Description
- Die vorliegende Erfindung betrifft den 4-Nitroxybutylester der 2-(S)-(6-Methoxy-2-naphthyl)propansäure (Naproxen) mit einem Enantiomeren-Überschuss der (S)-Form von höher als oder gleich 97% und vorzugsweise von höher als oder gleich 98%.
- Es ist im Stand der Technik gut bekannt, dass die enantiomere Form (S) die pharmakologisch aktive Form des oben genannten Produktes ist.
- Im Stand der Technik sind Syntheseverfahren von Nitroxyalkylestern der 2-(S)-(6-Methoxy-2-naphthyl)propansäure bekannt. In WO 98/25918 ist ein Syntheseverfahren von Naproxennitroxyalkylestern beschrieben, die in der Alkylkette einen gesättigten C3-8-Cycloalkylrest enthalten. Im genannten Verfahren werden die Säure oder eines ihrer funktionellen Derivate, z.B. das Chlorid oder Anhydrid, in einem inerten organischen Lösungsmittel mit einem Nitroalkanol umgesetzt, das den oben definierten Cycloalkylrest enthält. Die Reaktion erfolgt in der Gegenwart einer organischen Stickstoffbase, wie z.B. von 4-Dimethylaminopyridin, Morpholin, N-Methylmorpholin oder von Triethylamin. Von der Anmelderin durchgeführte Tests haben belegt, dass es mit diesem Verfahren des Standes der Technik nicht ermöglicht ist, Naproxennitroxyalkylester mit einem Enantiomeren-Überschuss im Bereich von 55 bis 80% zu erhalten, nur mit der spezifischen organischen Base 4-N,N-Dimethylaminopyridin werden 94% erhalten.
- Es bestand somit der Bedarf, Naproxennitroxyalkylester mit einem höheren Enantiomeren-Überschuss von mindestens 97% und vorzugsweise gleich oder höher als 98% zu erhalten.
- Gegenstand der vorliegenden Erfindung ist der 4-Nitroxybutylester der 2-(S)-(6-Methoxy-2-naphthyl)propansäure, welcher einen Überschuss der Enantiomeren von höher als oder gleich 97% und vorzugsweise von höher als oder gleich 98% aufweist. Diese Verbindung wird mit einem Verfahren erhalten, das dadurch gekennzeichnet ist, dass ein Halogenid der 2-(S)-(6-Methoxy-2-naphthyl)propansäure der Formel A-Hal, worin A der Acylrest der genannten Säure ist, in einem inerten organischen Lösungsmittel mit einem aliphatischen Nitroxyalkanol HO-Y-ONO2, worin Y ein lineares C4-Alkylen ist, in der Gegenwart einer organischen Base umgesetzt wird, um den entsprechenden 4-Nitroxybutylester der 2-(S)-(6-Methoxy-2-naphthyl)propan säure der Formel A-O-Y-ONO2 zu ergeben, worin A und Y wie oben definiert sind.
- Die Menge des aliphatischen Nitroxyalkohols liegt auf molarer Basis im Bereich von 1 bis 2 und vorzugsweise von 1,2 bis 1,5, bezogen auf diejenige des Säurehalogenids.
- Unter den anorganischen Basen sind Hydroxide, Oxide, Carbonate und Bicarbonate, Silikate und Alumosilikate der Alkali- und Erdalkalimetalle oder Hydroxide, Oxide, Carbonate und Bicarbonate von Metallen, die zur Gruppe IIB gehören, vorzugsweise Zink, oder zu den Gruppen IIIa oder IVa gehören, vorzugsweise Zinn, gemeint.
- Die Menge der anorganischen Base im molaren Verhältnis mit der Säurehalogenid-Menge liegt im Allgemeinen im Bereich von 1 bis 2 und vorzugsweise von 1,2 bis 1,5.
