Claims (14)
1. Genetski modifikovani mis koji eksprimira varijabilne domene teskog i lakog lanca coveka, pri cemu su varijabilni regioni lakog lanca coveka eksprimirani iz jedne rearanzirane sekvence gena varigbilnog regiona (VL/JL) lakog lanca imunoglobulina coveka koja je prisutan u germitivnoj liniji celija misa koja obuhvata sekvencu germitivne linje segmenta VL gena i sekvencu germitivne linije segmenta JL gena, gde varijabilni region lakog lanca imunoglobulina coveka je rearanziran VK1-39/JK, pri cemu: (a) mis bez lokusa endogenog varijabilnog regiona lakog lanca K imunoglobulina misa koji moze da rearanzira i formira gen koji kodira varijabilni region K misa; (b) mis eksprimira zajednicki laki lanac imunoglobulina coveka za veci broj teskih lanaca coveka, gde je zajednicki laki lanac izveden iz genske sekvence varijabilnog regiona lakog lanca imunoglobulina coveka; (c) mis sadr:Zi B celiju koja eksprimira antitelo koje se speficno vezuje za antigen od interesa, pri cemu antitelo sadrzi dati zajednicki laki lanac i teski lanac imunoglobulina izveden i rearanziranog varijabilnog genskog segmenta teskog lanca (VH) imunoglobulina coveka odabranog iz grupe koju cine segment VH2-5, VH3-23, VH3-30, VH4-59 i VH5-51 gena; i (d) 90-100% endogenih nerearanziranih genskih segmenata VH misa je zamenjeno vecim brojem nerearanziranih genskih segmenta VH coveka.
2. Genetski modifikovan mis iz zahteva 1, naznacen time, sto sadr:Zi zamenu od 90 do 100% nerearanziranih endogenih VH genskih segmenata misa sa: (i) najmanje 12 funkcionalnih nerearanziranih VH genskih segmenata coveka; (ii) najmanje 18 nerearanziranih VH genskih segmenata coveka; (iii) najmanje 39 nerearanziranih VH genskih segmenata coveka; (iv) najmanje 80 nerearanziranih VH segmenata coveka; iii (v) najmanje 81 nerearanziran VH segment coveka.
3. Genetski modifikovan mis prema jednom od prethodnih zahteva , nazncen time , sto mis eksprimira : (a) tezak lanac koji sadrzi varijabilni domen imunoglobulina coveka konstantan region imunoglobolina misa ; i/ili (b) antitelo koje sadrii laki lanac koji sadrii varijabilni domen lakog lanaca imunoglobulina coveka i konstantan dome lakog lanca imunoglobulina misa i tezak lanac koji sadrii varijabilni domen teskog lanca imunoglobulina coveka i konstantan domen teskog lanca imunoglobulina coveka .
4. Genetski modifikovan mis prema jednom od prethodnih zahteva , naznacentime , sto je genski segment V L coveka operablno vezan za promotor sekvencuimunoglobulina .
5. Genetski modifikovani mis iz zahteva 4, naznacen time , sto promotorsekvenca imunoglobulina je promotor sekvenca imunoglobulina coveka .
6. Postupak dobijanja antitela koje vezuje antigen od interesa , a koji obuhvataimunizaciju misa prema jednom od zahteva 1 do 5 , antigenom od interesa i primenusekvence gena varijabilnog regiona humanog imunoglobulina iz misa za dobijanjeantitela .
7. Postupak prema zahtevu 6 koji obuhvata : ekspresiju u jednoj celiji : (a) prvesekvence gena varijabilnog regiona teskog lanca misa prema jednom od zahteva 1do 5 koj i je imunizovan , pri cemu je sekvenca gena varijabilnog regionaimunoglobulina coveka fuzionisana sa sekvencom gena konstantnog reg iona teskoglanca imunoglobulina coveka ; i (b) sekvence gena varijabilnog regiona lakog lancahumanog imunoglobulina misa prema jednom od zahteva 1 do 5 koji je imunizovan ,pri cemu je sekvenca gena varijabilnog regiona lako lanca imunoglobulina covekafuzionisana sa sekvencom gena konstantnog regiona lakog lanca imunoglobulinacoveka ; odriavanje celije pod uslovima dovoljnim za ekspresiju potpuno humanogantitela ; i izolovanje antitela.
