Description
Claims (1)
- 【特許請求の範囲】 1.少なくとも1種の化合物を含む培地上または中で培養された真核細胞集団 から、形質転換の結果として代謝優勢を有する細胞を同定または選択するにあた り、i)前記細胞が(a)化合物の代謝に含まれるタンパク質をコードするか、 および/または(b)当該タンパク質をコードする遺伝子の活性を制御する領域 を含むヌクレオチド配列または共挿入ヌクレオチド配列で形質転換されているこ と、およびii)前記化合物がマンノースもしくはキシロースまたはそれらの誘導 体または前駆体、または前記タンパク質の基質であるか、または形質転換細胞に よりそのような基質に代謝されることができること(ただし、前記化合物は、前 記タンパク質がマンノース−6−リン酸イソメラーゼである場合、マンノースで はない。)を特徴とする方法。 2.少なくとも1種の化合物を含む培地上または中で培養された真核細胞集団 から、形質転換の結果として代謝優勢を有する細胞を同定または選択するにあた り、i)前記細胞が(a)化合物の代謝に含まれるタンパク質をコードするか、 および/または(b)当該タンパク質をコードする遺伝子の活性を制御する領域 を含むヌクレオチド配列または共挿入ヌクレオチド配列で形質転換されているこ と、およびii)前記化合物の細胞に対する毒性を低下させる薬剤を培地に添加す ることを特徴とする方法。 3.化合物はマンノースであり、ヌクレオチド配列または共挿入ヌクレオチド 配列はマンノース−6−リン酸イソメラーゼをコードするものである、請求項2 記載の方法。 4.ヌクレオチド配列(共挿入ヌクレオチド配列を含む。)の少なくとも1つ が、当該配列が挿入された生物によって奸まれるコドンが使用されて、当該配列 のタンパク質コード成分が内因性である生物中において非修飾DNAの発現で得 られたのと実質的に類似しているタンパク質を産生するように修飾されたDNA を含むものである、請求項3記載の方法。 5.当該化合物が非形質転換細胞中で不活性であるか、または形質転換細胞中 よりも非形質転換細胞中で活性が低いか、または非形質転換細胞よりも形質転換 細胞に対して毒性が低いものであり、ヌクレオチド配列の発現または転写が化合 物の細胞への利用を増加させるか、または細胞中に内因的に存在する酵素の活性 を高めるものであり、かくして形質転換細胞中の酵素活性の方が非形質転換細胞 中の酵素活性よりも大となるものであるか、またはヌクレオチド配列の発現また は転写が、形質転換細胞中の当該化合物の代謝阻害または当該化合物の合成阻害 を導くものであり、それにより形質転換細胞が非形質転換細胞から同定できる、 請求項1〜4の何れかに記載の方法。 6.細胞が、プロモーターにより制御され得る構造遺伝子の近くにあるDNA 配列と同種組換が可能な外因性プロモーターをコードしているヌクレオチド配列 で形質転換されている、請求項1〜5のいずれかに記載の方法。 7.化合物がマンノースである、請求項1〜6のいずれかに記載の方法。 8.化合物がマンノース−6−リン酸、キシロースまたはD−マンノースアミ ンである、請求項1〜6のいずれかに記載の方法。 9.タンパク質が酵素である、請求項1〜8のいずれかに記載の方法。 10.酵素が、ホスホシュガー−イソメラーゼ、ホスホシュガー−ムターゼ、 ホスファターゼ、シュガーエピメラーゼ、シュガー−パーミアーゼおよびホスホ シュガー−パーミアーゼからなる群から選択されるものである、請求項9に記載 の方法。 11.タンパク質がマンノース−6−ホスフェートイソメラーゼである、請求 項1〜6および8〜10のいずれかに記載の方法、またはタンパク質がキシロー スイソメラーゼである、請求項1〜10の何れかに記載の方法。 12.ムターゼがホスホマンノムターゼおよびホスファターゼがマンノース− 6−ホスファターゼである、請求項10記載の方法。 13.細胞が化合物に感受性の細菌で形質転換される、請求項1〜12の何れ かに記載の方法。 14.形質転換細胞が陽性選択および陰性選択を組み合わせて選択されたもの であり、当該形質転換細胞内のヌクレオチド配列は、毒素、抗生物質および除草 剤からなる群から選択された少なくとも1つに対する耐性をコードする更なるヌ クレオチド配列を共挿入されており、その中または上で細胞を培養する培地は形 質転換細胞が耐性を有する毒素、抗生物質および除草剤からなる群から選択され た少なくとも1つを含む、請求項1〜13のいずれかに記載の方法。 15.所望のヌクレオチド配列を、少なくとも2つの異なった選択遺伝子と共 挿入する、請求項1〜14のいずれかに記載の方法。 16.毒性軽減剤がメチル−3−O−グルコースおよびフロリジンからなる群 から選択されるものである、請求項1〜15の何れかに記載の方法。 17.請求項1〜16の何れかに記載の方法を使用して選択される、形質転換 細胞。 18.植物細胞である、請求項17記載の形質転換細胞、およびそのような細 胞由来の植物、子孫または種子。
