- Dosage regimens can be adjusted to provide the optimum therapeutic response. For example, several divided doses can be administered daily or the dose can be proportionally reduced as indicated by the exigencies of the therapeutic situation.
- Colonies were lifted onto Nytran filters and hybridized with 32 P- labeled oligonucleotides with the sequence 5'-CAGCTATGGTGGTGCCGACTACAA- 3' (SEQ ID NO:7), 5'-AGGTGCTAGGGGACAGTGTTAGACA-3' (SEQ ID NO:8) , and 5'-TCGCTTGTAGTCGGCACCACCATA-3' (SEQ ID NO:9). All oligos are from AA292201 sequence. Final wash conditions were 2 X SSC, 0.1 % SDS at 55° for 20 min. The two hybridizing colonies were picked and the sequence of the cDNA inserts was determined.
- the B7-4M form has approximately 21% amino acid identity to human B7-1 and about 20% amino acid identity to human B7-2 as calculated using the default Blosum62 matrix with gap penalties set at existence 11 and extension 1 (See http://www.ncbi.nlm.nih.gov), under conditions where B7-1 and B7-2 have about 26% identity.

- a minor 1.8 kb B7-4 mRNA species was also observed in interferon- ⁇ treated monocytes.
- the chromosomal localization of the B7-4 genes was determined using a monochromosomal blot kit commercially available from Quantum (Toronto, Canada). The blots were probed with a sequence that recognizes both B7-4S and B7-4M. Using this method, the B7-4 polypeptides have been localized to human chromosome 9, whereas B7-1 and B7-2 have been localized to human chromosome 3. The butyrophilins, which also share limited amino acid sequence identity with the B7-4 family have been localized to the major histocompatability complex on chromosome 6. The chromosomal location of B7-4 was confirmed using B7-4 specific primers in PCR amplification of monochromosomal somatic cell hybrid DNA templates available from Quantum Technologies (Canada).
- B7-4 expression was observed, inter alia, on heart tissue, tumor cells (including some lung tumor cells, some ovarian tumor cells, some breast tumor cells, some epithelial tumor cells, and some squamous cell carcinomas), placenta, and thymic epithelium.

- Antibodies having human variable regions which bind to B7-4 or PD-1 are detected by enzyme conjugates specific for human IgM and human IgG sub-classes with no cross reactivity to mouse immunoglobulin. Briefly, PVC microtiter plates are coated with B7-4 or PD-1 by coating wells overnight at 37°C with 5 ⁇ g/mL B7-4 in PBS.

- cells were stained separately with 5 ⁇ g/ml anti-IA d -PE or B7.2-PE (Pharmingen, San Diego, CA). Following each step, cells were washed three times with PBS/1% BSA/0.02% sodium azide. After the final incubation, cells were fixed with 1% paraformaldehyde. Ten thousand events were analyzed on a FACSCalibar (Becton Dickinson, Mountain View, CA). All isotype controls were all obtained from Pharmingen.
