Abstract
Description
本発明はまた、そのような多重特異性抗原結合分子を含む、抗腫瘍免疫応答の誘導用組成物やがんの治療または予防用組成物にも関する。本発明はさらに、そのような多重特異性抗原結合分子を投与する工程を含む、がんの治療または予防方法にも関する。
抗体は、血漿中での安定性が高く副作用が少ないことから、医薬品として注目されている。抗体は、抗原に結合する作用、アゴニスト作用やアンタゴニスト作用だけでなく、ADCC(Antibody Dependent Cytotoxicity:抗体依存性細胞傷害活性)、ADCP(Antibody Dependent Cell phagocytosis:抗体依存性細胞貪食作用)、CDC(補体依存性細胞傷害活性)といったエフェクター細胞による細胞傷害活性(エフェクター機能とも言う)を誘導する。このような抗体の機能を利用して、がん、免疫疾患、慢性疾患、感染症等の医薬品が開発されている(Nat Rev Drug Discov. 2018 Mar;17(3):197-223.(非特許文献1))。
〔1〕抗原提示細胞上の第一の抗原に結合する第一の抗原結合ドメイン、および、標的細胞上の第二の抗原に結合する第二の抗原結合ドメインを含む、多重特異性抗原結合分子。

また一態様において、本発明の多重特異性抗原結合分子は、標的細胞に破砕操作を加えずそのまま抗原提示細胞に貪食させることで、標的細胞の細胞表面抗原のみならず、細胞内抗原も抗原提示細胞に提示されるので、結果として幅広いバリエーションの抗原を抗原提示細胞に提示することができる。
本明細書において、用語「抗原結合分子」は抗原結合ドメインを含む分子を表す最も広義な意味として使用されており、具体的には、それらが抗原に対する結合活性を示す限り、様々な分子型が含まれる。例えば、抗原結合ドメインがFcRn結合ドメインと結合した分子の例として、抗体が挙げられる。抗体には、単一のモノクローナル抗体(アゴニストおよびアンタゴニスト抗体を含む)、ヒト抗体、ヒト化抗体、キメラ抗体等が含まれ得る。また抗体の断片として使用される場合としては、抗原結合ドメインおよび抗原結合断片(例えば、Fab、F(ab’)2、scFvおよびFv)が好適に挙げられ得る。既存の安定なα/βバレルタンパク質構造等の立体構造がスキャフォールド(scaffold;土台)として用いられ、その一部分の構造のみが抗原結合ドメインの構築のためにライブラリ化されたスキャフォールド分子も、本発明の抗原結合分子に含まれ得る。
本明細書において、「抗原結合ドメイン」は目的とする抗原に結合するかぎりどのような構造のドメインも使用され得る。そのようなドメインの例として、例えば、抗体の重鎖および軽鎖の可変領域、生体内に存在する細胞膜タンパクであるAvimerに含まれる35アミノ酸程度のAドメインと呼ばれるモジュール(WO2004044011、WO2005040229)、細胞膜に発現する糖タンパク質であるフィブロネクチン(fibronectin)中のタンパク質に結合するドメインである10Fn3ドメインを含むAdnectin(WO2002032925)、ProteinAの58アミノ酸からなる3つのヘリックスの束(bundle)を構成するIgG結合ドメインをスキャフォールドとするAffibody(WO1995001937)、33アミノ酸残基を含むターンと2つの逆並行ヘリックスおよびループのサブユニットが繰り返し積み重なった構造を有するアンキリン反復(ankyrin repeat:AR)の分子表面に露出する領域であるDARPins(Designed Ankyrin Repeat proteins)(WO2002020565)、好中球ゲラチナーゼ結合リポカリン(neutrophil gelatinase-associated lipocalin(NGAL))等のリポカリン分子において高度に保存された8つの逆並行ストランドが中央方向にねじれたバレル構造の片側を支える4つのループ領域であるAnticalin等(WO2003029462)、ヤツメウナギ、ヌタウナギなど無顎類の獲得免疫システムとしてイムノグロブリンの構造を有さない可変性リンパ球受容体(variable lymphocyte receptor(VLR))のロイシン残基に富んだリピート(leucine-rich-repeat(LRR))モジュールが繰り返し積み重なった馬てい形の構造の内部の並行型シート構造のくぼんだ領域(WO2008016854)が好適に挙げられる。本発明の抗原結合ドメインの好適な例として、抗体の重鎖および軽鎖の可変領域を含む抗原結合ドメインが挙げられる。
本発明における「抗原結合分子」とは、本発明の「抗原結合ドメイン」を含む分子であれば特に限定されず、さらに、5アミノ酸程度以上の長さを有するペプチドやタンパク質が含まれていてもよい。生物由来のペプチドやタンパク質に限定されず、例えば、人工的に設計された配列からなるポリペプチドであってもよい。また、天然ポリペプチド、あるいは合成ポリペプチド、組換えポリペプチド等のいずれであってもよい。
本発明の多重特異性抗原結合分子中の「抗原提示細胞上の第一の抗原に結合する第一の抗原結合ドメイン」の例として、
(1)配列番号14に示されるCDR1配列、配列番号15に示されるCDR2配列、および配列番号16に示されるCDR3配列を含む重鎖可変領域、ならびに配列番号18に示されるCDR1配列、配列番号19に示されるCDR2配列、および配列番号20に示されるCDR3配列を含む軽鎖可変領域;
