- TECHNISCHES GEBIET
- Die vorliegende Erfindung betrifft die Verwendung von antimikrobiellen Zusammensetzungen, die eine verbesserte unmittelbare und anhaltende antivirale Wirksamkeit bereitstellen, bei der Herstellung von Medikamenten zur Verhinderung und/oder Behandlung einer Erkältung und damit verbundener Atemwegserkrankungen, die von einem Rhinovirus verursacht werden.
- HINTERGRUND DER ERFINDUNG
- Die menschliche Gesundheit wird durch eine Vielfalt von mikrobiellen Organismen beeinflusst. Die Inokulation von Menschen und anderen Säugetieren durch diese Mikroorganismen verursacht verschiedene Krankheiten und Beschwerden. Das öffentliche Bewusstsein über derartige Verseuchungen ist aufgrund der zunehmenden Zahl an Fällen von Nahrungsmittelvergiftungen, Streptokokkeninfektionen usw. in der jüngsten Vergangenheit geschärft. Folglich kam es zu einem Vorstoß seitens der medizinischen Gemeinschaft, die Öffentlichkeit davon zu überzeugen, alle Flächen zu reinigen, die im Allgemeinen mit infizierten Oberflächen in Berührung kommen, wie Körperteile (z. B. Händewaschen), Nahrungsmittel (z. B. rohes Fleisch, Gemüse, Obst usw.), Kochgeräte, Kochflächen (z. B. Arbeitsplatten, Spülbecken usw.). Es hat sich herausgestellt, dass derartige Verfahren bei dem Bemühungen um eine Entfernung von pathogenen Mikroorganismen von der menschlichen Haut und von anderen Oberflächen wichtig sind.
- Zu den Arten von Mikroorganismen, die auf der Haut von Säugetieren gefunden werden, gehören Viren, Bakterien und Pilze. Unter Virologen herrscht allgemeine Einmütigkeit darüber, dass Rhinoviren, Grippeviren und Adenoviren mit größter Wahrscheinlichkeit die wichtigsten Viren sind, die Atemwegserkrankungen verursachen. Es wird angenommen, dass insbesondere Rhinoviren als Hauptursache für Erkältungen verantwortlich sind. Rhinoviren gehören zur Familie der Picornaviren. Als solche werden sie als „nackte Viren" bezeichnet, da ihnen eine äußere Hülle fehlt. Derartige Picornaviren lassen sich bekanntermaßen nur unter Schwierigkeiten durch üblicherweise verwendete Mittel, wie quartäre Ammoniumverbindungen, inaktivieren.
- Rhinovirusinfektionen werden über mit Viren verseuchte Sekretionen der Atemwege von Person zu Person verbreitet. Alles deutet darauf hin, dass der Hauptübertragungsweg über die direkte Berührung verläuft und nicht über das Einatmen von Virenpartikeln in der Luft. Es konnte nachgewiesen werden, dass erkrankte Personen dazu neigen, ihre Hände und Gegenstände in ihrer Umgebung zu kontaminieren. Rhinoviren wurden an 40% bis 90% der Hände von Personen gefunden, die an Erkältung leiden, und an 6% bis 15% verschiedener Gegenstände. Der Rhinovirus zeigt selbst Stunden nach der Verseuchung ein gutes Überleben auf zahlreichen Oberflächen in der Umgebung und eine Infektion ist schnell durch Kontakt zwischen zwei Fingern oder zwischen Finger und verseuchter Oberfläche in der Umgebung übertragen, wenn mit dem frisch verseuchten Finger anschließend ein Auge gerieben oder die Nasenschleimhaut berührt wird.
- Da ein wesentlicher Anteil an Rhinovirus-Erkältungen über den direkten Kontakt zwischen virenverseuchten Händen oder Gegenständen übertragen wird, ist es möglich, das Risiko einer Ansteckung durch Inaktivieren der Viren auf Händen oder Oberflächen zu senken. Ein übliches Phenol-/Alkohol-Haushaltsinfektionsmittel hat sich beim Desinfizieren verseuchter Oberflächen in der Umgebung als wirksam erwiesen, es fehlen aber anhaltende viruzide Wirkungen. Das Händewaschen ist zur Desinfektion von verseuchten Händen ausgesprochen wirksam, er neut fehlt aber die anhaltende Wirkung. Diese Mängel bieten gute Möglichkeiten für verbesserte viruzide Technologien gegen Rhinoviren mit anhaltender Wirkung.
- Es hat sich herausgestellt, dass Iod ein wirksames antivirales Mittel darstellt und auf der Haut eine anhaltende antirhinovirale Wirkung bereitstellt. In Studien über die experimentell verursachte und natürliche Übertragung von Erkältungen zeigten Probanden, die Iodprodukte verwendeten, signifikant weniger Erkältungen als Placebo-Benutzer. Dies lässt darauf schließen, dass Iod über längere Zeiträume beim Blockieren der Übertragung von rhinoviralen Infektionen wirksam ist. Somit wäre die Entwicklung von Handprodukten, -lotionen und -waschlösungen (ohne die mit Iod verbundenen negativen Farb- und Geruchsaspekte), die eine unmittelbare und anhaltende antivirale Wirkung bereitstellen, bei der Senkung von Erkältungsfällen wirksam. Ebenso wäre ein topisches Produkt, das eine antivirale Wirkung aufweist, zur Verhinderung und/oder Behandlung von Viruskrankheiten wirksam, die von anderen Viren, wie Adenoviren, Rotaviren, Herpes-Viren, Respiratory-syncytial-Viren, Coronaviren, Parainfluenza-Viren, Enteroviren, Influenza-Viren usw. verursacht werden.
