Description
Die Erfindung betrifft den Gegenstand der Ansprüche.
Verschiedene Carbostyrilverbindungen mit hypotensiven,
Blutplättchenkoagulation inhiberenden oder
antiallergischen Eigenschaften sind aus den JP-OSen
130 589/79, 135 785/79, 138 585/79, 141 785/79,
76 872/80, 49 319/80, 53 283/80, 53 284/80 und 83 781/80
bekannt.
Weiterhin werden in EP-A1-7 525 und EP-A1-8 014
Isochinolinverbindungen mit Herz- und Kreislaufaktivität
beschrieben.
Die Carbostyrilverbindungen der Erfindung sind
jedoch strukturverschieden von den üblichen Carbostyril-
und Isochinolinverbindungen.
Die Verbindungen der Formel (I) und deren pharmazeutisch
annehmbaren Salze haben eine
herzmuskelkontraktionsstimulierende Wirkung oder eine
positive ionotrope Wirkung und eine
coronarblutflußerhöhende Aktivität und sind als
cardiotonische Mittel zum Behandeln von Herzkrankheiten,
wie Kongestion-Herzversagen geeignet. Sie sind vorteilhaft,
weil sie nicht oder nur wenig, falls überhaupt, den
Pulsschlag erhöhen.
Die pharmakologische Aktivität der erfindungsgemäßen
Verbindungen wurde in nachfolgender Weise festgestellt.
Versuche wurden an erwachsenen Mischlingshunden beiderlei
Geschlechts durchgeführt. Die sinoatrialen
Knotenzubereitungen wurden von Hunden mit einem Gewicht
von 8 bis 13 kg, die mit Pentobarbitalnatrium
(30 mg/kg i. v.) anästhesiert worden waren und deren Heparinnatrium
(1000 U/kg i. v.) verabreicht worden war und
die dann ausgeblutet wurden, erhalten. Die Zubereitungen
bestanden im wesentlichen aus dem rechten Atrium
und wurden in eine kalte Tyrodes-Lösung gegeben. Die
Zubereitungen wurden in eine gläserne Wasserummantelung,
die bei etwa 38°C gehalten wurde, gegeben und
durch die rechte, mit einer Kanüle versehene Koronararterie
mit Blut von einem Spenderhund bei konstantem
Druck von 100 mmHg kreuzzirkuliert. Die als Spender
verwendeten Hunde hatten ein Körpergewicht von 18 bis
27 kg und waren mit Pentobarbitalnatrium (30 mg/kg i. v.)
anästhesiert. Heparinnatrium wurde in einer Dosis von
1000 U/kg i. v. verabreicht. Die vom rechten Atrium
entwickelte Spannung wurde mittels eines Spannungsmessers
gemessen. Das rechte Atrium wurde mit einem
Gewicht von etwa 1,5 g belastet. Die Sinusrate wurde
cardiotachometrisch mittels der an dem rechten Atrium
entwickelten Spannung gemessen. Der Blutdurchfluß
durch die rechte Koronararterie wurde mittels eines
elektromagnetischen Durchflußmessers gemessen. Es
wurden Aufzeichnungen der entwickelten Spannung, der
Sinusrate und des Blutdurchflusses mittels eines Tintenschreibers
auf Registrierstreifen durchgeführt.
Einzelheiten der Zubereitungen werden beschrieben von
Chiba et al (Japan. J. Pharmacol., 25, 433-439, 1975;
Naunyn-Schmiedberg′s Arch. Pharmacol. 289, 315-325,
1975). Die Verbindungen wurden innerhalb von 4 Sekunden
in Mengen von 10 bis 30 µl intraarteriell injiziert.
Die ionotrope Wirkung der Verbindungen wird
als Prozentsatz der entwickelten Spannung vor der Injizierung
der Verbindungen ausgedrückt. Die Wirkung
der Verbindungen auf die Sinusrate (Schläge/Minute)
oder den Blutdurchfluß (mm/Minute) wird als Unterschied
zwischen den Werten vor und nach dem Injizieren
der Verbindung ausgedrückt.
Die erzielten Ergebnisse werden in der nachfolgenden
Tabelle 1 gezeigt.

