Claims (11)
1. Vještački fuzijski protein Tip 1 CRISPR vezane proteinske podjedinice Cse1 i Fok I(jedan) endonukleaze.
2. Fuzijski protein prema zahtjevu 1, pri čemu fuzijski protein uključuje i signalni slijed za lokalizaciju.
3. Nukleinska kiselina čiji slijed nosi informaciju za sintezu fuzijskog proteina prema zahtjevu 1 ili 2.
4. Ekspresijski vektor koji sadrži nukleinsku kiselinu prema zahtjevu 3.
5. Kompleks proteina kaskade koji sadrži fuzijski protein prema zahtjevu 1 ili 2.
6. Ribonukleoproteinski kompleks koji sadrži fuzijski protein prema zahtjevu 1 ili 2 i molekulu CRISPR RNA (crRNA).
7. Ribonukleoproteinski kompleks prema zahtjevu 6, koji sadrži i proteinsku podjedinicu Cas6, proteinsku podjedinicu Cas5, proteinsku podjedinicu Cse2 i proteinsku podjedinicu Cas7.
8. Eukariotska stanica koja sadrži ribonukleoproteinski kompleks prema zahtjevu 6 ili 7, pri čemu ta stanica ne pripada humanoj germinativnoj liniji niti je dio humanog ili animalnog tijela.
9. Postupak za modifikaciju, vizualizaciju, aktivaciju ili represiju prepisivanja ciljne nukleinske kiseline in vitro koji uključuje kontakt ciljne nukleinske kiseline i ribonukleoproteinskog kompleksa prema zahtjevu 6 ili 7.
10. Postupak prema zahtjevu 9, pri čemu se postupak modifikacije sastoji iz vezanja i/ili cijepanja ciljne nukleinske kiseline.
11. Postupak prema zahtjevu 9 ili 10, pri čemu je ciljna nukleinska kiselina dvolančana DNA (dsDNA).