- Es besteht jedoch weiterhin ein Bedarf nach Zusammensetzungen und Produkten, die nicht nur eine verbesserte unmittelbare antivirale Wirksamkeit zeigen, sondern auch eine verbesserte anhaltende Wirksamkeit. Weiterhin besteht ein Bedarf nach Bereitstellung einer verbesserten unmittelbaren antiviralen (z. B. antirhinoviralen) Wirkung in Systemen auf Wasserbasis (d. H. nicht alkoholisch).

DE 2715711 lehrt die therapeutische, antivirale Verwendung von Metallsalzen, in erster Linie in Verbindung mit der Behandlung des Herpes-Virus. Sie enthält keine Lehre über die Verwendung gegen Rhinoviren oder zur Behandlung von Erkältungen. - US-Patent Nr. 4,975,217 betrifft viruzide Zusammensetzungen, die organische Säuren, wie Benzoesäure oder Salicylsäure, in Kombination mit einem Tensid enthalten. Die Zusammensetzungen können u. a. in Nasensprays eingearbeitet werden. Das Tensid kann ein anionisches Tensid sein, das mit einem zwei- oder dreiwertigen Metallkation assoziiert ist.
- US-Patent Nr. 5,508,282 betrifft Zusammensetzungen, z. B. Nasensprays, und Verfahren zum Lindern von Symptomen, und Verkürzen der Dauer, akuter oder chronischer Rhinosinusitis. Dessen Zusammensetzungen umfassen Vitamin C und Koffein und wahlweise Benzoesäure und Mineralstoffe, die Zink sein können. Dessen Zusammensetzungen sind vorzugsweise auf einen pH-Wert von 5,5 bis 6,5 eingestellt. Diese Schrift enthält keinen Hinweis darauf, dass deren Zusammensetzungen antimikrobielle oder antivirale Wirkung haben.
- WO 99/45771 offenbart wasserfreie Lotionen mit antiviraler und antimikrobieller Wirkung zur Behandlung von Gesichtstüchern. Die Lotionen enthalten ein antimikrobielles Mittel, das ein antivirales Mittel und ein antibakterielles Mittel oder eine Kombination sein kann. Salicylsäure ist als ein Beispiel für ein antivirales Mittel offenbart und Metallionen sind als ein Beispiel für ein antibakterielles Mittel offenbart. Die Schrift offenbart keine wässrigen Lösungen für die intranasale Anwendung.
- Die Anmelder haben festgestellt, dass die erfindungsgemäßen wässrigen Zusammensetzungen, die ein Benzoesäure-Analogon, ein Metallsalz und einen Träger umfassen, wobei die Zusammensetzung einen pH-Wert von 1,5 bis 5 aufweist, zur Herstellung von Medikamenten zur Verhinderung und/oder Behandlung einer Erkältung oder einer damit verbundenen Atemwegserkrankung, die von einem Rhinovirus verursacht wird, nützlich sind.
- ZUSAMMENFASSUNG DER ERFINDUNG
- Die vorliegende Erfindung betrifft die Verwendung eines Benzoesäure-Analogons, wie in Anspruch 1 beschrieben, zur Herstellung eines Medikaments zur Verhinderung und/oder Behandlung einer Erkältung oder damit verbundener Atemwegserkrankungen bei einem Säugetier, die von einem Rhinovirus verursacht wird.
- AUSFÜHRLICHE BESCHREIBUNG DER ERFINDUNG
- Wie hier verwendet, bedeutet „anhaltende antivirale Wirksamkeit" das Hinterlassen eines Rückstands oder das Vermitteln eines Zustands auf einem Keratingewebe (z. B. der Haut) oder anderen Oberflächen, der wirksam bleibt und über einen gewissen Zeitraum nach dem Aufbringen eine signifikante antivirale (spezifisch gegen Rhinovirus) Wirkung bereitstellt.
- Nachstehend sind die zentralen Bestandteile und Eigenschaften dieser Zusammensetzungen beschrieben. Ebenfalls eingeschlossen ist eine nicht einschränkende Beschreibung verschiedener fakultativer und bevorzugter Bestandteile, die in Ausführungsformen der vorliegenden Erfindung geeignet sind.
- Die vorliegende Erfindung kann jegliche hierin beschriebene erforderliche oder fakultative Bestandteile und/oder Einschränkungen umfassen, daraus bestehen oder im Wesentlichen daraus bestehen.
- Alle hier aufgeführten Prozentsätze und Verhältnisse sind, sofern nichts anderes angegeben wurde, auf das Gewicht bezogen. Alle Prozentsätze sind auf Grundlage der Gesamtzusammensetzung berechnet, sofern nicht anderweitig angegeben.
- Alle molaren Gewichtsangaben sind gewichtsgemittelte Molekulargewichte und in der Einheit Gramm/Mol angegeben.
- Alle Bestandteilkonzentrationen beziehen sich auf die Wirkstoffkonzentration des Bestandteils und schließen, sofern nichts anderes angegeben wurde, Lösungsmittel, Nebenprodukte oder andere Fremdstoffe, die in im Handel erhältlichen Quellen vorhanden sein können, aus.
- Alle Messungen wurden, sofern nichts anderes angegeben wurde, bei Raumtemperatur, die ungefähr 23°C (73°F) betrug, vorgenommen.
- Alle hierin genannten Schriften, einschließlich Patentschriften, Patentanmeldungen und Druckschriften, sind hiermit in ihrer Gänze durch Bezugnahme in diese Offenbarung eingeschlossen.
- Wie hier verwendet, bedeutet „sichere und wirksame Menge" eine Menge einer Verbindung, eines Bestandteils oder einer Zusammensetzung (abhängig davon, was jeweils zutreffend ist), die zum signifikanten Induzieren einer positiven Wirkung ausreicht, aber gering genug, um schwer wiegende Nebenwirkungen zu vermeiden (z. B. übermäßige Toxizität oder eine allergische Reaktion, d. h. die ein angemessenes Nutzen-Risiko-Verhältnis im Rahmen vernünftiger medizinischer Beurteilung bereitstellen.
