Description
本発明は、米国国立衛生研究所(NIH)、米国国立科学基金(NSF)および米空軍(Foster Millerからの外注契約)の基金番号第RO1EB003210、RO1DE13405-O1A1、DMR-0090384、F49620-01-C-0064により支援されており、米国政府は本発明に一定の権利を有する。
本発明は概して、濃縮された水性シルクフィブロイン溶液の調製法、ならびに線維、フィルム、スポンジ状多孔質フォーム、3次元足場、およびヒドロゲルなどのシルクフィブロイン材料の製造におけるこれらの溶液の使用法に関する。特にシルクフィブロイン溶液の調製のための全て水性の手段が説明される。
シルクは、カイコボンビックス・モリ(Bombyx mori)により生成されるよく説明された天然の線維であり、これは伝統的に数千年にわたり織物の糸の形で使用されている。このシルクは、糸コアを形成するフィブロイン(重鎖および軽鎖の両方)と称される線維状タンパク質、およびフィブロイン線維を取り囲み、それらを一緒に固めているセリシンと称される糊状(glue-like)タンパク質を含む。フィブロインは、線維内でβプリーツシートの形成につながるアミノ酸グリシン、アラニンおよびセリンを最大90%含有する、高度に不溶性のタンパク質である(Asakura, et al., Encylopedia of Agricultural Science, Arntzen, C. J., Ritter, E. M. Eds.;Academic Press:New York, NY, 1994;Vol.4, pp1-11)。
例えば、3次元多孔質シルク足場の、組織エンジニアリングにおける使用が説明されている(Meinel et al., Ann Biomed Eng, 2004 Jan;32(1):112-22;Nazarov, R., et al., Biomacromolecules、印刷中)。さらに再生されたシルクフィブロインフィルムは、酸素透過性膜および薬物透過性膜、酵素固定のための支持体、および細胞培養のための基体として研究されている。
加えてシルクヒドロゲルは、組織エンジニアリングに加え、ドラッグデリバリーにおいて多くの用途が認められる(Megeed et al., Pharm Res., 2002 Jul;19(7):954-9;Dinerman et al., J. Control Release., 2002 Aug 21;82(2-3):277-87)。
本発明は、濃縮された水性シルクフィブロイン溶液、および有機溶媒またはきつい化学物質の使用を避ける濃縮された水性フィブロイン溶液の調製のための全て水性の様式(all-aqueous mode)を提供する。本発明は、例えば線維、フィルム、フォーム、メッシュ、足場、およびヒドロゲルのような材料の製造における、これらの溶液の使用をさらに提供する。
有機溶媒またはきつい化学物質の非存在下で、濃縮された水性シルクフィブロイン溶液を調製する方法が説明される。この方法は、シルクフィブロインを含有する溶液を形成することを含む。好ましくは、この溶液は、臭化リチウムのような水性の塩である。その後この溶液は、吸湿性ポリマーに対して、10〜30wt%またはそれよりも大きい水性シルクフィブロイン溶液を生じるのに十分な時間透析される。好ましい吸湿性ポリマーは、ポリエチレングリコール(PEG)である。
(実施例1)電気紡糸による再生されたシルク溶液由来の水からの純粋なシルク線維の調製
方法
再生されたB・モリシルクフィブロイン溶液の調製
ボンビックス・モリシルクフィブロインは、本発明者らの初期の手法(Sofia, et al., Journal of Biomedical Materials Research, 2001, 54:139-148)の改変として、下記のように調製した。繭は、0.02M Na2CO3水溶液中で30分間煮沸し、その後水で入念にすすぎ、糊状のセリシンタンパク質を抽出した。抽出したシルクをその後、9.3M LiBr溶液中に室温で溶解し、20%(w/v)溶液を得た。この溶液を、Slide-A-Lyzer透析カセット(Pierce, MWCO 2000)を用い、水中で48時間透析した。水性シルク溶液の最終濃度は、8.0wt%であり、これは乾燥後の残留固形物を計量し決定した。
紡糸のため水性シルク溶液の粘度を8wt%を超えるよう増大するために、この溶液を、上記のPEG溶液法を用いて濃縮した。純粋なシルク溶液(8wt%)の粘度および表面張力が、キャピラリーの先端に安定した液滴を維持するのに十分ではないので、濃縮が必要があった。シルク溶液の増加は、電気紡糸に適した粘度および表面張力を生じた。新たなより濃縮された純粋なシルク溶液(10〜30%)により、直接紡糸が実行可能になる。先端と収集器との間の距離は10〜15cmであり、液体の流量は0.01〜0.05ml/分であった。キャピラリーの先端と対のアルミフォイル電極との間の電位差は、30kV(E=2〜3kV/cm)へ漸増したので、キャピラリー先端の末端での液滴は、半球形から円錐形へ伸長した。電気紡糸した線維の形態および直径は、SEMを用いて試験した。シルク/PEO配合液は、直径1.5μm〜25μmのマイクロサイズ線維を製造した。線維表面および液体窒素中の破砕された表面の形態は、天然のシルク線維に良く合致していた。
多孔質3次元足場を、シルクフィブロイン水溶液から塩浸出により調製した。シルクフィブロイン濃度および粒状NaClの粒度の調節により、足場の形態および機能の特性が制御される。これらの足場は高度に均一で、かつ相互に連結した孔を有し、調製の様式に応じて470〜940umの範囲の細孔径を示した。これらの足場は、>90%の多孔度を有した。足場の圧縮強度および圧縮弾性率は、各々、最大320±10KPaおよび3330±500KPaであった。足場は、プロテアーゼにより21日間に完全に分解された。これらの新規シルクベースの3次元マトリックスは、全て水性の様式の調製、細孔径の制御、細孔の接続性、分解性および有用な機械的特徴により、組織エンジニアリングのための生体材料マトリックスとして有用な特性を提供する。
