一般に及び本発明の文脈において、MAGEタンパク質はMAGE A1のアミノ酸195〜279と、そのコア領域において約50%、同一であろう。
本発明の一実施形態において、本誘導体は異種のパートナーに連結したMAGEタンパク質ファミリーからの抗原を含む融合タンパク質である。そのタンパク質は化学的にコンジュゲートされ得るが、好ましくは非融合タンパク質と比べて増加したレベルが発現系で生産されるのを許容する組換え融合として発現される。これにより、融合パートナーは、Tヘルパーエピトープ(免疫学的融合パートナー)、好ましくはヒトにより認識されるTヘルパーエピトープを供するのを補助し、又はネイティブ組換えタンパク質より高収率でタンパク質を発現するのを補助し得る(発現エンハンサー)。好ましくは融合パートナーは、免疫学的融合パートナー及び発現増強パートナーの両方であろう。
別の実施形態において、免疫学的融合パートナーはLYTAとして知られるタンパク質である。好ましくはその分子のC末端部分が用いられる。LYTAは、ペプチドグリカン骨格内の特定の結合を特異的に分解するオートリシンである(lytA遺伝子によりコードされる(Gene, 43 (1986) ページ265 〜272 ))N−アセチル−L−アラニンアミダーゼ、アミダーゼLYTAを合成するストレプトコッカス・ニューモニエ(Streptococcus pneumoniae)から得られる。LYTAタンパク質のC末端ドメインはコリンに対する又はDEAEのような特定のコリンアナログに対するアフィニティーの原因である。この特性は融合タンパク質の発現のために役立つ大腸菌C−LYTA発現プラスミドの開発のために活用されている。アミノ末端にC−LYTAフラグメントを含むハイブリッドタンパク質の精製は記述されている(Biotechnolosy: 10, (1992) ページ795 〜798 )。本明細書に用いる場合、好ましい実施形態は残基178で始まるC末端中に見い出されるLyta分子の反復部分を利用する。特に好ましい形態は残基188〜305を組み込む。
臨床セッティングにおけるこのような構成物は、本発明者により黒色腫を治療することができることが示されている。1つの場合、段階IVの黒色腫はアジュバントを添加しないlipo P 1/3 MAGE 3 His タンパク質の2回の投与後に転移物を取り除いた。
本発明は、本発明のタンパク質をコードする核酸も供する。このような配列は好適な発現ベクターに挿入してDNA/RNAワクチン接種のために用い又は好適な宿主内で発現させることができる。本核酸を発現する微生物ベクターはワクチンとして用いることができる。このようなベクターには、例えばポックスウイルス、アデノウイルス、アルファウイルス、リステリア及びモナルファージ(monarphage)がある。
特に、その方法は、
i)本タンパク質又はその免疫原性誘導体をコードするヌクレオチド配列を含むDNAポリマーを宿主内で発現することができる複製可能又は組込み可能発現ベクターを調製し;
ii)該ベクターで宿主細胞を形質転換し;
iii) 該形質転換された宿主細胞を、前記DNAポリマーの発現が前記タンパク質を生産することができる条件下で培養し;そして
iv)該タンパク質を回収すること
を含み得る。
複製可能な発現ベクターは宿主細胞に適合したベクターを開裂して完全なレプリコンを有する直鎖DNAセグメントを供し、そしてその直鎖セグメントを、その直鎖セグメントと一緒に要求される産物をコードする1又は複数のDNA分子、例えば本発明のタンパク質をコードするDNAポリマー又はその誘導体と、連結条件下で組み合わせることにより本発明に従って調製することができる。
ベクターの選択は、部分的に宿主細胞により決定されよう。それは原核生物でも真核生物であってもよいが、好ましくは大腸菌又はCHO細胞である。適切なベクターには、プラスミド、バクテリオファージ、コスミド及び組換えウイルスがある。
組換え宿主細胞は、本発明に従って、形質転換条件下で宿主細胞を本発明の複製可能発現ベクターで形質転換することにより調製される。適切な形質転換条件は慣用的であり、例えば上述のManiatisら又は“DNA Cloning¨ Vol.II, D.M.Glover ed., IRL Press Ltd, 1985に記載される。
その産物は、宿主細胞に従って及び(細胞内又は培養培地にもしくは細胞内ペリプラズムに分泌された)発現産物の局在化に従って、慣用的な方法により回収される。これにより、宿主細胞が細菌、例えば大腸菌である場合、それは例えば物理的に、化学的に又は酵素的に溶解し、生じたライゼートからタンパク質産物を単離することができる。宿主細胞が哺乳動物細胞である場合、その産物は一般に、栄養培地から又は無細胞抽出液から単離することができる。慣用的なタンパク質単離技術には、選択的沈殿、吸着クロマトグラフィー、及びモノクローナル抗体アフィニティーカラムを含むアフィニティークロマトグラフィーがある。
一般に、各々のヒトの投与量は、1〜1000μgのタンパク質、好ましくは30〜300μgを含むであろうと予想される。
本発明は、医薬として許容される賦形剤中に本発明のタンパク質を含む医薬組成物も供する。好ましいワクチン組成物は、少くともリポプロテインD−MAGE−3を含む。このようなワクチンは、任意に、1又は複数の他の腫瘍関連抗原を含み得る。例えばMAGE及びGAGEファミリーに属する他のメンバーがある。好適な他の腫瘍関連抗原には、MAGE−1,GAGE−1又はチロシナーゼタンパク質がある。