- In der Beschreibung der Erfindung sind verschiedene Ausführungsformen und/oder individuelle Merkmale offenbart. Wie für den Fachmann offensichtlich sein wird, sind alle Kombinationen solcher Ausführungsformen und Merkmale möglich und können zu bevorzugten Ausführungen der Erfindung führen.
- Benzoesäure-Analogon
- Die erfindungsgemäß nützliche antimikrobielle Zusammensetzung umfasst eine sichere und wirksame Menge eines Benzoesäure-Analogons ausgewählt aus Benzoesäure, Salicylsäure, 2-Nitrobenzoesäure, Thiosalicylsäure, 2,6-Dihydroxybenzoesäure, 3-Hydroxybenzoesäure, 5-Nitrosalicylsäure, 5-Bromsalicylsäure, 5-Iodsalicylsäure, 5-Fluorsalicylsäure, 3-Chlorsalicylsäure, 4-Chlorsalicylsäure, 5-Chlorsalicylsäure, Phthalsäure und Kombinationen davon. Ohne an eine Theorie gebunden zu sein, wird angenommen, dass diese bevorzugten Verbindungen eine verbesserte antibakterielle und antivirale unmittelbare und anhaltende Wirksamkeit zeigen.
- Die Benzoesäure-Analoga können als im Wesentlichen reine Stoffe oder als ein Extrakt enthalten sein, das mittels geeigneter physikalischer und/oder chemischer Isolierung aus natürlichen Quellen (z. B. Pflanzen, Nebenprodukte von Mikroorganismen) gewonnen wird.
- Am meisten bevorzugt ist das Benzoesäure-Analogon ausgewählt aus Salicylsäure, Benzoesäure und Kombinationen davon.
- Vorzugsweise umfassen die vorliegenden Zusammensetzungen 0,01 Gew.-% bis 20 Gew.-%, bezogen auf die Zusammensetzung, des Benzoesäure-Analogons, mehr bevorzugt 0,1 Gew.-% bis 10 Gew.-%, noch mehr bevorzugt 0,25 Gew.-% bis 5 Gew.-% und am meisten bevorzugt 1 Gew.-% bis 5 Gew.-%.
- Außerdem enthalten die erfindungsgemäßen Zusammensetzungen im Wesentlichen keine para-Aminosalicylsäure. Wie hier verwendet, bedeutet „im Wesentlichen kein", dass die nachweisbaren Konzentrationen an para-Aminosalicylsäure geringer als 0,01 Gew.-% der Zusammensetzung sind. Mehr bevorzugt enthalten die erfindungsgemäßen Zusammensetzungen praktisch keine para-Aminosalicylsäure. Wie hier verwendet, bedeutet „praktisch kein", dass para-Aminosalicylsäure in Mengen vorhanden ist, die durch üblicherweise zum Messen derartiger Konzentrationen verwendete Mittel nicht nachweisbar sind. Am meisten bevorzugt enthalten die erfindungsgemäßen Zusammensetzungen keine para-Aminosalicylsäure.
- Außerdem ist vorgesehen, dass der vorstehend beschriebene Säurebestandteil direkt zu den Zusammensetzungen gegeben werden kann oder dass die Säure nach topischer Anwendung der vorliegenden Zusammensetzungen in situ gebildet werden kann. Mit anderen Worten kann ein Vorläufer der beanspruchten Säure zu den Zusammensetzungen gegeben werden, der schließlich in den vorstehend beschriebenen Säurebestandteil umgewandelt wird, z. B. ein Ester der Säure.
- Metallsalze
- Die antimikrobiellen Zusammensetzungen umfassen eine sichere und wirksame Menge eines Metallsalzes ausgewählt aus Mn, Ag, Zn, Sn, Fe, Cu, Al, Ni, Co, Ti, Zr, Cr, La, Bi, K, Cd, Yb, Dy, Nd, Ce, Tl, Pr und Kombinationen davon. Noch mehr bevorzugt enthalten Metallsalze Salze von Metallen ausgewählt aus der Gruppe bestehend aus Mn, Ag, Zn, Sn, Fe, Cu, Al, Ni, Co, Ti, Zr, Cr, La und Kombinationen davon. Am meisten bevorzugt enthalten die Metallsalze Salze von Metallen ausgewählt aus der Gruppe bestehend aus Cu, Fe, Sn und Kombinationen davon.
- Genauer enthalten die Metallsalze, ohne darauf beschränkt zu sein, dermatologisch unbedenkliche Metallchelate und Salze, wie Bishistidinkomplexe, Bromide, Chondroitinsulfat, Chromite, Cyanide, Dipiocolinate, Ethylhexanoate, Glycerolatkomplex, Methoxide, Polyphosphonate, para-Phenolsulfonate, Perchlorate, Phenolsulfonate, Selenide, Stearate, Thiocyanate, Tripolyphosphate, Wolframate, Phosphate, Carbonate, para-Aminobenzoat, para-Dimethylaminobenzoate, Hydroxide, para-Methoxycinnamat, Naphthenate, Stearate, Caprate, Laurate, Myristate, Palmitate, Oleate, Picolinate, Pyrithione, Fluoride, Aspartate, Gluconate, Iodide, Oxide, Nitrite, Nitrate, Phosphate, Pyrophosphate, Sulfide, Mercaptopyridinoxide (z. B. - Mehr bevorzugt sind die Metallsalze ausgewählt aus der Gruppe, bestehend aus Phosphaten, Carbonaten, para-Aminobenzoat, para-Dimethylaminobenzoaten, Hydroxiden, para-Methoxycinnamat, Naphthenaten, Stearaten, Capraten, Lauraten, Myristaten, Palmitaten, Oleaten, Picolinaten, Pyrithionen, Fluoriden, Aspartaten, Gluconaten, Iodiden, Oxiden, Nitriten, Nitraten, Phosphaten, Pyrophosphaten, Sulfiden, Mercaptopyridinoxiden (z. B. Zinkpyrithion), Nicotinaten und Nicotinamiden, Hinokitiol, Acetaten, Ascorbaten, Chloriden, Benzoaten, Citraten, Fumaraten, Gluconaten, Glutaraten, Lactaten, Malaten, Maconaten, Salicylaten, Succinaten, Sulfaten, Undecylaten und Kombinationen davon.