シルクフィブロイン水溶液の調製
B・モリの繭を、0.02M Na2CO3水溶液中で20分間煮沸し、その後蒸留水で入念にすすぎ、糊状のセリシンタンパク質およびワックスを抽出した。抽出したシルクを次に、9.3M LiBr溶液中に60℃で4時間溶解し、20%(w/v)溶液を得た。この溶液を、Slide-A-Lyzer透析カセット(MWCO 3500, Pierce)を用いて、蒸留水中で2日間透析した。シルクフィブロイン水溶液の最終濃度は約8w/v%であり、これは乾燥後の残留固形物を計量して決定した。濃縮されたシルクフィブロイン溶液を調製するために、8w/v%シルクフィブロイン溶液10mlを、Slide-A-Lyzer透析カセット(MWCO 3500)を用い、1リットルの25wt%ポリエチレングリコール(PEG, 10,000g/mol)溶液に対し室温で透析した。必要な時間の後、濃縮されたシルクフィブロイン溶液を、過剰な剪断を避けるために、シリンジによりゆっくり収集し、濃度を決定した。濃度が8wt%未満のシルクフィブロイン水溶液を、蒸留水による希釈により調製した。全ての溶液を、早期沈殿を避けるために使用前に7℃で貯蔵した。8w/v%溶液から調製したシルクフィブロインフィルムを評価し、XPSによりLi+イオンの除去を証明した。残存するLi+イオンは検出されなかった。
粒状NaCl(粒度;300〜1180um)4gを、ディスク型テフロン容器中のシルクフィブロイン水溶液(4〜10wt%)2ml中に添加した(図2a)。容器に蓋をし、室温に放置した。24時間後、この容器を水中に含浸し、NaClを2日間抽出した。この方法で形成された多孔質シルクフィブロイン足場は、使用前に水中に7℃で貯蔵した。
40kVおよび40mAで作動中のRigaku RU-200BH回転式対陰極X線発生装置からのNiフィルターを通したCu-Kα照射(λ=0.15418nm)により、足場の凍結乾燥した試料のX線回折を得た。X線回折パターンは、真空カメラにおける点で視準された(point collimated)ビームおよびイメージングプレート(Fuji Film BAS-IP SR 127)により記録した。カメラ長は、NaFで較正した(d=0.23166nm)。
凍結乾燥した試料約1mgを、臭化カリウム200mgとペレットに圧縮し、フーリエ変換赤外(FTIR)スペクトルを、Nicolet Magna 860による64スキャンの蓄積および4cm-1の分解能で記録した。
シルク足場を、カミソリの刃を用い、蒸留水中で切片に切断し、その後凍結乾燥した。試料は金でスパッターコーティングした。足場の形態は、LEO Gemini 982電界放出ガンSEMで観察した。米国国立衛生研究所により開発されたImageJソフトウェアを用い、細孔径を得た。
シルク足場の密度および多孔度を、液体交換により測定した(Zhang, R. Y., et al., J. Biomed. Mater. Res., 1999, 44:446-455)。ヘキサンはマトリックスを膨潤または縮小することなくシルク足場を通して浸透するので、これを交換液として用いた。シルク足場(乾燥質量, W)は、メスシリンダー中の既知の容積(V1)のヘキサン中に5分間含浸した。ヘキサンおよびヘキサンを含浸した足場の総容積は、V2として記録した。その後ヘキサンを含浸した足場をシリンダーから取り除き、残存するヘキサン容積をV3として記録した。足場総容積は下記式である:
V=(V2-V1)+(V1−V3)=V2−V3
V2−V1は、ポリマー足場の容積であり、V1−V3は、足場内のヘキサン容積である。足場の多孔度(ε)は、下記式から得た:

シルクフィブロイン足場を、蒸留水中に室温で24時間含浸した。過剰な水を除去した後、足場の含水質量(Ws)を決定した。その後試料を、真空下65℃のオーブンの中で一晩乾燥し、足場の乾燥質量(Wd)を決定した。足場の膨潤比および足場の含水量は以下のように計算した:

足場(直径12mm、高さ10mm、ディスク)の力学的圧縮に対する抵抗を、0.1kNを装填した細胞を装加したInstron 8511上で室温で実行した。クロスヘッド速度は10mm/分であった。圧縮試験は、便宜上オープンサイド/拘束法(open-sided/confined method)で行った。4つの試料を、各組成について評価した。直径12mmおよび高さ10mmと測定された円柱形の試料を、ASTM法F451-95を基にした改変法に従い用いた。圧縮応力および圧縮ひずみをグラフ化し、平均圧縮強度に加え圧縮弾性率および標準偏差を決定した。弾性率は、応力−ひずみ曲線の最初の直線部分の勾配により定義した。圧縮強度は、1%ひずみから出発する、これに平行な直線を描くことにより決定した。この直線と応力−ひずみ曲線とが交差した点を、フォームの圧縮強度として定義した(Thomson RC et al., Biomaterials, 1998,19;1935-1943)。
シルクフィブロイン足場の分解は、活性5.6U/mgのプロテアーゼXIV(EC 3.4.24. 31, Sigma-Aldrich)を用いて評価した。試料(直径12mm、高さ5mm)を、プロテアーゼ(1U)を含有するリン酸緩衝生理食塩水5ml(pH7.4)に37℃で含浸した。特定時間の後に、試料をリン酸緩衝生理食塩水および蒸留水で洗浄し、凍結乾燥した。酵素溶液を、新たに調製した溶液と24時間毎に交換した。対照として、試料を酵素を含まないリン酸緩衝生理食塩水に含浸した。
水ベースの足場の調製
