Claims (20)
- 第二世代キメラ抗原受容体(CAR)をコードする核酸であって、
(a)CD171と特異的に結合することができるリガンド結合ドメインをコードするポリヌクレオチド;
(b)12アミノ酸長から119アミノ酸長の範囲にある長さを有するポリペプチドスペーサーをコードするポリヌクレオチド;
(c)膜貫通ドメインをコードするポリヌクレオチド;および
(d)(i)CD3ζドメインおよび4-1BB共刺激ドメインを含む細胞内シグナル伝達ドメイン、または
(ii)CD3ζドメインおよびCD28共刺激ドメインを含む細胞内シグナル伝達ドメイン
をコードするポリヌクレオチド
を含み:
前記CARを含む細胞を投与された対象が、CD28共刺激ドメインおよび4-1BB共刺激ドメインの両方を含むCARを含む細胞を投与された対象と比較して、向上された生存可能性を有することを特徴とする、核酸。 - 前記細胞内シグナル伝達ドメインがCD3ζドメインおよび4-1BB共刺激ドメインを含む、請求項1に記載の核酸。
- 前記ポリペプチドスペーサーが12アミノ酸長の長さを有し、かつ該ポリペプチドスペーサーがIgG4ヒンジドメインを含んでいてもよい、請求項1または2に記載の核酸。
- 前記ポリペプチドスペーサーが配列番号20、配列番号21、配列番号49、配列番号51および配列番号52のいずれかに示されるアミノ酸配列を含む、請求項3に記載の核酸。
- 前記ポリペプチドスペーサーが119アミノ酸長の長さを有し、かつ該ポリペプチドスペーサーが配列番号59に示されるアミノ酸配列と少なくとも95%の同一性を有するアミノ酸配列を有していてもよい、
請求項1または2に記載の核酸。 - 前記ポリペプチドスペーサーが配列番号59に示されるアミノ酸配列を含む、請求項5に記載の核酸。
- 前記リガンド結合ドメインが一本鎖可変フラグメントを含む、請求項1~6のいずれか一項に記載の核酸。
- 前記リガンド結合ドメインをコードするポリヌクレオチドが、配列番号55の2150~2875番目に示される核酸配列を含む、請求項7に記載の核酸。
- 選択可能なマーカーをコードするポリヌクレオチドをさらに含み、前記選択可能なマーカーが細胞の表面に発現することを特徴とする、請求項1~8のいずれか一項に記載の核酸。
- 前記選択可能なマーカーが、末端切断型EGF受容体(EGFRt)ポリペプチドを含む、請求項9に記載の核酸。
- 請求項1~10のいずれか一項に記載の核酸によりコードされる、キメラ抗原受容体(CAR)。
- 請求項1~10のいずれか一項に記載の核酸を含む、ベクター。
- ウイルスベクターである、請求項12に記載のベクター。
- アデノウイルスベクター、アデノ随伴ウイルスベクター、およびレンチウイルスベクターからなる群から選択される、請求項13に記載のベクター。
- 請求項1~10のいずれか一項に記載の核酸を含む、宿主細胞。
- 前記宿主細胞が、
T細胞前駆細胞;
造血幹細胞;
ナイーブCD8+T細胞、セントラルメモリーCD8+T細胞、エフェクターメモリーCD8+T細胞およびバルクCD8+T細胞からなる群から選択されるCD8+細胞傷害性Tリンパ球;または
ナイーブCD4+T細胞、セントラルメモリーCD4+T細胞、エフェクターメモリーCD4+T細胞およびバルクCD4+T細胞からなる群から選択されるCD4+ヘルパーTリンパ球
である、請求項15に記載の宿主細胞。 - CD171を発現しているがんの治療または抑制において使用するための、請求項15または16に記載の宿主細胞。
- 前記がんが、乳がん、脳がん、大腸がん、腎臓がん、膵臓がん、および卵巣がんからなる群から選択される、請求項17に記載の宿主細胞。
- 請求項15または16に記載の宿主細胞と薬学的に許容される添加剤を含む、医薬組成物。
- 請求項15または16に記載の宿主細胞を製造する方法であって、
(a)細胞に請求項1~10のいずれか一項に記載の核酸を導入すること;ならびに
(b)抗CD3抗体もしくはその抗原結合フラグメント、および/または抗CD28抗体もしくはその抗原結合フラグメントの存在下において前記細胞を培養すること
を含む、方法。
