Description
一方、セサミン及び/又はエピセサミンについては、本出願人によって自律神経調節作用を有することが示されている(特許文献6)。また、アルコールやタバコ中毒からの離脱の際の中毒症状の緩和する作用が示されている(特許文献7)。しかし、これらいずれの文献にも抗疲労作用については何ら示唆も開示もなされていなかった。

1. 次の一般式(I):
2. 前記ジオキサビシクロ〔3.3.0〕オクタン誘導体がセサミンである上記1に記載の抗疲労剤。
3. 疲労を伴う疾患を治療または予防するための、上記1または2に記載の抗疲労剤を含む医薬組成物。
4. 疲労を伴う疾患が、慢性疲労症候群である、上記3に記載の医薬組成物。
5. 上記1記載のジオキサビシクロ〔3.3.0〕オクタン誘導体の疲労の回復又は予防のための用途。
6. 前記ジオキサビシクロ〔3.3.0〕オクタン誘導体がセサミンである上記5に記載の用途。
7. 疲労を伴う疾患を治療または予防するための、上記5または6に記載の用途。
8. 疲労を伴う疾患が、慢性疲労症候群である、上記7に記載の用途。
本発明で使用するジオキサビシクロ〔3.3.0〕オクタン誘導体としては、セサミン、セサミノール、エピセサミン、エピセサミノール、セサモリン、2−(3,4−メチレンジオキシフェニル)−6−(3−メトキシ−4−ヒドロキシフェニル)−3,7−ジオキサビシクロ〔3.3.0〕オクタン、2,6−ビス−(3−メトキシ−4−ヒドロキシフェニル)−3,7−ジオキサビシクロ〔3.3.0〕オクタン、2−(3,4−メチレンジオキシフェニル)−6−(3−メトキシ−4−ヒドロキシフェノキシ)−3,7−ジオキサビシクロ〔3.3.0〕オクタン、2−(3−メトキシ−4−ヒドロキシフェニル)−6−(3,4−ジヒドロキシフェニル)−3,7−ジオキサビシクロ〔3.3.0〕オクタン、2,6−ビス(3,4−ジヒドロキシフェニル)−3,7−ジオキサビシクロ〔3.3.0〕オクタン、2−(3,4−メチレンジオキシフェニル)−6−(3,4−ジヒドロキシフェニル)−3,7−ジオキサビシクロ〔3.3.0〕オクタン等を挙げることができ、これらの立体異性体又はラセミ体を単独で、または混合して使用することができるが、中でも、セサミンを好適に用いることができる。
(抗疲労作用及び剤)
上記のセサミンなどのジオキサビシクロ〔3.3.0〕オクタン誘導体の摂取により、顕著な抗疲労作用が得られる。ここで、疲労とは、身体的あるいは精神的負荷を連続して与えたときにみられる一時的な身体的および精神的パフォーマンスの低下現象であり、パフォーマンスの低下は、身体的および精神的作業能力の質的あるいは量的な低下を意味する。また、本発明の「疲労」には、慢性疲労症候群や過労死をも包含するものとする。
実施例1.水浸断眠試験
被検サンプルとして、(セサミン/エピセサミン混合物、セサミン:エピセサミン=1:1(重量比))を用いた。
図1の結果から明らかなとおり、水浸飼育対照群の遊泳時間は、通常飼育対照群にくらべ有意に短縮した。一方、水浸飼育セサミン投与群は、遊泳時間の短縮が顕著に抑制された。水浸飼育対照群の遊泳時間を100%とした場合、セサミンを12.5mg/kg与えた群では遊泳時間は150%に、セサミンを50mg/kg与えた群では遊泳時間は300%に、セサミンを200mg/kg与えた群では遊泳時間は133%になり、遊泳時間の短縮が大きく抑制された。特に、セサミンを12.5〜50mg/kg与えた場合(より好ましくは50mg/kg与えた場合)に、遊泳時間の短縮が大きく抑制された。このことから、セサミンは、水浸断眠負荷による疲労を有意に軽減(疲れにくくする)し、疲労回復効果があることが明らかである。
実施例2.強制運動試験
被検サンプルとして、実施例1と同様のセサミンを用いた。

被検サンプルとして、実施例1と同様のセサミンを用いた。
実施例4.血中疲労物質の測定
生体の代謝物疲労をケトン体の変動を指標として検討した。マーカーへの影響を見るため、実施例1を若干改変して水浸断眠負荷動物を作成した。すなわち、被検動物としてBalb/c系雄性マウス(8週齢)を用い、平均体重が均等になるようにマウスを各群7−10匹からなる3群に分けた。そのうち、2群は水浸飼育群として、床敷(ペーパーチップ)のかわりに水温23℃の水道水を水深5mmになるように飼育ケージに入れて飼育することにより、マウスを水浸させた。3日間の水浸中に、被検サンプルとしてセサミンを1日1回、50mg/kg体重の用量を5ml/kg体重のオリーブ油に溶かして3日間強制経口投与した(水浸飼育セサミン投与群)。対照群にはオリーブ油(5ml/kg)をセサミン投与群と同じ頻度で強制経口投与した(水浸飼育対照群)。残りの1群は通常飼育群として、通常の床敷(ペーパーチップ)を敷いた飼育ケージで飼育し、オリーブ油(5ml/kg)をセサミン投与群と同じ頻度で強制経口投与した(通常飼育対照群)。
Claims (11)
- 前記抗疲労剤は、ジオキサビシクロ〔3.3.0〕オクタン誘導体を唯一の有効成分とする、請求項1に記載の使用。
- 前記抗疲労剤が疲労の回復または予防に用いられる、請求項1または2に記載の使用。
- 前記抗疲労剤は、液剤、錠剤、顆粒剤、散剤、カプセル剤、ドライシロップ剤、及び丸剤からなる群から選ばれる形態を有する、請求項1〜3のいずれか1項に記載の使用。
- 前記抗疲労剤は、その包装、容器または説明書に抗疲労効果の表示を有する、請求項1〜4のいずれか1項に記載の使用。
- 前記抗疲労剤は、ジオキサビシクロ〔3.3.0〕オクタン誘導体を唯一の有効成分とする、請求項6に記載の医薬組成物。
- 疲労を伴う疾患が、慢性疲労症候群である、請求項6または7に記載の医薬組成物。
- 前記抗疲労剤は、ジオキサビシクロ〔3.3.0〕オクタン誘導体を唯一の有効成分とする、請求項9に記載の方法。
- 前記抗疲労作用が疲労の回復または予防である、請求項9または10に記載の方法。
