Abstract

Oppfinnelsen vedrører en fremgangsmåte for fremstillingen av racemisk citalopram fri base eller et syreaddisjonssalt derav og/eller R- eller S-citalopram som den frie base eller et syreaddisjonssalt derav ved separasjon av en blanding av R- og S-citalopram med mer enn 50 % av en av enantiomerene til en fraksjon bestående av racemisk citalopram og/eller en fraksjon av S-citalopram eller R-citalopramat i) citalopram presipiteres fra et løsningsmiddel som den frie base eller som et syreaddisjonssalt derav; ii) det dannede presipitat separeres fra moderluten; iia) hvis presipitatet er krystallinsk, omkrystalliseres det eventuelt en eller flere ganger for å danne racemisk citalopram, og omdannes deretter eventuelt til et syreaddisjonssalt derav; iib) hvis presipitatet ikke er krystallinsk, gjentas eventuelt trinn i) og ii) inntil et krystallinsk presipitat oppnås og det krystallinske presipitat omkrystalliseres en eller flere ganger for å danne racemisk citalopram, og omdannes deretter eventuelt til et syreaddisjonssalt derav; iii) moderluten underkastes eventuelt ytterligere rensing og S-citalopram eller R- citalopram isoleres fra moderluten og omdannes eventuelt til et syreaddisjonssalt derav.
