Abstract
Description
本出願は、2009年12月21日出願の米国特許仮出願第61/288,666号、及び2010年6月2日出願の同第61/350,747号に基づく優先権を主張するものであり、これらの出願をいずれもその全容にわたって本出願に援用するものである。
本発明の組成物及び方法は、サーモビフィダ・フスカ(Thermobifida fusca)からクローニングされたリパーゼ、そのリパーゼをコードしたポリヌクレオチド、及びそれらの使用方法に関する。
以下に、サーモビフィダ・フスカよりクローニングされたリパーゼ(TfuLip2)に関連する組成物及び方法について述べる。当該組成物及び方法は、クローニング及び発現されたTfuLip2が、洗剤組成物の存在下でカルボン酸エステル加水分解酵素の活性を有するという観察に一部基づいたものである。TfuLip2はまた、洗剤組成物中でプロテアーゼが存在する場合であっても優れた安定性を示す。これらの特徴のため、TfuLip2は様々な洗浄用途における使用に極めて適したものであり、この酵素は、洗剤組成物中に見出される界面活性剤及び他の成分の存在下で脂質を加水分解することができる。
本発明の組成物及び方法を詳細に述べるのに先立って、理解を助けるため、以下の用語を定義する。定義されない用語及び略語は、当該技術分野において用いられるそれらの通常の意味を有するものとする。
A.TfuLip2ポリペプチド
一態様では、本発明の組成物及び方法は、組換えTfuLip2ポリペプチド又はその変異体を提供する。代表的なTfuLip2ポリペプチドとしては、サーモビフィダ・フスカ(GENBANKアクセッション番号YP_288944)から単離されたものがある。成熟したTfuLip2ポリペプチドは、配列番号3のアミノ酸配列を有する。同様のほぼ同一のTfuLip2ポリペプチドが、例えばT.フスカの他の株又は単離物など、自然に存在しうる。本発明の組成物及び方法には、これら及び他の組換えTfuLip2ポリペプチドが含まれる。
本組成物及び方法の別の態様は、本明細書において特定されるもののような異種生物内でTfuLip2ポリペプチドを発現させるための発現ベクターとの関連で提供される、TfuLip2ポリペプチド(その変異体及びフラグメントを含む)をコードするポリヌクレオチドである。TfuLip2ポリペプチドをコードするポリヌクレオチドは、コードされたポリペプチドの発現を助けるための調節要素(例えば、プロモーター、ターミネーター、エンハンサーなど)と機能的に連結させることができる。
本明細書において開示されるTfuLip2ポリペプチドは、広範なpH条件にわたって酵素活性を有しうる。特定の実施形態では、開示されるTfuLip2ポリペプチドは、約pH 4〜約pH 11.5で酵素活性を有する。好ましい実施形態では、TfuLip2は約pH 8〜約pH 10で活性である。本明細書において述べるpHの値は、±0.2だけ変化しうることに注意されたい。例えば、約8のpHの値は、pH 7.8〜pH 8.2で変化しうる。
本明細書において開示される組成物及び方法の一態様は、TfuLip2ポリペプチド(その変異体又はフラグメントを含む)を含む洗剤組成物、及び洗浄用途においてこうした組成物を使用するための方法である。洗浄用途としては、これらに限定されるものではないが、洗濯又は繊維製品のクリーニング、食器洗い(手洗い及び自動)、染みの前処理などが挙げられる。特定の用途は、脂質が除去すべき汚れ又は染みの成分であるような用途である。