本発明のタンパク質は、好ましくは、本発明のワクチン調製物中にアジュバントが添加される。好適なアジュバントアルミニウム塩、例えば水酸化アルミニウムゲル(アルム)又はリン酸アルミニウムを含むが、カルシウム、鉄又は亜鉛の塩であってもよく、またアシル化チロシン又はアシル化糖、カチオンに又はアニオンに誘導化したポリサッカライド又はポリホスファゼンの不溶性懸濁液であってもよい。他の周知のアジュバントには、CpG含有オリゴヌクレオチドがある。そのオリゴヌクレオチドは、CpGジヌクレオチドがメチル化されていないことを特徴とする。このようなオリゴヌクレオチドは公知であり、例えばWO96/02555に記載される。
水中油エマルション中のQS21,3D−MPL及びトコフェロールに関する特に能力のあるアジュバントはWO95/17210に記載され、好ましい製剤である。
好ましくは、そのワクチンは、サポニン、より好ましくはQS21を更に含む。
本発明の一態様において、組換え生産されたMAGE−タンパク質を精製するための方法を供する。その方法は、そのタンパク質を、例えば強カオトロピック剤(例えば尿素、塩酸グアニジウム)に、又は両性イオン界面活性剤、例えば(Empigen BB−n−ドデシル−N,N−ジメチルグリシン)に可溶化し、そのタンパク質の分子内及び分子間ジスルフィド結合を還元し、生じたチオールをブロッキングして酸化的再カップリングを防止し、そしてそのタンパク質を1又は複数のクロマトグラフィーステップにかけることを含む。
誘導化遊離チオール残基を有するMAGEタンパク質は新規であり本発明の一態様を形成する。特に、カルボキシアミド化又はカルボキシメチル化誘導体は本発明の好ましい実施形態である。
実施例I:
融合タンパク質リポプロテインD−MAGE−3−His(LPD 1/3−MAGE−3−His又はLpD MAGE−3−His)を発現する組換え大腸菌株の調製
1.大腸菌発現系:
リポプロテインDの生産のため、プロテインDをコードするDNAを発現ベクターpMG81にクローン化した。このプラスミドはラムダファージDNAからのシグナルを利用して挿入された外来遺伝子の転写及び翻訳を駆動する。そのベクターは、ラムダPLプロモーターPL、オペレーターOL及びNプロテインを供した時に転写極性効果を緩和するための2つの利用部位(Nu+L及びNu+R)を含む(Gross ら、1985, Mol. & Cell.Biol. 5; 1015 )。PLプロモーターを含むベクターはプラスミドDNAを安定化するため大腸菌溶原性宿主に導入される。溶原性宿主株は、ゲノム内に挿入された複製欠損ラムダファージDNAを含む(Shatzmanら、1983; In Experimental Manipulation of Gene Expression. Inouya (ed) pp 1 〜14, Academic Press NY )。ラムダファージDNAはベクターのOLリプレッサーに結合し、RNAポリメラーゼのPLプロモーターへの結合を防ぎ、それにより挿入された遺伝子の転写を防ぐcIリプレッサータンパク質の合成を指示する。発現株AR58のcI遺伝子は、PLに指示された転写が温度変化により制御され得るように、即ち培養温度の増加がリプレッサーを不活性化して外来タンパク質の合成が始まるように、温度感受性変異を含む。この発現系は、外来タンパク質、特に細胞に対して毒性であり得るものの合成を制御する(Shimataka & Rosenberg, 1981, Nature 292: 128)。
LPD−MAGE−3−Hisタンパク質の生産のために用いたAR58溶原性大腸菌株は標準的なNIH大腸菌K12株N99の誘導体(F-Su-galK2, lacZ-thr- )である。それは、欠損した溶原性ラムダファージを含む(galE::TN10, 1Kil-cI857 DH1)のKil−表現型は宿主高分子合成の遮断を防ぐ。cI857変異は、温度感受性障害をcIリプレッサーに与える。DH1欠失は、ラムダファージの右オペロン及び宿主のbio,uvr3、及びchlA座を除去する。AR58株は、SA500誘導体(galE::TN10, 1Kil-cI857 DH1)で先に増殖させたPラムダファージストックでのN99の導入により作り出した。N99への欠損溶原の導入は、隣接galE遺伝子中にテトラサイクリン耐性をコードするTN10トランスポゾンの存在によりテトラサイクリンで選択した。N99及びSA500は、National Institute of HealthのDr.Martin Rosenberg's laboratoryから得た大腸菌K12株である。
原理は、MAGE−3のN末端に連結した融合パートナーとして脂質化プロテインDのN末端1/3及びC末端に位置したいくつかのヒスチジン残基の配列(Hisテール)を用いて融合タンパク質としてMAGE3を発現させることであった。
次に、アミノ末端システイン残基に結合した脂肪酸は膜アンカーとして機能する。
4.LPD−MAGE−3−His融合タンパク質の形成のためのクローニングストラテジー(ベクターpRIT14477):
(Ludwig InstituteからのDr Thierry Boon から頂いた)MAGE−3遺伝子(Gaugler B ら、1994)のためのコーディング配列を含むcDNAプラスミド、及び(図2に概説するように調製した)Lipo−D−1/3コーディング配列のN末端部分を含むベクターPRIT14586を用いた。そのクローニングストラテジーは以下のステップを含む(図3)。
