Claims (12)
1. Kromatografisk separationsfremgangsmåde til genvinding af et polyumættet fedtsyre- (PUFA) produkt fra en foderblanding, der omfatter: (a) oprensning af foderblandingen i et første kromatografisk separationstrin ved anvendelse som eluent af en blanding af vand og et første organisk opløsningsmiddel for at opnå et mellemprodukt; og (b) oprensning af mellemproduktet i et andet kromatografisk separationstrin ved anvendelse som eluent af en blanding af vand og et andet organisk opløsningsmiddel for at opnå PUFA-produktet, hvor det andet organiske opløsningsmiddel adskiller sig fra det første organiske opløsningsmiddel, og hvor det første kromatografiske separationstrin omfatter indføring af foderblanding i et kromatografiapparat med simuleret eller aktuelt bevægeligt leje og det andet kromatografiske separationstrin omfatter indføring af mellemproduktet i et kromatografiapparat med stationært leje.
2. Fremgangsmåde ifølge krav 1, hvor det første og det andet organiske opløsningsmiddel er udvalgt fra alkoholer, ethere, estere, ketoner og nitriler.
3. Fremgangsmåde ifølge krav 2, hvor ketonen er acetone, methylethylketon eller methylisobutylketon (MIBK), fortrinsvis acetone.
4. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor ét af det første og det andet organiske opløsningsmiddel er methanol.
5. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor det andet organiske opløsningsmiddel er methanol.
6. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor det første organiske opløsningsmiddekvand-forhold er fra 99,9:0,1 til 75:25 volumendele, fortrinsvis fra 99,5:0,5 til 80:20 volumendele.
7. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor det andet organiske opløsningsmiddekvand-forhold er fra 99,9:0,1 til 75:25 volumendele, fortrinsvis fra 90:10 til 85:15 volumendele.
8. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor det andet organiske opløsningsmiddel er methanol, og methanol:vand-forholdet er fra 95:5 til 85:15 volumendele, fortrinsvis fra 93:7 til 90:10 volumendele.
9. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor PUFA-produktet er mindst ét ω-3-PUFA eller mindst ét ω-3-PUFA-derivat.
10. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor PUFA-produktet er eicosapentaensyre (EPA), docosahexaensyre (DFIA), EPA-triglycerid, DHA-triglycerid, EPA-ethylester eller DHA-ethylester.
11. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor PUFA-produktet er EPA eller EPA-ethylester.
12. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor PUFA-produktet opnås i det andet separationstrin ved en renhed, der er større end 95 vægt-%, fortrinsvis større end 97 vægt-%, mere fortrinsvis større end 98 vægt-%, endnu mere fortrinsvis større end 98,4 vægt-%.