洗剤組成物は、通常、TfuLip2又はその変異体の有効量、例えば、少なくとも0.0001重量%、約0.0001〜約1、約0.001〜約0.5、約0.01〜約0.1重量%、又は更には約0.1〜約1重量%、又はそれよりも多い量を含む。約0.4g/L〜約2.2g/L、約0.4g/L〜約2.0g/L、約0.4g/L〜約1.7g/L、約0.4g/L〜約1.5g/L、約0.4g/L〜約1g/L、約0.4g/L〜約0.8g/L、又は約0.4g/L〜約0.5g/Lの濃度を有する洗剤組成物を、TfuLip2リパーゼの有効量と混合することができる。洗剤組成物は、約0.4mL/L〜約2.6mL/L、約0.4mL/L〜約2.0mL/L、約0.4mL/L〜約1.5m/L、約0.4mL/L〜約1mL/L、約0.4mL/L〜約0.8mL/L、又は約0.4mL/L〜約0.5mL/Lの濃度で存在してもよい。
短鎖の脂質に対するTfuLip2の選択性のために、本発明のポリペプチドは、C4〜C16の基質が関与するエステル交換反応を行ううえで特に有用である。代表的な用途としては、ミルク脂肪の加水分解、構造化トリグリセリドの合成、ポリマーの合成及び分解、乳化剤及び界面活性剤の生成、香料及び芳香剤として使用されるエステルを製造する目的、バイオ燃料及び合成潤滑剤を製造する目的、過酸を生成する目的、並びに油脂化学産業における他の用途での、パーソナルケア製品、医薬品及び農薬の成分の合成がある。上記に述べた酵素の更なる使用については、米国特許出願公開第20070026106号、同第20060078648号、及び同第20050196766号、並びに国際特許出願公開第2005/066347号に述べられており、これらの文書を援用するものである。
サーモビフィダ・フスカリパーゼ2(TfuLip2)のクローニング及び発現
サーモビフィダ・フスカリパーゼ2(又はBTA加水分解酵素2)の遺伝子はこれまでに同定されており(Lykidisら、J.Bacteriol,189:2477〜2486,2007)、その配列は、GENBANKアクセッション番号YP_288944として公開されている。B.サブチリス発現ベクターp2JM103BBI(Vogtentanz,Protein Expr Purif,55:40〜52,2007)を制限酵素BssHII及びHindIIIで消化した。BCE103−BBI融合遺伝子配列を含まないDNAフラグメントを単離して、発現用のバックボーンとして使用した。このフラグメントを、TfuLip2酵素をコードした合成遺伝子にライゲートすることにより、TfuLip2ポリペプチドのN末端を、p2JM103BBIによってコードされるバチルス・サブチリスのAprEプロペプチドの3番目のアミノ酸に融合させた。ホスト内での天然のシグナルペプチダーゼによる切断後、このようにして生成された組換えTfuLip2タンパク質は、アミノ末端に3個の更なるアミノ酸(Ala−Gly−Lys)を有していた。
成熟したTfuLip2酵素のアミノ酸配列を配列番号2に示す。
3個のアミノ酸からなるアミノ末端の延長部分を有するTfuLip2酵素のアミノ酸配列を、配列番号3に示す。
TfuLip2タンパク質を、これまでに述べられている方法(Vogtentanz,Protein Expr Purif,55:40〜52,2007)を用いて、バチルス・サブチリス細胞(degUHy32,oppA,ΔspoIIE,ΔaprE,ΔnprE,Δepr,ΔispA,Δbpr,Δvpr,ΔwprA,Δmpr−ybfJ,ΔnprB,amyE::xylRPxylAcomK−ermC)内で産生させた。
TfuLip2の単離及び特性決定
限外濾過濃縮物を、発現用のバチルス・サブチリス株の14Lスケールの回分発酵より得た。清澄化したブロスを、組換えTfuLip2ポリペプチドの特性決定に用いた。
