Description
Den foreliggende oppfinnelse angår en fremgangsmåte for fremstilling av mikrokapsler, mikrokuler, nanokapsler og nanokuler. Mer spesielt angår den foreliggende oppfinnelse en emulsjons-basert fremgangsmåte for mikroinnkapsling av et middel under dannelse av et mikroinnkapslet produkt.
Mikrokapsler og mikrokuler er vanligvis pulvere som består av sfæriske partikler med diameter 2 millimeter eller mindre, vanligvis 500 nm eller mindre. Hvis partiklene er mindre enn 1/*m, omtales de ofte som nanokapsler eller nanokuler. Forskjellen mellom mikrokapsler og nanokapsler er hovedsakelig størrelsen av dem; deres indre struktur er omtrent den samme. Likeledes er forskjellen mellom mikrokuler og nanokuler størrelsen av dem; deres indre struktur er omtrent den samme.
En mikrokapsel (eller nanokapsel) har sitt innkapslede materiale, i det følgende omtalt som middel, plassert sen-tralt innenfor en unik membran, vanligvis en polymer-membran. Denne membran kan betegnes som et veggdannende materiale og er vanligvis et polymert materiale. På grunn av den indre struktur hos gjennomtrengelige mikrokapsler utformet for regulert-frigjørings-anvendelser, frigjør disse sitt middel med konstant hastighet (nulte ordens frigjøringshastighet) . Ugjennomtrengelige mikrokapsler kan også anvendes for brudd-frigjørings-anvendelser. I det følgende vil betegnelsen mikrokapsler innbefatte nanokapsler, mikrobobler (hule partikler), porøse mikrobobler og partikler generelt som omfatter en sentral kjerne omgitt av en unik ytre membran.
En mikrokule har sitt middel dispergert i hele partikkelen; det vil si at den indre struktur er en matriks av midlet og eksipiensen, vanligvis en polymer eksipiens. Vanligvis frigjør mikrokuler med regulert frigjøring sitt middel med avtakende hastighet (første orden). Men mikrokuler kan utformes slik at de frigjør midler med en nær nulte ordens hastighet. Det er en tendens til at mikrokuler er vanskeligere å bryte, sammenliknet med mikrokapsler, på grunn av at deres indre struktur er sterkere. I det følgende vil betegnelsen mikrokule innbefatte nanokuler, mikropartikler, nanopartikler, mikrosvamper (porøse mikrokuler) og partikler generelt, med en indre struktur som omfatter en matriks av middel og eksipiens.
Mange forskjellige fremgangsmåter for fremstilling av mikrokapsler og mikrokuler er beskrevet i litteraturen. Flere av disse fremgangsmåter gjør bruk av emulsjoner for fremstilling av mikrokuler, spesielt for fremstilling av mikrokuler med diameter under 2 millimeter. Som et generelt eksempel på slike fremgangsmåter kan man oppløse en polymer i et egnet organisk løsningsmiddel (polymer-løsningsmidlet), oppløse eller dispergere et middel i denne polymerløsning, dispergere den resulterende polymer/middel-blanding i en vandig fase (prosessmediet) under oppnåelse av en olje-i-vann-emulsjon med olje-mikrodråper dispergert i bearbeidelsesmediet, og fjerne løsningsmidlet fra mikrodråpene under dannelse av mikrokuler. Disse fremgangsmåter kan også utføres med vann-i-olje-emulsjoner og med dobbelt-emulsjoner.
Anvendelsen av emulsjonsbaserte fremgangsmåter som følger denne basis-fremgangsmåte, er beskrevet i flere US-patenter. For eksempel beskriver US-patent nr. 4 384 975 fremstilling av mikrokuler ved dannelse av en emulsjon og deretter langsom fjerning av polymerløsningsmidlet fra mikrodråpene i emulsjonen ved vakuumdestillasjon. Som et annet eksempel beskriver US-patent nr. 3 891 570 en fremgangsmåte hvor polymerløsningsmidlet fjernes fra mikrodråpene i emulsjonen ved anvendelse av varme eller ved reduksjon av trykket i fremstillingsbeholderen. I enda et annet eksempel, US-patent nr. 4 389 330, fjernes polymerløsningsmidlet delvis fra mikrodråpene i emulsjonen ved vakuumdestillasjon (fortrinnsvis 40-60% av polymerløsningsmidlet) , og deretter ekstraheres resten av polymerløsningsmidlet under størkning av mikrokulene.
I US-patent nr. 4 637 905 beskrives en fordampnings-faseseparasjonsprosess hvor løsningsmiddel fordampes fra en blanding av polymer, løsningsmiddel, ikke-løsningsmiddel og bioaktivt middel, noe som fører til at polymeren størkner og belegger midlet.
Ulempen ved de ovenfor beskrevne fremgangsmåter, som med andre emulsjonsbaserte fremgangsmåter, er at noen midler kan skilles ut i bearbeidelsesmediet, d.v.s. at midlene vandrer ut av mikrodråpene under polymerløsningsmiddel-fjernings-trinnet, hvilket resulterer i dårlig innkapslingseffektivi-tet. Videre gir alle de ovenfor beskrevne fremgangsmåter mikrokuler snarere enn mikrokapsler.