多孔質シルクフィブロイン足場を、コラーゲンおよびポリ乳酸などの他のポリマーからの多孔質足場の調製においてこれまでに使用されていた塩浸出法を用いて調製した。足場の細孔径および多孔度は、シルクフィブロイン水溶液への直径300〜1180μmの粒度を持つ粒状NaClの添加により調節した。この方法において、NaCl粒子の表面の一部は、シルクフィブロイン水溶液に溶解したが、この塩のほとんどは溶液の飽和のために固形物粒子として残存した。このシルクフィブロイン水溶液は、〜24時間後混合物中にヒドロゲルを形成し、これは水で安定的な多孔質マトリックスの形成を生じた。表1は、本試験において使用したシルクフィブロイン濃度およびNaClの粒度を示す。シルクフィブロイン濃度の増加に伴い、より大きい粒度のNaClの使用により、マトリックスは均質に形成された。粒度500〜600μmのNaClが8wt%シルクフィブロイン溶液に添加された場合、シルクフィブロイン水溶液の表面は、ヒドロゲルを迅速に形成した。
シルクフィブロインの構造の変化は、X線回折およびFTIR(図3)により決定した。シルクフィブロイン足場のX線回折は、20.8°の明瞭なピークおよび24.6°の小さいピークを示した。これらのピークは、天然のシルクフィブロインのβシート結晶構造(シルクII)のピークとほぼ同じであった(Asakura, T., et al., Macromolecules, 1985,18:1841-1845)。これらの結果は、20.8°および24.6°の反射に従い、各々、距離4.3および3.6Åのスペーシングのβ結晶を示している。シルクフィブロイン足場のFTIRスペクトルは、1701cm-1および1623cm-1(アミドI)にシルクIIの特徴的なピークを示した(Asakura, T., et al., Macromolecules, 1985,18:1841-1845)。水溶液中のシルクフィブロインは、中性pHで、ランダムコイル構造を示した。X線回折およびFTIR解析の結果より、これらの溶液からのシルクフィブロイン足場の形成は、ランダムコイルからβシートへの構造転移を誘導した。
様々なシルクフィブロイン濃度および様々なサイズのNaCl粒子から調製した凍結乾燥した足場のSEM画像は、高度に相互に連結した多孔質構造を示し、孔の分布は、足場の最表面の空気-水界面に形成された薄い層を除いて、足場全体で均質であった。足場は、多くのより小さい孔により高度に相互連結された粗い細孔表面を示した。直径1〜3μmの球状の構造が、細孔の表面上に観察された。シルクフィブロイン濃度が増加するにつれて、細孔壁は厚くなった。表2は、足場内の実際の細孔径を示し、これは350〜920μmの範囲である。
多孔度が>90%のシルクフィブロイン足場が形成され、かつ多孔度は、細孔径およびシルクフィブロイン濃度が減少するにつれて増加した(表3)。これらの値は、塩浸出またはガス発泡により調製されたHFIP由来のシルク足場のもの(84〜98%)と類似していた(Nazarov R, et al., Biomacromolecules, 印刷中)。足場の膨潤比および水取込みは、表4および5に示す。
足場は、本方法において使用したシルクフィブロイン濃度に依存する様々な硬さを有する、延性のあるスポンジ状の挙動を示した。弾性領域が、初期ひずみで観察され、それにピーク応力が続いた。表6は、シルクフィブロイン足場の力学特性を示す。足場の圧縮強度および圧縮弾性率は、シルクフィブロイン濃度の増加と共に増加した。
図4aは、分解期間21日間の、直径850〜1000μmの粒度を持つNaClで、4〜8wt%シルクフィブロインから調製された足場質量を経時的に示している。プロテアーゼを含まないリン酸緩衝液中の足場は、21日以内に分解を示さなかった。4wt%フィブロインで調製した足場は迅速に分解され、10日後に残存する質量は、わずかに2%であった。6および8wt%フィブロインから調製された足場は、経時的に徐々に分解し、21日後に、質量は各々30および20%に減少した。図4bは、様々な粒度のNaClで6wt%シルクフィブロインから調製した場合に、残存する足場の質量を示している。これらの分解パターンは、フィブロインの初期濃度の性質に関連して、細孔径は分解率に相関しないことを示した。
有機溶媒または化学架橋の完全な非存在下で、シルクフィブロイン水溶液からの塩浸出法により、多孔質シルクフィブロイン足場を直接調製した。足場の形成は、ランダムコイルからβシートへの構造転移を含んだ。この転移は、塩が疎水性ドメインからの水の喪失を促進し、これは鎖-鎖相互作用を増強し、その結果βシートの形成につながるので、転移に関する力学的基礎を提供する。足場の機能的および形態学的特性は、この方法において使用されるシルクフィブロイン溶液の濃度およびNaClの粒度により制御された。
浸透ストレスを介した水溶液中のシルクフィブロイン濃度の制御を、この方法に関連したゲル形成と構造、形態、および機能(力学的)の変化との関係を評価するために試験した。水性シルクフィブロインのインビボプロセッシングにおいて重要な可能性のある環境因子も試験し、この方法へのそれらの寄与を決定した。シルクフィブロイン水溶液のゲル化は、温度、Ca2+、pH、およびポリエチレンオキシド(PEO)により影響を受けた。ゲル化時間は、タンパク質濃度の増加、pHの低下、温度の上昇、Ca2+の添加、およびPEOの添加と共に減少した。K+の添加では、ゲル化時間に変化は認められなかった。ゲル化にあたり、シルクフィブロインのランダムコイル構造は、βシート構造に転移された。フィブロイン濃度が>4質量%のヒドロゲルは、走査型電子顕微鏡を基に、ネットワークおよびスポンジ状の構造を示した。凍結乾燥したヒドロゲルの細孔径は、シルクフィブロイン濃度またはゲル化温度が増加するにつれてより小さくなった。Ca2+の存在下で形成された凍結乾燥したヒドロゲルは、このイオン濃度が増加するにつれて、より大きい細孔を示した。ヒドロゲルの力学的圧縮強度および圧縮弾性率は、タンパク質濃度およびゲル化温度の増大と共に増加した。
シルクフィブロイン水溶液の調製
