Abstract
【解決手段】皮膚結合組織の成分と相互作用するためのナノトポグラフィベースの方法及びデバイスが提供される。本発明のデバイスは、ナノトポグラフィを形成するように表面上に作製された構造体を含む。サイズ及び/または形状の異なる構造体を含む複雑なパターンなど、構造体のランダムまたは非ランダムなパターンを作製することができる。細胞または組織への薬剤の送達にマイクロニードルを有効に利用することができる。作製されたナノトポグラフィと、細胞の細胞膜及び/または細胞外マトリックス成分との相互作用により、本発明のデバイスを用いて、細胞挙動を直接または間接的に変化させることができる。

1.透過性アッセイが行われる2週間前に、コラーゲンコーティングしたウェルインサート上にCaco−2細胞を播く。100%エタノールとコラーゲンの体積比を1:1にすることによって、コラーゲンコーティングプレートを作製する。無菌フード内で表面を乾燥するまで一晩乾燥させる。
2.フェノールレッド不含α−MEM培地において対象のFITC共役分子(BSA、IgGなど)の溶液を0.1mg/mL作る。アルミ箔にくるんで光から保護する。
3.抵抗を測定することによって、Caco−2細胞の密集度をチェックする。密集度に対して抵抗は約〜600Ωであるべきである。
4.頂端(アピカル)側及び基底外側においてセルカルチャーインサートから古い培地を吸引する。PBSでリンスして、残りのフェノールレッド色素を除去する。
5.各インサートの頂端側において0.5mLのFITC共役溶液を加える。
6.セルカルチャーインサートを備えた別の24ウェルプレートにおいて、0.5mLの加温したPBSを加える。
7.インサートをPBSの付着したプレートに移す。キムワイプでインサートの底部を拭き取ることにより、残りのフェノールレッドを除去する。
8.t=0の時点:インサートの基底外側から75mLを採取し、底部が黒色の96ウェルプレートに移す。上記体積を75mLの加温したPBSに置き換える。「チョップスティック型」電極を用いて各ウェルの抵抗を記録する。
9.メンブレンを適切にラベルを付けたウェルに注意深く加える。対照は、インプリントされていないメンブレン及び細胞単独である。顕微鏡下で、メンブレンが細胞と直接接触することをチェックする。細胞との接触を示すはっきりした円を見ることができるはずである。
10.t=0の時点:ステップ7を繰り返し、その後インキュベータ内に1時間載置する。
11.t=1の時点:ステップ7を繰り返し、その後インキュベータ内に1時間載置する。
12.t=2の時点:ステップ7を繰り返す。
13.分光蛍光光度計プレートリーダーを用いて蛍光シグナルを測定する。FITC(励起=490nm、発光=520nm)
使用したフィルム及び得られた結果を要約したものを下表7に示す。
Claims (21)
- 支持体から外向きに延出するマイクロニードルのアレイを含む医療デバイスであって、
前記マイクロニードルのうちの少なくとも1本の表面上に形成されかつ所定のパターンで配列された複数のナノ構造体を含むことを特徴とするデバイス。 - 前記ナノ構造体が、ピラーの形状を有することを特徴とする請求項1に記載の医療デバイス。
- 前記複数のナノ構造体のうちの少なくとも一部のナノ構造体が、約5nm〜500nmの断面寸法を有するか、または約100〜300nmの断面寸法を有するか、または略同一の断面寸法を有することを特徴とする請求項1または請求項2のいずれか1項に記載の医療デバイス。
- 前記パターンが、例えば約500nm超程度の断面寸法を有するマイクロ構造体をさらに含み、前記ナノ構造体が、前記マイクロ構造体よりも小さい断面寸法を有することを特徴とする請求項1ないし請求項3のいずれか1項に記載の医療デバイス。