- Noch mehr bevorzugt sind die Metallsalze ausgewählt aus der Gruppe, bestehend aus Fluoriden, Aspartaten, Gluconaten, Iodiden, Oxiden, Nitriten, Nitraten, Phosphaten, Pyrophosphaten, Sulfiden, Mercaptopyridinoxiden (z. B. Zinkpyrithion), Nicotinaten und Nicotinamiden, Hinokitiol, Acetaten, Ascorbaten, Chloriden, Benzoaten, Citraten, Fumaraten, Gluconaten, Glutaraten, Lactaten, Malaten, Malonaten, Salicylaten, Succinaten, Sulfaten, Undecylaten und Kombinationen davon.
- Noch mehr bevorzugt sind die Metallsalze und Komplexe: Acetate, Ascorbate, Chloride, Benzoate, Citrate, Fumarate, Gluconate, Glutarate, Lactate, Malate, Malonate, Salicylate, Succinate, Sulfate, Undecylate und Kombinationen davon.
- Am meisten bevorzugt sind die Metallsalze ausgewählt aus der Gruppe, bestehend aus Kupferpidolat, L-FER-Pidolat, Kupfer(I)sulfat, Eisen(III)chlorid, Kupfer(I)chlorid, Eisen(III)sulfat und Kombinationen davon.
- Ohne an eine Theorie gebunden zu sein, wird angenommen, dass das Benzoesäure-Analogon und das Metallsalz unter Bildung eines Metall-Säure-Komplexes komplexieren, der, wie sich herausgestellt hat, Oberflächen, auf die derartige Zusammensetzungen aufgebracht werden, eine unmittelbare und anhaltende antivirale und antibakterielle Wirksamkeit verleiht.
- In den Zusammensetzungen liegt das Metallsalz in einer Menge vor, dass das Metallion zum Schluss vorzugsweise 0,001 Gew.-% bis 20 Gew.-%, bezogen auf die Zusammensetzung, mehr bevorzugt 0,01 Gew.-% bis 10 Gew.-%, noch mehr bevorzugt 0,05 Gew.-% bis 5 Gew.-% und am meisten bevorzugt 0,05 Gew.-% bis 2 Gew.-% ausmacht.
- Als Alternative können das Benzoesäure-Analogon und das Metallsalz vor der Herstellung der erfindungsgemäßen Zusammensetzungen komplexiert werden, wobei sie einen Benzoesäure-Analogon-Metall-Komplex bilden. In diesem Fall liegt der Komplex vorzugsweise in einer Menge von 0,001 Gew.-% bis 20 Gew.-%, bezogen auf die Zusammensetzung, mehr bevorzugt von 0,01 Gew.-% bis 10 Gew.-% und am meisten bevorzugt von 0,1 Gew.-% bis 5 Gew.-% vor.
- Dermatologisch unbedenklicher Träger
- Die Zusammensetzungen umfassen einen dermatologisch unbedenklichen Träger für das Benzoesäure-Analogon und das Metallsalz. Der Ausdruck „dermatologisch unbedenklicher Träger", wie hier verwendet, bedeutet, dass der Träger für das In-Kontakt-Bringen mit oder die topische Anwendung auf Keratingewebe von Säugetieren (z. B. menschlichen Hände) geeignet ist, über gute ästhetisch Eigenschaften verfügt, mit den Wirkstoffen der vorliegenden Erfindung und jedem anderen Bestandteil kompatibel ist und keine unerwünschten Sicherheits- oder Toxizitätsbedenken verursacht. Eine sichere und wirksame Menge an Träger macht 50% bis 99,99%, vorzugsweise 80% bis 99,9%, mehr bevorzugt 90% bis 98% und am meisten bevorzugt 90% bis 95% der Zusammensetzung aus.
- Außerdem können die Zusammensetzungen in verschiedenen Produktformen für die intranasale Verwendung, einschließlich, aber nicht beschränkt auf, intranasaler Sprays, verwendet werden.
- Der erfindungsgemäße Träger ist eine wässrige Lösung. Eine derartige wässrige Lösung kann bis zu 98,8 Gew.-%, bezogen auf die Zusammensetzung, an Wasser umfassen.
- Zur Ausbildung eines Gels können vorzugsweise Verdickungsmittel zu den erfindungsgemäßen Lösungen gegeben werden. Zu Beispielen für geeignete Verdickungsmittel gehören, ohne darauf beschränkt zu sein, natürlich vorkommende Polymermaterialien, wie Natriumalginat, Xanthangummi, Quittensaatextrakt, Tragantgummi, Stärke und dergleichen, halb synthetische Polymermaterialien, wie Celluloseether (z. B. Hydroxyethylcellulose, Methylcellulose, Carboxymethylcellulose, Hydroxypropylmethylcellulose), Polyvinylpyrrolidon, Polyvinylalkohol, Guargummi, Hydroxypropylguargummi, lösliche Stärke, kationische Cellulosen, kationische Guare und dergleichen und synthetische Polymermaterialien, wie Carboxyvinylpolymere, Polyvinylpyrrolidon, Polyvinylalkohol, Polyacrylsäurepolymere, Polymethacrylsäurepolymere, Polyvinylacetatpolymere, Po lyvinylchloridpolymere, Polyvinylidenchloridpolymere und dergleichen. Anorganische Verdickungsmittel können ebenfalls verwendet werden, wie Aluminiumsilicate, wie zum Beispiel Bentonite, oder eine Mischung aus Polyethylenglycol und Polyethylenglycolstearat oder -distearat.