ボンビックス・モリの繭を、M. Tsukada(Institute of Sericulture, Tsukuba, Japan)およびM. Goldsmith(U. Rhode Island)のご厚意により入手し、0.02M Na2CO3の水溶液中で20分間煮沸し、その後蒸留水で入念にすすぎ、糊状のセリシンタンパク質およびワックスを抽出した。抽出したシルクフィブロインをその後、9.3M LiBr溶液中に60℃で4時間溶解し、20%(w/v)溶液を得た。この溶液を、Slide-A-Lyzer透析カセット(MWCO 3500 Pierce)を用い、蒸留水中で2日間透析した。シルクフィブロイン水溶液の最終濃度は約8w/v%であり、これは乾燥後の残留固形物を計量し決定した。8w/v%溶液から調製したシルクフィルムを評価し、XPSによりLi+イオンの除去を証明した。残存するLi+イオンは検出されなかった。
シルクフィブロイン水溶液(8wt%, 10ml)を、Slide-A-Lyzer透析カセット(MWCO 3500)を使用し、室温で10〜25wt%ポリエチレングリコール(PEG, 10,000g/mol)溶液に対して透析した。PEGのシルクフィブロイン溶液に対する容積比は100:1であった。浸透ストレスにより、シルクフィブロイン溶液中の水分子は、透析膜を通りPEG溶液へと移動した(Parsegian, V. A., et al., Methods in Enzymology, Packer, L., Ed.;Academic Press:1986;Vol.127, p400)。必要な時間の後、濃縮されたシルクフィブロイン溶液を、過剰な剪断を避けるために、シリンジによりゆっくり収集し、濃度を決定した。濃度が8wt%未満のシルクフィブロイン水溶液は、8wt%溶液を蒸留水で希釈することにより調製した。全ての溶液は、使用前は7℃で貯蔵した。
シルクフィブロイン水溶液0.5mlを、2.5mlの平底バイアル(直径:10mm)に入れた。これらのバイアルを密閉し、室温、37℃および60℃で維持した。試料が不透明な白色を示しかつ反転させたバイアルから30秒以内に落下しない時点で、ゲル化時間を決定した。この方法に対するイオンおよびイオン濃度の影響を調べるために、CaCl2またはKCl溶液を、シルクフィブロイン水溶液に添加し、最終塩濃度2.5〜30mMを生じた。シルクフィブロイン溶液のpHは、HClまたはNaOH溶液で調節した。シルクフィブロイン-ポリ(エチレン)オキシド(PEO, 900,000g/mol)溶液の調製のために、必要量のPEO溶液(5wt%)を、シルクフィブロイン溶液に、ゆっくり攪拌しながら5分間かけて添加した。シルクフィブロイン/PEOの配合比は、100/0、95/5、90/10、80/20および70/30(w/w)であった。
X線プロファイルを、Brucker D8 X線回折装置を用い、40kVおよび20mAで、NiフィルターCu-Kα照射により、凍結乾燥されたシルクフィブロイン溶液およびヒドロゲルについて記録した。
シルクフィブロイン溶液およびヒドロゲルを-80℃で凍結し、その後凍結乾燥した。これらの試料を、液体窒素中で破壊し、LEO Gemini 982電界放出ガンSEMを用いて試験した。凍結乾燥による人為的な形態変化をチェックするために、代替調製法では、室温で4時間のKarnovsky固定剤を使用した。固定剤入り、またはなしで、ヒドロゲルは、凍結乾燥時に形態学的変化をほとんど示さなかった。米国NIHにより開発されたImageJソフトウェアを用い、細孔径を得た。
ヒドロゲルの圧縮試験を、2.5kNを装填した細胞を装加したInstron 8511において室温で行った。クロスヘッド速度は10mm/分であった。試料の横断面は、直径12mmおよび高さ5mmであった。圧縮試験は、便宜上オープンサイド法で行った。圧縮限界は、装填細胞を保護するために98%ひずみであった。各組成物について5個の試料を評価した。
濃縮されたシルクフィブロイン溶液
初期濃度8wt%のシルクフィブロイン水溶液を、10〜25wt%PEG溶液に対し室温で透析した。シルクフィブロイン水溶液は、浸透ストレスにより経時的に濃縮され、25wt%PEG溶液に対する9時間の透析後、約21wt%の濃度を得た(図6)。より低い濃度のPEG溶液を使用する場合、より高い濃度のシルクフィブロイン水溶液を作製するには、より長い透析時間が必要であった。23〜33wt%のシルクフィブロインゲルは、濃縮法の間に透析カセット中に自然発生的に生成された。これらのゲルは、室温および60℃で乾燥後であっても透明であった。
シルクフィブロイン水溶液のゲル化に対する温度、Ca2+およびK+濃度、pH、ならびにPEO濃度の影響を調べた。図7は、シルクフィブロイン水溶液(pH6.5〜6.8)の様々な温度でのゲル化時間を示している。シルクフィブロイン水溶液のゲル化時間は、フィブロイン含量および温度の上昇と共に減少した。同時にランダムコイルからβシート構造への構造変化が観察され、ヒドロゲル内のβシート構造の形成は、以下に説明されるようなX線回折により確認された。図8は、様々なCa2+およびK+濃度でのシルクフィブロイン水溶液のゲル化時間を示している。Ca2+およびK+イオンを含むシルクフィブロイン溶液のpHは、各々、5.6〜5.9および6.2〜6.4であった。Ca2+は、比較的短いゲル化時間を生じたのに対し、K+の添加ではいずれの温度でもゲル化時間に変化はなかった。再生されたカイコフィブロインによるこれらの結果は、クモシルク溶液に添加されたK+イオンが、タンパク質の凝集および沈殿に影響を及ぼした先の試験とは異なったが、Ca2+イオンの添加後レオロジー変化はなかった。図9は、様々なpHでのシルクフィブロイン水溶液(4wt%)のゲル化時間を示す。ゲル化時間は、pHの低下と共に有意に短縮した。この挙動は、pH5.5でゲル化するがpH7.4では粘性の液体として挙動する、ニワオニグモ(Araneus diadematus)由来のシルクについて観察されたものに類似している(Vollrath, F., et al., Proc. R. Soc. London B, 1998, 265:817-820)。図10は、様々なポリエチレンオキシド(PEO)含量のシルクフィブロイン水溶液(4wt%)のゲル化時間を示す。PEO溶液の添加により、pHは、6.1〜6.4の範囲にわずかに低下した。ゲル化時間は、5%PEOのみの添加により有意に短縮したのに対し、濃度が5%を上回る場合のゲル化時間には差異はなかった。
