各ウェルが、上部開口及び底を画成する内面を有し、各ウェルの前記上部開口が、100マイクロメートル〜2000マイクロメートルの範囲の直径寸法を有することを特徴とする、実施形態1又は2に記載の細胞培養装置。
前記細胞培養装置が、互いに積層された複数の前記プレートを備えていることを特徴とする、実施形態1〜3のいずれかに記載の細胞培養装置。
入口及び出口をさらに備えており、前記複数の培地フローチャネルの各々が、前記入口及び前記出口と流体連通していることを特徴とする、実施形態1〜4のいずれかに記載の細胞培養装置。
前記入口を画成し、かつ、複数の入口フローチャネル開口を画成する、入口マニホルドをさらに備えており、前記複数の入口フローチャネル開口の各々が、それぞれの培地フローチャネルと流体連通していることを特徴とする、実施形態5に記載の細胞培養装置。
前記出口を画成し、かつ、複数の出口フローチャネル開口を画成する、出口マニホルドをさらに備えており、出口フローチャネル開口の各々が、それぞれの培地フローチャネルと流体連通していることを特徴とする、実施形態5に記載の細胞培養装置。
隣接する積層されたプレートのうちの少なくとも1つの対が、前記積層されたプレートのうちの1つのプレートの前記第1の主面によって画成された底面と、前記隣接する積層されたプレートの前記第2の主面によって画成された上面とを有する細胞培養チャンバを画成することを特徴とする、実施形態1〜7のいずれかに記載の細胞培養装置。
前記複数の積層されたプレートの最上部の細胞培養プレート上に配置された上部プレートをさらに備えることを特徴とする、実施形態1〜8のいずれかに記載の細胞培養装置。
少なくとも1つのスペーサが、前記スペーサの長さを延長する空気チャネルを備えており、該空気チャネルが、前記細胞培養装置の外部とガス連通し、かつ、前記スペーサによって画成された前記複数の培地フローチャネルの少なくとも1つとガス連通していることを特徴とする、実施形態1〜9のいずれかに記載の細胞培養装置。
前記プレートが、隣接するプレートのスペーサを位置合わせするように積層されており、該位置合わせされたスペーサが、最底部のプレートから最上部のプレートまで延びる壁を画成することを特徴とする、実施形態1〜10のいずれかに記載の細胞培養装置。
少なくとも1つの壁が、前記最底部のプレートから前記最上部のプレートまで延びる空気チャネルを画成しており、該空気チャネルが、前記細胞培養装置の外部とガス連通し、かつ、前記壁の前記スペーサによって画成されたそれぞれの培地フローチャネルとガス連通していることを特徴とする、実施形態11に記載の細胞培養装置。
前記複数のウェルが、六方最密充填されたウェルのアレイを含むことを特徴とする、実施形態1〜12のいずれかに記載の細胞培養装置。
前記ウェルが、前記上部開口から前記底まで画成された深さを有しており、該深さが、200マイクロメートル〜2000マイクロメートルの範囲内にあることを特徴とする、実施形態1〜13のいずれかに記載の細胞培養装置。
前記ウェルの各々が、細胞に対して非接着性であることを特徴とする、実施形態1〜14のいずれかに記載の細胞培養装置。
前記プレートが、前記ウェルを介してガス透過可能であることを特徴とする、実施形態1〜15のいずれかに記載の細胞培養装置。
実施形態1〜16のいずれかに記載の細胞培養装置;及び
前記入口に連結した酸素供給器
を備えた、細胞培養システム。
前記入口に連結したポンプをさらに備えていることを特徴とする、実施形態17に記載の細胞培養システム。
実施形態1に記載の細胞培養装置の前記ウェル内に細胞を導入する工程;及び
前記ウェル内で前記細胞を培養するために、細胞培養培地を第1の速度で前記細胞培養装置の前記培地フローチャネルを通じて灌流させる工程
を含む、方法。
前記ウェルから前記細胞を取り出すために、前記細胞培養培地を前記第1の速度より速い第2の速度で前記培地フローチャネルを通じて灌流させる工程をさらに含む、実施形態19に記載の方法。
a)実施形態1〜17のいずれかに記載の細胞培養装置の前記ウェル内でタンパク質を発現する細胞を培養する工程;及び
b)前記細胞から前記タンパク質を単離する工程
を含む、タンパク質を産生する方法。

Claims (18)
- 第1の主面及び反対側の第2の主面を各々有する互いに積層された複数のプレートであって、前記第1の主面が複数のマイクロウェルを備えている、プレート;及び
前記第1の主面から延びる複数のスペーサであって、前記プレートの長さに沿って前記第1の主面から延びており、複数の細胞培養培地フローチャネルが隣接するスペーサと、積層されたプレートとの間に画成され、少なくとも1つのスペーサが、前記スペーサの長さに沿って延びる空気チャネルを備えており、該空気チャネルが、前記細胞培養装置の外部とガス連通し、かつ、前記スペーサによって画成された前記複数の培地フローチャネルの少なくとも1つとガス連通している、スペーサ
を備えた、細胞培養装置。 - 各ウェルが、上部開口及び底を画成する内面を有しており、各ウェルの前記上部開口が、100マイクロメートル〜2000マイクロメートルの範囲の直径寸法を有することを特徴とする、請求項1に記載の細胞培養装置。
- 入口及び出口をさらに備えており、前記複数の培地フローチャネルの各々が、前記入口及び前記出口と流体連通していることを特徴とする、請求項1または2のいずれか一項に記載の細胞培養装置。
- 前記入口を画成し、かつ、複数の入口フローチャネル開口を画成する、入口マニホルドをさらに備えており、前記複数の入口フローチャネル開口の各々が、それぞれの培地フローチャネルと流体連通していることを特徴とする、請求項3に記載の細胞培養装置。
- 前記出口を画成し、かつ、複数の出口フローチャネル開口を画成する、出口マニホルドをさらに備えており、出口フローチャネル開口の各々が、それぞれの培地フローチャネルと流体連通していることを特徴とする、請求項3に記載の細胞培養装置。
- 隣接する積層されたプレートのうちの少なくとも1つの対が、前記積層されたプレートのうちの1つのプレートの前記第1の主面によって画成された底面と、前記隣接する積層されたプレートの前記第2の主面によって画成された上面とを有する細胞培養チャンバを画成することを特徴とする、請求項1に記載の細胞培養装置。
- 前記複数の積層されたプレートの最上部の細胞培養プレート上に配置された上部プレートをさらに備えることを特徴とする、請求項1に記載の細胞培養装置。
- 前記プレートが、隣接するプレートのスペーサを位置合わせするように積層されており、該位置合わせされたスペーサが、最底部のプレートから最上部のプレートまで延びる壁を画成することを特徴とする、請求項1〜7のいずれか一項に記載の細胞培養装置。
- 少なくとも1つの壁が、前記最底部のプレートから前記最上部のプレートまで延びる空気チャネルを画成しており、該空気チャネルが、前記細胞培養装置の外部とガス連通し、かつ、前記壁の前記スペーサによって画成されたそれぞれの培地フローチャネルとガス連通していることを特徴とする、請求項8に記載の細胞培養装置。

を含む、方法。 - 前記ウェルから前記細胞を取り出すために、前記細胞培養培地を前記第1の速度より速い第2の速度で前記培地フローチャネルを通じて灌流させる工程
をさらに含む、請求項16に記載の方法。 - a)請求項1〜15のいずれか一項に記載の細胞培養装置の前記ウェル内でタンパク質を発現する細胞を培養する工程;及び
b)前記細胞から前記タンパク質を単離する工程