Ved en annen emulsjonsbasert fremgangsmåte for fremstilling av mikrokuler beskrevet i US-patent nr. 3 737 337 anvendes regulert ekstraksjon av polymerløsningsmidlet fra mikrodråpene ved tilsetting av bearbeidelsesmedium til emulsjonen ved regulert hastighet. Dette patent lærer imidlertid annet enn den foreliggende oppfinnelse ved at det beskriver at ekstraksjonen må skje langsomt, idet det ellers ikke vil bli dannet noen sfæriske partikler. Likeledes beskriver US-patent 4 652 411 en fremgangsmåte for innkapsling av vann-løselige midler fra vann-i-olje-i-vann-emulsjoner, og lærer at en substans med høy viskositet og som holder tilbake legemidlet, må innarbeides i den indre vannfase for at legemidlet skal holdes i mikrodråpene under inndampning av polymer-løsningsmidlet. US-patent nr. 4 652 441 lærer også det mot-satte av den foreliggende oppfinnelse, ved at det antyder at det er umulig effektivt å innkapsle vannløselige midler uten anvendelse av substanser som holder legemidlene tilbake i emulsjonen.

(1988) og Mathiowitz et al., J. Appln. Polymer Science, bind 35, 755-774 (1988) beskriver på tilsvarende måte emulsjonsbaserte metoder for å fremstille mikrokapsler som inneholder bioaktive midler som ikke lett migrerer inn i prosessmediet mens midlet inkorporeres i mikrokapsler.
Ett formål med den foreliggende oppfinnelse er følgelig å tilveiebringe en emulsjonsbasert fremgangsmåte for fremstilling av mikrokuler med midler som har stor tilbøyelighet til å skilles ut i bearbeidelsesmediet, den kontinuerlige fase av emulsjonen, innen minutter. Et annet formål med den foreliggende oppfinnelse er en fremgangsmåte for fremstilling av mikrokapsler så vel som mikrokuler, ut fra en emulsjon. Enda et annet formål med oppfinnelsen er å tilveiebringe en fremgangsmåte for fremstilling av mikrokuler eller mikrokapsler som inneholder et middel som har en løselighet på over 10 milligram pr. milliliter i bearbeidelsesmediet. Enda et annet formål med oppfinnelsen er å regulere porøsiteten av veggen i mikrokapslene eller eksipiensen i mikrokuler ved å regulere ekstraksjonshastigheten for løsningsmidlet fra mikrodråpene i emulsjonen. Enda et annet formål med den foreliggende oppfinnelse er å tilveiebringe en fremgangsmåte for fremstilling av mikrokapsler og mikrokuler med diameter fra under 1 jum til over 2 millimeter. Enda et annet formål med den foreliggende oppfinnelse er å tilveiebringe en fremgangsmåte for fremstilling av mikrokuler og mikrokapsler som inneholder legemiddel, som resulterer i frittstrømmende pulvere av ikke-agglomererte sfæriske partikler egnet for parenteral så vel som andre måter for administrering av legemidler .
Kort angitt innbefatter denne oppfinnelse en emulsjons-basert fremgangsmåte for mikroinnkapsling av et middel under dannelse av et mikroinnkapslet produkt,karakterisert vedat den omfatter følgende trinn: a) en effektiv mengde av midlet dispergeres eller løses opp i et løsningsmiddel som inneholder et oppløst veggdannende materiale, under dannelse av en dispersjon; b) dispersjonen blandes med en effektiv mengde av et kontinuerlig-prosess-medium under dannelse av en
emulsjon som inneholder prosessmediet og mikro-
dråper omfattende midlet, løsningsmidlet og det
veggdannende materiale, og
c) emulsjonen tilsettes umiddelbart etter at den er dannet, hurtig til en effektiv mengde av et
ekstraksjonsmedium for å ekstrahere løsningsmidlet fra mikrodråpene slik at det mikroinnkapslede produkt dannes, idet løsemidlet har en løselighet i ekstraksjonsmediet fra én del pr. 100 til 25 deler pr. 100.
De spesielle trekk ved denne teknikk som viser at den er unik, er beskrevet nedenfor.
Ett av de viktige trekk ved denne mikroinnkapslings-teknikk er hastigheten av polymerløsningsmiddel-fjerning fra mikrodråpene i emulsjonen. Ved tilsetting av all emulsjonen til bearbeidelsesmediet på én gang og derved meget hurtig fjerning av mesteparten av polymerløsningsmidlet (innen 3 minutter), kan midler som er meget godt løselige i bearbeidelsesmediet, innkapsles, så vel som mindre løselige midler.
Den litteratur som finnes om mikroinnkapsling av vann-løselige midler, lærer at vannløselige midler, særlig hvis høye innhold så som mer enn 10 vekt%, og spesielt mer enn 3 0 vekt% er ønskelig, ikke lett kan innkapsles ved olje-i-vann-emulsjons-baserte fremgangsmåter på grunn av midlets tendens til å vandre ut av de organiske mikrodråpene i det vandige bearbeidelsesmedium. Denne vandring av middel er større med små emulsjons-mikrodråper på grunn av deres økte overflate-areal. Fordelen ved denne oppfinnelse i forhold til andre emulsjonsbaserte fremgangsmåter er at meget godt vannløselige midler så som midler med vannløselighet så høy som 2 gram pr. milliliter, kan innkapsles effektivt ved innhold på opp til 8 0 vekt%. Videre er de resulterende mikrokuler eller mikrokapsler frittstrømmende pulvere av sfæriske partikler. Avhengig av bearbeidelsesbetingelsene kan disse partikler ha diameter i området fra under ca. 1/xm til over 2 millimeter.