- Unter dem inerten organischen Lösungsmittel sind gemäß der vorliegenden Erfindung aromatische Kohlenwasserstoffe, wie z.B. Toluol und Xylol, chlorierte oder fluorierte organische Lösungsmittel, z.B. Methylenchlorid und Chlorbenzol, aliphatische Ester, z.B. C1-4-Säureester mit C1-5-Alkoholen, wie z.B. Ethylacetat und Butylacetat, usw. gemeint.
- Die Lösungsmittelmenge ist nicht kritisch, und es werden im Allgemeinen 1 bis 10 Volumina Lösungsmittel und vorzugsweise 2 bis 5 Volumina verwendet, bezogen auf das Gewicht des Säurehalogenids.
- Die Reaktion wird bei einer Temperatur im Bereich von –20 bis 50 und vorzugsweise von 0 bis 20°C durchgeführt.
- Der 4-Nitroxybutylester der 2-(S)-(6-Methoxy-2-naphthyl)propansäure wird am Ende der Reaktion nach Zugabe von Wasser zur organischen Phase, Trennung der Phasen und nach Verdampfen des Lösungsmittels gewonnen. Im Bedarfsfall kann eine weitere Reinigung durch Chromatografie an einer Kieselgel-Säule zur Erhöhung des Produkt-Titers durchgeführt werden.
- Alternativ dazu, kann die Verbindung auch durch Kristallisation aus einem geeigneten Lösungsmittel gereinigt werden.
- Die aliphatischen Nitroxyalkohole können gemäß im Stand der Technik bekannten Verfahren hergestellt werden. Siehe z.B. Gazzetta Chim. It. 1987, 117, 173, und WO 98/25 918.
- Die Anmelderin hat herausgefunden, dass es in überraschender Weise durch die Verwendung anorganischer Basen ermöglicht ist, den Überschuss der Enantiomeren von Naproxen-4-nitroxybutylester gegenüber den Verfahren des Standes der Technik zu verbessern, in denen, wie gesehen, organische Basen bei hohen Ausbeuten angewandt werden, wie oben erwähnt.
- Die folgenden Beispiele dienen einer weiteren Erläuterung der Erfindung und sollen sie nicht darauf einschränken.
- Beispiel 1 (Vergleich)
- Herstellung des 4-Nitroxybutylesters der 2-(S)-(6-Methoxy-2-naphthyl)propansäure gemäß WO 98/25 918
- Eine Mischung aus der 2-(S)-(6-Methoxy-2-naphthyl)propansäure (0,32 g, 1,4 mmol), aus 4-N,N-Dimethylaminopyridin (16 mg, 0,13 mmol) und aus 4-Nitroxybutan-1-ol (0,34 g, 2,5 mmol) in Dichlormethan (6 mL) wird bei einer Temperatur im Bereich von 0 bis 5°C unter Rühren zu einer Lösung von N,N'-Dicyclohexylcarbodiimid (0,29 g, 1,4 mmol) in Dichlormethan (6 mL) gegeben. Die Mischung wird bei der gleichen Temperatur 3 h lang gerührt, worauf das Lösungsmittel unter Vakuum verdampft wird. Der Rückstand wird durch Chromatografie an einer Kieselgel-Säule (Eluierungsmittel: Dichlormethan) gereinigt, um den 4-Nitroxybutylester der 2-(S)-(6-Methoxy-2-naphthyl)propansäure (0,41 g, 1,19 mmol, Ausbeute = 85%) in der Form eines Öls zu ergeben. HPLC-Reinheit: 98%

- Zu einer Lösung von 4-Nitroxybutan-1-ol (2,0 g, 14,8 mmol) in Dichlormethan (20 mL), gekühlt bei 0 bis 5°C, wird Kaliumcarbonat (3,21 g, 23,2 mmol) unter Rühren gegeben.