TfuLip2によるp−ニトロフェニルエステルの加水分解
エステル鎖の長さが異なる3種類の異なるp−ニトロフェニル(pNP)エステル基質に対してリパーゼ活性についてTfuLip2タンパク質をアッセイし、LipAの鎖長選択性を調べた。表3−1に、pNPエステル基質の詳細を示す。
洗剤の存在下及び非存在下でのTfuLip2によるトリグリセリドの加水分解
TfuLip2ポリペプチドを、洗剤の存在下及び非存在下でトリオクタン酸エステル及びトリオレイン酸エステル基質の加水分解についてアッセイした。トリオクタン酸グリセロール(CAS 538−23−8)及びトリオレイン酸グリセロール(CAS 122−32−7)基質をシグマ社(Sigma)より購入した。この実験では以下の市販の洗剤を使用した。(1)ユニリーバ社(Unilever)より販売されるOMO色物用液体洗剤、(2)プロクター・アンド・ギャンブル社(Procter & Gamble)より販売されるAriel色物用液体洗剤、(3)ブルモラー社(Blumoller)より販売されるBiotex色物用粉末洗剤、及び(4)プロクター・アンド・ギャンブル社より販売されるAriel色物用粉末洗剤。
OMO色物用液体洗剤組成物は、5〜15%のアニオン性界面活性剤及び非イオン性界面活性剤、<5%石鹸、カチオン性界面活性剤、ホスホン酸塩、香料、ブチルフェニルメチルプロピオネート、シトロネロール、酵素、及びベンゾイソチアゾリノンを含む。OMO色物用液体洗剤は、以下の界面活性剤を含んでいる。すなわち、C12〜C15パレス−7、ドデシルベンゼンスルホン酸ナトリウム、ラウレス硫酸ナトリウム、及び水添ヤシ脂肪酸ナトリウム。
Ariel色物用液体洗剤組成物は、5〜15%のアニオン性界面活性剤、<5%非イオン性界面活性剤、ホスホン酸塩、石鹸、酵素、香料、ブチルフェニルメチルプロピオネート、及びゲラニオールを含む。Ariel色物用液体洗剤は、以下の界面活性剤を含んでいる。すなわち、ドデシルベンゼンスルホン酸ナトリウム、C12〜C14パレス−7、ラウレス硫酸ナトリウム、及びC12〜C14パレス−4。
Biotex色物用粉末洗剤組成物は、15〜30%のゼオライト、5〜15%のアニオン性界面活性剤、<5%石鹸、ポリカルボキシレート、ホスホン酸塩、酵素、及び香料を含む。Biotex色物用粉末洗剤は、C12〜C15パレス−7界面活性剤を含んでいる。
Ariel色物用粉末洗剤組成物は、5〜15%のアニオン性界面活性剤、ゼオライト、<5%非イオン性界面活性剤、ポリカルボキシレート、ホスホン酸塩、酵素、香料、ヘキシルシンナマル、リモネン、及びブチルフェニルメチルプロピオネートを含む。Ariel色物用粉末洗剤は、以下の界面活性剤を含んでいる。すなわち、ドデシルベンゼンスルホン酸ナトリウム、C12〜C15パレス硫酸ナトリウム、及びC12〜C15パレス−7。
TfuLip2の洗浄性能
染みが付いた衣類に対するTfuLip2の洗浄性能について、微小布片アッセイ方式で試験を行った。24穴プレート方式(ヌンク社(Nunc)、デンマーク)で設定された脂質含有実験用染み(オランダの試験材料センター(Center for Testmaterials)より購入したCS−61布片)を使用して、染みの除去実験を行った。各アッセイウェルに、予め切断したCS−61布片の13mmの小片が入るように設定した。各布片は、リフレクトメータ(CR−400、コニカ・ミノルタ社)を使用して予め読み取ってから、24穴プレートに置いた。
TfuLip2を含む液体洗濯洗剤組成物