For fremstilling av mikrokapsler eller mikrokuler ved denne oppfinnelse blir først et egnet veggdannende materiale så som en polymer, oppløst eller på annen måte dispergert i et løsningsmiddel. Betegnelsen "veggdannende materiale" be-tegner også unike membraner og eksipienser. Løsningsmidlet som anvendes for oppløsing av veggmaterialet eller eksipiensen, kan velges fra forskjellige vanlige organiske løs-ningsmidler innbefattende halogenerte alifatiske hydrokarboner så som metylenklorid, kloroform og liknende; alko-holer; aromatiske hydrokarboner så som toluen og liknende; halogenerte aromatiske hydrokarboner; etere så som metyl-t-butyleter og liknende; cykliske etere så som tetrahydrofuran og liknende; etylacetat; dietylkarbonat,- aceton; cykloheksan; og vann. Disse løsningsmidler kan anvendes alene eller i kombinasjon. Det valgte løsningsmiddel må være et materiale som vil løse opp veggmaterialet eller eksipiensen, og det er best at det er kjemisk inert overfor det middel som innkapsles, og polymeren. Videre må løsningsmidlet ha begrenset løselighet i ekstraksjonsmediet. Vanligvis betyr begrenset løselighet en løselighet på fra ca. 1 del pr. 100 til ca. 25 deler pr. 100.
Egnede veggdannende materialer innbefatter, men er ikke begrenset til: poly(diener) så som poly(butadien) og liknende; poly(alkener) så som polyetylen, polypropylen og liknende; poly(akrylforbindelser) så som poly(akrylsyre) og liknende; poly(metakrylforbindelser) så som poly(metylmet-akrylat), poly(hydroksyetylmetakrylat) og liknende; poly-(vinyletere); poly(vinylalkoholer); poly(vinylketoner); poly-(vinylhalogenider) så som poly(vinylklorid) og liknende; poly(vinylnitriler); poly(vinylestere) så som poly(vinyl-acetat) og liknende; poly(vinylpyridiner) så som poly(2-vinylpyridin), poly(5-metyl-2-vinylpyridin) og liknende; poly(styrener); poly(karbonater); poly(estere); poly(orto-estere); poly(esteramider); poly(anhydrider); poly(uretaner); poly(amider); cellulose-etere så som metylcellulose, hydroksyetylcellulose, hydroksypropylmetylcellulose og liknende; cellulose-estere så som celluloseacetat, celluloseacetat-ftalat, celluloseacetatbutyrat og liknende; poly-(sakkarider), proteiner, gelatin, stivelse, gummiarter, harpikser og liknende. Disse materialer kan anvendes alene, som fysiske blandinger eller som kopolymerer. En foretrukket gruppe av veggdannende materialer innbefatter bionedbrytbare polymerer så som poly(laktid), poly(glykolid), poly(kaprolak-ton), poly(hydroksybutyrat), og kopolymerer derav, innbefattende, men ikke begrenset til, poly(laktid-ko-glykolid), poly(laktid-ko-kaprolakton) og liknende.
Det flytende eller faste middel som skal innkapsles, dispergeres eller oppløses så i løsningsmidlet som inneholder det oppløste veggdannende materiale eller eksipiens. Eksempler på biologiske midler som kan innkapsles ved denne teknikk, innbefatter, men er ikke begrenset til: smertestillende midler så som acetaminofen, acetylsalicylsyre og liknende; bedøvelsesmidler så som lidokain, xylokain og liknende; apetittnedsettende midler så som dexedrin, fendimetrazin-tartrat og liknende; antiartrittmidler så som metylpredni-solon, ibuprofen og liknende; antiastmatiske midler så som terbutalinsulfat, teofyllin, efedrin og liknende; antibiotika så som sulfisoxazol, penicillin G, ampicillin, cephalo- sporiner, amikacin, gentamicin, tetracykliner, kloramfenikol, erythromycin, clindamycin, isoniazid, rifampin og liknende; antisoppmidler så som amphotericin B, nystatin, ketoconazol og liknende; antivirusmidler så som acyclovir, amantadin og liknende; antikreftmidler så som cyklofosfamid, metotrexat, etretinat og liknende; antikoagulasjonsmidler så som heparin, warfarin og liknende; antikrampemidler så som phenytoin-natrium, diazepam og liknende; antidepressive midler så som isokarboxazid, amoxapin og liknende; antihistaminer så som difenhydramin-HCl, klorfeniramin-maleat og liknende; hormoner så som insulin, progestiner, østrogener, kortikoider, glyko-kortikoider, androgener og liknende; beroligende midler så som thorazin, diazepam, klorpromazin-HCl, reserpin, klor-diazepoxid-HCl og liknende; krampeløsende midler så som belladonna-alkaloider, dicyklomin-hydroklorid og liknende; vitaminer og mineraler så som essensielle aminosyrer, kalsium, jern, kalium, sink, vitamin B12og liknende; kardio-vaskulære midler så som prazosin-HCl, nitroglycerin, propranolol-HCl, hydralazin-HCl, verapamil-HCl og liknende; enzymer så som laktase, pankrelipase, ravsyredehydrogenase og liknende; peptider og proteiner så som LHRH, somatostatin, kalsitonin, veksthormon, veksthormon-frigjørende faktor, angiotensin, FSH, EGF, vasopressin, ACTH, humant serumalbumin, gammaglobulin og liknende; prostaglandiner; nuklein-syrer; karbohydrater; fettarter; narkotiske stoffer så som morfin, kodein og liknende; psykoterapeutika; antimalarie-midler; L-dopa; diuretika så som furosemid, spironolakton og liknende; antisår-legemidler så som ranitidin-HCl, cimetidin-HCl og liknende.