- 前記マイクロ構造体の前記断面寸法より小さくかつ前記第1のナノ構造体の前記断面寸法より大きい断面寸法を有する第2のナノ構造体をさらに含むことを特徴とする請求項4に記載の医療デバイス。
- 前記複数のナノ構造体のうちの少なくとも一部のナノ構造体が、約50nm〜1μmの中心間距離を有することを特徴とする請求項1ないし請求項5のいずれか1項に記載の医療デバイス。
- 互いに隣接する2つのナノ構造体の断面寸法と該2つの構造体間の中心間距離との比が、約1:1〜1:4であることを特徴とする請求項1ないし請求項6のいずれか1項に記載の医療デバイス。
- 前記複数のナノ構造体のうちの少なくとも一部のナノ構造体が、互いに等距離間隔で離間されていることを特徴とする請求項1ないし請求項7のいずれか1項に記載の医療デバイス。
- 前記複数のナノ構造体のうちの少なくとも一部のナノ構造体が、約10nm〜20μmまたは約100〜700nmの高さを有することを特徴とする請求項1ないし請求項8のいずれか1項に記載の医療デバイス。
- 前記複数のナノ構造体のうちの少なくとも一部のナノ構造体が、約0.15〜30または約0.2〜5のアスペクト比を有することを特徴とする請求項1ないし請求項9のいずれか1項に記載の医療デバイス。
- 前記パターンが、約1以上、例えば約1.5〜2.5程度のフラクタル次元を有することを特徴とする請求項1ないし請求項10のいずれか1項に記載の医療デバイス。
- 前記複数のナノ構造体を含む前記マイクロニードル表面が、以下の特性、すなわち、約10〜200nmの平均表面粗さ、約4〜320MPaの有効圧縮弾性率、約0.2〜50MPaの有効剪断率、約80°〜150°の水接触角のうちの1若しくは複数を有することを特徴とする請求項1ないし請求項11のいずれか1項に記載の医療デバイス。
- 例えば約100kDaより大きな分子量を有する薬剤化合物などの薬剤化合物を保持するための貯蔵部をさらに含むことを特徴とする請求項1ないし請求項12のいずれか1項に記載の医療デバイス。
- 前記薬剤化合物が、例えばTNF−α阻害薬などのタンパク質治療薬であることを特徴とする請求項13に記載の医療デバイス。
- 前記マイクロニードルのアレイのうちの少なくとも1本のマイクロニードルが、前記薬剤化合物を送達するためのチャンネルを含むことを特徴とする請求項1ないし請求項14のいずれか1項に記載の医療デバイス。
- 薬剤化合物を皮下位置に送達する方法であって、
表面上に形成されかつ或るパターンで配列された複数のナノ構造体を含みかつ前記薬剤化合物に流体連通しているマイクロニードルにより、角質層を穿刺するステップと、
前記マイクロニードルを介しかつ角質層を透過させて前記薬剤化合物を輸送するステップとを含むことを特徴とする方法。 - 細胞外マトリックスタンパク質、接着斑タンパク質などの原形質膜タンパク質、エンドサイトーシス媒介受容体、膜貫通タンパク質、またはこれらの組合せを、前記ナノ構造体と接触させるステップをさらに含むことを特徴とする請求項16に記載の方法。
- 前記ナノ構造体が、タイトジャンクションなどの細胞間結合の構造または細胞の膜伝導率の一方または両方を変更することを特徴とする請求項16または請求項17のいずれか1項に記載の方法。
- 前記薬剤化合物が、細胞の細胞質または細胞の核膜に送達されることを特徴とする請求項6ないし請求項18のいずれか1項に記載の方法。
- 医療デバイスを作製する方法であって、
マイクロニードルの表面上にナノ構造体のパターンを形成するステップを含むことを特徴とする方法。 - 前記ナノ構造体のパターンを、フォトリソグラフィ、電子ビームリソグラフィ、X線リソグラフィ、自己組織化技術、反応性イオンエッチング、湿式エッチング、膜堆積、スパッタリング、化学蒸着、エピタキシー、電気めっき、ナノインプリントリソグラフィ、及びそれらの組合せからなる群より選択される技術に従って作製することを特徴とする請求項20に記載の方法。