Die akute Toxizität wurde an männlichen Ratten bei oraler
Verabreichung bestimmt.
Die erfindungsgemäßen Verbindungen der allgemeinen Formel
(I) können durch verschiedene Verfahren hergestellt werden.
Diese Verfahren werden im Patentanspruch 13 angegeben.
Spezielle Ausführungsformen werden in den
Referenzbeispielen und Beispielen gezeigt.
Die erfindungsgemäßen Verbindungen der allgemeinen Formel
(I) bilden mit pharmazeutisch annehmbaren Säuren Salze.
Pharmazeutisch annehmbare Säuren, die zur Salzbildung
verwendet werden können, sind verschiedene anorganische
Säuren, z. B. Salzsäure, Phosphorsäure,
Bromwasserstoffsäure, und organische Säuren, wie Oxalsäure,
Maleinsäure, Fumarsäure, Apfelsäure, Weinsäure,
Zitronensäure und Benzoesäure.
Die Verbindungen der allgemeinen Formel (I) und deren Salze
können aus den jeweiligen Reaktionsmedien nach Beendigung
der Umsetzung gewonnen und in üblicher Weise gereinigt
werden, z. B. durch Lösungsmittelextraktion, nach der
Verdünnungsmethode, durch Ausfällen, Umkristallisieren
durch Säulenchromatografie oder durch präparative
Dünnschichtchromatografie.
Für den Fachmann ist es offensichtlich, daß die Verbindungen
der allgemeinen Formel (I) in optisch aktiven Formen
vorkommen können und solche optischen Isomere sind in die
Erfindung eingeschlossen.
Die Verbindungen der allgemeinen Formel (I) werden in
üblicher Weise zu Arzneimitteln formuliert. Geeignete
Dosierungsformen sind Tabletten, Pillen, Pulver,
Flüssigzubereitungen, Suspensionen, Emulsionen, Granulate,
Kapseln, Suppositorien und injizierbare Zubereitungen
(Lösungen, Suspensionen). Es werden die üblichen Träger
verwendet.
Die Dosierung erfolgt entsprechend den Symptomen. Im
allgemeinen liegt eine bevorzugte Dosis bei 1 bis 10 mg/kg
Körpergewicht pro Tag. Vorteilhaft ist es, wenn der aktive
Bestandteil in einer Einzeldosis in einer Menge von 50 bis
250 mg vorliegt.
Die Erfindung wird in den Referenzbeispielen und Beispielen
beschrieben.
8-Hydroxy-3,4-dihydrocarbostyril (20 g) wurden in Essigsäureanhydrid
(100 ml) suspendiert. Schwefelsäure (5
Tropfen) wurden zu der gebildeten Suspension gegeben,
und die Mischung wurde 1,5 Stunden bei 80°C gerührt.
Zum Reaktionsgemisch wurde Essigsäure (100 ml) gegeben
und die Mischung wurde eisgekühlt und dazu wurde unter
Rühren eine Lösung von 10,1 ml konzentrierte Salpetersäure
(d=1,38) in 30 ml Essigsäureanhydrid gegeben.
Das Rühren wurde während einer Nacht bei Raumtemperatur
fortgesetzt. Das Reaktionsgemisch wurde zu Eiswasser
zum Ausfällen der Kristalle gegossen und diese wurden
dann durch Filtrieren gesammelt und ergaben 21,74 g
6-Nitro-8-acetoxy-3,4-dihydrocarbostyril.