- Ferner hierin geeignet sind hydrophile Geliermittel wie die Acrylsäure/Acrylsäureethylester-Copolymere und die Carboxyvinylpolymere, die von der B. F. Goodrich Company unter der Marke „Carbopol Registered TM Resins" erhältlich sind. Diese Harze bestehen im Wesentlichen aus einem gallertartigen, wasserlöslichen, vernetzten Polyalkenylpolyetherpolymer von Acrylsäure, vernetzt mit von 0,75% bis 2,00% Vernetzungsmittel, wie Polyallylsaccharose oder Polyallylpentaerythrit. Beispiele umfassen Carbopol 934, Carbopol 940, Carbopol 950, Carbopol 980, Carbopol 951 und Carbopol 981. Carbopol 934 ist ein wasserlösliches Polymer von Acrylsäure, vernetzt mit etwa 1% Polyallylether von Saccharose mit durchschnittlich etwa 5,8 Allylgruppen für jedes Saccharosemolekül. Ebenfalls zur Verwendung hierin geeignet sind Carbomere, die unter den Handelsbezeichnungen CARBOPOL ULTREZ 10, CARBOPOL ETD2020, CARBOPOL 1382, CARBOPOL 1342, SALCARE SC96 (Polyquaternium-37 und Propylenglycoldicaprylat/-dicaprat und PPG-1 Trideceth-6), STABILEZE QM (Polyvinylmethacrylat/Methacrylsäuredecadien-Kreuzpolymer), STABYLEN 30 (Acrylat/Vinylisodecanoat-Kreuzpolymer) und PEMULEN TR-1 (CTFA-Bezeichnung: Acrylat/10-30-Alkylacrylat-Kreuzpolymer) vertrieben werden. Kombinationen der vorgenannten Polymere sind ebenfalls hierin geeignet. Zu weiteren Geliermitteln, die zur Verwendung hierin geeignet sind, gehören Oleogele, wie Trihydroxystearin und Aluminiummagnesiumhydroxystearat. Ein weiteres nützliches erfindungsgemäßes Verdickungsmittel ist das nichtionische Polymer mit der CTFA-Bezeichnung: Polyacrylamid und Isoparaffin und Laureth-7, erhältlich als SEPI-GEL von Seppic Corporation.
- Hydrophob modifizierte Cellulosen sind ebenfalls für die Verwendung in den Wasser- oder Alkohollösungen und -gelen geeignet. Diese Cellulosen sind in den US-Patenten 4,228,277 und 5,104,646, ausführlich beschrieben.
- Das Verdickungsmittel liegt vorzugsweise in einer Konzentration von 0,01% bis 10%, vorzugsweise von 0,1% bis 5% und am meisten bevorzugt von 0,1% bis 3% vor. Mischungen der vorstehend genannten Verdickungsmittel können gleichfalls verwendet werden.
- Weiter können lipophile Feuchthaltemittel/Weichmacher für die Haut eingearbeitet werden. Zu Beispielen für geeignete lipophile Feuchthaltemittel für die Haut gehören, ohne darauf beschränkt zu sein, Petrolatum, Mineralöl, mikrokristalline Wachse, Polyalkene, Paraffin, Cerasin, Ozokerit, Polyethylen, Perhydrosqualen, Dimethicone, Cyclomethicone, Alkylsiloxane, Polymethylsiloxane, Methylphenylpolysiloxane, hydroxyliertes Milchglycerid, Rizinusöl, Sojaöl, mit Maleinsäure behandeltes Sojaöl, Safloröl, Baumwollsaatöl, Maisöl, Walnussöl, Erdnussöl, Olivenöl, Lebertran, Mandelöl, Avokadoöl, Palmöl, Sesamöl, flüssige Saccharoseoctaester, Gemische aus flüssigen Saccharoseoctaestern und festen Polyolpolyestern, Lanolinöl, Lanolinwachs, Lanolinalkohol, Lanolinfettsäure, Isopropyllanolat, acetyliertes Lanolin, acetylierte Lanolinalkohole, Lanolinalkohollinoleat, Lanolinalkoholriconoleat, Bienenwachs, Bienenwachsderivate, Walrat, Myristylmyristat, Stearylstearat, Carnauba- und Candelillawachse, Cholesterin, Cholesterinfettsäureester und Homologe davon, Lecithin und Derivate, Sphingolipide, Ceramide, Glycosphingolipide und Homologe davon und Mischungen davon. Eine ausführlichere Besprechung nützlicher lipophiler Feuchthaltemittel für die Haut ist US-Patent Nr. 5,716,920, erteilt am 10. Februar 1998 an Glenn, Jr. et al., zu entnehmen.
- Bei Einarbeitung in die Lösungen liegt das lipophile Feuchthaltemittel für die Haut in Konzentrationen von 0,1 Gew.-% bis 20 Gew.-%, vorzugsweise von 1 Gew.-% bis 15 Gew.-%, mehr bevorzugt von 2 Gew.-% bis 10 Gew.-% vor.
