Description
実施例1:3つの抗体可変ドメインをもつ二重特異性抗体の設計、原核生物発現および精製
分子の抗EpCAM部分においてVLおよびVH、ならびに分子のCD3部分においてただ1つの抗体可変ドメイン(VH)を有する抗EpCAM x 抗CD3二重特異性抗体をコードするDNAが、pET-20b(+)ベクター(Novagen)の多重クローニング部位(MCS)にクローニングされる。ヒスチジン(x6)タグを有する二重特異性抗体の発現は、IPTGで誘導される。ベクターの選択は、組換えタンパク質の周辺質への輸送を促進する。pBAD-gIII(Invitrogen)、pET-32シリーズ(+)ベクター(Novagen)のような他のクローニングベクターもまた用いられうる。pBAD-gIIIに基づいた発現について、アラビノースは、IPTGの代わりに組換え遺伝子発現を誘導するために用いられる。いずれの場合においても、二重特異性抗体をコードするDNAは、組換えタンパク質の周辺質への輸送を媒介するシグナルペプチド(例えば、PelB、OmpA)をコードする配列とインフレームでクローニングされる。
ヒトEpCAM抗原を有する細胞の死滅をもたらすヒト細胞傷害性T細胞の細胞傷害性潜在能力を補充しうる、SEQ ID NO:1に示された配列をもつ二重特異性抗体の能力は、以下のように細胞傷害性アッセイで測定された。
Claims (7)
- 単一ポリペプチド鎖上に3つの抗体可変ドメインを含む二重特異性抗体であって、
該二重特異性抗体の第一部分が、ヒト免疫エフェクター細胞上に位置するエフェクター抗原に特異的に結合することによりヒト免疫エフェクター細胞の活性を補充する能力があり、該第一部分が1つの抗体可変ドメインからなり、かつ、該二重特異性抗体の第二部分が、エフェクター抗原以外の標的抗原に特異的に結合する能力があり、該標的抗原が該ヒト免疫エフェクター細胞以外の標的細胞上に位置しており、かつ該第二部分が2つの抗体可変ドメインを含み、
該エフェクター抗原がヒトCD3抗原であり、該標的抗原がヒトEpCAM抗原であり、かつ、SEQ ID NO:1に示された配列を含む、二重特異性抗体。 - SEQ ID NO:1のアミノ酸配列を含むポリペプチドをコードする核酸分子。
- 増殖性疾患、腫瘍性疾患、炎症性疾患、免疫不全、自己免疫疾患、感染性疾患、ウイルス性疾患、アレルギー反応、寄生反応、移植片対宿主疾患、もしくは宿主対移植片疾患の予防、治療または改善のための薬学的組成物の調製のための、請求項1記載の二重特異性抗体の、または、請求項2記載の核酸分子の、使用。
- 請求項1記載の二重特異性抗体または請求項2記載の核酸分子を含む、増殖性疾患、腫瘍性疾患、炎症性疾患、免疫不全、自己免疫疾患、感染性疾患、ウイルス性疾患、アレルギー反応、寄生反応、移植片対宿主疾患、もしくは宿主対移植片疾患の予防、治療または改善のために用いられる薬学的組成物。
- 免疫エフェクター細胞へ活性化シグナルを供給する能力があるタンパク性化合物をさらに含む、請求項4記載の薬学的組成物。
- 免疫エフェクター細胞へ活性化シグナルを供給する能力があるタンパク性化合物が、同時に、または非同時に投与される、請求項4記載の薬学的組成物。
- 請求項1記載の二重特異性抗体を含む、増殖性疾患、腫瘍性疾患、炎症性疾患、免疫不全、自己免疫疾患、感染性疾患、ウイルス性疾患、アレルギー反応、寄生反応、移植片対宿主疾患、もしくは宿主対移植片疾患の予防、治療または改善のために用いられるキット。