Salzsäure (200 ml) wurde zu 6-Nitro-8-acetoxy-3,4-
dihydrocarbostyril (20 g) gegeben und die Mischung
wurde 4 Stunden unter Rückfluß gehalten. Nach dem
Kühlen mit Eis wurden die ausgefallenen Kristalle
durch Filtrieren gesammelt und aus Methanol umkristallisiert,
wobei man 13,76 g 6-Nitro-8-hydroxy-3,4-
dihydrocarbostyril erhielt. F: 270°C, hellgelbe Schuppen.
6-Nitro-8-hydroxy-3,4-dihydrocarbostyril (10 g) und
Kaliumcarbonat (13,3 g) wurden mit Aceton (70 ml) und
Wasser (70 ml) vermischt und die Mischung wurde unter
tropfenweiser Zugabe von 12,1 g Dimethylsulfat rückflußbehandelt.
Nach 4stündiger Rückflußbehandlung
wurde das Reaktionsgemisch gekühlt und die ausgefallenen
Kristalle wurden durch Filtrieren gesammelt und
mit Wasser gewaschen, wobei man 8,22 g 6-Nitro-8-
methoxy-3,4-dihydrocarbostyril erhielt. F: 230°C, gelbe
Nadeln.
6-Nitro-8-hydroxy-3,4-dihydrocarbostyril (2 g) und
5% Palladium/Kohle (0,2 g) wurden zu Ethanol (50 ml)
gegeben und eine katalytische Reduktion wurde mit
einem Wasserstoffgasdruck von 3 kg/cm³ während einer Stunde
durchgeführt. Dann wurde das Reaktionsgemisch filtriert
und das Filtrat konzentriert. Der Rückstand
wurde aus Benzol umkristallisiert, wobei man 1,7 g
6-Amino-8-methoxy-3,4-dihydrocarbostyril erhielt.
F: 157-158°C, farblose Nadeln.
Zu einer Mischung aus 6-Amino-8-hydroxy-3,4-dihydrocarbostyril
(12 g), konzentrierter Salzsäure (16 ml)
und Wasser (50 ml) wurden Eis (30 g) und Natriumnitrit
(5 g) und Wasser (20 ml) allmählich zu der sich bildenden
Mischung unter Eiskühlung gegeben. Nach Beendigung
der Zugabe wurde die Mischung eine Stunde bei 0 bis
5°C umgesetzt. Weiterhin wurde eine erwärmte Lösung
aus Acetaldehyd-semicarbazid (10 g) in Wasser (50 ml)
und dann Natriumsulfit (0,25 g) und Kupfersulfat (3,2 g)
zu einer Lösung von Natriumacetat (34 g) in Wasser
(40 ml) gegeben, und die innere Badtemperatur wurde
auf eine Temperatur von 10 bis 20°C eingestellt. Die
Lösung wurde gerührt und unter die Oberfläche wurde
allmählich eine Lösung zugegeben, die man hergestellt
hatte durch Zugabe einer Lösung von Natriumacetat
(26 g) in Wasser (46 ml) zu der oben erwähnten Diazoniumsalzlösung.
Nach 2stündigem Rühren ließ man
die Reaktion über Nacht stehen. Der gebildete Niederschlag
wurde durch Filtrieren gesammelt und mit Wasser
gewaschen. Dazu wurden 200 ml 2N Salzsäure gegeben und
die Mischung wurde 2 Stunden rückflußbehandelt. Nach
dem Abkühlen wurde das Reaktionsgemisch 2mal mit
Chloroform (150 ml) extrahiert und die Chloroformschicht
wurde mit Wasser gewaschen. Die Lösung wurde
durch eine kurze, mit kieselgelaktivierter Kohle beladene
Kolonne filtriert. Diese Kolonne wurde mit 500 ml
Chloroform gewaschen. Die beiden Chloroformschichten
wurden vereint und das Chloroform abgedampft. Der Rückstand
wurde aus Benzol umkristallisiert, wobei man
5,1 g 6-Acetyl-8-methoxy-3,4-dihydrocarbostyril erhielt.
F: 150°C, hellgelbe Nadeln.
Zu einer Lösung von 6-Acetyl-8-methoxy-3,4-dihydrocarbostyril
(3,1 g) in Chloroform (30 ml) wurde tropfenweise
eine Lösung aus Brom (2,26 g) in Chloroform
(20 ml) unter Rühren bei Raumtemperatur gegeben. Nach
Beendigung der Zugabe wurde die Mischung 30 Minuten
bei Raumtemperatur gerührt und anschließend wurde das
Reaktionsgemisch konzentriert und der Rückstand wurde
aus Methanol umkristallisiert, wobei man 3,2 g (6-(α-
Bromoacetyl)-8-methoxy-3,4-dihydrocarbostyril erhielt.
F: 206-207°C, farblose Nadeln.
Zu einer Lösung aus Aluminiumchlorid (44 g) und α-
Bromopropionylchlorid (43 g) in Kohlendisulfid (150 ml)
wurde portionsweise 3,4-Dihydrocarbostyril (8 g) unter
Rückfluß und Rühren gegeben. Nach Beendigung der
Zugabe wurde die Mischung unter Rühren 2 Stunden
rückflußbehandelt. Nach dem Abkühlen wurde die Mischung
zu Eiswasser gegossen und die ausgefallenen
Kristalle wurden durch Filtrieren gesammelt und mit
Wasser gewaschen. Beim Umkristallisieren aus Ethanol
erhielt man 14,2 g 6-(α-Brompropionyl)-3,4-dihydrocarbostyril.
F: 192-193°C, hellgelbe Schuppen.
Zu einer Lösung aus Aluminiumchlorid (80 g) und Chloroacetylchlorid
(72 g) in Kohlendisulfid (200 ml) wurde
portionsweise innerhalb von 20 Minuten eine Suspension
von 3,4-Dihydrocarbostyril (14,7 g) in Kohlendisulfid
(100 ml) unter Rühren und Rückfluß gegeben.
Nach Beendigung der Zugabe wurde die Mischung 2 Stunden
unter Rückfluß gerührt. Nach dem Abkühlen wurde das
Reaktionsgemisch in Eiswasser gegossen und die ausgefallenen
Kristalle wurden durch Filtrieren gesammelt,
mit Wasser gewaschen und aus Ethanol umkristallisiert,
wobei man 20 g 6-Chloroacetyl-3,4-dihydrocarbostyril
erhielt. F: 230-231°C, farblose Nadeln.
In gleicher Weise wie in Referenzbeispiel 8, wobei
jedoch Carbostyril anstelle von 3,4-Dihydrocarbostyril
verwendet wurde, erhielt man 6-Chloroacetylcarbostyril.
F: 275-277°C, hellgrüne Nadeln.
8-Methyl-3,4-dihydrocarbostyril (60,1 g) wurde in
Kohlendisulfid (100 ml) suspendiert und α-Bromopropionylbromid
(35 g) wurde zu der Suspension unter Rühren und
Kühlen mit Eiswasser gegeben. Dann wurde wasserfreies
Aluminiumchlorid (30 g) portionsweise zu der Mischung
gegeben und dann 3 Stunden rückflußbehandelt. Das
Kohlendisulfid wurde unter vermindertem Druck abgedampft
und der Rückstand wurde durch Zugabe von Eiswasser
(500 ml) zersetzt.
Ein teeriges Produkt fiel aus und wurde abgetrennt und
mit Wasser gewaschen. Eine geringe Menge Methanol wurde
zu dem Produkt gegeben unter Ausbildung von Kristallen.
Die Kristalle wurden durch Filtrieren gesammelt und
aus Methanol umkristallisiert, wobei man 14,3 g 8-Methyl-
6-α-bromopropionyl-3,4-dihydrocarbostyril erhielt.
F: 232,5-233,5°C, farblose Nadeln.
Zu einer Lösung von α-Bromopropionylbromid (60 g)
und Aluminiumchlorid (40 g) in Kohlendisulfid (100 ml)
wurde 8-Chloro-3,4-dihydrocarbostyril (10 g) gegeben.
Nach 5stündigem Behandeln der Mischung unter Rückfluß
wurde Kohlendisulfid abgedampft und dann wurde
5 Stunden auf 70 bis 80°C erwärmt. Das Reaktionsgemisch
wurde zu Eiswasser gegossen und über Nacht stehen gelassen.
Die Mischung wurde mit Chloroform extrahiert
und die Chloroformschicht wurde mit Wasser gewaschen,
getrocknet und mit Aktivkohle behandelt und anschließend
wurde das Chloroform abgedampft. Der Rückstand
wurde mit Diethylether gewaschen und dann durch Filtrieren
gesammelt. Der Rückstand ergab nach der TLC
einheitliche rohe Kristalle. Die so erhaltenen Kristalle
wurden aus Methanol umkristallisiert, wobei man 12 g
8-Chloro-6-α-bromopropionyl-3,4-dihydrocarbostyril
erhielt. F: 180-182°C, hellgelbe Nadeln.
Zu einer Lösung aus 8-Hydroxycarbostyril (27 g) und
Chloroacetylchlorid (37 ml) in Nitrobenzol (250 ml)
wurde Aluminiumchlorid (85 g) portionsweise gegeben
und die Mischung wurde 20 Stunden bei 70°C gerührt.
Nach Zugabe von 10%iger Salzsäure (500 ml) wurde
Nitrobenzol durch Wasserdampfdestillation abgetrennt.
Nach dem Kühlen wurden die gebildeten Kristalle durch
Filtrieren gesammelt, mit 300 ml heißem Wasser gewaschen
und aus Methanol umkristallisiert, wobei man
4,0 g 5-Chloroacetyl-8-hydroxycarbostyril erhielt.
F: 285-287°C (Zersetzung), hellgelbe Kristalle.
6-(α-Bromopropionyl)-1-methyl-3,4-dihydrocarbostyril
(5 g), 2-Aminopyridin (4,77 g) und Acetonitril (20 ml)
wurden 1,5 Stunden unter Rückfluß behandelt. Das Reaktionsgemisch
wurde mit Eiswasser gekühlt und die ausgefallenen
Kristalle wurden durch Filtrieren gesammelt.
Die Kristalle wurden in Aceton gelöst und die Lösung
wurde durch Zugabe von 48%iger Bromwasserstoffsäure
auf einen pH von etwa 1 eingestellt. Die gebildeten
Kristalle wurden durch Filtrieren gesammelt und aus Wasser
umkristallisiert, wobei man 5,68 g 6-(3-methyl-
imidazo[1,2-a]pyridin-2-yl)-1-methyl-3,4-dihydrocarbostyrilmonohydro-bromid
erhielt.