Immunologiske midler som kan innkapsles ved denne fremgangsmåte, innbefatter: interleukiner, interferon, koloni-stimulerende faktor, tumornekrosefaktor og liknende; aller-gener så som katte-"flass", bjerkepollen, husstøvmidd, gress-pollen og liknende; antigener av slike bakterieorganismer som Streptococcus pneumoniae, Haemophilus influenzae, Staphylococcus aureus, Streptococcus pyrogenes, Corynebacterium diphtheriae, Listeria monocytogenes, Bacillus anthracis, Clostridium tetani, Clostridium botulinum, Clostridium perfringens, Neisseria meningitidis, Neisseria gonorrhoeae, Streptococcus mutans, Pseudomonas aeruginosa, Salmonella typhi, Haemophilus parainfluenzae, Bordetella pertussis, Francisella tularensis, Yersinia pestis, Vibrio cholerae, Legionella pneumophila, Mycobacterium tuberculosis, Mycobacterium leprae, Treponema pallidum, Leptospirosis interrogans, Borrelia burgdorferi, Campylobacter jejuni og liknende; antigener av slike virus som koppevirus, influensa A og B, respiratorisk syncytialvirus, parainfluensavirus, meslingvirus, HIV, varicella-zoster-virus, herpes simplex 1 og 2, cytomegalovirus, Epstein-Barr, rotavirus, rhinovirus, adenovirus, papillomvirus, poliovirus, kusmavirus, rabies-virus, rubellavirus, coxsackievirus, heste-hjernebetennelse, japansk hjernebetennelse, gulfeber, enzootisk hepatitt, lymfocyttisk choriomeningitt, hepatitt B og liknende; antigener av slike sopp-, protozo- og parasittorganismer som Cryptococcus neoformans, Histoplasma capsulatum, Candida albicans, Candida tropicalis, Nocardia asteroides, Rickettsia ricketsii, Rickettsia typhi, Mycoplasma pneumoniae, Chlamydial psittaci, Chlamydial trachomatis, Plasmodium falciparum, Trypanosoma brucei, Entamoeba histolytica, Toxoplasma gondii, Trichomonas vaginalis, Schistosoma mansoni og liknende. Disse antigener kan være i form av hele drepte organismer, peptider, proteiner, glykoproteiner, karbohydrater eller kombinasjoner av disse.
Eksempler på ikke-biologiske midler som kan innkapsles ved denne metode, innbefatter, men er ikke begrenset til, klebemidler, pesticider, duftmidler, antigromidler, farge-stoffer, salter, oljer, sverte, kosmetika, katalysatorer, vaskemidler, herdemidler, aromastoffer, matvarer, brenn-stoffer, herbicider, metaller, malinger, fotograferings-midler, biocider, pigmenter, plastifiseringsmidler, drivmidler, løsningsmidler, stabilisatorer, polymeradditiver og liknende.
Etter at midlet er tilsatt til veggmaterialet/eksipiens-løsningsmidlet, tilsettes midlet og veggmateriale/eksipiens-løsningsmiddelblandings-dispersjonen til et kontinuerlig prosessmedium under dannelse av mikrodråper. Dette prosess-medium er vanligvis vann, skjønt organiske løsningsmidler og oljer kan også anvendes når vann anvendes for oppløsing av veggmaterialet eller eksipiensen. Prosessmediet inneholder fortrinnsvis overflateaktive midler for at dannelse av en stabil emulsjon skal være mulig, og for forhindring av agglo-merering. Eksempler på kationiske, anioniske og ikke-ioniske forbindelser som kan anvendes som overflateaktive midler, innbefatter, men er ikke begrenset til,, poly(vinylalkohol) , karboksymetylcellulose, gelatin, poly(vinylpyrrolidon), Tween 80, Tween 20 og liknende. Konsentrasjonen av overflateaktivt middel i prosessmediet bør være tilstrekkelig til stabiliser-ing av emulsjonen. Konsentrasjonen av tilstedeværende overflateaktivt middel vil påvirke den endelige størrelse av mikrokapslene eller mikrokulene. Vanligvis vil konsentrasjonen av det overflateaktive middel i prosessmediet veggmateriale/eksipiens være fra 0,1 til ca. 20% avhengig av det anvendte overflateaktive middel, polymerløsningsmiddel og bearbeidelsesmedium.
Før tilsettingen av blandingen som inneholder det opp-løste veggmateriale/eksipiens, dets løsningsmiddel og midlet, mettes prosessmediet med det samme løsningsmiddel som anvendes for oppløsing av veggmaterialet/eksipiensen under forhindring av eventuell ekstraksjon av løsningsmiddel fra mikrodråpene under dannelse av emulsjonen. Prosessmediet blir så agitert mekanisk med anordninger så som homogenisa-torer, drivmidler eller liknende etter hvert som middel/veggmateriale/løsningsmiddelblandingen tilsettes til prosess-mediet. Under dette trinn i prosessen blir intet løsnings-middel fordampet eller fjernet fra mikrodråpene. Tempera-turen ved hvilken emulsjonen dannes, er ikke spesielt av-gjørende, bortsett fra at den må være innenfor et område som vil forhindre at løsningsmidlet koker eller at prosessmediet geléres eller fryser, eller at midlet eller veggmaterialet nedbrytes. Tiden som fordres for dannelse av en emulsjon, er meget kort. Vanligvis kan emulsjoner dannes innen fra 3 0 sekunder til 5 minutter, avhengig av det anvendte overflateaktive middel og metoden anvendt for agitering av prosess-mediet .