シルクフィブロインにおける構造変化を、X線回折により決定した。図11は、シルクフィブロイン水溶液から調製した凍結乾燥されたシルクフィブロイン溶液およびヒドロゲルのX線プロファイルを示している。シルクフィブロイン溶液がガラス転移点以下の低温(-34〜-20℃)で凍結された場合、その構造は有意に変化しなかった(Li, M., et al., J Appl. Polym. Sci., 2001, 79:2185-2191)。凍結乾燥したシルクフィブロイン試料は、シルクフィブロイン濃度に関わりなく、20°近傍で、幅広のピークを示し、これはアモルファス構造を示している。中性pH水溶液中のシルクフィブロインはランダムコイル構造を示した(Magoshi, J., et al., Polymeric Materials Encyclopedia;Salamone, J. C., Ed.;CRC Press:NewYork, 1996;Vol.1, p.667;Magoshi, J., et al., Polymeric Materials Encyclopedia;Salamone, J. C., Ed.;CRC Press: New York, 1996;Vol.1, p.667)。シルクフィブロイン溶液から調製した全てのヒドロゲルは、明確なピークを20.6°に、ならびに9°および24°付近に2個の小さいピークを示した。これらのピークは、シルクフィブロインのβシート結晶構造のものとほぼ同じであった(Ayub, Z. H., et al., Biosci. Biotech. Biochem., 1993, 57:1910-1912;Asakura, T., et al., Macromolecules, 1985, 18:1841-1845)。これらのピークは、9°、20.6°、および24°に従い各々、β結晶スペーシング距離9.7、4.3、および3.7Åを示している。X線回折の結果から、シルクフィブロイン溶液のゲル化は、先に報告されたような、ランダムコイルからβシートへのコンホメーション転移を誘導した(Ayub, Z. H., et al., Biosci. Biotech. Biochem., 1993, 57:1910-1912;Hanawa, T., et al., Chem. Pharm. Bull., 1995, 43:284-288;Kang, G. D., et al., Marcromol. Rapid Commun., 2000, 21:788-791)。
シルクフィブロイン溶液およびヒドロゲルの形態学的特徴を、-80℃で凍結乾燥した後、SEMにより観察した。凍結乾燥した4〜12wt%のシルクフィブロイン溶液は、葉状の形態を示した。凍結乾燥した16wt%および20wt%のシルクフィブロイン溶液は、各々、細孔径5.0±4.2μmおよび4.7±4.0μmのネットワークおよびスポンジ状構造を示した。SEM画像により、4wt%シルクフィブロイン溶液から調製した凍結乾燥したヒドロゲルは、温度とは無関係に葉状形態および相互に連結した細孔を示し、4wt%よりも高いフィブロイン濃度では、スポンジ状構造が観察されたことが決定された。凍結乾燥したヒドロゲルの細孔径(<1.1±0.8μm)は、凍結乾燥したシルクフィブロイン溶液試料について観察されるものよりも小さかった。凍結乾燥されたヒドロゲルの細孔径は、シルクフィブロイン濃度の増加と共に減少し、かつ細孔径は、同じシルクフィブロイン濃度では温度が上昇するにつれ減少した。Ca2+イオンを含む4wt%の凍結乾燥されたヒドロゲルは、ネットワークおよびスポンジ状構造を示したが、K+イオンを含む4wt%の凍結乾燥されたヒドロゲルは、葉状形態を有した。フィブロイン濃度>4wt%の凍結乾燥されたヒドロゲルにおいて、Ca2+を含む凍結乾燥されたヒドロゲルの細孔径は、Ca2+イオンを含まないシルクフィブロイン水溶液から調製された凍結乾燥されたヒドロゲルのものよりも大きかった。興味深いことに、細孔径は、同じシルクフィブロイン濃度で、Ca2+濃度が増加するにつれて、凍結乾燥されたヒドロゲルにおいてより大きくなった。Ca2+を含む凍結乾燥されたヒドロゲルとは対照的に、K+を含む凍結乾燥されたヒドロゲルの細孔径は、シルクフィブロイン水溶液から調製した凍結乾燥されたヒドロゲルのものと類似したサイズを示した。これらの結果は、Ca2+は、シルクフィブロイン鎖間で相互作用を誘導する点で、K+よりもより有効であることを暗示している。この結果は、Ca2+がK+よりもより短いゲル化時間を生じた先行するデータとも一致する。
シルクフィブロイン水溶液から調製したヒドロゲルの圧縮強度および圧縮弾性率は、シルクフィブロイン濃度の増加と共に増加した(図12aおよび12b)。力学特性の改善は、細孔径の減少を伴うポリマー濃度の増加に起因する。同じシルクフィブロイン濃度で、より高い温度で調製されたヒドロゲルは、減少した細孔径のために、より高い圧縮強度および圧縮弾性率を示した。4〜8wt%フィブロインのヒドロゲルが55%未満のひずみを示したのに対し、12〜16wt%フィブロインのヒドロゲルは75%〜96%の範囲のより大きいひずみを示した(図12c)。より小さな細孔径は、ヒドロゲル内の応力をより均一に分配し、応力の集中に抵抗するため、細孔径の影響が考慮された。より小さい細孔径および増加した数の細孔は、亀裂の広がりに対する障壁としても機能する。
ゲル化は、疎水性相互作用および水素結合を含む、タンパク質鎖間の分子内および分子間相互作用の形成により生じる(Ayub, Z. H., et al., Biosci. Biotech. Biochem., 1993, 57:1910-1912;Hanawa, T., et al., Chem. Pharm. Bull., 1995, 43:284-288;Kang, G. D., et al., Marcromol. Rapid Commun., 2000, 21:788-791)。フィブロイン含量および温度の増加と共に、フィブロイン鎖間の相互作用は増加する。これによりシルクフィブロイン分子は、より迅速に相互作用することができ、このことは物理的架橋につながる。