(4 g), 2-Aminopyridin (3,57 g) und Acetonitril (20 ml)
wurden 3 Stunden rückflußbehandelt. Das Reaktionsgemisch
wurde zur Trockne konzentriert und der Rückstand
(öliges Produkt) mit Wasser gewaschen. Dann wurde
der Rückstand in Aceton gelöst und zu der Lösung
wurde 48%ige Bromwasserstoffsäure unter Einstellung
des pH-Wertes auf etwa 1 gegeben und die sich bildenden
Kristalle wurden durch Filtrieren gesammelt. Die
Rohkristalle wurden aus Wasser umkristallisiert, wobei
man 2,74 g 8-Chloro-6-(3-methylimidazol[1,2-a]pyridin-
2-yl)-3,4-dihydrocarbostyrilmonohydrobromid-hemihydrat
erhielt.

Zu einer Suspension von 6-(Imidazo[1,2-a]pyridin-2-yl)-
3,4-dihydrocarbostyril (5 g) in Dioxan (50 ml) wurde
2,3-Dichlor-5,6-dicyan-1,4-benzochinon (6,47 g) unter Rühren bei 70°C gegeben. Das Reaktionsgemisch
wurde 5 Stunden unter Rühren erhitzt. Nach
Beendigung der Reaktion wurde das Lösungsmittel abgedampft.
Der Rückstand wurde durch Zugabe von Chloroform
und 0,5N Natriumhydroxid extrahiert. Die Chloroformschicht
wurde mit 0,5N Natriumhydroxid gewaschen, dann mit
Eiswasser gewaschen und getrocknet. Nach dem Abdampfen
des Chloroforms wurde der erhaltene Rückstand isoliert
und durch Kieselgel-Säulenchromatographie gereinigt.
Das gebildete ölige Produkt wurde in Aceton gelöst
und die Lösung wurde auf einen pH von etwa 1 mit
konzentrierter Salzsäure eingestellt. Die gebildeten
Kristalle wurden durch Filtrieren gesammelt. Die erhaltenen
Rohkristalle wurden aus Wasser umkristallisiert,
wobei man 2,4 g 6-(Iimidazo[1,2-a]pyridin-2-yl)-carbostyril-
monohydrochlorid erhielt.

Zu einer Lösung von 8-Methyl-6-(3-methylimidazo[1,2-a]
pyridin-2-yl)-carbostyril (2 g) in 40 ml Methanol wurde
10% Palladium/Kohle (0,2 g) gegeben und die Mischung
wurde einer katalytischen Reduktion von 2 bis 3 kg/cm²
unterworfen. Nach Beendigung der Reaktion wurde
der Katalysator abfiltriert und das Filtrat zur Trockne
konzentriert. Der Rückstand wurde in Aceton gelöst und
die Lösung wurde auf einen pH von etwa 1 mit konzentrierter
Salzsäure eingestellt. Die ausgefallenen Kristalle
wurden durch Filtrieren gesammelt. Die so gebildeten
Rohkristalle wurden aus Ethanol umkristallisiert,
wobei man 1,2 g 8-Methyl-6-(3-methylimidazo[1,2-a]
pyridin-2-yl)-3,4-dihydrocarbostyrilmonohydrochlorid-
monohydrat erhielt.

