Abstract
La presente invención está relacionada a composiciones y métodos para generar una célula T modificada con un ácido nucleico capaz de regular a la baja la expresión genética endógena de un gen seleccionado del grupo que consiste en una cadena TCR a, una cadena TCR ß, beta-2 microglobulina, una molécula HLA, CTLA-4, PD1, y FAS y adicionalmente comprende un ácido nucleico que codifica para un receptor de células T modificado (TCR) que comprende afinidad para un antígeno de superficie sobre una célula objetivo o un ácido nucleico electroporado que codifica para un receptor de antígeno quimérico (CAR); también incluidos están métodos y composiciones farmacéuticas que comprenden la célula T modificada para terapia adoptiva y tratar una condición, tal como una enfermedad autoinmune.
