Claims (36)
1. Гетеродимерное антитело, содержащее:
а) первую тяжелую цепь, содержащую:
i) первый вариантный Fc-домен; а также
ii) одноцепочечный Fv-участок (scFv), в котором указанный scFv-участок содержит первый вариабельный домен тяжелой цепи, первый вариабельный домен легкой цепи и заряженный линкер scFv, причем указанный заряженный линкер scFv ковалентно присоединяет упомянутый первый вариабельный домен тяжелой цепи и указанный первый вариабельный домен легкой цепи;
b) вторую тяжелую цепь, содержащую мономер VH-CH1-шарнир-CH2-CH3, в котором VH является вторым вариабельным доменом тяжелой цепи, а CH2-CH3 является вторым вариантным Fc-доменом; и
c) легкую цепь, содержащую второй вариабельный домен легкой цепи и константный домен легкой цепи;
причем указанный второй вариантный Fc-домен содержит аминокислотные замены N208D/Q295E/N384D/Q418E/N421D,
при этом указанные первый и второй вариантный Fc-домены каждый содержат аминокислотные замены E233P/L234V/L235A/G236del/S267K,
причем указанный первый вариантный Fc-домен содержит аминокислотные замены S364K/E357Q и указанный второй вариантный Fc-домен содержит аминокислотные замены L368D/K370S,
причем указанный второй вариабельный домен тяжелой цепи содержит SEQ ID NO: 1071, а указанный второй вариабельный домен легкой цепи содержит SEQ ID NO: 1076, в которых нумерация соответствует индексу EU, как у Кабата.
2. Гетеродимерное антитело по п. 1, оличающееся тем, что указанный scFv связывает CD3.
3. Гетеродимерное антитело по п. 1, оличающееся тем, что указанный первый вариабельный домен тяжелой цепи и указанный первый вариабельный домен легкой цепи выбирают из наборов, содержащих: SEQ ID NO: 1 и SEQ ID NO: 5; SEQ ID NO: 10 и SEQ ID NO: 14; SEQ ID NO: 19 и SEQ ID NO: 23; SEQ ID NO: 28 и SEQ ID NO: 32; SEQ ID NO: 37 и SEQ ID NO: 41; и SEQ ID NO: 46 и SEQ ID NO: 50, соответственно.
4. Гетеродимерное антитело по п. 3, оличающееся тем, что указанный первый вариабельный домен тяжелой цепи содержит SEQ ID NO: 1, и указанный первый вариабельный домен легкой цепи содержит SEQ ID NO: 5.
5. Гетедомерное антитело по п. 1, оличающееся тем, что компонент CH1-шарнир-CH2-CH3 второй тяжелой цепи содержит SEQ ID NO: 1108, указанный первый вариантный Fc-домен содержит SEQ ID NO: 1109, и указанный константный домен легкой цепи содержит SEQ ID NO: 1110.
6. Гетеродимерное антитело по п. 1, оличающееся тем, что указанная первая тяжелая цепь содержит SEQ ID NO: 1080, указанная вторая тяжелая цепь содержит SEQ ID NO: 1070, а указанная легкая цепь содержит SEQ ID NO: 1075.
7. Композиция нуклеиновых кислот, содержащая:
а) первую нуклеиновую кислоту, кодирующую указанную первую тяжелую цепь по п. 1;
b) вторую нуклеиновую кислоту, кодирующую указанную вторую тяжелую цепь по п. 1; и
с) третью нуклеиновую кислоту, кодирующую указанную легкую цепь по п. 1.
8. Композиция экспрессионного вектора, содержащая:
а) первый экспрессионный вектор, содержащий указанную первую нуклеиновую кислоту по п. 7;
b) второй экспрессионный вектор, содержащий указанную вторую нуклеиновую кислоту по п. 7; и
c) третий экспрессионный вектор, содержащий указанную третью нуклеиновую кислоту по п. 7.
9. Клетка-хозяин, содержащая указанную композицию экспрессионного вектора по п. 8.
10. Способ получения гетеродимерного антитела по п. 1, включающий культивирование указанной клетки-хозяина по п. 9 в условиях, в которых указанное антитело экспрессируется, и выделение указанного антитела.
11. Способ лечения нейроэндокринного рака у субъекта, нуждающегося в этом, включающий введение указанному субъекту гетеродимерного антитела по п. 1.
12. Композиция, содержащая домен, связывающий рецептор соматостатина 2-го типа (SSTR2), указанный SSTR2-связывающий домен, содержащий вариабельный домен тяжелой цепи, содержащий SEQ ID NO: 958 и вариабельный домен легкой цепи, содержащий SEQ ID NO: 962.
13. Композиция нуклеиновых кислот, содержащая:
а) первую нуклеиновую кислоту, кодирующую указанный вариабельный домен тяжелой цепи по п. 12; и
b) вторую нуклеиновую кислоту, кодирующую указанный вариабельный домен легкой цепи по п. 12.
14. Композиция экспрессионного вектора, содержащая:
а) первый экспрессионный вектор, содержащий указанную первую нуклеиновую кислоту по п. 13; и
b) второй экспрессионный вектор, содержащий указанную вторую нуклеиновую кислоту по п. 13.
15. Клетка-хозяин, содержащая указанную композицию экспрессирующего вектора по п. 14.
16. Способ получения композиции по п. 12, включающий культивирование указанной клетки-хозяина по п. 15 в условиях, в которых указанное антитело экспрессируется, и выделение указанного антитела.
17. Способ лечения нейроэндокринного рака у субъекта, нуждающегося в этом, включающий введение указанному субъекту композиции по п. 12.
