Description
- Die vorliegende Erfindung betrifft Verfahren zur Herstellung von Paroxetinhydrochloridanhydrat.
- EP-B-223403 (Beecham Group plc) beschreibt Paroxetinhydrochloridhemihydrat und seine Verwendung bei der Behandlung bestimmter Erkrankungen. Beispiel 8 in diesem Dokument beschreibt die Herstellung von Paroxetinhydrochloridanhydrat als bei 118°C schmelzende Plättchen und mit IR-Banden bei 890, 1200, 1490, 3400 und 3640 cm–1 durch Kristallisation aus einem wasserhaltigen Lösungsmittel. Dieses Material wird nachstehend als Form Z bezeichnet. Die nachfolgende Wiederholung des in Beispiel 8 beschriebenen Verfahrens führte jedoch nicht zu Paroxetinhydrochloridanhydrat jeglichen Typs, und es gibt auch keine eindeutige Lehre an anderer Stelle im Dokument über alternative Wege oder Abwandlungen des Verfahrens, die dann zum Anhydrat führen würden.
- Paroxetinhydrochloridanhydrat soll auch im International Journal of Pharmaceutics 42 (1988) 135–143, veröffentlicht von Elsevier, offenbart sein. Es heißt, dass das Anhydrat durch Kristallisieren von Paroxetinhydrochlorid aus wasserfreiem Propan-2-ol erzeugt wird. Die nachfolgende Wiederholung dieses Verfahrens führte zu einem Propan-2-ol-Solvat von Paroxetinhydrochlorid. Das heißt, dass Propan-2-ol in dem Produkt gebunden ist. Dieses gebundene Propan-2-ol kann durch herkömmliche Trocknungsverfahren, wie das Trocknen in einem Vakuumofen, nicht entfernt werden.
- In einem ersten Aspekt stellt die vorliegende Erfindung ein Verfahren zur Herstellung von Paroxetinhydrochloridanhydrat bereit, das weniger als 2% gebundenes organisches Lösungsmittel umfasst, welches das Verdrängen des solvatisierten Lösungsmittels von einem Paroxetinhydrochloridsolvat unter Verwendung eines Verdrängungsmittels umfasst, wobei das solvatisierte Lösungsmittel Aceton und das Verdrängungsmittel Wasser ist.
- Folglich beschreibt die vorliegende Erfindung Paroxetinhydrochloridanhydrat, das im wesentlichen frei von Propan-2-ol ist.
- Die vorliegende Erfindung beschreibt auch Paroxetinhydrochloridanhydrat, das im wesentlichen frei von gebundenem organischen Lösungsmittel ist.
- Im wesentlichen frei von gebundenem organischen Lösungsmittel ist so zu verstehen, dass es weniger als die Menge von Propan-2-ol ist, die solvatisiert, d. h. gebunden, im Kristallgitter des Produktes unter herkömmlichen Bedingungen der Trocknung im Vakuumofen zurückbleiben würde.
- Die vorliegende Erfindung beschreibt auch andere Paroxetinhydrochloridsolvate als Propan-2-olsolvate als Vorläufer bei der Herstellung von Paroxetinhydrochloridanhydrat, das im wesentlichen frei von gebundenen organischen Lösungsmitteln ist. Beispiele solcher Solvate schließen Ketonsolvate wie Aceton ein.
- Vorzugsweise wird Paroxetinhydrochloridanhydrat, das im wesentlichen frei von gebundenem Propan-2-ol ist, in im wesentlichen reiner Form bereit gestellt. Geeigneterweise wird Paroxetinhydrochloridanhydrat, das im wesentlichen frei von gebundenem Propan-2-ol ist, in einer Reinheit des Paroxetinhydrochloridanhydrats von mehr als 50%, vorzugsweise von mehr als 60%, stärker bevorzugt von mehr als 70%, noch stärker bevorzugt von mehr als 80% und sogar noch stärker bevorzugt von mehr als 90% bereitgestellt. Am stärksten bevorzugt wird das Paroxetinhydrochloridanhydrat in im wesentlichen reiner Form bereitgestellt, d. h. Paroxetinhydrochloridanhydrat, das im wesentlichen frei von gebundenem Propan-2-ol ist, ist zu mehr als 95% rein.
