Claims (24)
1. Miš obuhvaća u svojoj zametnoj lozi prvi varijabilni segment gena nepresloženog ljudskog kappa lakog lanca (Vκ) i povezujući segment gena nepresloženog ljudskog kappa lakog lanca (Jκ) operabilno povezan s endogenom mišjom konstantnom regijom teškog lanca (CH) na endogenom lokusu mišjeg teškog lanca, pri čemu prvi nepresloženi ljudski Vκ segment gena i nepresloženi ljudski Jκ segment gena zamjenjuju sve funkcionalne endogene mišje varijabilne (VH) segmente gena teškog lanca, sve funkcionalne endogene mišje raznovrsne (DH) segmente gena i sve funkcionalne endogene mišje povezujuće (JH) segmente gena, pri čemu prvi nepresloženi ljudski Vκ segment gena i nepresloženi ljudski Jκ segment gena sudjeluju u preslaganju kako bi tvorili presloženi Vκ/Jκ slijed operabilno povezan s endogenom mišjom konstantnom regijom teškog lanca u miša, i pri čemu miš nadalje sadrži u svojoj zametnoj lozi drugi varijabilni segment gena ljudskog lakog lanca (VL) i segment gena ljudskog lakog lanca J (JL) operabilno povezan s genom mišjeg konstantnog lakog lanca (CL).
2. Miš iz patentnog zahtjeva 1, obuhvaća B stanicu koja sadrži u svojem genomu presloženi slijed nukleinske kiseline varijabilne regije kappa lakog lanca ljudskog imunoglobulina operabilno povezan s endogenom mišjom konstantnom regijom teškog lanca na endogenom lokusu teškog lanca mišjeg imunoglobulina.
3. Miš iz patentnog zahtjeva 1, pri čemu je drugi VL segment ljudskog gena segment ljudskog Vκ gena.
4. Miš iz patentnog zahtjeva 3, pri čemu je konstantni gen lakog lanca konstantni gen κ lakog lanca.
5. Miš iz patentnog zahtjeva 1, pri čemu je drugi VL segment ljudskog gena segment ljudskog Vλ gena.
6. Miš iz patentnog zahtjeva 5, pri čemu je konstantni gen lakog lanca konstantni gen λ lakog lanca.
7. Miš iz patentnog zahtjeva 1 koji eksprimira antigen-vezujući protein, pri čemu antigen-vezujući protein obuhvaća prvu varijabilnu domenu ljudskog kappa lakog lanca spojenu s konstantnom regijom teškog lanca mišjeg imunoglobulina, i drugi polipeptid koji obuhvaća drugu varijabilnu domenu ljudskog lakog lanca spojenu s konstantnom regijom lakog lanca mišjeg imunoglobulina.
8. Miš iz patentnog zahtjeva 7, pri čemu je druga varijabilna domena ljudskog lakog lanca odabrana između ljudske Vκ varijabilne domene i ljudske Vλ varijabilne domene.
9. Miš iz patentnog zahtjeva 1, koji je homozigot ili heterozigot za navedeni endogeni lokus teškog lanca koji obuhvaća prvi nepresloženi segment ljudskog Vκ gena i nepresloženi segment ljudskog Jκ gena operabilno povezan s endogenom konstantnom regijom lokusa mišjeg teškog lanca, pri čemu prvi nepresloženi segment ljudskog Vκ gena i nepresloženi segment ljudskog Jκ gena zamjenjuju sve funkcionalne endogene segmente gena mišjeg VH, DH i JH.
10. Uporaba miša u skladu s bilo kojim od prethodnih patentnih zahtjeva za proizvodnju antigen vezujućeg proteina koji obuhvaća prvi polipeptid koji sadrži prvu varijabilnu domenu ljudskog kappa lakog lanca spojenu s konstantnom regijom teškog lanca mišjeg imunoglobulina, i drugi polipeptid koji sadrži drugu varijabilnu domenu ljudskog lakog lanca spojenu s konstantnom regijom lakog lanca mišjeg imunoglobulina.