Så snart det dannes en emulsjon, overføres alt prosess-mediet som inneholder de organiske mikrodråper så hurtig som mulig til et ekstraksjonsmedium slik at mer enn 20-30% av løsningsmidlet umiddelbart fjernes fra mikrodråpene (d.v.s. innen 3 minutter). Normalt anvendes vann som ekstraksjonsmedium, men andre løsningsmidler eller oljer kan også anvendes. Dessuten kan salter tilsettes til ekstraksjonsmediet for justering av dets ionestyrke eller pH. Mengden anvendt ekstraksjonsmedium er noe avgjørende på den måte at det må være tilstede tilstrekkelig medium til at omtrent umiddelbar ekstraksjon av løsningsmidlet ut av mikrodråpene blir mulig. Følgelig vil ekstraksjonsmediets volum avhenge av det løs-ningsmiddel som anvendes for oppløsing av veggmaterialet, og dets løselighet i ekstraksjonsmediet. Generelt bør ekstraksjonsmediets volum være minst det volum som trenges for opp-løsing av alt løsningsmidlet ut fra mikrodråpene, fortrinnsvis et 10-dobbelt volum eller mer.
Etter ekstraksjon av alt, eller nesten alt, løsnings-middel fra mikrodråpene (vanligvis innen 15-30 minutter), oppsamles de størknede mikrokapsler eller mikrokuler ved sentrifugering, filtrering eller liknende. Én fordel ved denne prosess er at den kan være en diskontinuerlig eller kontinuerlig prosess.
Idet oppfinnelsen er blitt beskrevet generelt, vil det nå bli beskrevet noen bearbeidelsesparametere som påvirker strukturen og egenskapene hos sluttproduktet. Når faste forbindelser og i visse tilfeller væsker mikroinnkapsles, er vanligvis det resulterende oppnådde produkt mikrokuler. Når væsker innkapsles, flyter væsken vanligvis sammen inne i mikrodråpen, hvilket resulterer i et mikrokapselprodukt. Hvis væsken fjernes, for eksempel ved vakuumtørking, fra mikrokapselproduktet, kan det fås mikrobobler.
Én av fordelene ved den foreliggende oppfinnelse er at faste midler kan innkapsles slik at sluttproduktet omfatter mikrokapsler som viser nulte ordens eller nær nulte ordens frigjørings-kinetikk. Dette oppnås ved innkapsling av meget godt vannløselige midler. Særlig under dannelse av emulsjonen tiltrekker meget godt vannløselige midler vann inn i mikrodråpen, som flyter sammen og bevarer det veggdannende materiale fra å utfelles som en matriks i hele mikrodråpen. For oppnåelse av en mikrokapsel må selvfølgelig det faste middel som innkapsles, ha tilstrekkelig vannløselighet til at
vann trekkes inn i mikrodråpen. Hvis det aktive middel ikke har tilstrekkelig løselighet, kan ko-innkapsling av midlet med en meget godt vannløselig hjelpeforbindelse så som et sukker eller salt, resultere i dannelse av mikrokapsler. Eller hvis bare sukkeret eller saltet innkapsles og deretter fjernes fra mikrokapslene, kan det oppnås mikrobobler. •
På grunn av at vannløselige midler så som peptider og proteiner ikke diffunderer gjennom hydrofobt veggdannende materiale så som laktid/glykolid-kopolymerene, må det dannes porer i mikrokapsel- eller mikrokulemembranen for at disse midler skal kunne diffundere ut for regulert-frigjørings-anvendelser. Flere faktorer vil påvirke den oppnådde porøsi-tet. Mengden av middel som innkapsles, påvirker mikrokulers porøsitet. Selvfølgelig vil mikrokuler med større mengde middel (d.v.s. mer enn ca. 20 vekt%, og fortrinnsvis mellom 2 0 og 80 vekt%) være mer porøse enn mikrokuler som inneholder mindre mengder middel (d.v.s. under ca. 20 vekt%) på grunn av at flere områder med legemiddel finnes i hele mikrokulene. Forholdet mellom middel og veggdannende materiale som kan innarbeides i mikrokulene, kan være så lavt som 0,1% opp til så høyt som 80%. Selvfølgelig vil den mengde som kan oppnås av spesifikke midler i en viss grad avhenge av midlets fysiske egenskaper og den ønskede anvendelse for mikrokule-preparatet.
Løsningsmidlet som anvendes for oppløsing av det veggdannende materiale, vil også påvirke membranens porøsitet. Mikrokuler eller mikrokapsler fremstilt ut fra et løsnings-middel så som etylacetat vil være mer porøse enn mikrokuler eller mikrokapsler fremstilt ut fra kloroform. Dette skyldes den høyere løselighet av vann i etylacetat enn i kloroform. Mer spesifikt fjernes intet løsningsmiddel fra mikrodråpene under emulgeringstrinnet på grunn av at prosessmediet er mettet med løsningsmiddel. Vann kan imidlertid oppløses i løsningsmidlet i mikrodråpene under prosessens emulgerings-trinn. Ved utvelgelse av det hensiktsmessige løsningsmiddel eller ko-løsningsmidler, kan mengden av kontinuerlig-prosess-mediet som vil oppløses i mikrodråpene, reguleres, hvilket vil påvirke den endelige porøsitet av membranen og den indre struktur av mikrokulene eller mikrokapslene.
En annen faktor som vil påvirke membranens porøsitet, er begynnelseskonsentrasjonen av veggmateriale/eksipiens i løs-ningsmidlet. Høye konsentrasjoner av veggmateriale i løs-ningsmidlet resulterer i mindre porøse membraner enn lave konsentrasjoner av veggmateriale/eksipiens. Dessuten for-bedrer høye konsentrasjoner av veggmateriale/eksipiens i løsningsmidlet innkapslingseffektiviteten av vannløselige forbindelser fordi løsningens viskositet er høyere. Vanligvis vil konsentrasjonen av veggdannende materiale/eksipiens i løsningsmidlet være i området fra ca. 3 til ca. 4 0%, avhengig av de fysiske/kjemiske egenskaper hos veggmateriale/eksipiens så som molekylvekten for det veggdannende materiale og anvendt løsningsmiddel.