c)プラスミドpRIT14647からのNcoI XbaIフラグメントの切り出し、及びベクターpRIT14586へのクローニング。
d)宿主株AR58の形質転換。
実施例II:
LPD1/3−MAGE−3−His抗原の調製:
1.細菌株の増殖及び誘導−LPD1/3−MAGE−3−Hisの発現:
プラスミドpRIT14477で形質転換したAR58の細胞を、各々イーストエキス(6.4g/L)及びカナマイシンスルフェート(50mg/L)を補給したLY12培地400mLを含む2リッターフラスコ内で増殖させた。振とうテーブル上で、30℃で8+/−1時間、インキュベートした後、顕微鏡検査のため各々のフラスコから少量のサンプルを除去した。2つのフラスコの内容物をプールして20リッター発酵槽のための接種物を供した。
42時間後に、発酵槽中の温度を39℃に迅速に増加させ、供給速度を、LPD−MAGE−3−Hisの細胞内発現が最大レベルに達する期間である更なる22〜23時間、誘導期の間、0.005mL/g DCW/分の一定値に維持した。
発酵の終りに、その培養物の光学密度は(48〜72gの DCW/Lの細胞濃度に相当する)80〜120であり、全液体量は約12リッターであった。その培養物を迅速に6〜10℃に冷却し、ECK32の細胞を、5000×gで4℃で30分の遠心により培養液から分離した。ECK32の濃縮細胞を迅速にプラスチックバッグに保存し、直ちに−80℃に凍結した。
ECK32の凍結した濃縮細胞を4℃に解凍した後、細胞破壊緩衝液に再度懸濁して(約36gの DCW/Lの細胞濃度に相当する)60の最終的な光学密度にした。
その細胞を高圧ホモジナイザー(1000bar )に2回、通すことにより破壊した。その破壊した細胞懸濁液を遠心し(×10,000g、4℃、30分)、そのペレット画分をTriton X100(1%w/v)+EDTA(1mM)で2回、洗い、次にリン酸緩衝塩類溶液(PBS)+Tween20(0.1%v/v)で洗い、そして最後にPBSで洗った。各々の洗浄段階の間に、懸濁液を×10,000gで30分、4℃で遠心し、その上清を捨てて、ペレット画分を保持した。
融合タンパク質LipoD−MAGE3のキャラクタリゼーション:
1.精製:
LPD−MAGE−3−Hisを以下に記載のステップの順番を用いて細胞ホモジネートから精製した:
a)細胞破壊物からの洗浄したペレット画分の可溶化。
c)粒子の除去及びエンドトキシンの削減のための反応混合物の精密ろ過。
d)ポリヒスチジンテールと亜鉛装填Chelating Sepharose との間のアフィニティー相互作用の利用によるLPD−MAGE−3−Hisの捕獲及び最初の精製。
その精製したLPD−MAGE3−Hisはいくつかの仕上げ段階にかけた:
f)Superdex75を用いるサイズ排除クロマトグラフィーによる緩衝液交換/尿素除去。
g)製造過程のろ過。
これらのステップの各々を以下により詳細に記載する。
1.1)細胞ホモジネートペレットの可溶化
(上述の)最終洗浄段階からのペレット画分を、塩酸グアニジン(6M)及びリン酸ナトリウム(0.1M、pH7.0)の溶液800mL中で4℃で一晩、再び可溶化した。
可溶化した材料(うすい黄色、濁った懸濁液)にアルゴンを流して残った酸素を一掃し、2−メルカプトエタノールの保存溶液(14M)を加えて4.3Mの最終濃度(溶液1mL当り0.44mLの2−メルカプトエタノールに相当)を供した。
1.3)精密ろ過
その混合物を、Minikros中空繊維カートリッジ(ret. No M22M-600-01N;面積5,600cm2 、0.2μm)を備えたAmicon Proflux M12接線フロー(tangenital-flow )ユニット内で精密ろ過した。その透過液を後のクロマトグラフィー精製のために保持した。
金属キレートクロマトグラフィーを、BPG 100/500カラム(Pharmacia Biotechnology Cat No.18-1103-01 )に充填したChelating Sepharose FF (Pharmacia Biotechnology Cat. No.17-0575-01)で行った。充填したベッドの寸法は、直径10cm;断面積79cm2 ;ベッドの高さ19cm;充填容量1, 500mLであった。空のカラムを水酸化ナトリウム(0.5M)で浄化して精製水で洗った。
金属キレートクロマトグラフィーの次の段階は、60mL/分の溶出流速で行った。カラムを洗い、最初に塩酸グアニジン(6M)及びリン酸ナトリウム(0.1M、pH7.0)を含む溶液で、次に尿素(6M)及びリン酸ナトリウム(0.1M、pH7.0)を含む溶液で、カラム溶出液がOD280nm で0の吸光度に達する(ベースライン)まで洗った。
1.5)アニオン交換クロマトグラフィー
アニオン交換クロマトグラフィーを続ける前に、半純粋なLPD−MAGE−3−Hisタンパク質画分のコンダクタンスを、尿素(6M)及びTris−HCl(20mM、pH8.0)を含む溶液で希釈することにより約4mS/cmに減少させた。
精製したLPD−MAGE−3−Hisを、尿素(6M)、Tris−HCl(20mM、pH8.0)及びNaCl(0.25M)を含む溶液でカラムから溶出した。
