Abstract
La presente invencion comprende un proceso de seleccion para incrementar el numero de células de planta transformadas a partir de un mezcla de células de planta transformadas y no transformadas cultivadas bajo condiciones de cultivo heterotroficas. De acuerdo con este proceso, se cultiva una mezcla de células de planta transformadas y no transformadas bajo condiciones de cultivo heterotroficas en un medio de cultivo que contiene nutrientes minerales requeridos para la proliferacion y crecimiento de células de planta no transformadas excepto por una fuente de carbono que sustenta el crecimiento y la proliferacion, y aproximadamente 1.5 a 3 veces la cantidad estándar de fosforo. La fuente de carbono utilizada se sustituye por una fuente de carbono oculta que no sustenta el crecimiento y la proliferacion de dichas células no transformadas. Las células de planta transformadas tienen un segmento de ADN heterologo insertado dentro de su genoma, que contiene por lo menos un casete de expresion. El casete de expresion contiene un segmento marcador seleccionable de ADN heterologo que incluye un primer gen heterologo que codifica una enzima heterologa, que en la expresion convierte a la fuente de carbono oculta en una fuente de carbono que sustenta el crecimiento y la proliferacion de las células de planta transformadas bajo condiciones de cultivo heterotroficas. Adicionalmente, la presente invencion también contempla un proceso de seleccion para incrementar el numero de células de planta transformadas a partir de una mezcla de células de planta transformadas y no transformadas cultivadas bajo condiciones retardadas de cultivo selectivo.