- Es sollte klar sein, dass das Paroxetinhydrochloridanhydrat, das im wesentlichen frei von gebundenem Propan-2-ol ist, ungebundenes Wasser enthalten kann, also Wasser das kein Kristallwasser ist.
- Typischerweise ist die Menge des gebundenen organischen Lösungsmittels auf einer Gewicht/Gewicht-Basis weniger als 2,0%, vorzugsweise weniger als 1,8%, stärker bevorzugt weniger als 1,5%, noch stärker bevorzugt weniger als 1,0%, sogar noch stärker bevorzugt 0,5% und am stärksten bevorzugt weniger als 0,1%.
- Im Allgemeinen werden vorwiegend alle Prozentangaben auf einer Gewicht/Gewicht-Basis angegeben, soweit nicht anders vermerkt.
- Eine Form von Paroxetinhydrochloridanhydrat, das im wesentlichen frei von gebundenem Propan-2-ol oder im wesentlichen frei von gebundenem organischen Lösungsmittel ist, ist Paroxetinhydrochloridanhydrat in der Form A (wie nachstehend definiert).
- Diese Form von Paroxetinhydrochloridanhydrat kann unterschieden werden von dem Material, das aus den Verfahren hervorgeht, die in EP-B 0 223 403 und im „Journal of Pharmaceutics 42", (1988), 135 bis 143, beschrieben werden, durch Kristallform, Lösungsmittelanalyse oder Techniken wie z. B. IR, Schmelzpunkt, Röntgenbeugung, NMR, DSC, mikroskopische und jede andere analytische Technik, die eine Form von der anderen unterscheidet.
- Die Form A, die im wesentlichen frei von Lösungsmittel ist, kann z. B. durch die folgenden Analysendaten von anderen Formen unterschieden werden. Form A weist einen Schmelzpunkt von etwa 123°C–125°C auf, wenn sie in ähnlicher Reinheit wie das in Beispiel 1 beschriebene Material erhalten wird, die durch herkömmliche Verfahren, wie HPLC, bestimmt werden kann, und weist signifikante IR-Banden (
1 ) bei etwa 513, 538, 571, 592, 613, 665, 722, 761, 783, 806, 818, 839, 888, 906, 924, 947, 966, 982, 1006, 1034, 1068, 1091, 1134, 1194, 1221, 1248, 1286, 1340, 1387, 1493, 1513, 1562, 1604, 3402, 3631 cm–1 auf. - Die bei 10°C pro Minute gemessene DSC-Exotherme zeigt in offenen Schalen ein Maximum bei 126°C und in geschlossenen Schalen ein Maximum bei etwa 121°C. Die Form A zeigt auch ein im wesentlichen ähnliches Röntgenbeugungsdiagramm wie das in
4 gezeigte, zum Beispiel liegen charakteristische Signale bei 6,6, 8,0, 11,2, 13,1° 2θ vor, und ein im wesentlichen ähnliches Festkörper-NMR-Spektrum wie das in7 , zum Beispiel mit charakteristischen Signalen bei 154,3, 149,3, 141,6, 138,5 ppm. - Die Frage, welche spezielle Form eine spezielle Paroxetinhydrochloridanhydratprobe hat, kann leicht unter Verwendung herkömmlicher Techniken durch einen Fachmann beantwortet werden, mit Bezug auf die oben angegebenen Daten, die Daten in den Beispielen und auf jede andere herkömmliche Art.
- Die Form A liegt bevorzugt als Nadeln vor.
- Die vorliegende Erfindung stellt auch ein Verfahren zur Herstellung von Paroxetinhydrochloridsolvaten bereit, die sich vom Propan-2-ol unterscheiden, das das Kristallisieren von Paroxetinhydrochlorid in einem organischen Lösungsmittel oder einem Gemisch aus Lösungsmitteln, die ein Solvat mit dem Paroxetinhydrochloridanhydrat bilden und die nicht durch herkömmliche Trocknungsverfahren zu entfernen sind, umfasst.
