Abstract
Niniejszy wynalazek dotyczy wyizolowanego kwasu nukleinowego obejmującego sekwencję kodującą fuzję polipeptydową mającą aktywność ß(1,4)-galaktozylotransferazy (GalT), przy czym fuzja polipeptydowa obejmuje domenę katalityczną GalT i domenę lokalizacji w aparacie Golgiego ß(1, 2)-N-acetyloglukoza-minylotransferazy I (GnTI) lub mannozydazy II (Man II), przy czym domena katalityczna GalT obejmuje sekwencję aminokwasową co najmniej w 90% identyczną z sekwencją aminokwasową o SEK.ID.NR: 22, z konserwatywnymi podstawieniami aminokwasowymi, a domena lokalizacji Man II w aparacie Golgiego obejmuje sekwencję aminokwasową co najmniej w 90% identyczną z sekwencją aminokwasową o SEK. ID. NR: 23, z konserwatywnymi podstawieniami aminokwasowymi, a domena GnTI lokalizacji w aparacie Golgiego obejmuje sekwencję aminokwasową co najmniej w 90% identyczną z sekwencją aminokwasową o SEK.ID. NR:24, z konserwatywnymi podstawieniami aminokwasowymi. Ponadto w zakres wynalazku wchodzą ssaczy wektor ekspresyjny, fuzje polipeptydowe, komórki gospodarza, sposób wytwarzania fuzji polipeptydowych, sposoby in vitro i ex vivo modyfikowania profilu glikozylacji oraz wyizolowana cząsteczka kwasu nukleinowego.