この実施例では、液体洗濯洗剤組成物の異なる配合を提供する。これらの配合のそれぞれにおいて、TfuLip2は約0.0001〜約10重量%の濃度で含まれる。いくつかの代替的な実施形態では、配合者によってニーズに応じて決定される他の濃度も有用であろう。
TfuLip2を含む液体食器手洗い用洗剤組成物
この実施例では、異なる食器手洗い用液体洗剤の配合を提供する。これらの配合のそれぞれにおいて、TfuLip2は約0.0001〜約10重量%の濃度で含まれる。いくつかの代替的な実施形態では、配合者によってニーズに応じて決定される他の濃度も有用であろう。
TfuLip2を含む液体自動食器洗い用洗剤組成物

TfuLip2を含む粒状及び/又は錠剤洗濯組成物
この実施例では、粒状及び/又は錠剤洗濯洗剤の異なる配合を提供する。これらの配合のそれぞれにおいて、TfuLip2は約0.0001〜約10重量%の濃度で含まれる。いくつかの代替的な実施形態では、配合者によってニーズに応じて決定される他の濃度も有用であろう。
TfuLip2を含む更なる液体洗濯洗剤
この実施例では、液体洗濯洗剤の更なる配合を提供する。これらの配合のそれぞれにおいて、TfuLip2は約0.0001〜約10重量%の濃度で含まれる。いくつかの代替的な実施形態では、配合者によってニーズに応じて決定される他の濃度も有用であろう。
TfuLip2を含む高密度食器洗い用洗剤
この実施例では、高密度食器洗い用洗剤の異なる配合を提供する。これらの圧縮型配合のそれぞれにおいて、TfuLip2は約0.0001〜約10重量%の濃度で含まれる。いくつかの代替的な実施形態では、配合者によってニーズに応じて決定される他の濃度も有用であろう。
TfuLip2を含む錠剤食器洗い用洗剤組成物
この実施例は、異なる錠剤食器洗い用洗剤の配合を提供する。以下の本開示の錠剤洗剤組成物は、12個のヘッドを有する標準的なロータリープレスを使用して、13KN/cm2(130MPa)の圧力で粒状食器洗い用洗剤組成物を圧縮することによって調製される。これらの配合のそれぞれにおいて、TfuLip2は約0.0001〜約10重量%の濃度で含まれる。いくつかの代替的な実施形態では、配合者によってニーズに応じて決定される他の濃度も有用であろう。
TfuLip2を含む液体硬質表面洗浄用洗剤
この実施例では、液体硬質表面洗浄用洗剤の異なる配合を提供する。これらの配合のそれぞれにおいて、TfuLip2は約0.0001〜約10重量%の濃度で含まれる。いくつかの代替的な実施形態では、配合者によってニーズに応じて決定される他の濃度も有用であろう。
プロテアーゼの存在下及び非存在下での洗剤中のTfuLip2の安定性
洗剤中のTfuLip2の安定性を、市販の洗剤においてプロテアーゼの存在下又は非存在下で調べ、同様の条件下における市販の基準酵素Lipex(登録商標)(サーモマイセス・ラヌギノーサスLip3リパーゼ(ノボザイムズ社(Novozymes)、コペンハーゲン、デンマーク))の安定性と比較した。
異なる温度におけるTfuLip2の洗浄性能
染み付きの布地に対するTfuLip2の洗浄性能を、市販の加熱不活性化したAriel色物用液体洗剤及びAriel色物用粉末洗剤中で、微小布片アッセイ方式で15℃、20℃、30℃、及び40℃で試験した。アッセイは、プレートを37℃ではなく、それぞれ15、20、30、及び40℃で振盪する変更を行った以外は実施例5で述べたのと同様にして行った。0.2又は0.7U/mLのTfuLip2を添加し、トリオレイン酸エステル(pH 8.2)から放出される遊離脂肪酸(μmol/分)を実施例4で述べたのと同様にして測定した。結果を表15−1、15−2、及び15−3に示す。