(2)配列番号17に示されるアミノ酸配列を有する重鎖可変領域および配列番号21に示されるアミノ酸配列を有する軽鎖可変領域;または
(3)配列番号1に示されるアミノ酸配列を有する重鎖および配列番号2に示されるアミノ酸配列を有する軽鎖

本発明の多重特異性抗原結合分子中の「標的細胞上の第二の抗原に結合する第二の抗原結合ドメイン」の例として、
(4)配列番号22に示されるCDR1配列、配列番号23に示されるCDR2配列、および配列番号24に示されるCDR3配列を含む重鎖可変領域、ならびに配列番号26に示されるCDR1配列、配列番号27に示されるCDR2配列、および配列番号28に示されるCDR3配列を含む軽鎖可変領域;

本明細書において、抗体とは、天然のものであるかまたは部分的もしくは完全合成により製造された免疫グロブリンをいう。抗体はそれが天然に存在する血漿や血清等の天然資源や抗体を産生するハイブリドーマ細胞の培養上清から単離され得るし、または遺伝子組換え等の手法を用いることによって部分的にもしくは完全に合成され得る。抗体の例としては免疫グロブリンのアイソタイプおよびそれらのアイソタイプのサブクラスが好適に挙げられる。ヒトの免疫グロブリンとして、IgG1、IgG2、IgG3、IgG4、IgA1、IgA2、IgD、IgE、IgMの9種類のクラス(アイソタイプ)が知られている。本発明の抗体には、これらのアイソタイプのうちIgG1、IgG2、IgG3、IgG4が含まれ得る。
「破砕されていない」細胞とは、細胞が細片(debris)になっていないことを指す。細胞が細片になる例として、がん細胞の細胞死が挙げられる。細胞死は、その形態学的特徴および生化学的特徴などからネクローシスとアポトーシスに大別される。アポトーシスを起こした細胞は縮小し、アポトーシス小体と呼ばれる細胞断片を形成した後、生体内で食細胞によって貪食除去される。一方で、ネクローシスは細胞が膨張し、最終的に細胞内容物が細胞外に漏出することで周辺細胞に炎症を惹起する。アポトーシスおよびネクローシスは、いずれもがん細胞が抗がん剤や放射線治療によって、がん細胞が消失するメカニズムとされている(YAKUGAKU ZASSHI 137(11) 1315―1321 (2017))。
細胞を破砕する一般的な方法としては、超音波処理、界面活性剤の使用、浸透圧ショック法、ホモジナイザーによる粉砕、ガラスビーズによる粉砕などが知られている(上記方法は、例えば下記ウエブサイトに例示されている:https://www.gelifesciences.co.jp/technologies/protein_preparation/lysis.html)。
好ましい一態様として、貪食させる標的細胞は破砕されていない、細胞として数をカウントすることができる細胞を、抗原提示細胞に貪食させることが望ましい。破砕されていない標的細胞をそのまま抗原提示細胞に貪食させることで、標的細胞が有する抗原は、その細胞表面抗原のみならず、その細胞内抗原に至るまで、幅広いバリエーションの抗原が抗原提示細胞に提示させることができる。
抗原提示細胞内に取り込まれた標的細胞に由来する複数種の抗原がプロセシングされ、生成された抗原ペプチドが抗原提示細胞により提示される。(epitope spreading)。これらの抗原ペプチドは、MHCクラスII分子との複合体として提示されてCD4+ T細胞を活性化する他、MHCクラスI分子との複合体として提示(クロスプレゼンテーション)されてCD8+ T細胞を活性化することができる。抗原提示細胞のMHC分子に提示された標的細胞由来の抗原ペプチドを検出/同定/測定する方法としては、任意のペプチド抗原-MHC複合体反応性TCRを有するT細胞を抗原提示細胞が活性化出来るかを評価する方法(例えば、本明細書の実施例の項に記載のOT-Iマウス(OVA反応性T細胞を持つ)を用いた系)、任意の抗原ペプチド-MHC複合体を特異的に認識する抗体を用いてFACSで検出/測定する方法、MHCに提示されている抗原ペプチドをMSで直接検出/測定する方法等がある。T細胞活性化の検出/測定方法としては、活性化した際に放出されるサイトカインをELISAやFACSで検出/測定する方法、活性化マーカーをFACSで検出/測定する方法、細胞数の増加を検出/測定する方法等がある。
こうして活性化されたT細胞は、抗原提示細胞により提示された抗原ペプチドに特異的なT細胞である。本発明の多重特異性抗原結合分子を用いることにより、標的細胞に由来する複数種の抗原ペプチドが抗原提示細胞において提示されることから、複数種のがん抗原ペプチドを標的とするT細胞の活性化が誘導され得る。当該複数種のがん抗原ペプチドには、標的細胞上の第二の抗原とは異なる抗原に由来するものも含まれる。
このような機能を担う抗原提示細胞の好適な例として、樹状細胞を挙げることができる。
クロスプレゼンテーションを担う樹状細胞サブセットの細胞表面に発現するCLEC9Aは、壊死細胞上に露出したF-アクチンに結合し、ITAM(Immunoreceptor tyrosine-based activation motif)を有する細胞内ドメインとSykとの相互作用を通して細胞内シグナル伝達経路を活性化し、樹状細胞によるクロスプレゼンテーションを促進することが知られている(Nat Rev Immunol. 2010 Jun;10(6):387-402)。クロスプレゼンテーションを担う樹状細胞サブセットにおいてはDEC205等の発現も知られているが、DEC205よりもCLEC9Aの方が高い特異性で当該サブセットにおいて発現することが報告されている(Blood,2012, vol.119: pp.2284-2292)。