Idet oppfinnelsen er blitt beskrevet generelt, kan man få ytterligere forståelse ved referanse til noen spesifikke eksempler som er tilveiebrakt i det foreliggende kun for illustrasjonsformål og som ikke er påtenkt å være begrensende dersom ikke annet er spesifisert.
EKSEMPEL 1
Følgende fremgangsmåte ble anvendt for innkapsling av cholinklorid i polystyren under anvendelse av et vandig prosessmedium. Løseligheten av cholinklorid i vann er større enn 3 gram pr. milliliter.
Først ble det tillaget en polymerløsning ved oppløsing av 1,0 g polystyren (Type 6850, Dow Chemical Co., Midland, MI) i 9,0 g metylenklorid. Deretter ble 1,0 g cholinklorid oppløst i 250 fil avionisert vann. Polystyrenløsningen ble overført til et 100 x 20 mm testrør. Mens polystyrenløs-ningen ble blandet ved virvling, ble cholinkloridløsningen tilsatt dråpevis i testrøret. Cholinkloridet ble homogent dispergert i polystyrenløsningen ved homogenisering under anvendelse av en Brinkmann Polytron (Modell 10, PTA-10S-probe, hastighetsinnstilling 5, Brinkmann Instruments Co., Westbury, NY).
En100 ml harpiksbeholder ble utstyrt med en "truebore"-rører og en 3 8 mm TEFLON turbin-rører. Deretter ble 50 ml 4 vekt% vandig poly(vinylalkohol) (PVA) mettet med 0,8 g metylenklorid og overført til harpiksbeholderen. Poly styren/cholinklorid-dispersjonen ble helt direkte i PVA-bearbeidelsesmediet. Under denne overføring ble PVA-løs-ningen omrørt med ca. 550 rpm (omdreininger pr. minutt). Etter at den resulterende olje-i-vann-emulsjon var blitt omrørt i harpiksbeholderen i 1 minutt, ble alt innholdet i harpiksbeholderen overført på én gang til 3,5 1 avionisert vann i et 4 1 begerglass som ble omrørt med ca. 750 rpm med en 51 cm rører av rustfritt stål. De resulterende mikrokuler ble omrørt i det avioniserte vann i ca. 25 min., oppsamlet over en sikt av rustfritt stål med diameter 20 cm og en maskestørrelse på 45fim, skyllet med41 avionisert van og tørket i 4 8 timer ved omgivelsestemperatur i et vakuumkammer.
Det endelige mikrokuleprodukt besto av frittstrømmende sfæriske partikler med diameter ca. 45-250fim og som inneholdt ca. 40-45 vekt% cholinklorid.
EKSEMPEL 2
En 15 vekt% polymerløsning ble tillaget ved oppløsing av 0,75 g 50:50 poly(DL-laktid-ko-glykolid) (DL-PLG) i 4,25 g metylenklorid. Deretter ble 3 0 mg formalinisert stafylokokk-enterotoksin B (SEB) oppløst i 110 fil avionisert vann. Den organiske polymerløsning ble overført til et 16 x 100 mm testrør, og SEB-toksoidløsningen ble så innført dråpevis i polymerløsningen mens sistnevnte ble agitert med en virvel-blander. Denne blanding ble så homogenisert med en Polytron-homogenisator for å sikre at SEB-toksoidet var homogent dispergert i DL-PLG-løsningen.
I en atskilt beholder ble 300 ml 1,5 vekt% vandig karboksymetylcellulose som var blitt mettet med metylenklorid, ekvilibrert til 19,0 +/- 1,0°C. Standardhodet i en Silverson laboratorieblander, utstyrt med emulsjonssikten, ble anbrakt under overflaten av karboksymetylcellulose-løsningen, og blanderens omrøringshastighet ble justert til ca. 4200 rpm.
SEB-toksoid/DL-PLG-blandingen ble dispergert som mikrodråper i den vandige karboksymetylcellulose. Den resulterende olje-i-vann-emulsjon ble omrørt i ca. 3 minutter, hvoretter all emulsjonen ble overført på én gang til 3,5 1 avionisert vann i et glassbeger, som ble omrørt med ca. 500 rpm med en 51 mm rører av rustfritt stål. De resulterende mikrokuler ble omrørt i det rensede vann i ca. 2 0 min., oppsamlet over et 0,22fim filter og tørket i 48 timer i et vakuumkammer.
Det resulterende mikrokule-produkt besto av sfæriske partikler på ca. 1-10 fim omfattende 2,7 vekt% SEB-toksoid i poly(DL-laktid-ko-glykolid).
EKSEMPEL 3
Ca. 2,5 g poly(DL-laktid) (DL-PL) ble oppløst i en passende mengde metylenklorid for frembringelse av en 11,1 vekt% polymerløsning. Etter at polymeren var fullstendig oppløst, ble en for-bestemt mengde testosteronpropionat tilsatt og fikk oppløses. Denne polymer/legemiddel-løsning ble så helt i en 1 1 harpiksbeholder som inneholdt 400 g 5,0 vekt% PVA. PVA-løsningen ble omrørt med ca. 750 rpm med en 64 mm TEFLON-rører drevet av en Fisher Stedi-Speed-motor. PVA-løsningen ble også mettet med 7 ml metylenklorid før tilsettingen av polymer/legemiddel-løsningen. Den resulterende emulsjon fikk omrøres i 7 minutter, hvoretter alt harpiksbeholder-innhold ble overført på én gang til12,0 1 avionisert vann som ble omrørt. Mikrokulene ble omrørt i det avioniserte vann i ca. 3 0 minutter og ble deretter oppsamlet over 45fim og 212^m rustfrie ståltrådduk-sikter anordnet i serie. Mikrokulene ble skyllet med ytterligere avionisert vann og fikk lufttørkes.