Zu einer Lösung von 6-(3-Methylimidazo[1,2-a]pyridin-
2-yl)-3,4-dihydrocarbostyril (3 g) in Dimethylformamid
(50 ml) wurde 50%iges öliges Natriumhydrid (590 mg)
gegeben und die Mischung wurde 2 Stunden bei Raumtemperatur
gerührt. Nach Zugabe von Methyljodid (1,85 g)
wurde die Mischung weitere 3 Stunden bei Raumtemperatur
gerührt. Nach Beendigung der Umsetzung wurde das Dimethylformamid
abgedampft. Chloroform und 0,5N Natriumhydroxid
wurden zum Extrahieren des Rückstands zugegeben.
Die Chloroformschicht wurde gründlich mit Wasser
gewaschen und getrocknet und anschließend wurde
Chloroform abgedampft. Der Rückstand wurde durch Kieselgelsäulenchromatografie
gereinigt und das so erhaltene
ölige Produkt in Aceton gelöst. Die Lösung wurde durch
Zugabe von 48%iger Bromwasserstoffsäure auf einen pH
von etwa 1 eingestellt und die gebildeten Kristalle
wurden durch Filtrieren gesammelt. Die so erhaltenen
Rohkristalle wurden aus Wasser umkristallisiert, wobei
man 2,9 g 6-(3-Methylimidazo[1,2-a]pyridin-2-yl)-1-
methyl-3,4-dihydrocarbostyrilmonohydrobromid erhielt.

(5 g), 2-Aminopyridin (4,33 g) und Acetonitril (20 ml)
wurden 6 Stunden unter Rückfluß umgesetzt. Das Reaktionsgemisch
wurde zur Trockne konzentriert und der
Rückstand wurde durch Zugabe von Wasser kristallisiert.
Die Kristalle wurden durch Filtrieren gesammelt und
mit Wasser gewaschen. Die Kristalle wurden in Aceton
gelöst und die Lösung wurde auf einen pH von 1 bis 2
durch Zugabe von 48%iger Bromwasserstoffsäure eingestellt.
Die gebildeten Kristalle wurden durch Filtrieren
gesammelt. Die so erhaltenen Rohkristalle wurden
aus Methanol-Ether umkristallisiert, wobei man 3,73 g
5-(3-Ethylimidazo[1,2-a]pyridin-2-yl)-8-methoxy-3,4-
dihydrocarbostyrilmonohydrobromid erhielt.

5-Chloroacetyl-8-methoxycarbostyril (5 g), 2-Amino-
5-picolin (6,45 g) und Acetonitril (40 ml) wurden 3
Stunden unter Rückfluß umgesetzt und das Umsetzungsgemisch
wurde mit Eiswasser gekühlt. Die gebildeten
Kristalle wurden durch Filtrieren gesammelt. Die Kristalle
wurden in Aceton-Methanol suspendiert und die
Suspension wurde durch Zugabe von konzentrierter
Salzsäure auf einen pH von etwa 1 eingestellt. Die
gebildeten Kristalle wurden durch Filtrieren gesammelt
und aus Methanol umkristallisiert, wobei man 4,0 g
5-(7-Methylimidazo[1,2-a]pyridin-2-yl)-8-methoxycarbostyril-
monohydrochlorid-3/2-hydrat erhielt.

bei Raumtemperatur tropfenweise eine Lösung von Brom
(2,22 g) in Essigsäure (5 ml) gegeben. Nach Beendigung
der Zugabe wurde die Mischung 3 Stunden gerührt.
Die ausgefallenen Kristalle wurden durch Filtrieren
gesammelt und mit Diethylether gewaschen. Die erhaltenen
Kristalle wurden in Aceton gelöst und die Lösung
wurde durch Zugabe von 48%iger Bromwasserstoffsäure
auf einen pH von etwa 1 eingestellt. Die ausgefallenen
Kristalle wurden durch Filtrieren gesammelt und aus
Methanol-Ether umkristallisiert, wobei man 5 g 5-(3-
Bromoimidazo[1,2-a]pyridin-2-yl)-8-methoxycarbostyril-
monohydrobromid-monohydrat erhielt.

Zu einer Suspension von 5-(Imidazo[1,2-a]pyridin-2-yl)-
8-methoxy-3,4-dihydrocarbostyril (2,5 g) in 300 ml
Wasser wurden 1,71 ml konzentrierte Salzsäure gegeben.
Die Mischung wurde erwärmt und die Lösung wurde auf
50°C gekühlt und dann wurden 10 ml einer wäßrigen Lösung,
in welcher 0,65 g Natriumnitrit gelöst waren,
tropfenweise zu der Lösung bei 50°C unter Rühren zugegeben.
Nach Beendigung der Zugabe wurde die Umsetzung
2 Stunden weitergeführt und das Reaktionsgemisch wurde
über Nacht bei Raumtemperatur stehen gelassen. Die
ausgefallenen Kristalle wurden durch Filtrieren gesammelt
und aus Methanol umkristallisiert, wobei man 1,9 g
5-(3-Nitrosoimidazo[1,2-a]pyridin-2-yl)-8-methoxy-
3,4-dihydrocarbostyril erhielt.
F: 236,5-238°C (Zersetzung), grüne