精製したLPD−MAGE−3−Hisからの尿素の除去及び緩衝液交換は、両方とも、サイズ排除クロマトグラフィーにより行った。これは、XK 50/100カラム(Pharmacia Biotechnology Cat.No.18-8753-01 )に充填したSuperdex 75 (Pharmacia Biotechnology Cat.No.17-1044-01)を用いて行った。充填したベッドの寸法は、直径5cm;断面積19.6cm2 ;ベッドの高さ90cm;充填容量1,800mLであった。
その精製したLPD−MAGE−3−His画分(脱塩ラン当り最大500mL)を20mL/分の溶出流速でカラムに充填した。その脱塩した精製LPD−MAGE−3−Hisを、Tween80(0.1%v/v)を含む3リッターのPBSでカラムから溶出した。
1.7)製造過程のろ過
サイズ排除クロマトグラフィーからのバルクLPD−MAGE−3−Hisを、層流フード中0.22μm膜(クラス10.000)を介してろ過した。そのろ過したバルクは−80℃に凍結し、脱塩ステップまで保存した。
最終的なバルクのオスモル濃度は400mOsM未満であるべきであるので、塩濃度を減少させるために更なる緩衝液交換が必要とされた。これは、BPG 100/950カラム(Pharmacia Biotechnology Cat.No.18-1103-03 )に充填したSephadex G25 (Pharmacia Biotechnology Cat.No.17-0033-02 )を用いて脱塩クロマトグラフィーにより行った。充填したベッドの寸法は直径10cm;断面積78.6cm2 ;ベッドの高さ85cm;充填容量6,500mLであった。
そのカラムを6リッターの水酸化ナトリウム(0.5M)で浄化し、次にリン酸ナトリウム(10mM、pH6.8)、NaCl(20mM)及びTween80(0.1%v/v)を含む10リッターの溶液で平衡化した。
最終的なバルクタンパク質を+4℃に解凍した後、容器に分取してラクトース賦形剤中に凍結乾燥した。
LPD−MAGE−3−His精製抗原を、12.5%アクリルアミドゲル上で還元条件下でSDS−PAGEにより分析した。
タンパク質充填量はクーマシーブルー染色について50μg及び硝酸銀染色について5μgであった。臨床用lot 96K19及びパイロットlot 96J22を分析した。60kDa の分子量に相当する1つの主要バンドを可視化した。約45kDa 及び35kDa の2つのマイナーな更なるバンドも見られた。
LPD−MAGE−3−Hisタンパク質のSDS−PAGE分析により表されるペプチドを、マウスモノクローナル抗体を用いてウェスタン・ブロットにより同定した。これらの抗体は、MAGE−3−Hisタンパク質(このタンパク質はLPD−MAGE−3−HisのLPD部分を含まない)の精製した調製物を用いて企業内で開発した。
実施例IV:
1.LPD−MAGE−3−Hisタンパク質を用いるワクチン調製物:
これらの実験に用いたワクチンは、アジュバントを加えた又は加えなかった、株AR58からの大腸菌で発現されたリポプロテインD1/3−MAGE−3−Hisをコードする組換えDNAから作り出す。アジュバントとして、その製剤は、油/水エマルション中、3de−O−アシル化モノホスホリル脂質A(3D−MPL)及びQS21の混合物を含む。アジュバントシステムSBAS2は、先にWO95/17210に記載されている。
油/水エマルションは、2種の油(トコフェロール及びスクアレン)から構成される有機相、及び乳化剤としてTween80を含むPBSの水性相から構成される。そのエマルションは、5%のスクアレン、5%のトコフェロール、0.4%のTween80を含み、180nmの平均粒径を有し、SB62として知られる(WO95/17210を参照のこと)。
2.エマルションSB62(2倍濃縮物)の調製:
Tween80をリン酸緩衝塩類溶液(PBS)に溶かしてPBS中2%溶液を供する。100mLの2倍濃縮エマルションを供するために、5gのDL−α−トコフェロール及び5mLのスクアレンをボルテキシングして完全に混合する。90mLのPBS/Tween溶液を加え、完全に混合する。次に生じたエマルションをシリンジに通して、M110Sマイクロフルーディクスマシンを用いることにより最終的にミクロな液体にする。得られた油滴は約180nmの大きさを有する。
アジュバントを油/水エマルション中で、MPL及びQS21の組合せとして調剤する。この調製物を0.7mlの容器に入れて、凍結乾燥した抗原(容器は30〜300μgの抗原を含む)と混合する。
抗原を含まないアジュバント対照は、タンパク質をPBSで置換することにより調製した。
4.ワクチン抗原:融合タンパク質リポプロテインD1/3−MAGE−3−His:
リポプロテインDはグラム陰性細菌ヘモフィルス・インフルエンゼの表面上に露出したリポタンパク質である。
1:マウス及びサルにおけるLPD−MAGE−3−Hisの免疫原性:
ヒトMAGE−3タンパク質の抗原性及び免疫原性をテストするため、候補ワクチンを、遺伝的バックグラウンド及びMHC対立遺伝子が異なる2つの異なるマウス株(C57BL/6及びBalb/c)に注入した。両方のマウス株について、潜在的なMHCクラスI及びMHCクラスIIペプチドモチーフは、LPD−MAGE−3−His融合タンパク質のMAGE部分のためのものであると理論的に予測された。
各々の株の5匹のマウスに、ヒトに用いた濃度の10分の1でSBAS2中に調剤した5μgのLPD−MAGE−3−His又はそれのないものを、足のひらに、2週間間隔で2回、注入した。
b)増殖アッセイ:
リンパ球を、最後の注入後2週間に、マウスからの脾臓又は膝窩リンパ節を破壊することにより調製した。2×105 細胞を96ウェルプレートに3回重複して入れ、細胞を試験管内で72時間、それ自体で又はラテックスマイクロビーズ上にコートした異なる濃度(1〜0.1μg/ml)のHis−Mage3で再刺激した。
c)結論
LPD−MAGE−3−Hisはマウスにおいて免疫原性であり、この免疫原性は、SBAS2アジュバント製剤の使用により増加させることができる。
a)免疫化プロトコル
Balb/c又はC57BL/6マウスを、PBS、又はSBAS2、又は5μGのLPD−MAGE−3−His、又は5μGのLPD−MAGE−3−His+SBAS2のいずれかで、2週間間隔で足のひら内注入により2回、免疫化した。
b)間接ELISA:
2回目の注入後2週に、個々の血清を採取し、間接ELISAにかけた。
2μG/mlの精製His MAGE−3をコートした抗原として用いた。PBS+1%新生ウシ血清中で、37℃で1時間の飽和の後、その血清を飽和緩衝液中で逐次的に希釈(1/1000で開始)し、4℃で一晩、又は37℃で90分、インキュベートした。PBS/Tween20、0.1%で洗った後、ビオチニル化ヤギ抗マウス全IgG(1/1000)又はヤギ抗マウスIgG1,IgG2a,IgG2b抗血清(1/5000)を二次抗体として用いた。37℃で90分のインキュベーションの後、ペルオキシダーゼに連結したストレプトアビジンを加え、TMB(テトラメチルベンジジンペルオキシド)を基質として用いた。10分後、その反応をH2 SO4 0.5Mを加えることによりブロックし、そのO.D.を測定した。
図9は、マウスの異なるグループ(N=5/グループ)間で、曲線の中点に達するために必要とされる平均希釈にある、血清の相対的平均中点タイターを比較する。
これらの結果は、テストした両方のマウス株においてLPD−MAGE−3−Hisのみの2回の注入の後に弱いAb応答がマウントされるが、LPD−MAGE−3−HisをSBAS2の存在下で注入した時にはより高い抗MAGE−3 Ab濃度が形成されることを示す。これにより、2週間隔でのLPD−MAGE−3−Hisの2回の注入のみで観察される高いAb応答を作り出すのに十分である。
アジュバントSBAS2中のLPD−MAGE−3−Hisをワクチン接種したマウスからでさえ、血清サンプルのいずれにおいてもIgAもIgMも検出されなかった。
異なるIgGサブクラス濃度の分析は、テストした全てのIgGサブクラス(IgG1,IgG2a,IgG2b)のレベルが、Ag又はアジュバントのみを注入したマウスにおいてのものよりアジュバントを加えたAgをワクチン接種したマウスにおいて高かったので、混合されたAb応答がマウスにおいて誘導されたことを示す。
3.アカゲザルにおけるリポプロテインD1/3 MAGE3−His+SBAS2アジュバントの免疫原性
5匹のアカゲザル(Macaca Mulatta)の3つのグループを選択した。RTS,S及びgp120を陽性対照として用いた。

左足:GP120/SB26T
グループ3 右足:LipoD1/3 Mage3 His/SBAS2
0日目に動物にワクチンを与え、28日目にブーストし、84日目にMAGE3及びプロテインD成分の両方に対する抗体応答を決定するために採血した。ワクチンを、右足の後部にボーラス注入(0.5ml)で筋内に投与した。
血清を除去して−20℃で凍結し、特異的ELISAによる抗体レベルの測定のために用いた。
ELISAによりMAGE3に対する抗体応答の速度を追跡するために、14日毎に少量の血液サンプルを採取した。結果は、LPD1/3 Mage3 His+SBAS2の1回の注入後に、Mage3特異的全Igタイターが低くなることを示し、同じサルでのLipoD1/3 Mage3+アジュバントの2回目及び3回目の注入後に、5の動物のうち3つで見られた。少い応答のものは、3回の注入後でさえ陰性であり続けた。II後28日又はIII 後に、抗体タイターは基底レベルに戻った。抗体のサブクラスはIgMでなく主要なIgGとして決定した。IgGへのスイッチは、Tヘルパー応答が誘発されていることを示唆する。プロテインD特異的抗体応答は、弱いか、Mage3抗体応答と正確に平行している。
1.LPD−MAGE1 His
同様に、LPD−MAGE−1−Hisを調製した。そのアミノ酸及びDNA配列を配列番号:3及び4に示す。得られたタンパク質は、LPD−MAGE−3−Hisタンパク質と同様に精製した。要約すると、細胞培養物をホモジナイズし、0.5% Empigen界面活性剤の存在下で4MグアニジンHCl及び0.5M β−メルカプトエタノールで処理した。その産物をろ過し、浸透物を0.6Mヨードアセトアミドで処理した。そのカルボキシアミド化画分をIMAC(亜鉛キレート−セファロースFF)クロマトグラフィーにかけた。そのカラムを平衡化し、4Mグアニジン、HCl及びリン酸ナトリウム(20mM、pH7.5)及び0.5% Empigenを含む溶液で洗い、次にそのカラムをリン酸ナトリウム(20mM、pH7.5)0.5% Empigen緩衝液中4M尿素を含む溶液で洗った。タンパク質を同じ緩衝液であるが、イミダゾールの濃度を増加させて(20mM,400mM及び500mM)溶出した。
発現プラスミドpRIT14426の作製及びNS1−MAGE−3 Hisを生産するための宿主株AR58の形質転換:
タンパク質デザイン:
大腸菌内で発現させるべき融合タンパク質NS1,MAGE−3−Hisのデザインを図12に示す。
上述のタンパク質デザインに対応するコーディング配列(配列番号:6)を大腸菌発現プラスミド中のλpLプロモーターの制御下においた。
NS1 −MAGE−3−His融合タンパク質の形成のためのクローニングストラテジー
出発材料は、MAGE−3遺伝子のためのコーディング配列を含む、Ludwig InstituteからのDr.Tierry Boonから受け取ったcDNAプラスミド、及びインフルエンザからのNS1 (非構造タンパク質)コーディング領域の81aaを含むベクターPMG81であった。
a)オリゴヌクレオチドセンス:5′gc gcc atg gat ctg gaa cag cgt agt cag cac tgc aag cct、及びオリゴヌクレオチドアンチセンス:5′gcg tct aga tta atg gtg atg gtg atg gtg atg acc gcc ctc ttc ccc ctc tct caa を用いて、プラスミドcDNA MAGE−3内に供される配列のPCR増幅。
b)先に増幅したフラグメントのインビトロゲンのTAクローニングベクターへのクローニング及び中間体ベクターpRIT14647の調製。
d)宿主株AR58の形質転換。
e)NS1−MAGE3−His融合タンパク質を発現するプラスミドpRIT14426(図14を参照)を含む大腸菌株形質転換体の選択及びキャラクタリゼーション。
細菌を、50μg/mlのカナマイシンを補給したLB培地で、30℃で増殖させた。その培養がOD=0.3(620nm)に達した時に、温度を42℃に上げることにより熱誘導を行った。
ウサギ/マウス免疫化のためのNS1−MAGE3−His(大腸菌)の精製。

ペレットの1/3を34mlの100mM PO4 −6M GuHCl pH7中4℃でO/Nに再び可溶化した。JA20ローターで15,000rpm で30分の遠心の後、ペレットを捨てて、上清をIMACにより更に精製した。
カラム容量:15mL(16mm×7.5cm)
パッキング緩衝液:0.1M PO4 −6M GuHCl pH7
サンプル緩衝液:同上
洗浄緩衝液:0.1M PO4 −6M GuHCl pH7
0.1M PO4 −6M尿素 pH7
溶出:6M尿素を補給した0.1M PO4 緩衝液 pH7中のイミダゾール勾配(0→250mM)。
a.濃縮:
IMAC溶出物の抗原陽性画分(160mL)をプールし、Filtron膜(Omega型カットオフ10,000)でAmicon撹拌セル内で5mLに濃縮した。この段階の純度はSDS−PAGEにより評価して約70%である。
2.4mLの濃縮サンプルを0.8mLの還元サンプル緩衝液中で煮沸し、10%アクリルアミドゲルに充填した。その抗原を、4% SDSを補給したTris−Glycine緩衝液 pH8.3中に溶出し、Ns1 −MAGE3 His陽性画分をプールした。
抗原をTCAで沈殿させ、JA20ローターで15,000rpm で20℃、遠心した後、上清を捨てた。そのペレットをPBS緩衝液 pH7.4に再び溶かした。
そのタンパク質は、凍結/解凍後にPBSに可溶性であり、37℃で3時間、保存した後にいずれの分解も示さず、そしてSDS(12.5% PAGE)により測定して約50,000ダルトンの見掛け分子量を有する。
融合タンパク質CLYTA−MAGE−1−Hisテールを発現する大腸菌株の調製
1.発現プラスミドpRIT14613の作製及び宿主株AR58の形質転換:
タンパク質デザイン:
大腸菌内で発現させる融合タンパク質Clyta−Mage−1−Hisのデザインを図15に示す。
上述のタンパク質デザインに相当するコーディング配列(配列番号:8を参照のこと)を大腸菌発現プラスミド中のλpLプロモーターの制御下においた。
クローニング:
出発材料は、ストレプトコッカス・ニューモニエからのLytAコーディング領域の117C末端コドンを含むベクターPCUZ1、及び我々が、Ludwig InstituteからDr.Thierry Boon から頂いたプラスミドからの先にサブクローニングしたMAGE−1遺伝子cDNAを有するベクターpRIT14518であった。
2.CLYTA−Mage−1−Hisコーディング配列モジュールの調製:
a)最初のステップは、CLYTA配列をNdeI−AflIII 制限部位に隣接させることを予定したPCR増幅であった。そのPCR増幅は、プラスミドPCUZ1テンプレート及びプライマーとしてオリゴヌクレオチドセンス:5′tta aac cac acc tta agg agg ata taa cat atg aaa ggg gga att gta cat tca gac 、及びオリゴヌクレオチドアンチセンス:5′GCC AGA CAT GTC CAA TTC TGG CCT GTC TGC CAG を用いて行った。これは、378ヌクレオチド長のCLYTA配列の増幅を導く。
d)プラスミドpRIT14613を含む大腸菌形質転換体(KAN耐性)の選択及びキャラクタリゼーション(図16を参照)。
1.組換えタンパク質CLYTA−MAGE−1−His(pRIT14613)のキャラクタリゼーション:
細菌を、30℃で50μg/mlカナマイシンを補給したLB培地で増殖させた。その培養物がOD=0.3(620nm)に達した時に、温度を38℃に上げることにより熱誘導を行った。
