Abstract

"MéTODOS PARA DETERMINAR PELO MENOS UMA PRODUçãO DE SUBSTáNCIA QUE AFETA FLUXO METABóLICO USANDO CéLULAS, PARA CRIAR UMA CEPA BACTERIANA APRESENTANDO UMA CAPACIDADE DE PRODUZIR SUBSTáNCIA, E PARA PRODUZIR UMA SUBSTáNCIA, PROGRAMA PARA DETERMINAR PELO MENOS UMA PRODUçãO DE SUBSTáNCIA QUE AFETA FLUXO METABóLICO USANDO CéLULAS, E, MEIO DE REGISTRO QUE PODE SER LIDO POR COMPUTADOR". Uma produção de substância que afeta fluxo metabólico empregando células é determinada por meio de 1) criação de uma matriz estequiométrica com base em fórmulas de reações bioquímicas de um substrato por meio de uma substância produzida desejada, 2) seleção do mesmo número de fluxos metabólicos independentes de todos os fluxos metabólicos que o grau de liberdade da matriz estequiométrica como fluxos livres, 3) criação de um número suficiente de combinações randómicas dos fluxos livres para uma análise estatística e cálculo de uma distribuição de fluxo metabólico de cada combinação criada com base na matriz estequiométrica, 4) obtenção de uma equação de regressão incluindo um número mínimo de fluxos livres que mostra uma correlação com produção de substância das distribuições calculadas de fluxos metabólicos por meio de uma análise estatística multivariável, e 5) determinação de pelo menos uma produção de substância que afeta fluxo metabólico com base em um coeficiente na equação de regressão obtida.
