む、請求項55〜59のいずれか1項に記載の方法。
- 前記遺伝子操作されたメガヌクレアーゼが、請求項16〜27のいずれか1項に記載の遺伝子操作されたメガヌクレアーゼである、請求項62に記載の方法。
- 前記対象に請求項47〜54のいずれか1項に記載の医薬組成物を投与することを含む、請求項55〜63のいずれか1項に記載の方法。
- 前記遺伝子操作されたメガヌクレアーゼ又は前記遺伝子操作されたメガヌクレアーゼをコードする前記核酸が、標的肝細胞に送達される、請求項55〜64のいずれか1項に記載の方法。