長いPLプロモーター及びCLYTA配列の一部を含むEcoRI−NCO1 制限フラグメントをプラスミドpRIT DVA6から調製し、プラスミドpRIT14613のEcoRI−NCO1 部位の間に挿入した。
融合タンパク質CLYTA−Mage−1−Hisをコードする組換えプラスミドpRIT14614(図17を参照)を用いて大腸菌AR120を形質転換した。Kan耐性候補株を選択し、キャラクタライズした。
組換えタンパク質のキャラクタリゼーション:
細菌を50mg/mlのカナマイシンを補給したLB培地で30℃で増殖させた。その培養物がOD=400(620nm)に達した時に、ナリジクス酸を60mg/mlの最終濃度まで加えた。
CLYTA−MAGE−3−HIS
A:腫瘍拒絶組換え抗原:C−lytA融合パートナーが可溶性タンパク質の発現を導く融合タンパク質CLYTA−Mage−3−Hisはアフィニティータグとして機能し、有用なT−ヘルパーを供する。
発現プラスミドpRIT14646の作製及び宿主株AR120の形質転換:
タンパク質デザイン:
大腸菌内で発現させる融合タンパク質Clyta−Mage−3−Hisのデザインを図18に示す。
上述のタンパク質デザインに相当するコーディング配列を大腸菌発現プラスミド内のλpLプロモーターの制御下においた。
クローニング:
出発材料は、Gene 43 (1986) p 265-272に記載されるストレプトコッカス・ニューモニエからのLytAコーディング領域の117C末端コドンを含むベクターPCUZ1、及び我々が、Ludwig InstituteからDr.Tierry Boonから頂いたプラスミドからのMAGE−3遺伝子cDNAを先にサブクローニングしたベクターpRIT14426であった。
1.CLYTA−MAGE−3−Hisコーディング配列モジュールの調製:
1.1.最初のステップはAflII及びAflIII 制限部位にCLYTAを隣接させることを予定したPCR増幅であった。PCR増幅は、テンプレートとしてプラスミドPCUZ1及びプライマーとしてオリゴヌクレオチドセンス:5′tta aac cac acc tta agg agg ata taa cat atg aaa ggg gga att gta cat tca gac 、及びオリゴヌクレオチドアンチセンス:5′ccc aca tgt cca gac tgc tgg cca att ctg gcc tgt ctg cca gtg を用いて行った。これは、427ヌクレオチド長CLYTA配列の増幅を導く。先に増幅したフラグメントをInvitrogenのTAクローニングベクターにクローン化して、中間体ベクターpRIT14661を得た。
短いpLプロモーター及びCLYTA−Mage−3−Hisコーディング配列を含むBglII−XbaI制限フラグメントをプラスミドpRIT14662から調製し、プラスミドTCM67(国際出願PCT/EP92/01827に記載される、アンピシリンに対する耐性、及び長いλpLプロモーターを含むrBR322誘導体)のBglII−XbaI部位の間に挿入した。プラスミドpRIT14607を得た。
3.プラスミドpRIT14646の調製:
最後に、pRIT14607 に類似するがカナマイシン選択を有するプラスミドを作製した(pRIT14646)。
細菌を50mg/mlのカナマイシンを補給したLB培地で30℃で増殖させた。その培養物がOD=400(600nm)に達した時に、ナリジクス酸を60g/mlの最終濃度まで加えた。
4時間の誘導の後、細胞を収集し、PBSに懸濁し、分解(分解CLS“ワン・ショット”型)により溶解した。遠心の後、ペレット上清及び全抽出物をSDS−PAGEにより分析した。タンパク質をクーマシーブルーで染色したゲルで可視化し、ここでその融合タンパク質は全大腸菌タンパク質の約1%を示した。その融合タンパク質は、ウサギ抗Mage−3ポリクローナル抗体を用いてウェスタンブロット分析により同定した。組換えタンパク質は約58kDの見掛けMWの単一バンドとして表れた。
組換えタンパク質CLYTA−Mage−3 Hisの精製:
組換え細菌AR120(pRIT14646)を30℃で、フェド・バッチ条件下で20リッター発酵槽中で増殖させた。組換えタンパク質の発現は、ナリジクス酸を60g/mlの最終濃度まで加えることにより誘導した。発酵の終りに細胞を収集し、フレンチプレス破砕機(20,000psi )に2回、通すことにより60 OD/600に溶解した。溶解した細胞を4℃で15,000gで20分、ペレット化した。組換えタンパク質を含む上清を、0.3M NaCl、20mM Tris HCl pH7.6(緩衝液A)で予め平衡化した交換DEAE Sepharose CL6B 樹脂(Pharmacia )に充填した。緩衝液Aでのカラム洗浄の後、融合タンパク質を緩衝液A中2%コリンにより溶出した。抗Mage−3抗体を用いてウェスタン・ブロット分析により表した陽性抗原画分をプールした。DEAEで溶出した抗原を0.5% Empigen BB(両性イオン界面活性剤)に及び0.5M NaClにし、次に0.5% Empigen BB、0.5M NaCl、50mMリン酸緩衝液 pH7.6(緩衝液B)で予め平衡化したIon Metal Affinityクロマトグラフィーカラムに充填した。
0.090〜0.250Mイミダゾール画分をプールし、10kDa Filtronオメガ膜で濃縮し、次にPBS緩衝液に対して透析した。