本発明の多重特異性抗原結合分子と抗原提示細胞の活性化剤とのコンジュゲートは、様々な二官能性タンパク質連結剤を用いて作製され得る。例えばN-スクシンイミジル-3-(2-ピリジルジチオ)プロピオネート (SPDP)、スクシンイミジル-4-(N-マレイミドメチル)シクロヘキサン-1-カルボキシレート (SMCC)、イミノチオラン (IT)、イミドエステルの二官能性誘導体(例えば、アジプイミド酸ジメチルHCl)、活性エステル(例えば、スベリン酸ジスクシンイミジル)、アルデヒド(例えば、グルタルアルデヒド)、ビス-アジド化合物(例えば、ビス(p-アジドベンゾイル)ヘキサンジアミン)、ビス-ジアゾニウム誘導体(例えば、ビス-(p-ジアゾニウムベンゾイル)-エチレンジアミン)、ジイソシアネート(例えば、トルエン2,6-ジイソシアネート)、およびビス活性フッ素化合物(例えば、1,5-ジフルオロ-2,4-ジニトロベンゼン)である。リンカーは、細胞内での抗原提示細胞の活性化剤の放出を促進する「切断可能なリンカー」であり得る。例えば、酸不安定性リンカー、ペプチダーゼ感受性リンカー、光不安定性リンカー、ジメチルリンカー、またはジスルフィド含有リンカー(Chari et al., Cancer Res. 52:127-131 (1992)が使用され得る。
本発明のコンジュゲートは、市販(例えば、Thermo Fisher Scientific Inc.)されているBMPS、EMCS、GMBS、HBVS、LC-SMCC、MBS、MPBH、SBAP、SIA、SIAB、SMCC、SMPB、SMPH、スルホ-EMCS、スルホ-GMBS、スルホ-KMUS、スルホ-MBS、スルホ-SIAB、スルホ-SMCC、およびスルホ-SMPB、ならびにSVSB(スクシンイミジル-(4-ビニルスルホン)ベンゾエート)を含むがこれらに限定されない架橋試薬を用いて調製されるコンジュゲートを考慮するが、これらに限定されない。
抗原提示細胞の活性化剤としては、抗原提示細胞を直接活性化する薬剤(図2A、B)を用いることができる他、抗原提示細胞以外の免疫細胞を活性化する薬剤(図2C、D)を用いることもでき、後者を用いた場合、それによって活性化されたエフェクター細胞等の免疫細胞や傷害を受けた死細胞から放出される炎症性サイトカイン等によって抗原提示細胞を間接的に活性化することができる。これらの活性化剤の例としては、インターフェロンα(IFNα)、ポリI:C(poly-I:C)またはT細胞活性化剤等が挙げられる。
一態様として、これら活性化剤を添加することで、添加しない場合に比べて、破砕されていない標的細胞をそのまま効率よく抗原提示細胞に貪食させることができる。破砕されていない標的細胞が抗原提示細胞に提示されると、標的細胞が有する抗原はその細胞表面抗原のみならず、その細胞内抗原に至るまで、幅広いバリエーションとして抗原提示細胞に提示させることができる。その結果として、抗原提示細胞をより効果的に活性化させることができる。
また、樹状細胞を活性化させるアジュバントとして、死菌マイコバクテリアおよびLPS(Lipopolysaccharide)、水酸化アルミニウム、リン酸アルミニウム(分子細胞免疫学、第7版、エルゼビア・ジャパン、2014年)が、そして、IFNα、IFNγ、TNFα、IL-1β、IL-6、LL37-DNA complex(N Engl J Med 2009; 361:496-509)が知られている。また、TLR(Toll-like receptor)、NLR(nucleotide-binding oligomerization domain-like (NOD-like) receptor)、RLR(RIG-I-like receptor)、およびCタイプレクチン受容体のそれぞれに対するアゴニストも、樹状細胞の活性化剤となることが知られている(Front Immunol. 2014; 5: 255)
抗原提示細胞を直接活性化する薬剤を本発明の多重特異性抗原結合分子にコンジュゲートすることによって、本発明の多重特異性抗原結合分子が結合する抗原提示細胞を特異的に活性化させることができる(図2B)。当該薬剤は、例えば、本発明の多重特異性抗原結合分子が有するFc領域またはFab領域にコンジュゲートされてもよい。
本発明の多重特異性抗原結合分子が結合するがん特異的抗原を発現する細胞において特異的に発現される抗原に対する結合部位と、T細胞表面抗原(例えばCD3)に対する結合部位とを有する抗原結合分子をT細胞活性化剤として本発明の多重特異性抗原結合分子と併用した場合、本発明の多重特異性抗原結合分子が結合するがん細胞が、当該T細胞活性化剤によって当該がん細胞の近傍に動員されたT細胞による傷害作用を受けて炎症性サイトカイン等を放出し、本発明の多重特異性抗原結合分子が結合する抗原提示細胞を特異的に活性化させることができる(図2C)。そのようなT細胞活性化剤としては、例えばWO2012073985に記載されているようなT細胞とがん細胞を架橋することで当該がん細胞に対する細胞傷害活性を誘導する二重特異性抗体が挙げられる。
また、エフェクター細胞(細胞傷害性T細胞)を活性化する抗体フラグメントや薬剤をT細胞活性化剤として本発明の多重特異性抗原結合分子にコンジュゲートすることによって、本発明の多重特異性抗原結合分子が結合するがん細胞が、当該抗原結合分子によって当該がん細胞の近傍に動員されたエフェクター細胞による傷害作用を受けて炎症性サイトカイン等を放出し、本発明の多重特異性抗原結合分子が結合する抗原提示細胞を特異的に活性化させることができる(図2D)。当該T細胞活性化剤は、例えば、本発明の多重特異性抗原結合分子が有するFc領域またはFab領域にコンジュゲートされてもよい。