Uporaba miša u skladu s bilo kojim od patentnih zahtjeva 1-9 za proizvodnju protutijela koje sadrži prvi polipeptid koji sadrži prvu varijabilnu domenu ljudskog kappa lakog lanca spojenu s konstantnom regijom teškog lanca mišjeg imunoglbulina, i drugi polipeptid koji sadrži drugu varijabilnu domenu ljudskog lakog lanca spojenu s konstantnom regijom lakog lanca mišjeg imunoglobulina.
12. Uporaba miša u skladu s bilo kojim od patentnih zahtjeva 1-9 za proizvodnju hibridoma ili kvadroma za proizvodnju protutijela kako je definirano u patentnom zahtjevu 11.
13. Uporaba u skladu s patentnim zahtjevom 11, pri čemu je protutijelo bispecifično protutijelo.
14. Mišja stanica obuhvaća u svojem genomu prvi nepresloženi segment ljudskog Vκ gena i nepresloženi segment ljudskog Jκ gena operabilno povezan s endogenom konstantnom regijom teškog lanca na endogenom lokusu mišjeg teškog lanca, pri čemu prvi nepresloženi segment ljudskog gena Vκ i nepresloženi segment ljudskog Jκ gena zamjenjuju sve funkcionalne endogene segmente gena mišjeg VH, DH i JH, i pri čemu mišja stanica nadalje obuhvaća drugi segment ljudskog VL gena i segment ljudskog JL gena operabilno povezan s konstantnim genom mišjeg lakog lanca.
15. Stanica u skladu s patentnim zahtjevom 14 koja je dobivena iz miša u skladu s bilo kojim od patentnih zahtjeva 1 do 9.
16. Stanica u skladu s patentnim zahtjevom 14 ili 15, pri čemu je stanica ES stanica.
17. Tkivo dobiveno iz miša u skladu s bilo kojim od patentnih zahtjeva 1-9, koje sadrži stanice definirane u patentnom zahtjevu 14.

Metoda za izradu genetski modificiranog miša u skladu s bilo kojim od patentnih zahtjeva 1 do 9, obuhvaća zamjenu na endogenom lokusu mišjeg teškog lanca svih funkcionalnih segmenata gena VH, DH i JH miša prvim nepresloženim segmentom ljudskog Vκ gena i nepresloženi segmentom ljudskog Jκ gena, na taj način operabilno povezivanje prvog nepresloženog segmenta ljudskog Vκ gena i nepresloženog segmenta ljudskog Jκ gena s endogenom konstantnom regijom teškog lanca i također umetanje u zametnu liniju miša drugog segmenta gena ljudskog VLi ljudskog segmenta gena JL operabilno povezanog s mišjom konstantnom regijom gena lakog lanca.

Antigen-vezujući protein koji se može dobiti od miša u skladu s bilo kojim od patentnih zahtjeva 1 do 9, pri čemu antigen vezujući protein obuhvaća varijabilnu domenu lakog lanca ljudskog imunoglobulina spojenu s konstantnom domenom mišjeg lakog lanca i varijabilnu domenu kappa lakog lanca ljudskog imunoglobulina spojenu s konstantnom domenom mišjeg teškog lanca.

23. Metoda za proizvodnju antigen-vezujućeg proteina obuhvaća ljudsku Vκ domenu, pri čemu navedena metoda obuhvaća izlaganje miša u skladu s bilo kojim od patentnih zahtjeva 1 do 9 antigenu od interesa, dozvoljavajući mišu razvijanje imunog odgovora na antigen od interesa i izoliranje navedenog antigen-vezujućeg proteina, ili izoliranje ljudske Vκ domene navedenog antigen-vezujućeg proteina.
24. Metoda za dobivanje ljudskog slijeda Vκ gena, pri čemu navedena metoda obuhvaća izlaganje miša u skladu s bilo kojim od patentnih zahtjeva 1 do 9 antigenu od interesa i izoliranje iz navedenog miša presloženog slijeda ljudskog Vκ gena, pri čemu je presloženi slijed ljudskog Vκ gena spojen sa slijedom nukleotida koji kodira CH regiju u navedenog miša.
