Description
Oppfinnelsens bakgrunn
Foreliggende oppfinnelse er rettet mot eri fremgangsmåte for acylering av en C(10)-hydroksygruppe i et taksan som har C(7)- og C(10)-hydroksygrupper.
10-DAB (1) som ekstraheres fra nålene fra Taxus bac-cata L., vanlig barlind, er blitt et nøkkelutgangsmateriale ved fremstillingen av taksol og taxoter, som begge er sterke antikreftmidler. Omdannelse av 10-DAB til taksol, Taxoter og andre taksaner med antitumoraktivitet krever beskyttelse eller derivatisering av C(7)- og C(10)-hydroksylgruppene, etterfulgt av forestring av C(13)-hydroksylgruppen for å feste en passende sidekjede i den stillingen.
Hittil har strategier for fremstillingen av taksol og taxolanaloger vært basert på iakttagelsen til Senilh et al.
(CR. Acad. Sei. Paris, II, 1981, 293,. 501) om at den relative reaktivitet til de fire hydroksylgruppene i 10-DAB mot eddiksyreanhydrid i pyridin er C(7)-OH > C(10)-OH > C(13)-OH > C(l)-OH. Denis, et al. (J. Am. Chem. Soc, 1988, 110, 5917) rapporterte selektiv silylering av C(7)-hydroksylgruppen i 10-DAB med trietylsilylklorid i pyridin, hvorved man får 7-trietylsilyl-10-deacetylbaccatin (III) (2) i 85 % utbytte. Basert på disse rapportene må, i de fremgangsmåtene hvor differensiering mellom C(7)- og C(10)-hydroksylgruppene er nødvendig (f eks fremstilling av taksol 10-DAB), C(7)-hydroksylgruppen beskyttes (eller derivatiseres) før C(10)-hydroksylgruppen beskyttes eller derivatisering. F eks kan taksol fremstilles ved å behandle 10-DAB med trietylsilyl-
klorid for å beskytte C(7)-hydroksylgruppen, acetylere C(10)-hydroksylgruppen, feste sidekjeden ved forestring av C(13)-hydroksylgruppen og til sist fjerne beskyttelsesgrupper.
Det er kjent at taksaner med forskjellige substituenter bundet til enten C(10)- eller C(7)-oksygenatomene oppviser antikreftaktivitet. For å tilveiebringe mer virkningsfull syntese av disse materialene, ville det vært nyttig å ha fremgangsmåter som muliggjør mer virkningsfull og mer høyselektiv beskyttelse eller derivatisering av C(10)- og C(7)-hydroksylgruppene.
Oppsummering av oppfinnelsen
Formålet ved foreliggende oppfinnelse er derfor tilveiebringelsen av en svært virkningsfull fremgangsmåte for fremstilling av taksol og andre taksaner gjennom selektiv acylering av C(10)-hydroksylgruppen i 10-DAB og andre taksaner, særlig en fremgangsmåte hvor C(10)-hydroksylgruppen acyleres før C(7)-hydroksylgruppen.
Foreliggende oppfinnelse er derfor rettet mot en. fremgangsmåte for acylering av en C(10)-hydroksygruppe i et taksan som har C(7)- og C(10)-hydroksygrupper. Fremgangsmåten er kjennetegnet ved at taksanet behandles med et acyleringsmiddel valgt fra gruppen bestående av anhydrider, dikarbonater, tiodikarbonater og isocyanater, i en reaksjons-blanding inneholdende en Lewis-syre og mindre enn én ekvivalent av en base for hver ekvivalent taksan, for selektivt å acylere C(10)-hydroksygruppen.
Nærmere beskrivelse av de foretrukne utførelsesformer
Blant andre ting muliggjør den foreliggende
oppfinnelse den selektive acylering av C(10)-hydroksylgruppen i et taksan uten først å beskytte C(7)-hydroksylgruppen. Sagt på en annen måte er det blitt oppdaget at reaktivitetene tid-ligere rapportert for C(7)- og C (10)-hydroksylgruppene kan
reverseres, det vil si at reaktiviteten til C(10)-hydroksyl-gruppen blir større enn reaktiviteten til C(7)-hydroksyl-gruppen under visse betingelser.
Den foreliggende oppfinnelse byr på særlige fordeler ved den selektive acylering av taksaner som har hydroksy-grupper i C(7) og C(10), dvs. 7,10-dihydroksytaksaner. Generelt tilsvarer 7,10-dihydroksytaksaner som kan acyleres selektivt i overensstemmelse med foreliggende oppfinnelse, den følgende formel:

Selektiv C(10)-derivatisering
I overensstemmelse med fremgangsmåten ifølge foreliggende oppfinnelse er det blitt oppdaget at C(10)-hydroksylgruppen i et taksan kan acyleres selektivt i fravær av en base, fortrinnsvis i fravær av en aminbase. Aminbaser, som pyridin, trietylamin, dimetylaminopyridin og 2,6-lutidin, er derfor, dersom de i det hele tatt er til stede, fortrinnsvis til stede i reaksjonsblandingen ved forholdsvis lav konsentrasjon. Sagt på en annen måte er, dersom en base er til stede i reaksjonsblandingen, det molare forholdet mellom aminbasen og taksanet fortrinnsvis mindre 1:1, mer foretrukket mindre enn 10:1, og mest foretrukket mindre enn 100:1.
Acyleringsmidler som kan anvendes til den selektive acylering av C(10)-hydroksylgruppen i et taksan, omfatter anhydrider, dikarbonater, tiodikarbonater og isocyanater. Generelt tilsvarer anhydridene, dikarbonatene og tiodikarbonatene formel 4, og isocyanatene tilsvarer formel 5:

For eksempel omfatter egnede karboksyisyreanhydridacylerende midler eddiksyreanhydrid, kloreddiksyreanhydrid, propionsyreanhydrid, benzosyreanhydrid og andre karboksylsyreanhydrider som inneholder substituerte eller usubstituerte hydrokarbyl- eller heteroarylrester; egnede dikarbonatacyleringsreagenser omfatter dibenzyldikarbonat, diallyldikarbonat, dipropyldikarbonat og andre dikarbonater som inneholder substituerte eller usubstituerte hydrokarbyl- eller heteroarylrester; og egnede isocyanatacyleringsmidler omfatter fenylisocyanat og andre isocyanater. som inneholder substituerte eller usubstituerte hydrokarbyl- eller heteroarylrester. Selv om anhydridene, dikarbonatene og tiodikarbonatene som anvendes som acyleringsmidler kan være blandet, er det generelt foretrukket at de er symmetriske; dvs. at R<1> og R2 er valgt slik at molekylet er symmetrisk (f eks dersom R<1> er Ra, R<2> er -OC(0)R<b >idet Ra er det samme som Rb) .
Selv om acyleringen av C(10)-hydroksygruppen i taksanet vil forløpe ved en passe hastighet for mange acyleringsmidler, er det blitt oppdaget at reaksjonshastigheten kan økes ved å inkludere en Lewis-syre i reaksjonsblandingen. Konsentrasjonen av Lewis-syren synes ikke å være spesielt avgjørende; forsøksbevis oppnådd hittil tyder på den kan være til stede enten ved en støkiometrisk eller en katalytisk mengde. Generelt omfatter Lewis-syrer som kan anvendes, triflater og halogenider av grunnstoffene i gruppene IB, HB, UIB, IVB, VB, VIB, VIIB, VIII, HIA, IVA, lantanider og actinider i den periodiske tabell (formatet til American Chemical Society). Foretrukne Lewis-syrer omfatter sinkklorid, tinnklorid, ceriumtriklorid, kobberklorid, lantanumtriklorid, disprosiumtriklorid og ytterbiumtriklorid. Sinkklorid eller ceriumtriklorid er særlig foretrukket når acyleringsmidlet er et anhydrid eller dikarbonat. Kobberklorid er særlig foretrukket når acyleringsmidlet er et isocyanat.
Oppløsningsmidlet for den selektive acylering er fortrinnsvis et slikt eteriske oppløsningsmiddel som tetrahydrofuran. Alternativt kan det imidlertid anvendes andre oppløsningsmidler, slik som eter eller dimetoksyetan.
Temperaturen hvorved den C(10)-selektive acylering utføres, er ikke spesielt avgjørende. Generelt utføres den imidlertid foretrukket ved romtemperatur eller høyere for at reaksjonen skal forløpe ved en tilstrekkelig høy hastighet.
For illustrasjonsformål er acyleringsreaksjoner som omfatter dibenzyldikarbonat, diallyldikarbonat, eddiksyreanhydrid, kloreddiksyreanhydrid og fenylisocyanat, illustrert i reaksjonsskjemaene 1-5 nedenunder. I denne serien av reaksjonsskjemaer er taksanet som er selektivt acylert i C (10)-stillingen, 10-deacetylbaccatin .1.11. Det skal imidlertid forstås at disse reaksjonsskjemaene. kun er.illustrerende og at andre taksaner med en C(10)-hydroksygruppe generelt, og andre 7,10-dihydroksytaksaner spesielt, selektivt kan acyleres med disse og andre acyleringsmidler i overensstemmelse med foreliggende oppfinnelse.
Definisjoner
Slik de her er brukt skal uttrykkene "selektiv" og "selektiv acylering" bety at det ønskede produkt fortrinnsvis er dannet over hvilket som helst annet produkt. Det ønskede produkt er fortrinnsvis til stede i et molart forhold på minst 9:1 i forhold til ethvert annet biprodukt og, mer foretrukket, er det til stede ved et molart forhold på minst 20:1 i forhold til ethvert annet biprodukt.

C. Selektiv acylering av en C (10)-hydroksylgruppe under anvendelse av CeCl3: Generell fremgangsmåte: Til en oppløsning av 10-DAB i THF (20 ml pr mmol 10-DAB) under N2 ble det tilsatt CeCl3 og det passende anhydrid eller pyrokarbonat (mengder angitt i tabell 1). Reaksjonsblandingen ble omrørt ved 25 °C og over-våket ved hjelp av TLC-analyse. Da analysen indikerte full-stendig omsetning (tid angitt i tabell 1), ble reaksjonsblandingen fortynnet med EtOAc og vasket 3 ganger med mettet vandig natriumbikarbonatoppløsning. De kombinerte bikarbonat-vaskinger ble ekstrahert tre ganger med EtOAc, de organiske lagene ble slått sammen og tørket over natriumsulfat og opp-løsningsmidlet ble avdampet. Råproduktet ble renset ved hjelp av hurtigkolonnekromatografi. Videre rensing ble om nødvendig utført ved rekrystallisasjon fra EtOAc/heksan.

1. Fremgangsmåte for acylering av en C(10)-hydroksygruppe i et taksan som har C(7)- og C(10)-hydroksygrupper, karakterisert ved at taksanet behandles med et acyleringsmiddel valgt fra gruppen bestående av anhydrider, dikarbonater, tiodikarbonater og isocyanater, i en reaksjons-blanding inneholdende en Lewis-syre og mindre enn én ekvivalent av en base for hver ekvivalent taksan, for selektivt å acylere C(10)-hydroksygruppen.