別の観点においては、本発明は、本発明の多重特異性抗原結合分子を有効成分として含む医薬組成物(薬学的製剤)を提供する。また、本発明は、当該抗原結合分子を有効成分として含有する、抗腫瘍免疫応答の誘導用組成物、細胞傷害性T細胞の活性化を誘導する医薬組成物(細胞傷害性T細胞活性化剤)、細胞傷害を誘導する医薬組成物(細胞傷害誘導治療剤)、細胞増殖抑制剤および抗がん剤に関する。本発明の医薬組成物は、がん治療剤またはがん予防剤(がんの治療用組成物またはがんの予防用組成物)として用いることもできる。本発明の抗腫瘍免疫応答の誘導用組成物、細胞傷害性T細胞活性化剤、細胞傷害誘導治療剤、細胞増殖抑制剤および抗がん剤は、がんを罹患している対象または再発する可能性がある対象に投与されることが好ましい。本発明の組成物との関連において、抗腫瘍免疫応答の誘導は、好ましくは細胞傷害性T細胞(Cytotoxic T Lymphocyte; CTL)の活性化であってもよい。
他の局面において、本発明は、本発明の多重特異性抗原結合分子を用いて、抗原提示細胞を標的細胞に標的化する工程、および当該標的細胞に由来する抗原ペプチドを当該抗原提示細胞のMHCクラスIタンパクに提示させる工程を含む、がんに由来する抗原ペプチドをスクリーニングする方法を提供する。また他の局面において、本発明は、本発明の多重特異性抗原結合分子を用いて、抗原提示細胞を標的細胞に標的化する工程、および当該抗原提示細胞のMHCクラスIタンパクに提示された当該標的細胞に由来する抗原ペプチドを検出する工程を含み、検出された抗原ペプチドを同定する工程を含んでいてもよい、標的細胞に由来する抗原ペプチドをスクリーニングする方法を提供する。当該スクリーニング方法によって得られたペプチドを使用することによって、当該ペプチドを含むMHC/ペプチド複合体に特異的な細胞傷害性T細胞を誘導することができる。
抗原提示細胞のMHCクラスIタンパクに提示された標的細胞に由来する抗原ペプチドを検出する方法としては、任意のペプチド抗原-MHC複合体反応性TCRを有するT細胞を抗原提示細胞が活性化出来るかを評価する方法(例えば、本明細書の実施例の項に記載のOT-Iマウス(OVA反応性T細胞を持つ)を用いた系)、任意の抗原ペプチド-MHC複合体を特異的に認識する抗体を用いてFACSで検出/測定する方法、MHCに提示されている抗原ペプチドをMSで直接検出/測定する方法等がある。T細胞活性化の検出/測定方法としては、活性化した際に放出されるサイトカインをELISAやFACSで検出/測定する方法、活性化マーカーをFACSで検出/測定する方法、細胞数の増加を検出/測定する方法等がある。
検出された抗原ペプチドを同定する方法としては、例えばヒト抗原提示細胞に提示された抗原ペプチドを同定する方法の一例として、以下の方法等が挙げられる。なお、以下の方法例に記載されている抗HLA-DRビーズの代わりに抗マウスMHC抗体ビーズを用いて同様の実験を行うことにより、マウス抗原提示細胞に提示された抗原ペプチドを同定することもできる。
(1)抗HLA-DRビーズの生成
1. 抗HLA-DR抗体G46-6(BD Biosciences,Cat.:555809)を、CNBr-activated Sepharose beads(GE Healthcare,Cat.:17-0430-01)に終濃度1mg/mLで固層化し、抗HLA-DR抗体固層化ビーズとする。
2. 抗HLA-DR抗体固層化ビーズについては0.02%アジ化ナトリウム(Wako,Cat.:190-14901)を含むPBS(Wako,Cat.:041-20211)中で保管する。
(2)HLA-DR-ペプチド複合体のナノスケール精製
1. Tris(SIGMA,Cat.:T1503-1KG)20mM, MgCl2 (MERCK,Cat.:1.05833.0250)5mMを加え、HCl(MERCK,Cat.:1.00316.1000)を用いてpH7.8に調製した超純水(Wako,Cat.:210-01303)溶液に、10% TritonX-100(Roche Diag,Cat.:11332481001)を1/10倍、protease inhibitor mix (11.6mg/mL PMSF(nakalai,Cat.:27327-94)、1.7mg/mL pepstatin A(SIGMA,Cat.:P4265-25MG)、1.7mg/mL chymostatin(Roche Diag,Cat.:11004638001)、0.8mg/mL leupeptin(SIGMA,Cat.:L9783-25MG)、及び133mg/mLアジ化ナトリウム(Wako,Cat.:190-14901)のmixture)を17/5000倍となるよう加え、Lysis Bufferを調製する。
2. 抗原提示細胞(例えば樹状細胞)のペレットにLysis Bufferを氷冷条件下で10倍量加え、Thermomixer Confort(Eppendorf)にて1100rpm、1時間、4℃で振とうし、ライセートを取得する。
3. 14000rpm、10分、4℃でスピンダウンし、ライセートを細胞片や細胞核と分離する。
4. 抗HLA-DR抗体固層化ビーズをライセート100μLに対して5-10μL加え、水平振とう機(horizontal shaker)で1100rpm、4℃、一晩振とうし、ライセート中のHLA-DR-ペプチド複合体を抗HLA-DR抗体固層化ビーズに結合させた。
