Abstract

Ένα σύμπλεγμα συνδεδεμένο με στοχευόμενη τακτική βραχεία παλινδρομική επανάληψη (CRISPR) για προσαρμοστική αντιιική άμυνα (Cascade)• όπου το σύμπλεγμα πρωτεΐνης Cascade περιλαμβάνει τουλάχιστον τις υπομονάδες της CRISPR-συνδεδεμένης πρωτεΐνης cas7, cas5, και cas6 που περιλαμβάνει τουλάχιστον μία υπομονάδα με μία επιπλέον αλληλουχία αμινοξέων που διαθέτει δραστικότητα τροποποίησης, εμφάνισης, ενεργοποίησης μεταγραφής ή καταστολής μεταγραφής νουκλεϊκού οξέος ή χρωματίνης. Το σύμπλεγμα Cascade με επιπλέον δραστικότητα ενώνεται με ένα μόριο RNA για να παράγει ένα σύμπλεγμα ριβονουκλεοπρωτεΐνης. Το μόριο RNA επιλέγεται να έχει ουσιαστική συμπληρωματικότητα προς μία αλληλουχία στόχο. Οι στοχευόμενες ριβονουκλεοπρωτεΐνες μπορούν να χρησιμοποιούνται ως εργαλεία γενετικής μηχανίκευσης για κοπή ακριβείας νουκλεϊκών οξέων σε ομόλογο ανασυνδυασμό, μη ομόλογη σύνδεση άκρου, τροποποίηση γονιδίου, ενσωμάτωση γονιδίου, επιδιόρθωση μετάλλαξης ή για την εμφάνιση, μεταγραφική ενεργοποίηση ή καταστολή αυτών. Ένα ζεύγος ριβονουκλεοτιδίων συγχωνευμένων με διμερή Fokl μπορούν να χρησιμοποιούνται για την παραγωγή δίκλωνων θραύσεων στο DNA ώστε να διευκολύνονται οι εφαρμογές αυτές κατά έναν τρόπο ειδικής αλληλουχίας.