一次配列から、カイコシルクフィブロイン重鎖は、鎖端に2個の大きい親水性ブロックを有する7個の内部疎水性ブロック、および7個のはるかに小さい内部親水性ブロックから構成されている(Zhou, C. Z., et al., Nucleic Acids Res., 2000,28:2413-2419;Jin, H. J., et al., Nature, 2003,424:1057-1061)。シルクフィブロイン中の疎水性残基の割合は79%であり(Braun, F. N., et al., Int. J. Biol. Macromol., 2003,32:59-65)、疎水性残基中の反復配列は、シルクフィブロイン線維およびフィルム内の結晶領域を形成するβシート構造を支配するGAGAGSペプチドからなる(Mita, K., et al., J. Mol. Evol., 1994, 38:583-592)。これらβシートの形成は、水中での不溶性をもたらす(Valluzzi, R., et al., J. Phys. Chem. B, 1999, 103:11382-11392)。水溶液中のシルクフィブロインの疎水性領域は、疎水性相互作用により物理的に集合し、最終的にはヒドロゲルに組織化される(Jin, H. J., et al., Nature, 2003,424:1057-1061)。シルクフィブロイン濃度、温度、Ca2+、pHおよびPEOは、シルクフィブロイン水溶液のゲル化に影響を及ぼす。フィブロイン含量および温度の増加に伴い、シルクフィブロイン分子間の物理的架橋は、より容易に形成される。Ca2+イオンは、おそらく鎖末端の親水性ブロックを介して、これらの相互作用を促進する。pHの低下および親水性ポリマーの添加は、シルクフィブロイン分子間の反発力を低下させ、タンパク質からの水の脱離を促進し、より短いゲル化時間をもたらす。ゲル化にあたり、ランダムコイルからβシート構造への構造転移が誘導され、水中のシルクフィブロインヒドロゲルの不溶性および安定性が促進される。シルクフィブロインヒドロゲルは、ネットワークおよびスポンジ状構造を有する。細孔径は、シルクフィブロイン濃度およびゲル化温度が増加するにつれて、より小さくなった。凍結乾燥されたヒドロゲルは、Ca2+濃度の増加に伴い、同じフィブロイン含量のシルクフィブロイン水溶液から調製された凍結乾燥されたヒドロゲルよりもより大きい細孔径を示した。イオンを含まないシルクフィブロイン水溶液から調製されたヒドロゲルの圧縮強度および圧縮弾性率は、タンパク質濃度およびゲル化温度の増加に伴い、増加した。
本発明者らは、本発明の水性シルク溶液からの3次元シルク足場上でのヒト骨髄幹細胞の骨再生を試験した。シルク足場の骨髄幹細胞の増殖および分化を支持する能力を試験するために、本発明者らは、いかなる修飾もしないシルク足場を使用した。
材料
ウシ血清、ダルベッコ変法イーグル培地(DMEM)、最小必須培地α改変型(αMEM)、塩基性線維芽細胞増殖因子(bFGF)、ペニシリン-ストレプトマイシン(Pen-Strep)、ファンギゾン、非必須アミノ酸、トリプシンは、Gibco(Carlsbad, CA)から得た。アスコルビン酸リン酸塩、Histopaque-1077、デキサメタゾン、およびβグリセロリン酸塩は、Sigma(St. Lois, MO)から得た。他の物質は全て、分析用または医薬品グレードであり、Sigmaから得た。カイコ繭は、M. Tsukada(Institute of Sericulture, Tsukuba, Japan)およびMarion Goldsmith (University of Rhode Island, Cranston, RI)のご厚意により入手した。
水性由来のシルク足場は、粒状NaCl(粒度;1000〜1180μm)4gを、ディスク型テフロン容器中の8wt%シルクフィブロイン溶液2mlに添加して調製した。この容器に蓋をし、室温に放置した。24時間後、容器を水に含浸し、NaClを2日間抽出した。HFIP由来のシルク足場は、4gの粒状NaCl(粒度;850〜100μm)をHFIP中の8wt%シルクフィブロイン2mlに添加することにより調製した。この容器に蓋をしHFIPを減少させ、この溶液のより均一な分布に十分な時間を提供した。溶媒を室温で3日間揮発させた。シルク/ポロゲンの複合体を、メタノール中で30分間処理し、βシート構造および水溶液中の不溶性を誘導した後、この複合体をNaClを除去するために2日間水に含浸した。この多孔質シルク足場を風乾した。
全骨髄(25cm3、Clonetics, Santa Rosa, CA.)を、分離培地(RPMI 1640培地中5%FBS)100ml中に希釈した。細胞を、密度勾配遠心分離法により分離した。簡単に述べると、骨髄懸濁液の20mlアリコートを、ポリショ糖勾配(1,077g/cm3, Histopaque, Sigma, St. Louis, MO)上に積層し、800xgで30分間、室温で遠心分離した。この細胞層を慎重に取り出し、分離培地10mlで洗浄し、Pure-Gene溶解液5ml中でペレット化し夾雑している赤血球を溶解した。細胞をペレット化し、増殖培地(DMEM、10%FBS、1ng/ml bFGF、100U/mlペニシリン、100μg/mlストレプトマイシン、0.25μg/mlファンギゾン、非必須アミノ酸)中に懸濁し、75cm2フラスコ中に、密度5x104個細胞/cm2で播種した。接着細胞は、約80%のコンフルエンスに到達させた(初代継代12〜17日間)。細胞をトリプシン処理し、再度播種し、継代2(P2)細胞(6〜8日後に80%コンフルエント)を、実験に使用した。