5. 抗HLA-DR抗体固層化ビーズに結合したHLA-DR-ペプチド複合体を3000rpm、1分、4℃でスピンダウンした後、Lysis buffer 500μLで1回、さらに0.1% Zwittergent 3-12(Calbiochem,Cat.:693015)を含むPBS 500μLで2回洗浄する。
(3)HLA-DR-関連ペプチドの溶出
1. HLA-DR抗体固層化ビーズに結合したHLA-DR-ペプチド複合体を400μLの超純水に懸濁し、Ultrafree-MC filter(Durapore PVDF, 0.22um)(Millipore)に移し、14000rpm、10秒、4℃でスピンダウンする。
2. チューブ底に落とした超純水を除去し、400μLの超純水をフィルタ上に加え14000rpm、10-30秒、4℃でスピンダウンする洗浄操作を10回繰り返し実施する。
3. 0.1% trifluoracetic acid(Thermo Fisher Scientific,Cat.:28904)を含む超純水60uLを加え、37℃、30分インキュベートし、HLA-DR-ペプチド複合体よりペプチド混合物を溶出させた後、14000rpm、3分、18℃でスピンダウンし、vacuum centrifuge 5305C(Eppendorf)により溶出したペプチド混合物を乾燥する。
(4)イオントラップMS/MS質量分析によるペプチドの配列解析
1. 乾燥したペプチド混合物を2% acetonitrile(Wako,Cat.:018-19853)、0.5% acetic acid(MERCK,Cat.:1.00066.0250)、1% formic acid(MERCK,Cat.:1.11670.1000)を含む超純水15μLに再溶解し、そのうち5μLをMSに接続したnano-LC Ultimate 3000 RSLCnano system(Dionex)に注入する。LC分析条件としては、EP1715343A1に記載の条件、又はこれに類似する当業者に公知の条件で、逆相材料及びイオン交換材料を組み合わせたカラム、あるいは逆相材料単独のカラムを用い、適切な緩衝液を用いて行うことができる。HPLCカラムをナノ-LC エレクトロスプレーイオン化源を設置したOrbitrap Elite(Thermo)に連結し、製造元のプロトコルに従い、full scan精密質量分析及びMS-MSによる質量分析を実施する。
2. ペプチドの配列解析をSEQUESTアルゴリズムにより実施する。
(1)抗原提示細胞に標的細胞抗原を提示させるコンセプト
既存の抗原提示細胞を標的とした種々の治療法には以下のような課題がある。
1.体外で培養した抗原提示細胞を移入する治療法の場合、生体内で存在する抗原提示細胞と同等の機能を持つ細胞を体外で培養することが極めて困難である。また培養に設備や時間等のコストがかかる。
2.抗原ペプチドやmRNA等を投与する治療法の場合、免疫する抗原の解析・調製が必要となる。
この課題を解決するためには以下の条件を満たす事が重要であると考えた。
1.生体内に存在する自然な状態の抗原提示細胞を標的とする。
2.抗原提示細胞による標的細胞の貪食、抗原提示を促進する。
上記条件を満たす医薬組成物として抗原提示細胞と標的細胞を架橋させる分子を考案した。
図1では抗原提示細胞と標的細胞を架橋させる抗体の例を示している。抗原提示細胞上の抗原と標的細胞上の抗原を認識する二重特異性抗体を用いて抗原提示細胞と標的細胞を架橋し、抗原提示細胞による標的細胞の貪食を促進する。標的細胞を貪食した抗原提示細胞は標的細胞内抗原も含めた様々な抗原をMHCに提示し、標的細胞反応性免疫応答を惹起することが出来る。
例えば樹状細胞においては未成熟な樹状細胞よりも成熟な樹状細胞の方が抗原提示能は高いとされている(Front Immunol. 2013 Apr 3;4:82.)。抗原提示細胞の活性化方法は、抗原提示細胞を直接活性化する方法と、抗原提示細胞以外の免疫細胞を活性化し、その活性化細胞や傷害を受けた死細胞から放出される炎症性サイトカイン等によって間接的に活性化する方法が知られている。そこで、抗原提示細胞と標的細胞を架橋させる二重特異性抗体を、抗原提示細胞を活性化する因子と併用することで、(1)に示した抗原提示効率を高めることができると考えた(図2(A))。また、抗原提示細胞と標的細胞を架橋させる二重特異性抗体を、抗原提示細胞を直接活性化する分子とコンジュゲートした抗体(図2(B))によっても、抗原提示効率を高めることが可能だと考えた。さらに、例えばWO2012073985に記載されているようなT細胞とがん細胞を架橋することで細胞傷害活性を誘導する二重特異性抗体との併用(図2(C))、または細胞傷害性T細胞を活性化する抗体や薬剤のコンジュゲート(図2(D))によって、傷害を受けた死細胞から放出される炎症性サイトカイン等によって間接的に抗原提示細胞を活性化し、(1)に示した抗原提示効率を高めることが出来ると考えた。