- Zur Verwendung in den Lösungen sind ebenfalls emulgierende Tenside mit einem HLB-Wert von weniger als 12 oder etwa 12 bevorzugt, wie Steareth-2, PEG-5-Sojasterolöl, PEG-10-Sojasterolöl, Diethanolamincetylphosphat, Sorbitanmonostearat (SPAN 60), Diethylenglycolmonostearat, Glycerylmonostearat und Mischungen davon; emulgierende Tenside mit einem HLB-Wert von 12 oder mehr, wie Steareth-21, Polyoxyethylensorbitantristearat (TWEEN 65), Polyethylenglycol-20-Sorbitanmonostearat, Polyethylenglycol-60-Sorbitanmonostearat, Polyethylenglycol-80-Sorbitanmonostearat, Steareth-20, Ceteth-20, PEG-100-Stearat, Natriumstearoylsarcosinat, hydriertes Lecithin, Natriumcocoylglycerylsulfat, Natriumstearylsulfat, Natriumstearoyllactylat, PEG-20-Methylglucosidsesquistearat, PEG-20-Glycerylmonostearat, Saccharosemonostearat, Saccharosepolystearate (mit einem hohen Anteil an Saccharosemonostearat), Polyglyceryl-10-Stearat, Polyglyceryl-10-Myristat, Steareth-10, DEA-Oleth-3-Phosphat, DEA-Oleth-10-Phosphat, PPG-5-Ceteth-10-Phosphat-Natriumsalz, PPG-5-Ceteth-10-Phosphat-Kaliumsalz und Mischungen davon; und Mischungen davon. Vorzugsweise umfassen die erfindungsgemäßen Zusammensetzungen wenigstens ein emulgierendes Tensid mit einem HLB-Wert von weniger als 12 und wenigstens ein emulgierendes Tensid mit einem HLB-WERT von 12 oder mehr. „HLB-Wert" ist dem Fachmann ein Begriff und bedeutet Hydrophilie-/Lipophilie-Gleichgewicht. Siehe „The HLB System, A Time-Saving Guide to Emulsifier Selection" ICI Americas Inc., August (1984) und McCutcheon's Detergents and Emulsifiers, North American Edition (1987); in denen mehrere hier nützliche Emulgatoren aufgeführt sind.
- Das emulgierende Tensid ist zu etwa 0% bis 20%, vorzugsweise 0,1% bis 10%, mehr bevorzugt 0,25% bis 5%, am meisten bevorzugt 0,25% bis 2,5% enthalten.
- Geeignete Träger können auch eine wasserhaltige (d. H. nicht auf der Grundlage von Alkohol) Emulsion umfassen, wie Öl-in-Wasser-Emulsionen, Wasser-in-Öl-Emulsionen und Wasser-in-Silikon-Emulsionen. Wie der Fachmann weiß, verteilt sich eine bestimmte Komponente in erster Linie entweder in der Wasser- oder in der Öl/Silikon-Phase, abhängig von der Wasserlöslichkeit/dispergierbarkeit der Komponente in der Zusammensetzung.
- Erfindungsgemäße Emulsionen enthalten generell eine Lösung, wie vorstehend beschrieben, und ein Lipid oder Öl. Lipide können von Tieren, Pflanzen oder Erdöl abgeleitet sein und können natürlich oder synthetisch (d. h. künstlich) sein. Bevorzugte Emulsionen enthalten auch ein Feuchthaltemittel, wie Glycerin. Emulsionen enthalten vorzugsweise ferner von 1 Gew.-% bis 10 Gew.-%, mehr bevorzugt von 2 Gew.-% bis 5 Gew.-%, bezogen auf den Träger, eines Emulgators. Emulgatoren können nicht-ionisch, anionisch oder kationisch sein. Geeignete Emulgatoren sind beispielsweise im US-Patent 3,755,560, erteilt am 28. August 1973 an Dickert et al., im US-Patent 4,421,769, erteilt am 20. Dezember 1983 an Dixon et al., und in McCutcheon's Detergents and Emulsifiers, North American Edition, Seiten 317–324 (1986) offenbart.
- Die Emulsion kann auch ein Schaum unterdrückendes Mittel enthalten, um das Schäumen beim Auftragen auf die zu behandelnde Oberfläche zu minimieren. Schaum unterdrückende Mittel schließen hochmolekulare Silikone und andere Materialien ein, die in der Technik für diesen Zweck bekannt sind.
- Geeignete Emulsionen können einen breiten Viskositätsbereich aufweisen, abhängig von der gewünschten Produktform. Beispielhafte niedrig viskose Emulsionen, die bevorzugt sind, haben eine Viskosität von ungefähr 50 mm2s–1 (Centistoke) oder weniger, mehr bevorzugt 10 mm2s–1 (Centistoke) oder weniger, am meisten bevorzugt 5 mm2s–1 (Centistoke) oder weniger.
- pH
- Die erfindungsgemäß nützlichen antimikrobiellen Zusammensetzungen weisen einen pH-Wert von 1,5 bis 5 auf. In der am meisten bevorzugten Ausführungsform beträgt der pH-Wert der Zusammensetzungen 2 bis 4.
- Ohne an eine Theorie gebunden zu sein, wird angenommen, dass eine derartige saure Umgebung die virale Kapsidhülle protoniert, was eine Konformationsänderung einleitet, die das Virus irreversibel inaktiviert und damit das Virus unfähig macht, eine Infektion einzuleiten. Diese Wirkung tritt in Synergie mit dem Metallsalz und der Säurestruktur auf, um die gewünschte unmittelbare und anhaltende antivirale und antibakterielle Wirksamkeit zu erzeugen, die der Schlüssel der vorliegenden Zusammensetzungen ist.
- Fakultative Bestandteile
- Die Zusammensetzungen können eine Vielfalt von weiteren Bestandteilen enthalten, wie sie herkömmlich in einem beliebigen Produkttyp verwendet werden, mit der Maßgabe, dass diese die Vorteile der Erfindung nicht maßgeblich verändern.