- Nach einem bevorzugten Aspekt der Erfindung wird die Kristallisation von Paroxetinhydrochloridanhydrat durch das Inkontaktbringen einer Lösung der freien Base Paroxetin in einem organischen Lösungsmittel oder Lösungsmitteln mit trockenem Chlorwasserstoffgas erreicht.
- Alternativ kann das Wasser vor der Kristallisation von Paroxetinhydrochlorid durch azeotrope Destillation entfernt werden.
- Deshalb wird, nach einem anderen Aspekt der Erfindung, Paroxetinhydrochloridanhydrat kristallisiert, indem Paroxetinhydrochloridhemihydrat in einem geeigneten Lösungsmittel gelöst wird, das im wesentlichen frei von Wasser ist und das mit Wasser ein Azeotrop bildet. Geeigneterweise wird das Lösungsmittel durch Destillation entfernt und ein frisches Lösungsmittel, das im wesentlichen frei von Wasser ist, wird hinzugefügt bis das ganze Wasser entfernt ist.
- Paroxetinhydrochloridhemihydrat oder die freie Base davon kann gemäß den allgemein in EP-B-0223403 angegebenen Verfahren hergestellt werden.
- Die organischen Lösungsmittel sollten im wesentlichen bis zu dem Grad wasserfrei sein, dass zum Zeitpunkt der Kristallisation nicht genügend Wasser vorliegt, um die Umwandlung in das Hydrochloridhemihydrat zu bewirken. Organische Lösungsmittel, die im wesentlichen wasserfrei sind, können auf herkömmliche Art und Weise erhalten werden. Sie können zum Beispiel unter Verwendung herkömmlicher Verfahren, wie das Trocknen über Molekularsieben, getrocknet werden, oder sie können gekauft werden.
- Faktoren, welche die Form beeinflussen, in der das Produkt erhalten wird, schliessen die jeweilige Wahl des zu verwendenden organischen Lösungsmittels oder der Lösungsmittel ein und werden von der jeweiligen Form des gewünschten Produkts abhängen.
- Es sollte auch verständlich sein, dass das Verfahren zum Entfernen des Lösungsmittels auch von der jeweiligen Form des gewünschten Produkts abhängt.
- Das Paroxetinhydrochloridsolvat, das mit dem im ersten Aspekt der Erfindung beschriebenen Verfahren hergestellt wird, wird geeigneterweise durch herkömmliche Verfahren isoliert und getrocknet wie z. B. Vakuumtrocknen, um etwas oder das gesamte freie oder ungebundene Lösungsmittel zu entfernen. Es sollte verständlich sein, dass es vorzuziehen und unerwartet ist, dass der Trocknungsgrad so kontrolliert wird, dass nur freies Lösungsmittel entfernt wird. Das gebundene Lösungsmittel wird dann mit einem Verdrängungsmittel wie z. B. Wasser verdrängt. Es ist möglich andere Verdrängungsmittel zu verwenden, die durch Routineexperimente ausgewählt werden können.
- Vorzugsweise kann gasförmiges oder flüssiges Wasser als Verdrängungsmittel verwendet werden. Es ist wichtig, dass das Paroxetinhydrochloridsolvat mit ausreichend Wasser über eine ausreichend lange Zeit in Kontakt kommt um das Lösungsmittel zu verdrängen, jedoch nicht lange genug um die Umwandlung zu Hydrochloridhemihydrat zu bewirken.
- Die Wassermenge, die Wasserform, z. B. flüssig oder gasförmig, und der Zeitraum während dem das Paroxetinhydrochloridsolvat mit dem Wasser in Kontakt ist, unterscheidet sich von Solvat zu Solvat. Dies hängt größtenteils von der Löslichkeit des jeweiligen Solvats ab.
- Spezielle Paroxetinhydrochloridsolvat/Wasser-Verhältnisse werden in den nachstehend beschriebenen Beispielen umrissen (Beispiel 1 und 2). Die Verwendung des allgemeinen Ioneneffekts bei Verwendung von verdünnter Salzsäure kann dazu beitragen das Lösen des Solvats und die darauffolgende Umwandlung zu Hydrochloridhemihydrat zu verhindern.