CLEC9Aは、取り込んだ抗原をMHC-Iへ提示する機能を有するCross presenting DC特異的な抗原であり、GPC3はある種のがん細胞の細胞膜上に発現することが知られている(Trends Immunol. 2013 Aug; 34(8): 361-370.、European Journal of Cancer Volume 47, Issue 3, February 2011, Pages 333-338)。これらの抗原を用いて樹状細胞とがん細胞を架橋することで、樹状細胞によるがん細胞の貪食、および、がん細胞の保持する抗原の樹状細胞による提示を促進できることが考えられた。そこで本発明者らは、抗CLEC9A抗体と抗GPC3抗体から構成される二重特異性抗体を作製した。公知の抗CLEC9A抗体である10B4 (重鎖:10B4H-mF18mP4dGK (配列番号:1)、軽鎖:10B4L-mk1 (配列番号:2))、および、抗GPC3抗体であるGCH065(重鎖:GCH065-mF18mN4dGK (配列番号:3)、軽鎖:L0011-k0a (配列番号:4)) の発現ベクターを当業者公知の方法で調製し、Expi293 (Thermo Fisher Scientific ) 細胞を用いてこれらの抗体を発現した。また、ネガティブコントロール抗体(重鎖: NCHn(配列番号:5)、軽鎖:NCL(配列番号:6))を用いた。培養上清からの抗体の精製、および、二重特異性抗体の調製は当業者公知の方法にておこなった。以下、抗CLEC9A抗体と抗GPC3抗体の二重特異性抗体を10B4//GCH065、抗CLEC9A抗体とネガティブコントロール抗体の二重特異性抗体を10B4//NCと表記する。
実施例2で作製した抗体を用いて、樹状細胞とがん細胞を架橋させることにより、樹状細胞によるがん細胞の取り込みが促進されるかを評価した。標的とするがん細胞は、ヒトGPC3(hGPC3)とオボアルブミン(OVA)を発現するLL/2(LLC1)(CRL-1642、ATCC)トランスフェクタント細胞(以下LLC1/hGPC3/OVA)を使用した。また、評価に用いる樹状細胞は、C57BL/6NCrlマウス(雌、6〜8週齢、チャールスリバー)の骨髄から既報(Blood 2014 124:3081-3091)の方法で誘導したBMDC (Bone marrow dendritic cell)を使用した。誘導したBMDCにおいて抗原発現を表1のフローサイトメトリー用蛍光標識抗体を用いて当業者公知の方法で染色を行い、Fortessa (BD Biosciences) を用いてCLEC9A発現量を評価した結果を図3に示す。図3に示すように、CD103陽性樹状細胞はCLEC9A陽性、CD103陰性樹状細胞集団はCLEC9A陰性であった。
低吸着平底 96well plate (Cat:3474, Corning)に上記の操作を施した標的細胞を2.0×104 cells/well、各抗体を1 μg/well、BMDCを2.0×104 cells/wellとなるように加え、1時間反応させた。反応は5%炭酸ガス、37℃条件下で行われた。1時間の反応後、表2のフローサイトメトリー用蛍光標識抗体を用いて当業者公知の方法で染色を行い、Fortessa (BD Biosciences) を用いて測定した。CLEC9A陽性樹状細胞(CD103陽性樹状細胞)集団とCLEC9A陰性樹状細胞(CD103陰性樹状細胞)集団のそれぞれ何%が標識細胞を貪食したかを評価した。その結果を図4に示す。10B4//GCH065はネガティブコントロール抗体である10B4//NCと比較してCLEC9A陽性樹状細胞の標的細胞貪食割合を増加させた。一方でCLEC9A陰性樹状細胞の標的細胞割合には影響を与えなかった。
同様に、実施例2に記載の方法に従って調製したCD103陽性樹状細胞上の別抗原であるDEC205に結合する公知の抗体NLDC145 (重鎖:NLDC145VH-mF18mP4dGK (配列番号:7)、軽鎖:NLDC145VL-mk1 (配列番号:8))と標的細胞抗原GPC3を認識するGCH065(重鎖:GCH065-mF18mN4dGK (配列番号:3)、軽鎖:L0011-k0a (配列番号:4))の二重特異性抗体NLDC145//GCH065においても、ネガティブコントロール抗体(重鎖NCHp(配列番号:9)、軽鎖NCL((配列番号:6))と抗GPC3抗体GCH065(重鎖GCH065-mF18mN4dGK(配列番号:3)、軽鎖L0011-k0a(配列番号:4))の二重特異性抗体NC//GCH065と比較して貪食割合が促進した。試験条件は、標的細胞LLC1/hGPC3/OVA 5.0×104 cells/well、BMDC 2.0×104 cells/well、30分間の反応時間で実施した。その結果を図5に示す。
以上より、標的抗原と樹状細胞抗原を認識する二重特異性抗体によって、樹状細胞による標的細胞の貪食を誘導できることが示された。
実施例2で作製した二重特異性抗体を用いて細胞を貪食した抗原提示細胞が標的細胞の内在性抗原を提示して免疫細胞を活性化出来るかを評価した。免疫細胞としては、OVAペプチド(257-264)-MHC class I複合体を認識するCD8+T細胞を有するOT-Iマウス(Stock No: 003831、The Jackson Laboratory)の脾臓からCD8a+T Isolation kit (Cat: 130-104-075, Miltenyi Biotec)により分離したCD8+T細胞を使用した。