シルク足場上のインビトロにおける細胞の増殖および分化を試験するために、BMSC(5x105個細胞/足場、継代2)を、予め湿らせた(α-MEM、一晩)シルク足場上に播種した。24時間後、この培地を除去し、6ウェルプレートの個々のウェル中で培養物を維持した。骨形成性の培地は、ペニシリンおよびストレプトマイシンおよびファンギゾンの存在する、10%FBS、非必須アミノ酸、50μg/mlアスコルビン酸-2-リン酸塩、10nMデキサメタゾン、および7mMβグリセロリン酸を補充したαMEMであった。培養物は、5%CO2を補充した加湿したインキュベーター内、37℃で維持した。培地の半分を、2〜3日おきに交換した。
足場を、骨形成性培地中で2週間および4週間培養し、生化学分析および組織学のために処理した。DNA分析のために、1群1時点あたり3〜4足場を、分解した。DNA含量(n=3〜4)を、PicoGreenアッセイ(Molecular Probes, Eugene, OR)を製造業者のプロトコールに従い使用し、測定した。試料を、励起波長480nmおよび放出波長528nmで蛍光定量的に測定した。総カルシウム含量については、試料(n=4)を、5%トリクロロ酢酸0.5mlで2回抽出した。カルシウム含量は、o-クレゾールフタレインコンプレキソン(Sigma, St. Louis, MO)を使用する比色アッセイ法により決定した。カルシウム複合体は、575nmで分光光度法により測定した。アルカリホスファターゼ活性は、405nmで分光光度法により測定されるp-ニトロフェニルリン酸のp-ニトロフェノールへの転換を基にした、Sigma(St. Louis, MO)の生化学アッセイを用いて測定した。
新たな足場(n=3〜4/群)を、2mlプラスチック製チューブに移し、1.0mlトリゾールを添加した。スチールボールおよびMicrobeaterを用い、足場を分解した。チューブを、12000gで10分間遠心し、上清を新たなチューブに移した。クロロホルム(200μl)をこの溶液に添加し、室温で5分間インキュベートした。チューブを12000gで15分間再度遠心し、上側水層を新たなチューブに移した。70%エタノール(v/v)1容量を添加し、RNeasyミニスピンカラム(Quiagen, Hilden, Germany)に装加した。RNAを洗浄し、製造業者のプロトコールに従い溶離した。RNA試料を、製造業者のプロトコールに従いオリゴ(dT)選択を用いcDNAに逆転写した(Superscript Preamplification System, Life Technologies, Gaithersburg, MD)。I型コラーゲン、II型コラーゲン、アルカリホスファターゼ、骨シアロタンパク質およびオステオポンチンの遺伝子発現を、ABI Prism 7000 Real Time PCRシステム(Applied Biosystems, Foster City, CA)を用い定量した。PCR反応条件は、50℃で2分間、95℃で10分間、その後95℃で15秒を50サイクル、および60℃で1分間であった。発現データは、ハウスキーピング遺伝子グリセルアルデヒド-3-リン酸-デヒドロゲナーゼ(GAPDH)の発現に対して標準化した。GAPDHプローブは、5'末端を蛍光色素VICで、および3'末端を消光色素TAMRAで標識した。ヒトGAPDH遺伝子のプライマー配列は以下であった:フォワードプライマー

。アルカリホスファターゼ、骨シアロタンパク質(BSP)、オステオポンチンのプライマーおよびプローブは、Applied Biosciencesから購入した(Assay on Demand #, Hs 00240993 ml(ALP), Hs 00173720 ml(BSP), Hs 00167093 ml(オステオポンチン))。
総タンパク質抽出のために、細胞を、プロテアーゼインヒビターおよびホスファターゼインヒビターを含有する、RIPA緩衝液[50mM Tris-HCl、pH8.0、150mM NaCl、1% Nonidet P-40(NP-40)、0.2%SDS、5mM NaF]中で溶解した。タンパク質含量を、Bradfod法により測定した。タンパク質を、3〜8%SDS-PAGEにより分解し、メンブレンに移した。ブロットを、一次抗体で4℃で12時間プロービングし、洗浄し、適当なペルオキシダーゼで標識した二次抗体と共に、室温で1時間インキュベートした。タンパク質バンドを、ECL(Armersham-Pharmacia)により顕在化した。
細胞接着の前後に、ポリマー表面を走査型電子顕微鏡(SEM)により試験した。マトリックスは、Karnovsky固定剤を用いて24時間固定し、CMPBS中で3回洗浄し、残存する固定剤を除去した。その後これらの試料を、一連の段階的エタノール(50〜100%)を15分間隔で用いて乾燥した。乾燥後、試料を金でスパッタコーティングし、LEO Gemini 982電界放出ガンSEMで調べた。
4%リン酸緩衝ホルムアルデヒドで少なくとも24時間固定した後、標本をパラフィンに包埋し、切片とした(4μm)。標準の組織化学的技術を用い、連続切片をヘマトキシリン・エオシンおよびアルシアンブルーで染色した。
SEM分析
3Dシルク足場の特徴を、細孔径分布および表面トポグラフィーの分析のためのSEMによる構造的評価により決定した。SEM分析は、水-シルク足場が、平均細孔径920±50μmである相互に連結された多孔質ネットワークを有することを示した。細孔表面は、非均質の微小孔を含む粗い構造の外観を有した。しかしHFIP-シルク足場は、平均細孔径900±40μmである、相互連結の乏しい多孔質ネットワークを有し、かつ滑らかな表面構造を示した。BMSC(継代2)を、水-シルクスポンジおよびHFIP-シルクスポンジに播種した。HFIP-シルクスポンジよりも、水-シルクスポンジに、より多くの細胞が接着した。水-シルクスポンジは、細胞播種を促進した。かつBMSCは、水-シルクスポンジ全体に均一に分布された。対照的に、HFIP-シルクスポンジ上のBMSCの分布は均一ではなかった。BMSC増殖は、水-シルクスポンジ上で認められた。SEMは、水-シルクスポンジ上でのBMSCの広範囲の増殖、その後最大4週間の増殖を確認した。