Zu einer Suspension von 5-(3-Nitrosoimidazo[1,2-a]
pyridin-2-yl)-8-methoxy-3,4-dihydrocarbostyril (20,85 g)
in Methanol (500 ml) wurden 12 ml konzentrierte Salzsäure
zugegeben und die Suspension sauer eingestellt.
10%ige Palladium/Kohle (2 g) wurde zu der Suspension
gegeben und die katalytische Umsetzung wurde bei
Raumtemperatur unter Atmosphärendruck durchgeführt.
Nach Beendigung der Umsetzung wurde Wasser zum Auflösen
zu dem Reaktionsgemisch gegeben und die Mischung
wurde erwärmt. Nach dem Abfiltrieren des Katalysators
wurde das Filtrat zur Trockne konzentriert. Zum Rückstand
wurde Aceton gegeben und die ausgefallenen Kristalle
wurden durch Filtrieren gesammelt und aus Wasser
umkristallisiert, wobei man 21,20 g 5-(3-Aminoimidazo
[1,2-a]pyridin-2-yl)-8-methoxy-3,4-dihydrocarbostyril-
monohydrochlorid-hemihydrat erhielt.
F: 254,5-257,0°C (Zersetzung), hellgelbe

Zu einer Lösung von 5-(Imidazo[1,2-a]pyridin-2-yl)-8-
methoxycarbostyril (5 g) in Essigsäure (50 ml) wurde
eine 50%ige wäßrige Lösung von Dimethylamin (1,7 g)
und eine 35%ige wäßrige Lösung von Formalin (1,62 g)
gegeben. Die Mischung wurde 6 Stunden bei 60°C gerührt.
Nach dem Abdampfen des Lösungsmittels wurde der
Rückstand durch Zugabe von Chloroform und 0,5N Natriumhydroxid
extrahiert. Die Chloroformschicht wurde mit
Wasser gewaschen, getrocknet und dann wurde das Lösungsmittel
abgedampft. Der Rückstand wurde in Methanol
gelöst und die Lösung wurde durch Zugabe von konzentrierter
Salzsäure auf einen pH von etwa 1 eingestellt
und anschließend zur Trockne konzentriert. Aceton
wurde zum Ausfällen der Kristalle zu dem Rückstand gegeben
und die Kristalle wurden durch Filtrieren gesammelt.
Die so erhaltenen Rohkristalle wurden aus Methanol-
Aceton umkristallisiert, wobei man 5-(3-Dimethylaminomethylimidazo
[1,2-a]pyridin-2-yl)-8-methoxycarbostyrildihydrochlorid-
trihydrat erhielt.

Zu einer Suspension von 5-(3-Dimethylaminomethylimidazo
[1,2-a]pyridin-2-yl)-8-methoxycarbostyril (1,5 g) in
Acetonitril (20 ml) wurde Methyljodid (1,5 ml) gegeben
und die Mischung wurde eine Stunde bei 40°C gerührt. Die
so gebildeten Kristalle wurden durch Filtrieren gesammelt
und aus Wasser umkristallisiert, wobei man 1,7 g
5-(3-Trimethylaminomethylimidazo[1,2-a]pyridin-2-yl)-
8-methoxycarbostyriljodid-trihydrat erhielt.