En liknende sats av testosteronpropionat-mikrokuler ble laget med en 20,6 vekt% polymerløsning. In vitro-fri-gjøringshastighetene for disse to satser er vist nedenfor, og disse viser at konsentrasjonen av polymerløsningen kan anvendes til manipulering av mikrokulers frigjøringsegenskaper. Det vil si at høyere polymerkonsentrasjon ga mikrokuler med mer langsom frigjøring.

EKSEMPEL 4
En 0,5 g mengde av etretinat [(All-E)-9-(4-metoksy-2,3,6-trimetyl)fenyl-3,7-dimetyl-2,4,6,8-nonatetraensyre-etylester] og 0,33 g 50:50 DL-PLG ble oppløst i 12,4 g metylenklorid. (På grunn av etretinats lysfølsomhet, ble alle trinn i prosessen utført i mørke). Den organiske løsning ble dispergert som mikrodråper i 300 g 10 vekt% vandig poly(vinylalkohol). Emulsjonen ble oppnådd ved tilsetting av den organiske løsning til en løsning av vandig poly(vinyl)alkohol) som ble hurtig omrørt i en glassbeholder. En ekstra kraftig laboratorieblander av typen Silverson ble anvendt til omrøring av emulsjonen.
Etter at de organiske mikrodråpene var blitt omrørt i poly(vinylalkohol)løsningen i 5 minutter under dannelse av en stabil olje-i-vann-emulsjon, ble emulsjonen overført til 4 1 avionisert vann under omrøring. De resulterende mikrokuler ble omrørt i det avioniserte vann i 3 0 minutter, skilt fra poly(vinylalkoholen) ved sentrifugering og oppsamlet ved lyo-filisering.
Sluttproduktet besto av frittstrømmende partikler med diametrer fra 0,5 til 5^m inneholdende 4 0 vekt% etretinat i poly(DL-laktid-ko-glykolid).
EKSEMPEL 5
En 12 vekt% polymerløsning ble tillaget ved oppløsing av1,0 g 50:50 DL-PLG i 7,3 g metylenklorid. Deretter ble 0,4 g finpulverisert cefazolin-natrium dispergert i polymerløs-ningen. Cefazolin/polymer-blandingen ble dispergert som mikrodråper i 100 g 6 vekt% vandig poly(vinylalkohol) mettet med 2,4 g metylenklorid. Emulsjonen ble oppnådd ved til setting av cefazolin/polymer-blandingen til den vandige poly-(vinylalkohol)løsning under omrøring av PVA-løsningen med ca. 1000 rpm i en harpiksbeholder. En TEFLON-turbinrører drevet av en Fisher Stedi-Speed-motor ble anvendt for omrøring av emulsjonen. Mens emulsjonen ble omrørt, kom vann inn i mikrodråpene (ifølge observasjon under mikroskop) og fløt sammen. Etter at det var blitt dannet en stabil olje-i-vann-emulsjon, ble alt innholdet i harpiksbeholderen overført på én gang til 3,5 1 vann som ble omrørt med 600 rpm, under ekstraksjon av metylenkloridet fra mikrokapslene. Etter at ekstraksjonen var fullført, fikk mikrokapslene bunnfelles. Mikrokapslene ble oppsamlet over sikter og ble deretter vasket med minst 3 1 vann. Mikrokapslene ble anbrakt i vakuum ved omgivelsestemperatur for tørking i minst 24 timer.
Det resulterende mikrokapselprodukt besto av sfæriske partikler- med en sentral kjerne av cefazolin-natrium innkapslet i en ytre DL-PLG-membran.
EKSEMPEL 6
En 15 vekt% polymerløsning ble tillaget ved oppløsing av 3 g 50:50 DL-PLG i 17 g metylenklorid. Deretter ble 0,4 g LHRH dispergert i polymerløsningen mens sistnevnte ble agitert med en homogenisator av typen Polytron. LHRH/DL-PLG-blandingen ble dispergert som mikrodråper i 200 g 5 vekt% poly(vinylalkohol) (PVA) som på forhånd var blitt mettet ved tilsetting av 3,6 g metylenklorid til PVA-løsningen. Emulsjonen ble oppnådd ved tilsetting av LHRH/DL-PLG-blandingen til PVA-prosessmediet som ble omrørt med 1060 rpm og som befant seg i en harpiksbeholder. En TEFLON-turbinrører drevet av en Fisher Stedi-Speed-motor ble anvendt til om-røring av emulsjonen.
Etter at det var blitt dannet en stabil olje-i-vann-emulsjon, ble all emulsjonen overført på én gang til 7 1 avionisert vann som ble omrørt, under ekstrahering av metylenkloridet. De resulterende mikrokuler fikk størkne i vannbadet i 15 minutter, ble oppsamlet over 45- og 150 fin-sikter, vasket med ca. 2 1 avionisert vann under fjerning av eventuelt rest-PVA, og lufttørket i 4 8 timer.
Sluttproduktet besto av et frittstrømmende pulver med diametere i området fra 45 til 150fxm omfattende 8,2 vekt% LHRH innkapslet i DL-PLG.