- Eine andere Klasse antimikrobieller Wirkstoffe (insbesondere antibakterieller Wirkstoffe), die in der vorliegenden Erfindung nützlich sind, sind die so genannten „natürlichen" antibakteriellen Wirkstoffe, bezeichnet als natürliche ätherische Öle. Diese Wirkstoffe haben ihre Namen von ihrem natürlichen Vorkommen in Pflanzen. Typische antibakterielle Wirkstoffe aus natürlichen ätherischen Ölen umfassen Öle aus Anis, Zitrone, Orange, Rosmarin, Moosbeere, Thymian, Lavendel, Gewürznelken, Hopfen, Teebaum, Citronella, Weizen, Gerste, Zitronengras, Thuja, Zedernholz, Zimt, Flohsegge, Geranie, Sandelholz, Veilchen, Preiselbeere, Eukalyptus, Eisenkraut, Pfefferminze, Benzoeharz, Basilikum, Fenchel, Tanne, Balsambaum, Menthol, Ocmea Origanum, Hydastis Carradensis, Berberidaceae Daceae, Ratanhiae und Curcuma longa. In diese Klasse der natürlichen, ätherischen Öle eingeschlossen sind auch die wichtigsten chemischen Bestandteile der Pflanzenöle, von denen festgestellt wurde, dass sie den antimikrobiellen Vorzug bieten. Zu diesen Chemikalien gehören, ohne darauf beschränkt zu sein, Anethol, Catechin, Camphen, Thymol, Eugenol, Eucalyptol, Ferulasäure, Farnesol, Hinokitiol, Tropolon, Limonen, Menthol, Methylsalicylat, Carvacol, Terpineol, Verbenon, Berberin, Ratanhiaeextrakt, Caryophyllenoxid, Citronellasäure, Curcumin, Nerolidol und Geraniol.
- Antioxidationsmittel/Radikalfänger
- Die Zusammensetzungen können eine sichere und wirksame Menge an Antioxidationsmittel/Radikalfänger enthalten. Das Antioxidationsmittel/der Radikalfänger ist besonders geeignet zur Bereitstellung eines Schutzes gegen UV-Strahlung, die eine verstärkte Abschilferung oder Strukturveränderungen in der Hornhaut bewirken kann, und gegen andere Umwelteinflüsse, die eine Hautschädigung bewirken können.
- Eine sichere und wirksame Menge eines Antioxidationsmittels/Radikalfängers kann zu den Zusammensetzungen der beanspruchten Erfindung gegeben werden, vorzugsweise von 0,1% bis 10%, mehr bevorzugt von 1% bis 5%, bezogen auf die Zusammensetzung.
- Antioxidationsmittel/Radikalfänger wie Ascorbinsäure (Vitamin C) und deren Salze, Ascorbylester von Fettsäuren, Ascorbinsäurederivate (z. B. Magnesiumascorbylphosphat), Tocopherol (Vitamin E), Tocopherolsorbat, Tocopherolacetat, andere Ester von Tocopherol, butylierte Hydroxybenzoesäuren und deren Salze, 6-Hydroxy-2,5,7,8-tetramethylchroman-2-carbonsäure (unter der Handelsbezeichnung Trolox® im Handel erhältlich), Gallussäure und deren Alkylester, insbesondere Propylgallat, Harnsäure und deren Salze und Alkylester, Sorbinsäure und deren Salze, Liponsäure, Amine (z. B. N,N-Diethylhydroxylamin, Aminoguanidin), Sulfhydrylverbindungen (z. B. Glutathion), Dihydroxyfumarsäure und deren Salze, Lycinpidolat, Argininpilolat, Nordihydroguajaretsäure, Bioflavonoide, Lysin, Methionin, Prolin, Superoxiddismutase, Silymarin, Teeextrakte, Traubenhaut-/-kernextrakte, Melanin und Rosmarinextrakte können verwendet werden. Bevorzugte Antioxidationsmittel/Radikalfänger werden aus Tocopherolsorbat und anderen Estern von Tocopherol ausgewählt, mehr bevorzugt Tocopherolsorbat. Die Verwendung von Tocopherolsorbat in topischen Zusammensetzungen und auf die vorliegende Erfindung anwendbar ist beispielsweise in US-Patent Nr. 4,847,071, erteilt am 11. Juli 1989 an Donald L. Bissett, Rodney D. Bush und Ranjit Chatterjee, beschrieben.
- Chelatoren
- Die Zusammensetzungen können auch eine sichere und wirksame Menge eines Komplexbildners oder Chelatbildners umfassen, sofern dieser nicht die Wirkung des Benzoesäure-Analogons und des Metallsalzes stört. Wie hierin verwendet, bedeutet „Chelator" oder „Chelatisierungsmittel" einen Wirkstoff, der in der Lage ist, ein Metallion aus einem System zu entfernen, und zwar durch Bilden eines Komplexes, sodass das Metallion nicht ohne Weiteres an chemischen Reaktionen teilnehmen oder diese katalysieren kann. Das Einbinden eines Komplexbildners ist besonders geeignet zur Bereitstellung eines Schutzes gegen UV-Strahlung, die zu einem übermäßigen Abschilfern oder zu Strukturveränderungen der Haut führen kann, und gegen andere Umwelteinflüsse, die eine Hautschädigung bewirken können.