5-(3-Trimethylammoniummethylimidazo[1,2-a]pyridin-2-yl)-
8-methoxycarbostyriljodid (8 g) und Natriumcyanid
(3,2 g) wurden zu Wasser (100 ml) gegeben und die Mischung
wurde 5 Stunden rückflußbehandelt. Die ausgefallenen
Kristalle wurden durch Filtrieren gesammelt.
Nach dem Waschen mit Methanol wurde die Verbindung in
das salzsaure Salz in Methanol mit konzentrierter Salzsäure
überführt und aus Methanol-Ether umkristallisiert,
wobei man 3,6 g 5-(3-Carbamoylmethylimidazo
[1,2-a]pyridin-2-yl)-8-methoxycarbostyril-monohydrochlorid-
5/4-hydrat erhielt.
F: 250,5-251,5°C (Zersetzung), farblose
Nadeln
Eine Lösung von 5-(3-Trimethylammoniummethylimidazo
[1,2-a]pyridin-2-yl)-8-methoxycarbostyriljodid (4,39 g)
in Wasser (40 ml) wurde rückflußbehandelt und dabei
wurden tropfenweise 10 ml einer wäßrigen Lösung, in
welcher 440 mg Natriumcyanid gelöst waren, zugegeben.
Nach Beendigung der Zugabe wurde die Mischung 30 Minuten
umgesetzt und dann abkühlen gelassen. Die ausgefallenen
Kristalle wurden durch Filtrieren gesammelt und
durch Durchleiten durch eine Kieselgelsäule isoliert.
Die so gebildeten Rohkristalle wurden aus Methanol
umkristallisiert, wobei man 0,85 g 5-(3-Cyanomethylimidazo
[1,2-a]pyridin-2-yl)-8-methoxycarbostyril erhielt.
F: 261-263°C (Zersetzung), farblose
Prismen.
Eine Mischung aus 5-(3-Carbamoylmethylimidazo[1,2-a]
pyridin-2-yl)-8-methoxycarbostyril (1,6 g), Kaliumhydroxid
(2,6 g), Wasser (3 ml) und Ethanol (9 ml)
wurde eine Stunde unter Rückfluß behandelt. Nach Beendigung
der Umsetzung wurde Wasser zum Reaktionsgemisch
gegeben, welches anschließend mit Aktivkohle behandelt
wurde und dann durch Zugabe von konzentrierter
Salzsäure auf einen pH von etwa 1 eingestellt wurde
und anschließend über Nacht stehen gelassen wurde. Die
ausgefallenen Kristalle wurden durch Filtrieren gesammelt
und aus verdünnter Salzsäure umkristallisiert,
wobei man 0,9 g 5-(3-Carboxymethylimidazo[1,2-a]pyridin-
2-yl)-8-methoxycarbostyril-hemihydrat erhielt.
F: 259-260,5°C (Zersetzung) farblose
Nadeln
5-(Imidazo[1,2-a]pyridin-2-yl)-8-methoxy-3,4-dihydrocarbostyril
(1,4 g) und 2,3-Dichlor-5,6-dicyan-1,4-benzochinon (3,5 g) wurden zu Dioxan
(30 ml) gegeben und die Mischung wurde 5 Stunden rückflußbehandelt.
Das Reaktionsgemisch wurde unter vermindertem
Druck konzentriert und der Rückstand wurde
durch Zugabe von Chloroform und 0,5N Natriumhydroxid
extrahiert. Die Chloroformschicht wurde mit 0,5N Natriumhydroxid
und anschließend 2mal mit Wasser gewaschen.
Nach dem Trocknen wurde das Chloroform abgedampft. Der
Rückstand wurde isoliert und über einer Kieselgelsäule
gereinigt. Die erhaltenen Rohkristalle wurden in Aceton
gelöst und zu der erhaltenen Lösung wurde Salzsäure
gegeben. Die ausgefallenen Kristalle wurden durch Filtrieren
gesammelt und aus Wasser umkristallisiert, wobei
man 410 mg 5-(Imidazo[1,2-a]pyridin-2-yl)-8-
methoxycarbostyrilmonohydrochlorid-monohydrat erhielt.
F: 256-257,5°C (Zersetzung), farblose
flauschige Kristalle
Zu einer Lösung von 5-(3-Ethylimidazo[1,2-a]pyridin-
2-yl)-8-methoxycarbostyril (2 g) in Methanol (50 ml)
wurde 10%ige Palladium/Kohle (0,2 g) gegeben und
die katalytische Reduktion wurde während 6 Stunden bei
50 bis 60°C unter einem Wasserstoffgasdruck von 2
bis 3 kg/cm² durchgeführt. Der Katalysator wurde abfiltriert
und das Filtrat wurde zur Trockne konzentriert.
Der Rückstand wurde in Aceton gelöst und zu
der Lösung wurde Bromwasserstoffsäure gegeben. Die ausgefallenen
Kristalle wurden durch Filtrieren gesammelt
und die so erhaltenen Rohkristalle wurden aus Methanol-
Ether umkristallisiert, wobei man 1,5 g 5-(3-Ethylimidazo
[1,2-a]pyridin-2-yl)-8-methoxy-3,4-dihydrocarbostyril-
monohydrobromid erhielt.
F: 254-256,5°C, farbloses Pulver
Zu einer Lösung von 5-(Imidazo[1,2-a]pyridin-2-yl)-8-
methoxy-3,4-dihydrocarbostyril (3 g) in Dimethylformamid
(100 ml) wurde 50%iges öliges Natriumhydrid
(600 mg) gegeben und die Mischung wurde 2 Stunden bei
Raumtemperatur gerührt. Nach der Zugabe von Allylbromid
(1,48 g) wurde die Mischung 3 Stunden bei Raumtemperatur
umgesetzt. Das Reaktionsgemisch wurde zur Trockne
konzentriert und der Rückstand wurde durch Zugabe von
Chloroform und 0,5N Natriumhydroxid extrahiert. Die
Chloroformschicht wurde mit Wasser gewaschen, getrocknet
und anschließend eingedampft. Der Rückstand wurde
mittels Kieselgelsäulenchromatografie gereinigt und
das ölige Produkt wurde in Aceton gelöst und in das
salzsaure Salz durch Zugabe von Salzsäure überführt.
Die ausgefallenen Kristalle wurden durch Filtrieren gesammelt
und aus Methanol-Ether umkristallisiert, wobei
man 1,76 g 1-Allyl-5-(imidazo[1,2-a]pyridin-2-yl)-8-
methoxy-3,4-dihydrocarbostyril-monohydrochlorid erhielt.
F: 250-252°C (Zersetzung), farblose