- Zu der Mischung wird eine Lösung von 2-(S)-(6-Methoxy-2-naphthyl)propansäurechlorid (3,86 g, 15,5 mmol; Enantiomeren-Überschuss: 98%) in Dichlormethan (22 mL) gegeben, wobei die Temperatur im Bereich von 10 bis 15°C gehalten wird. Nach Beendigung der Zugabe wird die Temperatur angehoben und 10 h lang bei einem Wert im Bereich von 15 bis 20°C gehalten, worauf die Lösung filtriert wird. Das Lösungsmittel wird unter Vakuum verdampft. Der Rückstand wird durch Chromatografie an einer Kieselgel-Säule (Eluierungsmittel: Dichlormethan) gereinigt, um den 4-Nitroxybutylester der 2-(S)-(6-Methoxy-2-naphthyl)propansäure (4,4 g, 12,6 mmol, Ausbeute = 85%) in der Form eines Öls zu ergeben. HPLC-Reinheit: 99%

- Beispiel 2 wird mit Toluol als Lösungsmittel wiederholt. Die Nitroxyester-Ausbeute beträgt 76%, die (HPLC)-Reinheit > 99%. Der Entantiomeren-Überschuss ist gleich 98%.
- Beispiel 4
- Beispiel 2 wird wiederholt, wobei aber als Base Calciumcarbonat verwendet wird. 4,6 g, gleich 13,3 mmol Nitroxyester (Ausbeute = 90%) werden erhalten. HPLC-Reinheit > 99%, Enantiomeren-Überschuss: 98%.
- Beispiel 5
- Beispiel 2 wird wiederholt, wobei aber als Base Calciumalumosilikat verwendet wird, 4,6 g, gleich 13,3 mmol Nitroxyester (Ausbeute = 90%) werden erhalten, HPLC-Reinheit > 99%, Enantiomeren-Überschuss: 98%.
- Beispiel 6
- Zu einer Lösung von 4-Nitroxybutan-1-ol (2,0 g, 14,8 mmol) in Dichlormethan (20 mL), gekühlt bei einer Temperatur im Bereich von 0 bis 5°C, wird Kaliumcarbonat (3,21 g, 23,2 mmol) unter Rühren gegeben.
- Zu der Mischung wird eine Lösung von 2-(S)-(6-Methoxy-2-naphthyl)propansäurechlorid (3,86 g, 15,5 mmol, Enantiomeren-Überschuss: 98%) in Dichlormethan (22 mL) gegeben, wobei die Temperatur im Bereich von 10 bis 15°C gehalten wird. Nach Beendigung der Zugabe wird die Temperatur auf einen Wert im Bereich von 15 bis 20°C 10 h lang angehoben, worauf die Lösung filtriert wird. Wasser (1 mL) und N,N-Dimethylformamid (2 mL) werden zur Lösung gegeben, worauf das Ganze bei Raumtemperatur 3 h lang gerührt wird. Am Ende wird die organische Phase abgetrennt, mit Wasser gewaschen und durch eine Kaliumcarbonat-Schicht filtriert. Das Lösungsmittel wird unter Vakuum verdampft, und es werden 4,1 g, gleich 11,8 mmol Ester (Ausbeute = 80%) in der Form eines Öls erhalten. HPLC-Reinheit > 99%, Enantiomeren-Überschuss: 98%.
- Beispiel 7 (Vergleich)
- Beispiel 2 wird wiederholt, wobei aber als Base Triethylamin verwendet wird. Die nach der Reaktion erhaltene Mischung wird zur Ermittlung des Enantiomeren-Überschusses analysiert, welcher 80% beträgt.
- Beispiel 8 (Vergleich)
- Beispiel 2 wird wiederholt, wobei aber als Base Diisopropylethylamin verwendet wird. Die nach der Reaktion erhaltene Mischung wird zur Ermittlung des Enantiomeren-Überschusses analysiert, welcher 76% beträgt.
- Beispiel 9 (Vergleich)
- Beispiel 2 wird wiederholt, wobei aber als Base N,N-Methylmorpholin verwendet wird. Die nach der Reaktion erhaltene Mischung wird zur Ermittlung des Enantiomeren-Überschusses analysiert, welcher 56% beträgt.

- 9-Nitroxybutylester der 2-(S)-(6-Methoxy-2-naphthyl)propansäure mit einem Überschuss der Enantiomeren der (S)-Form von höher als oder gleich 97%.