8. Postupak prema zahtevu 7, naznacen time , sto celija sadrii drugu genskusekvencu varijabilnog regiona teskog humanog imunoglobulina misa prema jednomod zahteva 1 do 5 koji je imunizovan, pri cemu je druga genska sekvenca teskogvarijabilnog regiona imunoglobulina coveka fuzionisana sa genskom sekvencomkonstantnog regiona teskog lanca imunoglobulina coveka, pri cemu prva genskasekvenca varijabilnog regiona teskog lanca kodira varijabilni region teskog lancahumanog imunoglobulina koji prepoznaje prvi epitop i druga genska sekvencavarijabilnog regiona teskog lanca imunoglobulin coveka kodira varijabilni domenteskog lanca imunoglobulina coveka koji prepoznaje drugi epitop i pri cemu prviepitop drugi epitop nisu identicni.
9. Upotreba misa prema jednom od zahteva 1 do 5 za dobijanje antitela.
10. Upotreba prema zahtevu 9, gde antitelo je humano bispecificno antitelo.
11 . Postupak za dobijanje humanog bispecificnog antitela, koji obuhvatadobijanje genske sekvence varijabilnog regiona imunoglobulina coveka iz B celijamisa prema jednom od zahteva 1 do 5 i dobijanje antitela sa datim sekvencama.
12. Upotreba prema zahtevu 12, gde celija sadrzi:(a) gensku sekvencu varijabilnog regiona lakog lanca imunoglobulina covekakoja kodira varijabilni dome lakog lanca imunoglobulina coveka izvedan izsegmenta VK1-39JK gena coveka, gde je sekvenca gena za varijabilni regionlakog lanca imunoglobulina coveka je fuzionisan , direktno iii preko linkera, zagensku sekvencu konstantnog regiona lakog lanca imunoglobulina coveka, stoza rezultat daje gen koji kodira laki lanac humanog imunoglobulina;(b) prvu gensku sekvencu za varijabilni region lakog lanca imunoglobulinacoveka koja kodira prvi varijabilni domen teskog lanca imunoglobulina coveka,pri cemu je prva genska sekvenca za varijabilni region teskog lancaimunoglobulina coveka fuzionisana, direktno iii preko linkera, za genskusekvencu konstantnog regiona teskog lanca imunoglobulina coveka, pri cemunastaje gen koji kodira prvi tezak lanac imunoglobulina coveka; pri cemu prvivarijabilni domen teskog lanca prepoznaje prvi epitop; i(c) drugu gensku sekvencu za varijabilni region teskog lanca imunoglobulinacoveka koja kodira drugi varijabilni dimen teskog lanca imunoglobulina coveka,pri cemu je druga genska sekvenca za varijabilni region teskog lancaimunoglobulina coveka fuzionisana, direktno iii preko linkera, za genskusekvencu varijabilnog regiona teskog lanca imunoglobulina coveka, dajuci takagen koji kodira drugi tezak lanac imunoglobulina coveka; pri cemu drugivarjabilni domen teskog lanca humanog imunoglobulina prepoznaje drugiepitop i specificno ne prepoznaje prvi epitop,pri cemu i prvi i drugi tezak lanac imunoglobulina su povezani sa lakim lancem iz (a)imunoglobulina coveka.
14. Upotreba misa prema jednom od zahteva 1 to 5 za identifikaciju varijabilnogdomena teskog lanca imunoglobulina coveka prisutnog u teskom lancuimunoglobulina koji je povezan sa jednim lakim lancem.