細胞足場構築体は、5%CO2雰囲気下、37℃で、骨形成性培地において培養した。構築体は、6ウェルプレートにおいて最大28日間培養した。BMSC水-シルク構築体は、培養後丸くなり始めたが、BMSCHFIP-シルク構築体は当初平坦で、培養後も変化しなかった。水-シルク足場中に形成された組織は、白っぽく、手および外科用鉗子で触ると固かった。しかし、HFIP-シルク足場に播種されたBMSCは、白っぽい組織を形成しなかった。2週間および4週間の標本は、肉眼による試験では有意差は認められなかった。水-シルク足場内のBMSCの均一な細胞分布は、足場の表面上および中心全体にわたる均一なマトリックス染色(生存細胞によるMTT転換の指標)により定性的に明らかであった。しかしHFIP-シルク足場は、構築体の表面に沿った強力な染色、および構築体内部の弱い染色領域を示した。
3Dマトリックスの多孔度は、水-シルクおよびHFIP-シルクの両方において約92%であった。水-シルク足場の圧縮強度および圧縮弾性率は、100±10kPaおよび1300±40kPaであった。HFIP足場のこれらの値は、50±5kPaおよび210±60kPaであった。
BMSCにより生成された骨様組織を特徴付けるために、いくつかの骨形成性の分化および軟骨形成性の分化のマーカー遺伝子の発現を、リアルタイムRT-PCRアッセイ法を用いて定量した。分析した遺伝子は、骨形成性分化マーカーI型コラーゲン(Col I)、アルカリホスファターゼ(ALP)、オステオポンチン(OP)、骨シアロタンパク質(BSP)、および軟骨形成性分化マーカーII型コラーゲン(Col II)を含む。足場の種類間の転写レベル(増幅の直線範囲内でGAPDHに対して標準化)の差異は、有意であった。Col I、ALP、およびOP転写レベルは、HFIP-シルク足場と比べ、水-シルク足場において増加した。水-シルク足場において28日間培養後、Col I、ALPおよびBSPの遺伝子発現は、1日培養後と比べ、各々、190%、1100%、および10500%有意に増加した。しかしOPおよびCol IIの発現は、有意に減少した。BSP発現は、水-シルク足場およびHFIP-シルク足場において、同様に調節された。足場の種類間の差異に統計学的有意性はなかった。
3D水-シルク足場培養条件において、ヒト骨髄幹細胞は、骨芽細胞マーカーを発現した。HFIP-シルク構築体と比べて、Col Iの発現は、水-シルク培養条件下で、2週間培養後、タンパク質レベルの有意な増加を示した。しかしCol Iの発現は両方の条件下で、4週間の培養後に減少した。28日間の培養後、OPの発現は水-シルク構築体において増加した。このタンパク質は2本のバンドを示し、そのうち最も高い分子量は、高度にグリコシル化、硫酸化、またはリン酸化されていると推定された(Singh et al., J. Biol. Chem., 1990,65:18696-18701)。骨の他のタンパク質であるBSPは、水-シルク足場およびHFIP-シルク足場の両方で培養された細胞において発現された。しかしその発現は、28日間培養後、HFIP-シルク構築体において増加した。
これらの標本のヘマトキシリン・エオシン染色を用いる組織学的試験は、それらの立方体または円柱状の形態の骨芽細胞様細胞の割合が、水-シルク構築体における培養期間の延長と共に増加したことを明らかにした。14日間の培養後、ほぼ全ての細孔が、結合組織、線維芽細胞、および立方体の骨芽細胞様細胞により充填された。28日後、これらの細孔は細胞外マトリックス、骨芽細胞様細胞、およびわずかな線維芽細胞様形態の細胞で充填された。しかしHFIP-シルク足場の組織学的切片は、細胞のわずかな分布が存在し、その大半は足場の表面に細胞層を形成していることを明らかにした。28日間の培養後、HFIP-シルク構築体内の細胞の大半は、平坦な線維芽細胞形態を示した。
シルクタンパク質ベースのマトリックス足場は、骨組織エンジニアリングに関して現在関心がもたれている。これらの足場は、PGA-PLAコポリマーおよびコラーゲンのような、他の一般的生分解性合成ポリマーおよび天然ポリマーよりも、より高い力学特性を示した。HFIPを使用して、多孔質シルクフィブロイン材料が調製されてきた。HFIP-シルク足場は、それらの独特な力学特性について知られているが、これらの天然のポリマーは、細胞認識シグナルを欠いているため不充分な細胞接着を生じる。この問題点を克服するために、アルギニン-グリシン-アスパラギン酸(RGD)による表面修飾、および天然の生分解性ポリマーとのハイブリッドを含む、多くのアプローチが開発されてきた。
Claims (46)
- 有機溶媒を含まない、少なくとも10wt%のフィブロイン濃度を有する水性シルクフィブロイン溶液であり、
吸湿性ポリマーに対して少なくとも10wt%の水性フィブロイン溶液を生じるのに十分な時間、溶解したカイコシルクまたはクモシルクのシルクフィブロイン溶液を透析する段階を含む方法によって製造される、水性シルクフィブロイン溶液。 - フィブロイン濃度が少なくとも15wt%、少なくとも20wt%、少なくとも25wt%、または少なくとも30wt%である、請求項1記載の水性シルクフィブロイン溶液。
- 治療用物質をさらに含む、請求項1記載の水性シルクフィブロイン溶液。
- 溶液がシルクフィブロインおよび水からなる、請求項1記載の水性溶液。
- 吸湿性ポリマーが、ポリエチレングリコール、アミラーゼ、およびセリシンからなる群より選択される、請求項1記載の水性シルクフィブロイン溶液。
- 吸湿性ポリマーが、分子量8,000〜10,000g/molのポリエチレングリコール(PEG)である、請求項1記載の水性シルクフィブロイン溶液。
- PEGの濃度が25〜50%である、請求項6記載の水性シルクフィブロイン溶液。
- 請求項1記載の溶液を、線維の形成のために加工する工程を含む、線維を製造する方法。
- 線維をメタノール/水溶液に含浸する工程をさらに含む、請求項8記載の方法。
- 線維を水中で洗浄する工程をさらに含む、請求項9記載の方法。
- 加工が、電気紡糸または湿式紡糸を含む、請求項8記載の方法。