低吸着平底 96well plate (Cat:3474, Corning)に標的細胞であるLLC1/hGPC3/OVAを5.0x104 cells/well、各抗体を1 μg/well、CD8+T細胞を1.0×105 cells/well、BMDCを1.0x105 cells/well、poly I:C (Cat:P1530, SIGMA)またはIFNα(Cat:752806, Biolegend)をそれぞれ20 μg/wellまたは10000 U/well加え、48時間反応させた。反応は5%炭酸ガス、37℃条件下で行われた。反応終了後に上清を回収し、上清中のmouse IFNγ(mIFNγ)量をMouse IFN-gamma DuoSetELISA(R&D、Cat:DY485)を用いて測定した。図6(A)、図6(B)に示すように、10B4//GCH065抗体により、樹状細胞の標的細胞内抗原の提示能が促進されている事が確認された。これらの結果は、標的抗原と樹状細胞抗原を認識する二重特異性抗体によって、樹状細胞に標的細胞を貪食させることで、標的細胞内の抗原をpMHC-Iに提示させるcross presentationを誘導できることを示している。
皮下移植モデルマウスを用いて、実施例2に記載の方法に従って調製した抗CLEC9A抗体10B4(重鎖10B4H-mF18mP4dGK(配列番号:1)、軽鎖10B4L-mk1(配列番号:2))と抗GPC3抗体GCH065(重鎖GCH065-mF18mN4dGK(配列番号:3)、軽鎖L0011-k0a(配列番号:4))の二重特異性抗体10B4//GCH065、及びネガティブコントロール抗体(重鎖NCHp(配列番号:9)、軽鎖NCL(配列番号:6))と抗GPC3抗体GCH065(重鎖GCH065-mF18mN4dGK(配列番号:3)、軽鎖L0011-k0a(配列番号:4))の二重特異性抗体NC//GCH065を用いて、抗腫瘍効果を誘導出来得るかを評価した。まず、標的細胞であるLLC1/hGPC3/OVAを2×106 cellsずつC57BL/6NCrlマウス(雌、8週齢、チャールスリバー)に移植した。移植後7日目に腫瘍径と体重を測定し、結果を基にランダマイズを実施して2群(6匹/群)に分けた。その後、ランダマイズされた各群において下記のように移植後8日目と11日目に薬剤を投与した。
群1 NC//GCH065 5mg/kg+poly I:C 50μg
群2 10B4//GCH065 5mg/kg+poly I:C 50μg
各抗体は静脈投与、poly I:C(tlrl-pic、InvivoGen)は腫瘍内投与を行った。腫瘍径は週2回測定した。腫瘍径変化のグラフを図7に示す。図7に示すように標的抗原と樹状細胞抗原を認識する二重特異性抗体によって抗腫瘍効果が誘導されることが確認された。
実施例1(3)に示したように抗原提示細胞を活性化する事によって、抗原提示効率を高めることが出来ると考えられる。その方法としては抗原提示細胞を直接活性化する方法と、抗原提示細胞以外の免疫細胞を活性化し、その活性化細胞や傷害を受けた死細胞から放出される炎症性サイトカイン等によって間接的に活性化する方法がある。その例を図8、9に示した。
実施例3と同様の系を用いてhEpCAM陽性標的細胞貪食効率を評価した。標的細胞としては、ヒトEpCAM(hEpCAM)を発現するcolon 38 (MC38)((公財)がん研究会)トランスフェクタント細胞(以下MC38/hEpCAM)を使用した。なお、WO2010142990A1に記載されている公知の抗hEpCAM抗体の重鎖及び軽鎖可変領域を用いた。
まず標的細胞であるMC38/hEpCAMをPKH26 Red Fluorescent Cell Linker Kit for General Cell Membrane Labeling (PKH26GL, SIGMA)を用いて標識した。次に標識細胞を液体窒素で凍結した後に、37℃で融解する操作を2回行った。
低吸着平底 96well plate (Cat:3474, Corning)に上記の操作を施した標的細胞を5.0×104 cells/well、各抗体を1 μg/well、BMDCを5.0×104 cells/wellとなるように加え、200 μl/wellで1時間反応させた。反応は5%炭酸ガス、37℃条件下で行われた。1時間の反応後、表2のフローサイトメトリー用蛍光標識抗体を用いて当業者公知の方法で染色を行い、Fortessa (BD Biosciences) を用いて測定した。CLEC9A陽性樹状細胞(CD103陽性樹状細胞)集団とCLEC9A陰性樹状細胞(CD103陰性樹状細胞)集団のそれぞれ何%が標識細胞を貪食したかを評価した。その結果を図10に示す。実施例2に記載の方法で調製した抗CLEC9A抗体10B4(重鎖10B4H-mF18mP4dGK(配列番号:1)、軽鎖10B4L-mk1(配列番号:2))と抗hEpCAM結合抗体3171(重鎖3171H-mF18mN4dGK(配列番号:54)、軽鎖3171L-KT0(配列番号:55))の二重特異性抗体10B4//3171は、ネガティブコントロール抗体である10B4//NCと比較してCLEC9A陽性樹状細胞の標的細胞貪食割合を増加させた。一方でCLEC9A陰性樹状細胞の標的細胞割合には影響を与えなかった。
実施例2に記載の方法で作製した、DEC205に結合する公知の抗体NLDC145 (重鎖:NLDC145VH-mF18mP4dGK (配列番号:7)、軽鎖:NLDC145VL-mk1 (配列番号:8))と標的細胞抗原GPC3を認識するGCH065(重鎖:GCH065-mF18mN4dGK (配列番号:3)、軽鎖:L0011-k0a (配列番号:4))の二重特異性抗体NLDC145//GCH065を用いて、細胞を貪食した抗原提示細胞が標的細胞の内在性抗原を提示して免疫細胞を活性化出来るかを評価した。免疫細胞としては、実施例4と同様にOT-Iマウス(Stock No: 003831、The Jackson Laboratory)の脾臓からCD8a+T Isolation kit (Cat: 130-104-075, Miltenyi Biotec)により分離したCD8+T細胞を使用した。標的細胞としては、4回凍結融解を行ったLLC1/hGPC3/OVAを用いた。