- Nach dem Kontakt mit Wasser um das gebundene Lösungsmittel zu verdrängen, wird das Produkt geeigneterweise getrocknet, z. B. im Vakuum bei erhöhter Temperatur. Geeignete Trocknung kann über Trockenmitteln wie z. B. Phosphorpentoxid stattfinden.
- Das Verfahren der Erfindung wird vorzugsweise verwendet um Form A herzustellen.
- Die Kristallisation des Vorläufersolvats von Paroxetinhydrochloridanhydrat der Form A kann vorzugsweise unterstützt werden durch die Zugabe von Impfkristallen vom Vorläufersolvat von Paroxetinhydrochloridanydrat der Form A.
- Alternativ können Impfkristalle von Paroxetinhydrochloridanhydrat der Form A verwendet werden, um die Kristallisation des Vorläufersolvats von Paroxetinhydrochloridanhydrat der Form A zu unterstützen.
- Impfkristalle der Form A können gemäß den hier beschriebenen Verfahren hergestellt werden oder sind auf Nachfrage von Corporate Intellectual Property, SmithKline Beecham plc im New Frontiers Science Park, Third Avenue, Harlow, Essex, CM19 5AW, GB frei erhältlich. Die Form A ist BRL 29060F.
- Die nachstehenden Beispiele 1 und 2 veranschaulichen die vorliegende Erfindung:
- Referenzbeispiel
- Kristallines, im wesentlichen von gebundenem Propan-2-ol freies Paroxetinhydrochloridanhydrat (Form A)
- i) Paroxetinhydrochloridpropan-2-olsolvat
- 150 g Paroxetinhydrochloridhemihydrat wurden mit 1000 ml Propan-2-ol und 300 ml Toluol in einem Rundkolben gerührt und zum Sieden erhitzt. Das Lösungsmittel wurde durch Destillation entfernt, wobei das Gesamtvolumen durch die Zugabe von frischem Propan-2-ol aufrechthalten wurde, bis der Siedepunkt etwa 82°C erreicht hatte, was zeigte, dass das gesamte Wasser entfernt war.
- Man ließ das Gemisch auf ungefähr 50°C abkühlen, worauf es spontan kristallisierte. Der Kolbeninhalt verfestigte sich schnell zu einem dicken Brei, der mit ungefähr 500 ml Propan-2-ol verdünnt und kräftig gerührt wurde. Man ließ die entstandene Suspension auf ungefähr 30°C abkühlen und filtrierte unter Vakuum, wobei man darauf achtete, dass die Ab sorption von atmosphärischer Feuchtigkeit vermieden wurde. Der Lösungsmittelnasse Kuchen wurde im Hochvakuum über Phosphorpentoxid getrocknet.
Ausbeute des solvatisierten Paroxetinhydrochlorids: 151 g, Propan-2-olgehalt: 13,0% (durch NMR geschätzt). - Das Infrarotspektrum (Nujolfilm) zeigte unter anderem eine charakteristische Bande bei 667 cm–1.
- ii) Paroxetinhydrochloridanhydrat (Form A)
- 110 g Paroxetinhydrochloridpropan-2-olsolvat [Propan-2-olgehalt 13,0%] wurden 20 Minuten mit 275 ml Wasser in einem Becherglas gerührt. Das Gemisch wurde unter Vakuum filtriert, und der feuchte Feststoff wurde im Vakuum über Phosphorpentoxid bis zur Gewichtskonstanz getrocknet.
Ausbeute des Paroxetinhydrochloridanhydrats Form A: 91,0 g
Wassergehalt: 0,13% (KF), Propan-2-olgehalt: 0,05% (durch NMR geschätzt)
Schmelzpunkt: 123–125°C - Die bei 10°C pro Minute gemessene DSC-Exotherme zeigte unter Verwendung einer offenen Schale ein Maximum bei etwa 126°C und unter Verwendung einer geschlossenen Schale ein Maximum bei etwa 121°C.