EKSEMPEL 7
En 8 vekt% etylcelluloseløsning ble tillaget ved opp-løsing av 1 g Ethocel (høy kvalitet, standard-etoksyinnhold, 20 viskositet, Dow Chemical Co., Midland, MI) i 11,5 g metylenklorid. Deretter ble 0,5 g mannitol oppløst i 3 ml avionisert vann. Etylcelluloseløsningen ble overført til et 100 x 20 mm testrør. Mens etylcelluloseløsningen ble agitert med en vortex-blander, ble mannitol-løsningen tilsatt dråpevis i røret. En Brinkmann Polytron (modell 10, PTA-10S-probe, hastighetsinnstilling på 5, Brinkmann Instruments Co., Westbury, NY) ble deretter anvendt til homogenisering av løsningen.
En 473 ml krukke med vid åpning ble anvendt til opp-bevaring av 300 ml av en 5 vekt% vandig løsning av (PVA). Denne løsning ble mettet med 4,8 g metylenklorid. Gjennom hele prosessen ble PVA-løsningen holdt på 19°C. En laboratorieblander-emulgator av typen Silverson (modell L2R, utstyrt med en emulsjons-mellomsikt, Silverson Machines Limited, Waterside, Chesham, Buckinghamshire, England) ble anvendt til omrøring av PVA-løsningen med 4 00 0 rpm. Ved anvendelse av en trakt med 10 mm hull, ble etylcellulose/- mannitol-løsningen tilsatt til den omrørte PVA-løsning. Etter 4 minutter ble hele krukkens innhold overført på én gang i 3 1 avionisert vann som ble omrørt med ca. 750 rpm. Metylenkloridet ble ekstrahert i vannet sammen1med mannitolen under dannelse av mikrobobler. Mikroboblene ble omrørt i 1 time for å sikre at all mannitol og metylenklorid ble fjernet. Mikroboblene ble så oppsamlet.
Slutt-mikrobobleproduktet består av sfæriske partikler med diameter 1-10^m som var hule inni.
EKSEMPEL 8
En 11,9 vekt% polymerløsning ble tillaget ved oppløsing av 0,5 g 52:48 poly(DL-laktid-ko-glykolid) (DL-PLG) (iboende viskositet 0,73 dl/g, målt ved en polymerkonsentrasjon på0,5g/dl i heksafluorisopropanol ved 30°C under anvendelse av et viskosimeter av typen Cannon) i 3,7 g metylenklorid. Deretter ble 0,125 g av en blanding som omfattet 1 vektdel interleukin-2 konjugert til en polyol-polymer (PEG-IL-2) og 2 0 vektdeler humant serumalbumin, veiet i et 16 x 75 mm test-rør. DL-PLG-løsningen ble tilsatt i testrøret og blandingen ble homogenisert tre ganger i 3 0 sekunder, med 15 sekunders mellomrom mellom homogeniseringene. Homogeniseringen ble ut-ført med en Brinkman Polytron (modell 10, PTA-10S probe, hastighetsinnstilling på 6).
En 20 0 ml harpiksbeholder ble forsynt med en "truebore"-rører og en 3 8 mm TEFLON-turbinrører. Deretter ble 150 ml 6 vekt% vandig poly(vinylalkohol) mettet med 2,4 g metylenklorid og overført til harpiksbeholderen. Den homogeniserte organiske blanding ble dispergert som mikrodråper i poly-(vinylalkoholen). Dispersjonen ble oppnådd ved tilsetting av den organiske blanding under overflaten av poly(vinyl-alkohol) -løsningen. Under denne overføring ble poly(vinyl-alkoholen) omrørt med ca. 1000 rpm. Dispersjonen ble omrørt i harpiksbeholderen i 5 minutter, hvilket resulterte i dannelse av en stabil olje-i-vann-emulsjon.
Etter at det var blitt fremstilt en stabil olje-i-vann-emulsjon, ble innholdet i harpiksbeholderen hurtig overført til 10 1 avionisert vann i et 12 1 begerglass som ble omrørt med ca. 800 rpm med en 51 mm rører av rustfritt stål. De resulterende mikrokuler ble omrørt i det avioniserte vann i ca.15 minutter, oppsamlet over en 45fim rustfri stålsikt med diameter 2 0,3 cm, skyllet med 4 1 avionisert vann og tørket i 48 timer ved omgivelsestemperatur i et vakuumkammer. Sluttproduktet besto av frittstrømmende partikler med diametere fra 45 til 200 jxm omfattende 15,6 vekt% av PEG-IL-2/HSA-blandingen i poly(DL-laktid-ko-glykolidet).
Claims (23)
1. Emulsjonsbasert fremgangsmåte for mikroinnkapsling av et middel under dannelse av et mikroinnkapslet produkt,karakterisert vedat den omfatter følgende trinn: a) en effektiv mengde av midlet dispergeres eller løses opp i et løsningsmiddel som inneholder et oppløst veggdannende materiale, under dannelse av en dispersjon,- b) dispersjonen blandes med en effektiv mengde av et kontinuerlig-prosess-medium under dannelse av en emulsjon som inneholder prosessmediet og mikrodråper omfattende midlet, løsningsmidlet og det veggdannende materiale, og c) emulsjonen tilsettes umiddelbart etter at den er dannet, hurtig til en effektiv mengde av et ekstraksjonsmedium for å ekstrahere løsningsmidlet fra mikrodråpene slik at det mikroinnkapslede produkt dannes, idet løsemidlet har en løselighet i ekstraksjonsmediet fra én del pr. 100 til 25 deler pr. 100.
2. Fremgangsmåte ifølge krav 1,karakterisert vedat løsningsmidlet som anvendes ikke er blandbart med prosessmediet.