我々は、融合タンパク質LPD−MAGE3−Hisがマウスにおいて免疫原性であること、及びこの免疫原性(増殖性応答及び抗体応答)は、上述のアジュバントの使用により更に増加させることができることを証明した。精製は、ジスルフィド結合を形成するチオールを誘導化することにより増強することができる。
ヒトにおいてアジュバントを加えない製剤中のLPD−MAGE3−Hisで患者を臨床的に処置すると、黒色腫を除去した。

- (i) ヘモフィルス・インフルエンゼB(Heamophilus influenzae B)由来のプロテインD、又はプロテインDの最初の約1/3もしくはプロテインDのN末端109もしくは100〜110アミノ酸を含むそのフラグメント;
(ii) インフルエンザ由来のNS1プロテイン、又はNS1プロテインのN末端81アミノ酸を含むそのフラグメント;あるいは
(iii) ストレプトコッカス・ニューモニエ(Streptococcus pneumoniae)由来のLytA、又は(a)Lyt−AのC末端部分;(b)残基178で始まるC末端中に発見されるLytA分子の反復部分;及び/又は(c)LytAのアミノ酸残基188〜305を含むそのフラグメント、から選択される融合パートナーに連結したMAGE遺伝子のファミリーによりコードされる抗原を含む融合タンパク質。 - アフィニティータグを更に含む、請求項1に記載の融合タンパク質。

- 前記ワクチンが、アジュバント、及び/又は免疫刺激性サイトカインもしくはケモカインを更に含む、請求項15〜20のいずれか1項に記載のタンパク質。
- 前記タンパク質が、水中油又は油中水エマルションビヒクルに供される、請求項21に記載のタンパク質。
- 前記アジュバントが、3D−MPL,QS21又はCpGオリゴヌクレオチドを含む、請求項21又は22に記載のタンパク質。
- 1又は複数の他の抗原を更に含む、請求項21〜23のいずれか1項に記載のタンパク質。
- MAGEタンパク質の精製又は製造方法であって、該タンパク質のジスルフィド結合を還元すること、及び得られた遊離チオール基をブロッキング基でブロッキングすることを含み、そして1又は複数のクロマトグラフィーステップを更に含み、ここで該タンパク質は強カオトロピック剤又は両性イオン界面活性剤を使用して可溶化される、方法。
- 前記カオトロピック剤が、尿素又は塩酸グアニジウムである、請求項25に記載の方法。
- 前記両性イオン界面活性剤が、エンピゲンBB−n−ドデシル−N,N−ジメチルグリシンである、請求項25に記載の方法。
- 前記ブロッキング剤がアルキル化剤である、請求項25〜27のいずれか1項に記載の方法。
- 前記ブロッキング剤がα−ハロ酸及び/又はα−ハロアミドである、請求項28に記載の方法。
- 前記ブロッキング剤が、1又は複数の以下の群:ヨード酢酸;ヨードアセトアミド;N−エチルマレイミド;クロロアセチルホスフェート;O−メチルイソウレア;及びアクリロニトリルである、請求項28又は29に記載の方法。
- 前記タンパク質がアフィニティータグを含む、請求項25〜30のいずれか1項に記載の方法。
- 前記アフィニティータグが、CLytA又はポリヒスチジンテールである、請求項31に記載の方法。
- 前記アフィニティータグを含むタンパク質が、ブロッキングステップ後にアフィニティークロマトグラフィーにかけられる、請求項31又は32に記載の方法。
- 前記アフィニティークロマトグラフィーが、固定化金属イオンアフィニティークロマトグラフィー(IMAC)である、請求項33に記載の方法。
- 前記金属イオンが、亜鉛、ニッケル、鉄、マグネシウム又は銅から選択される、請求項34に記載の方法。
- 前記アフィニティークロマトグラフィーが、両性イオン界面活性剤エンピゲンBBを含有する緩衝液を使用して行われる、請求項33〜35のいずれか1項に記載の方法。
- 前記タンパク質が、MAGE抗原及び融合パートナーを含む融合タンパク質である、請求項25〜36のいずれか1項に記載の方法。
- 前記融合パートナーが、ヘモフィルス・インフルエンゼB(Heamophilus influenzae B)由来のプロテインDもしくはプロテインDの最初の1/3を含むそのフラグメント、インフルエンザ由来のNS1プロテインもしくはNS1のN末端81アミノ酸を含むそのフラグメント、又はストレプトコッカス・ニューモニエ(Streptococcus pneumoniae)由来のLytAもしくはLytAの残基188〜305を含むそのフラグメントである、請求項37に記載の方法。
- 前記融合パートナーがプロテインDの脂質化形態である、請求項38に記載の方法。
- 前記タンパク質が、LytAもしくは残基188〜305を含むそのフラグメントを含み、そしてここで該タンパク質がコリン又はコリンアナログ、例えばDEAEに対するアフィニティークロマトグラフィーにより精製される、請求項32〜39のいずれか1項に記載の方法。
- 前記MAGE抗原が、MAGE A1,MAGE A2,MAGE A3,MAGE A4,MAGE A5,MAGE A6,MAGE A7,MAGE A8,MAGE A9,MAGE A10,MAGE A11,MAGE A12,MAGE B1,MAGE B2,MAGE B3,MAGE B4,MAGE C1及びMAGE C2から選択される、請求項25〜40のいずれか1項に記載の方法。
- ワクチンの製造方法であって、請求項25〜41のいずれか1項に記載の方法によりMAGEタンパク質を産生又は精製する工程、及び得られたタンパク質をワクチンとして調剤する工程を含む方法。
