Claims (8)
1. Antitijelo koje sadrži modificiranu konstantnu regiju teškog lanca, pri čemu modificirana konstantna regija teškog lanca sadrži CH1 domenu, zglob, CH2 domenu, i CH3 domenu redom od N- do C- terminusa, pri čemu:
(a) zglob sadrži aminokiselinsku sekvencu SEQ ID NO: 21;
(b) CH1 domena sadrži aminokiselinsku sekvencu SEQ ID NO: 7;
(c) CH2 domena sadrži aminokiselinsku sekvencu SEQ ID NO: 4 ili aminokiselinsku sekvencu SEQ ID NO: 24; i
(d) CH3 domena sadrži aminokiselinsku sekvencu SEQ ID NO: 5 ili SEQ ID NO: 5 sa aminokiselinskim supstitucijama E356D i M358L;
ili pri čemu modificirana konstantna regija teškog lanca koji sadrži CH1 domenu, zglob, CH2 domenu, i CH3 domenu kao što je definirano pod (a), (b), (c) i (d) nema C-terminalni GK ili K.
2. Antitijelo prema patentnom zahtjevu 1, pri čemu antitijelo sadrži modificiranu konstantnu regiju teškog lanca odabranu iz grupe SEQ ID NO: 34, 35, 37, , 81, i 84.
3. Antitijelo prema patentnom zahtjevu 2, koje se (i) specifično vezuje za kostimulatorni receptor; (ii) specifično vezuje za molekulu stanične površine i pokreće internalizaciju posredovanu antitijelom molekula stanične površine; (iii) specifično vezuje za inhibitorni receptor; (iv) specifično vezuje za molekulu stanične površine i pokreće unutarstaničnu signalizaciju; (v) specifično vezuje za molekulu stanične površine i pokreće formiranje kompleksa antitijelo- molekulu stanične površine velike molekulske mase; ili (vi) specifično vezuje za molekulu stanične površine i pokreće klasteriziranje ili oligomerizaciju molekula stanične površine.
4. Antitijelo prema patentnom zahtjevu 3, pri čemu kostimulatorni receptor je GITR, OX40, 4- 1BB, CD28, ICOS, CD40L, CD27 ili bilo koji drugi član TNFR superfamilije; molekula stanične površine je CD73; inhibitorni receptor je CTLA-4, PD-1, LAG-3, TIM-3, Galektin 9, CEACAM- 1, BTLA, CD69, Galektin-1, TIGIT, CD113, GPR56, VISTA, 2B4, CD48, GARP, PD1H, LAIR1, TIM-1 i TIM-4; ili unutarstanična signalizacija posreduje u agonističkoj aktivnosti, antagonističkoj aktivnosti, internalizaciji molekula stanične površine, ili ADCC.
5. Antitijelo prema patentnom zahtjevu 3 ili 4, pri čemu antitijelo ispoljava poboljšanu ili izmijenjenu agonističku aktivnost u odnosu na antitijelo koje ima iste varijabilne regije i laki lanac, ali koje sadrži konstantnu regiju teškog lanca IgG1; antitijelo posjeduje poboljšana ili izmijenjena svojstva internalizacija u odnosu na antitijelo koje ima iste varijabilne regije i laki lanac, ali koje sadrži konstantnu regiju teškog lanca IgG1; antitijelo ispoljava potentniju ili izmijenjenu antagonističku aktivnost ili uvodi novu aktivnost u odnosu na isto antitijelo koje ima konstantnu regiju teškog lanca IgG1; antitijelo pokreće potentniju unutarstaničnu signalizaciju u odnosu na antitijelo koje ima iste varijabilne regije i laki lanac, ali koje sadrži konstantnu regiju teškog lanca IgG1; antitijelo pokreće formiranje kompleksa veće molekulske mase u odnosu na antitijelo koje ima iste varijabilne regije i laki lanac, ali koje sadrži konstantnu regiju teškog lanca IgG1; ili antitijelo pokreće veće klasteriziranje ili oligomerizaciju molekula stanične površine u odnosu na antitijelo koje ima iste varijabilne regije i laki lanac, ali koje sadrži konstantnu regiju teškog lanca IgG1.
6. Bispecifična molekula koji sadrži antitijelo prema bilo kojem od prethodnih patentnih zahtjeva, povezan sa molekulom koji ima drugu specifičnost vezivanja.
7. Imunokonjugat koji sadrži antitijelo prema bilo kojem od prethodnih patentnih zahtjeva, povezan sa drugim agensom.
8. Kompozicija koja sadrže antitijelo, bispecifičnu molekulu ili imunokonjugat, prema bilo kojem od patentnih zahtjeva 1-7, nosač, i po izboru jedan ili više dodatnih terapijskih agenasa.