Claims (23)
- サーモビフィダ・フスカから得られるリパーゼと、
界面活性剤と、を含む洗剤組成物であって、
洗浄しようとする表面から油性の染みを除去するうえで、前記リパーゼを含まない同等の洗剤組成物よりも効果的である、洗剤組成物。 - 前記リパーゼが、TfuLip2リパーゼである、請求項1に記載の洗剤組成物。
- 前記リパーゼが、配列番号2又は配列番号3と少なくとも90%のアミノ酸配列同一性を有するアミノ酸配列を含む、請求項1又は2に記載の洗剤組成物。
- 前記リパーゼが、配列番号2又は配列番号3と少なくとも95%のアミノ酸配列同一性を有するアミノ酸配列を含む、請求項3に記載の洗剤組成物。
- 前記リパーゼが、組換えリパーゼである、請求項1〜4のいずれか一項に記載の洗剤組成物。
- 前記リパーゼが、バチルス内で発現される組換えリパーゼである、請求項1〜5のいずれか一項に記載の洗剤組成物。
- 前記界面活性剤が、イオン性又は非イオン性界面活性剤である、請求項1〜6のいずれか一項に記載の洗剤組成物。
- 前記界面活性剤が、アニオン性界面活性剤、カチオン性界面活性剤、双性イオン性界面活性剤、及びこれらの組み合わせからなる群から選択される1以上の界面活性剤である、請求項1〜7のいずれか一項に記載の洗剤組成物。
- 前記界面活性剤が、ドデシルベンゼンスルホン酸ナトリウム、水添ヤシ脂肪酸ナトリウム、ラウレス硫酸ナトリウム、C12〜14パレス−7、C12〜15パレス−7、C12〜15パレス硫酸ナトリウム、及びC14〜15パレス−4からなる群から選択される1以上の界面活性剤を含む、請求項1〜8のいずれか一項に記載の洗剤組成物。
- 約8.0〜約10.0のpHで配合される、請求項1〜9のいずれか一項に記載の洗剤組成物。
- 約8.2〜約10.0のpHで配合される、請求項1〜10のいずれか一項に記載の洗剤組成物。
- 前記洗剤組成物が、洗濯洗剤、食器洗い用洗剤、及び硬質表面洗浄用洗剤からなる群から選択される、請求項1〜11のいずれか一項に記載の洗剤組成物。
- 前記組成物の形態が、液体、粉末、粒状固形物、及び錠剤からなる群から選択される、請求項1〜12のいずれか一項に記載の洗剤組成物。
- 前記洗剤組成物が、約30℃〜約40℃の温度で効果的に脂質を加水分解する、請求項1〜13のいずれか一項に記載の洗剤組成物。
- 前記洗剤組成物が、サーモビフィダ・フスカのリパーゼの代わりにシュードモナス・シュードアルカリゲネスのリパーゼ変異体M21L(LIPOMAX(商標))を含む同等の洗剤組成物と比較して、C4〜C16の基質をより効果的に加水分解する、請求項1〜14のいずれか一項に記載の洗剤組成物。
- プロテアーゼを更に含む、請求項1〜15のいずれか一項に記載の洗剤組成物。
- サブチリシンプロテアーゼを更に含む、請求項16に記載の洗剤組成物。
- サーモビフィダ・フスカリパーゼの安定性が、サーモビフィダ・フスカリパーゼの代わりにサーモマイセス・ラヌギノーサのLip3リパーゼ(LIPEX(登録商標))を含む同等の洗剤組成物中におけるサーモマイセス・ラヌギノーサLip3リパーゼの安定性よりも高い、請求項16又は17に記載の洗剤組成物。
- 安定性が、最終的な洗浄媒質中で測定される、請求項18に記載の組成物。
- 表面の汚れ又は染みに存在する脂質を加水分解するための方法であって、前記表面を、請求項1〜19のいずれか一項に記載の洗剤組成物と接触させる工程を含む、方法。
- エステル交換反応を行うための方法であって、ドナー分子を、請求項1〜19のいずれか一項に記載の洗剤組成物と接触させる工程を含む、方法。