- Das Infrarotspektrum (Nujolfilm) zeigte unter anderem charakteristische Banden bei 665, 3631 und 3402 cm–1 (siehe
1 ). - Die Probe wurde auch durch Pulver-Röntgenbeugung (siehe
4 ) und durch Festkörper-13C-NMR (siehe7 ) untersucht. - Beispiel 1
- Paroxetinhydrochloridanhydrat, im wesentlichen frei von gebundenem Aceton (Form A)
- i) Paroxetinhydrochloridacetonsolvat
- 10,51 g der freien Base Paroxetin wurden in 40 ml Aceton [getrocknet mit 4A Molekularsieben] gelöst, und eine Lösung von 1,31 g Chlorwasserstoffgas in 10 ml trockenem Aceton wurde unter Rühren zugegeben. Die Kristallisation trat spontan innerhalb 1 Minute auf und die Mischung konnte schnell nicht mehr gerührt werden. Nach ungefähr einer halben Stunde wurde das Produkt filtriert, in einen Exsikkator mit Phosphorpentoxid gegeben, und bei Raumtemperatur über Nacht getrocknet.
- Gewicht des Produkts: 11,24 g. Acetongehalt (geschätzt mit NMR) 4% Gew./Gew. Das Infrarotspektrum zeigte eine charakteristische Bande bei 667 cm–1.
- Ungefähr die Hälfte des Produkts wurde in einen auf 50°C eingestellten Vakuumofen gegeben und weiter bis zu einem konstanten Gewicht getrocknet. NMR-Analysen des resultierenden Produkts zeigten die Gegenwart von 1,2% Aceton Gew./Gew.
- ii) Paroxetinhydrochloridanhydrat (Form A)
- Eine Probe des Acetonsolvats (5,18 g) wurde 10 Minuten in 20 ml Wasser gerührt, filtriert und im Vakuumofen bei 50°C über Phosphorpentoxid getrocknet.
- Gewicht des Produkts: 4,63 g. Die Analyse durch NMR zeigte die Gegenwart von 0,6% Aceton Gew./Gew. Das Infrarotspektrum entsprach dem Spektrum von Paroxetinhydrochloridanhydrat der Form A und zeigte eine charakteristische Bande bei 665 cm–1.
- Beispiel 2
- Paroxetinhydrochloridanhydrat, im wesentlichen frei von gebundenem Aceton (Form A)
- i) Herstellung von Paroxetinhydrochloridacetonsolvat
- Eine Suspension von 5,0 g Paroxetinhydrochloridanhydrat der Form C (Prismen) in 75 ml Aceton wurde bis zum Siedepunkt erwärmt, um eine Masse an feinen Nadeln zu ergeben. Der Kolben wurde verschlossen und bei Raumtemperatur über Nacht stehen gelassen. Das Lösungsmittel wurde unter Verwendung eines Rotationsverdampfers bei niedrigen Temperaturen entfernt und durch 100 ml Hexan ersetzt. Das Lösungsmittel wurde erneut bei niedrigen Temperaturen entfernt, um das Acetonsolvat als kristallinen Rückstand zu ergeben. NMR-Analysen zeigten die Gegenwart von 12,2 Gew.-% Aceton und das Infrarotspektrum (Nujolfilm) zeigte charakteristische Banden bei 667 und 1714 cm–1.
- ii) Herstellung von Paroxetinhydrochloridanhydrat (Form A) aus dem Acetonsolvat
- 5,3 g Paroxetinhydrochlorid der Form C wurde durch ein ähnliches Verfahren wie oben beschrieben in Acetonsolvat umgewandelt. 50 ml Wasser wurden zu gegeben und die resultierende Suspension wurde für 10 Minuten leicht geschüttelt. Der weiße Feststoff wurde durch Filtration aufgenommen, gründlich entwässert und bei 50°C in einem Vakuumofen getrocknet. Ausbeute: 4,6 g. Acetongehalt: 0,1 Gew.-% (NMR). Das Infrarotspektrum (Nujolfilm) entsprach dem einer Standardprobe von Paroxetinhydrochloridanhydrat der Form A.
Claims (1)
- Verfahren zur Herstellung von Paroxetinhydrochloridanhydrat, umfassend weniger als 2% gebundenes organisches Lösungsmittel, welches Verdrängen des solvatisierten Lösungsmittels aus einem Paroxetinhydrochloridsolvat unter Verwendung eines Verdrängungsmittels umfasst, wobei das solvatisierte Lösungsmittel Aceton ist und das Verdrängungsmittel Wasser ist.