Claims (14)
1. Imidazo-[1,2-a]pyridin-2-yl-carbostyril-Derivate der
allgemeinen Formel (I)
worin R¹ ein Wasserstoffatom oder die Methyl-,
Allyl- oder Propargylgruppe bedeutet,
R² ein Wasserstoffatom oder die Methylgruppe, die Methoxygruppe, ein Chloratom oder die Hydroxygruppe bedeutet,
R³ ein Wasserstoffatom, die Methyl- oder Ethylgruppe, ein Bromatom, eine Nitrosogruppe, eine Aminogruppe, eine N,N-Dimethylaminomethyl- oder eine Trimethylammoniummethylgruppe, eine Carbamoylmethylgruppe, eine Cyanomethylgruppe oder eine Carboxymethylgruppe bedeutet,
R⁴ und R⁵, die gleich oder verschieden sein können, jeweils ein Wasserstoffatom, ein Chlor- oder Bromatom, eine Methylgruppe, eine Hydroxygruppe, eine Methoxygruppe oder eine Nitrogruppe bedeuten, die Verbindung zwischen der 3- und 4-Stellung des Carbostyrilkerns eine Einfachbindung oder eine Doppelbindung bedeutet, und
wobei die Stellung, in welcher die Imidazopyridylgruppe der Formel am Carbostyrilkern gebunden ist, die 5- oder 6-Stellung ist,
mit der Maßgabe, daß dann, wenn die Imidazopyridylgruppe in der 5-Stellung des Carbostyrilkerns gebunden ist, R² kein Wasserstoffatom, keine Methylgruppe und kein Chloratom bedeutet,
oder deren pharmazeutisch annehmbares Salz.
R² ein Wasserstoffatom oder die Methylgruppe, die Methoxygruppe, ein Chloratom oder die Hydroxygruppe bedeutet,
R³ ein Wasserstoffatom, die Methyl- oder Ethylgruppe, ein Bromatom, eine Nitrosogruppe, eine Aminogruppe, eine N,N-Dimethylaminomethyl- oder eine Trimethylammoniummethylgruppe, eine Carbamoylmethylgruppe, eine Cyanomethylgruppe oder eine Carboxymethylgruppe bedeutet,
R⁴ und R⁵, die gleich oder verschieden sein können, jeweils ein Wasserstoffatom, ein Chlor- oder Bromatom, eine Methylgruppe, eine Hydroxygruppe, eine Methoxygruppe oder eine Nitrogruppe bedeuten, die Verbindung zwischen der 3- und 4-Stellung des Carbostyrilkerns eine Einfachbindung oder eine Doppelbindung bedeutet, und
wobei die Stellung, in welcher die Imidazopyridylgruppe der Formel am Carbostyrilkern gebunden ist, die 5- oder 6-Stellung ist,
mit der Maßgabe, daß dann, wenn die Imidazopyridylgruppe in der 5-Stellung des Carbostyrilkerns gebunden ist, R² kein Wasserstoffatom, keine Methylgruppe und kein Chloratom bedeutet,
oder deren pharmazeutisch annehmbares Salz.

13. Verfahren zur Herstellung der Verbindungen nach Anspruch
1, dadurch gekennzeichnet, daß man in
an sich bekannter Weise
- (a) eine Verbindung der allgemeinen Formel (II) worin X¹ ein Halogenatom und R⁶ ein Wasserstoffatom oder eine Methyl- oder Ethylgruppe bedeutet und R¹ und R² und die Bindung zwischen der 3- und 4-Stellung im Carbostyrilkern die vorher angegebenen Bedeutungen haben, mit einer Verbindung der allgemeinen Formel (III) worin R⁴ und R⁵ die vorher angegebenen Bedeutungen haben, umsetzt,
- (b) daß man eine Verbindung der allgemeinen Formel (Ie) worin R¹, R², R⁴, R⁵ und die Bindung zwischen der 3- und 4-Stellung im Carbostyrilkern die vorher angegebenen Bedeutungen haben, bromiert, (R³=Br),
- (c) daß man eine Verbindung der allgemeinen Formel (Ie) worin R¹, R², R⁴, R⁵ und die Bindung zwischen der 3- und 4-Stellung im Carbostyrilkern die vorher angegebenen Bedeutungen haben, nitrosiert (R³=NO),
- (d) daß man eine Verbindung der allgemeinen Formel (Ig) worin R¹, R², R⁴, R⁵ und die Bindung zwischen der 3- und 4-Stellung im Carbostyrilkern die vorher angegebenen Bedeutungen haben, reduziert (R³=NH₂),
- (e) daß man eine Verbindung der Formel (Ie) worin R¹, R², R⁴, R⁵ und die Bindung zwischen der 3- und 4-Stellung im Carbostyrilkern die vorher angegebenen Bedeutungen haben, mit Dimethylamin der Formel (XXI) in Gegenwart von Formalin umsetzt (R³=CH₂N(CH₃)₂),
- (f) daß man eine Verbindung der Formel (Im) worin R¹, R², R⁴, R⁵ und die Bindung zwischen der 3- und 4-Stellung im Carbostyrilkern die vorher angegebenen Bedeutungen haben, mit einem Methylhalogenid der Formel (XXII)H₃C-X² (XXII)worin X² ein Halogenatom bedeutet, umsetzt (R³=CH₂-N(CH₃)₃⁺),
- (g) daß man eine Verbindung der Formel (In) worin R¹, R², R⁴, R⁵ und die Bindung zwischen der 3- und 4-Stellung im Carbostyrilkern die vorher angegebenen Bedeutungen haben, mit einer Verbindung der Formel (XXIII)MCN (XXIII)worin M ein Alkalimetall bedeutet, umsetzt (R³=CH₂-CN),
- (h) daß man eine Verbindung der Formel (Io) worin R¹, R², R⁴, R⁵ und die Bindung zwischen der 3- und 4-Stellung im Carbostyrilkern die vorher angegebenen Bedeutungen haben, hydrolysiert (R³=CH₂-COOH),
- (i) oder daß man eine Verbindung der Formel (Ip) worin R¹, R², R⁴, R⁵ und die Bindung zwischen der 3- und 4-Stellung im Carbostyrilkern die vorher angegebenen Bedeutungen haben, hydrolysiert (R³=CH₂-COOH), und gewünschtenfalls das Produkt gemäß einer der vorhergehenden Stufen (a) bis (i) in ein pharmazeutisch annehmbares Salz überführt.
14. Arzneimittel, enthaltend eine Verbindung nach Anspruch 1,
neben einem pharmazeutisch annehmbaren Träger.