- Eine sichere und wirksame Menge eines Komplexbildners kann zu den Zusammensetzungen der beanspruchten Erfindung gegeben werden, vorzugsweise von 0,1% bis 10%, mehr bevorzugt von 1% bis 5%, bezogen auf die Zusammensetzung. Beispiele für Komplexbildner, die hierin geeignet sind, sind im US-Patent Nr. 5,487,884, erteilt am 30.1.96 an Bissett et al.; in der internationalen Veröffentlichung Nr. 91/16035, Bush et al., veröffentlicht am 31.10.95; und in der internationalen Veröffentlichung Nr. 91/16034, Bush et al., veröffentlicht am 31.10.95, offenbart. Bevorzugte Komplexbildner, die in den Zusammensetzungen der beanspruchten Erfindung geeignet sind, sind Furildioxim und Derivate davon.
- Die Hautempfindung betreffende Mittel
- Die antimikrobiellen Zusammensetzungen können ferner die Hautempfindung betreffende Mittel enthalten. In der vorliegenden Erfindung können die Hautempfindung betreffende Mittel in einer Konzentration von 0,01% bis 10%, typisch von 0,1% bis 5% und vorzugsweise von 0,2% bis 1% vorliegen. Die Konzentration wird derart gewählt, dass das gewünschte Ausmaß der vom Verbraucher wahrgenommenen Empfindung erreicht wird und nach Bedarf verändert werden kann. Zu geeigneten die Hautempfindung betreffenden Technologien gehören Menthol, Eucalyptus, 3-1-Menthoxypropan-1,2-diol, N-substituierte p-Menthan-3-carboxamide und acyclische Carboxamide.
- Verfahren zur Verwendung der antimikrobiellen Zusammensetzungen
- Die antimikrobiellen Zusammensetzungen sind zur Verhinderung und/oder Behandlung einer Erkältung oder einer damit verbundenen Atemwegserkrankung bei einem Säugetier geeignet, falls diese von einem Rhinovirus verursacht wird. Die Verhinderung und/oder Behandlung umfasst den Schritt des topischen Aufbringens der Zusammensetzungen in die Nasenwege.
- BEISPIELE
- Die folgenden Beispiele beschreiben und zeigen weiter Ausführungsformen im Rahmen der vorliegenden Erfindung.
- Beispiele 1 und 2
- Durch Vereinigen der folgenden Bestandteile unter Verwendung herkömmlicher Mischtechniken, die den nachstehend Beschriebenen ähnlich sind, wird eine intranasale Zubereitung hergestellt.
- Die vorstehend aufgeführten Zutaten werden nacheinander unter Rühren in ein Gefäß passender Größe in Wasser eingebracht, wobei sich eine erst vollständig löst, bevor die nächste zugegeben wird. Nach Zugabe aller Zutaten wird die Charge mit gereinigtem Wasser auf das passende Gewicht gebracht. Die Lösung wird in einen flexiblen Verbundstoffbehälter gegeben und der Behälter in eine elektrostatische Sprüheinrichtung eingepasst. Das Nasenstück der Einrichtung wird an das Nasenloch gehalten und die Einrichtung so geführt, dass der Spraykopf in das Nasenloch eingeführt wird. Die abgegebene Flüssigkeit stellt eine unmittelbare und anhaltende antirhinovirale Wirksamkeit in der Nase bereit und lindert die Symptome einer Erkältung.
- Andere Zusammensetzungen werden auf die vorstehend beschriebene Weise unter Verwendung von NiSO4, SnCl2 oder Silbernitrat anstatt des Kupfersalzes hergestellt und intranasal wie vorstehend beschrieben verwendet.
Claims (6)
- Verwendung eines Benzoesäure-Analogons, ausgewählt aus Benzoesäure, Salicylsäure, 2-Nitrobenzoesäure, 2,6-Dihydroxybenzoesäure, 2-Hydroxybenzoesäure, 5-Nitrosalicylsäure, 5-Bromsalicylsäure, 5-Iodsalicylsäure, 5-Fluorsalicylsäure, 3-Chlorsalicylsäure, 4-Chlorsalicylsäure, 5-Chlorsalicylsäure, Phthalsäure und Kombinationen davon, zur Herstellung eines intranasalen Medikaments zur Verhinderung und/oder Behandlung einer Erkältung oder einer damit verbundenen Atemwegserkrankung, die vom Rhinovirus bei einem Säugetier verursacht wird; wobei a) das Medikament 0,01% bis 5% des Benzoesäure-Analogons umfasst; b) das Medikament in Form einer wässrigen Lösung mit einem pH-Wert von 1,5 bis 5 vorliegt; c) das Medikament ferner umfasst i) eine sichere und wirksame Menge eines Metallsalzes, wobei das Metallsalz ein Metall ausgewählt aus Mn, Ag, Zn, Sn, Fe, Cu, Al, Ni, Co, Ti, Zr, Cr, La, Bi, K, Cd, Yb, Dy, Nd, Ce, Tl, Pr und Kombinationen davon umfasst; ii) einen dermatologisch unbedenklichen Träger für das Benzoesäure-Analogon und das Metallsalz; d) die Zusammensetzung im Wesentlichen frei von p-Aminosalicylsäure ist.
- Verwendung nach Anspruch 1, wobei das Benzoesäure-Analogon und das Metallsalz in Form eines Benzoesäure-Analogon-Metall-Komplexes vorliegen.
- Verwendung nach Anspruch 1, wobei das Metallsalz Zink umfasst.
- Verwendung nach Anspruch 1, wobei das Medikament 0,001 Gew.-% bis 20 Gew.-%, vorzugsweise 0,05 Gew.-% bis 5 Gew.-%, bezogen auf die Zusammensetzung, des Metallions umfasst.
- Verwendung nach Anspruch 1, wobei das Benzoesäure-Analogon ausgewählt ist aus Benzoesäure, Salicylsäure und Kombinationen davon.
- Verwendung nach Anspruch 1, wobei die Zusammensetzung ferner ein emulgierendes Tensid mit einem HLB-Wert von 12 oder mehr umfasst.