低吸着平底 96well plate (Cat:3474, Corning)に標的細胞であるLLC1/hGPC3/OVAを5.0x104 cells/well、各抗体を1 μg/well、CD8+T細胞を1.0×105 cells/well、BMDCを1.0x105 cells/well、poly I:C (Cat:P1530, SIGMA)4μg/well加え、200 μl/wellで48時間反応させた。反応は5%炭酸ガス、37℃条件下で行われた。反応終了後に上清を回収し、上清中のmouse IFNγ(mIFNγ)量をMouse IFN-gamma DuoSetELISA(R&D、Cat:DY485)を用いて測定した。図11に示すように、NLDC145//GCH065はネガティブコントロール抗体であるNC//GCH065と比較してmIFNγの産生を促し、樹状細胞の標的細胞内抗原の提示を促進している事が確認された。従って、NLDC145//GCH065は樹状細胞による標的細胞の貪食を促進し、抗原提示を促進することが示された。
実施例2に記載の方法で調製した、CLEC9A抗体10B4(重鎖10B4H-mF18mP4dGK(配列番号:1)、軽鎖10B4L-mk1(配列番号:2))と抗hEpCAM結合抗体3171(重鎖3171H-mF18mN4dGK(配列番号:54)、軽鎖3171L-KT0(配列番号:55))の二重特異性抗体10B4//3171を用いて、細胞を貪食した抗原提示細胞が標的細胞の内在性抗原を提示して免疫細胞を活性化出来るかを評価した。免疫細胞としては、実施例4と同様にOT-Iマウス(Stock No: 003831、The Jackson Laboratory)の脾臓からCD8a+T Isolation kit (Cat: 130-104-075, Miltenyi Biotec)により分離したCD8+T細胞を使用した。標的細胞としては、MC38/hEpCAMにMyc-OVAプラスミドをFuGENE (Cat:E2321A、promega)を用いて導入し一過性にOVAを発現させた後に、4回凍結融解を行ったMC38/hEpCAM/OVAを用いた。
低吸着平底 96well plate (Cat:3474, Corning)に標的細胞であるMC38/hEpCAM/OVAを1.0x104 cells/well、各抗体を1 μg/well、CD8+T細胞を1.0×105 cells/well、BMDCを1.0x105 cells/well、poly I:C (Cat:P1530, SIGMA)を4 μg/well加え、200 μl/wellで48時間反応させた。反応は5%炭酸ガス、37℃条件下で行われた。反応終了後に上清を回収し、上清中のmouse IFNγ(mIFNγ)量をMouse IFN-gamma DuoSetELISA(R&D、Cat:DY485)を用いて測定した。図12に示すように、10B4//3171が樹状細胞の標的細胞内抗原の提示を促進している事が確認された。従って、10B4//3171は樹状細胞による標的細胞の貪食を促進し、抗原提示を促進することが示された。
Claims (15)
- 抗原提示細胞上の第一の抗原に結合する第一の抗原結合ドメイン、および、標的細胞上の第二の抗原に結合する第二の抗原結合ドメインを含む、多重特異性抗原結合分子。
- 前記標的細胞ががん細胞である、請求項1に記載の多重特異性抗原結合分子。
- 前記抗原提示細胞および前記標的細胞を前記多重特異性抗原結合分子を介して架橋することで前記抗原提示細胞による前記標的細胞の貪食作用(Phagocytosis)を誘導し得る、請求項1または2に記載の多重特異性抗原結合分子。
- 前記標的細胞が、破砕されていない標的細胞である、請求項3に記載の多重特異性抗原結合分子。
- 前記抗原提示細胞内に取り込まれた前記標的細胞に由来する複数種の抗原ペプチドが前記抗原提示細胞に提示される、請求項1〜4のいずれかに記載の多重特異性抗原結合分子。
- 前記抗原提示細胞のMHCクラスIタンパク質に提示される前記標的細胞に由来する抗原ペプチドが、前記標的細胞上の第二の抗原とは異なる抗原に由来するまたは前記標的細胞上の第二の抗原に由来する、請求項1〜5のいずれかに記載の多重特異性抗原結合分子。
- 前記抗原提示細胞上の第一の抗原が、樹状細胞表面上の抗原であり、樹状細胞クロスプレゼンテーションを誘導し得る抗原である、請求項1〜6のいずれかに記載の多重特異性抗原結合分子。
- 前記抗原提示細胞表面上の抗原が、Cタイプレクチン受容体またはインテグリン受容体である、請求項1〜7のいずれかに記載の多重特異性抗原結合分子。
- 前記抗原提示細胞上の第一の抗原が、CLEC9A、DEC205およびCD207からなる群から選択される抗原である、請求項1〜8のいずれかに記載の多重特異性抗原結合分子。
- 前記標的細胞上の第二の抗原が、がん抗原である、請求項1〜9のいずれかに記載の多重特異性抗原結合分子。
- 前記がん抗原が、GPC3、IL6R、およびEpCAMからなる群から選択される抗原である、請求項10に記載の多重特異性抗原結合分子。
- 前記抗原結合分子が抗体である、請求項1〜11のいずれかに記載の多重特異性抗原結合分子。
- 請求項1〜12のいずれかに記載の多重特異性抗原結合分子を含む、抗腫瘍免疫応答の誘導用組成物。
