4. Arzneimittel, enthaltend als Wirkstoff mindestens eine Verbindung nach Anspruch 1 bis 3 zusammen
mit physiologisch unbedenklichen Trägerstoffen.
5. Verfahren zur Herstellung der Verbindungen gemäß Anspruch 1, dadurch gekennzeichnet, daß man ein
Sulfid der Formel
in der R1, A!k und A die angegebener. Bedeutungen besitzen, in an sich bekannter Weiss rnii H:.Q-, in. Gegenwart
von Essigsäure oxidiert und gegebenenfalls die freie Base in an sich bekannter Weise in das entsprechende
nichttoxische Additionssalz überführt.
Die Erfindung betrifft Phenylsulfinylalkylamidine, ein Verfahren zu ihrer Herstellung und diese enthaltende
Arzneimittel.
Im folgenden werden unter dem Ausdruck »Amidine und ihre Derivate« nicht nur die Verbindungen mit einer
Amidingruppe verstanden, sondern auch die Verbindungen, die eine von der Amidingruppe abgeleitete Gruppe
oder ein Homolog enthalten, das heißt die Amidoximgruppe, Hydroxylamidgruppe und zyklische Amidin-
gruppen wie 2-/l2-Imidazolinyl und 2-(l,4,5,6-Tetrahydropyrimidinyl-Gruppen), wobei die letzteren zyklischen
so Gruppen auch substituiert sein können, wie unten weiter ausgeführt wird.
Die erfindungsgemäßen Verbindungen sind die Phenylsulfinylalkylamidine und ihre Derivate der allgemeinen
Formel
in der
R, ein H-, F- oder Cl-Atom oder eine CF3-Gruppe,
Alk einen geradkettigen CrKohlenwasserstoffrest, und
A Cl=NH)NHj. Q=NH)NHOH, C(=0)NH0H oder
bedeutet, wobei
Z CH1CH2 oder CH2CH1CH1 und
R CH2COOH, CH(CH3)COOH und C(CHj)2COOH sind,
sowie ihre nichttoxischen Additionssalze.
Unter Additionssalze werden hiersowohl die Ammoniumsalze als auch die durch Anlagerung von Säuren entstandenen
Salze verstanden.
Die Verbindungen der Formel I können nach einem an sich bekannten Verfahren hergestellt werden. Das
erfindungsgemäße Verfahren zur Herstellung der Verbindungen der Formel I besteht darin, daß man mit Hilfe
von H2O2 in Anwesenheit von Essigsäure ein Sulfidderivat der Formel
in der R, Alk und A die angegebenen Bedeutungen besitzen, oxydiert.
Vorzugsweise wird die Oxydation des Sulfids II mit konzentriertem Wasserstoffperoxid, das heißt mindestens
33 Gew.-% H2O2, ausgeführt. Im Verlauf dieser Oxydation muß man die Bildung einer relativ großen Menge dtr
entsprechenden Suifonylverbindung vermeiden. Praktisch erhält man im wesentlichen nur das genannte Sulfonylderivat,
wenn man bei 100°C 1 Stunde oder mehr mit Wasserstoffperoxidlösung von 110 bis 120 Volumenanteilen
arbeitet, um nur das Suifinyiderivat zu erhalten, arbeitet man entweder 1 Stunde bei 500C, kühlt dann
gegebenenfalls ab und hält das Reaktionsgemisch einige Stunden (insbesondere 1 bis 3 Stunden) bei Raumtemperatur
(15 bis 25°C) oder man arbeitet bei 37 bis 45°C (einer Temperatur, die im allgemeinen im Reaktionsgemisch sich selbst einstellt, da die Reaktion exotherm ist), kühlt dann ab und hält 3 bis 15 Stunden auf Raum-
temperatur (15 bis 25°C). Man ersieht aus den unten gegebenen Beispielen, daß man etwa stöchiometrische
Mengen an Sulfid II und H2O2 einsätzen kann.
Die Additionssalze von Säuren, die aus den Basen der Formel I hergestellt werden können, werden nach an
sich bekanntem Verfahren erhalten, beispielsweise durch Reaktion der freien Base mit einer Mineralsäure oder
einer organischen Säure. Unter den anwendbaren Säuren seien insbesondere Salzsäure, Bromwasserstoffsäure,
Jodwasserstoffsäure, Schwefelsäure, Ameisensäure, Maleinsäure, Fumarsäure^ Oxalsäure, Ascorbinsäure,
Zitronensäure, Essigsäure, Methansulfonsäure, p-Toluolsulfonsäure, Milchsäure, Bernsteinsäure, Benzoesäure,
Salicylsäure, Acetylsalicylsäure, Apfelsäure, Weinsäure, Glutaminsäure und Asparaginsäure genannt.
Die erfindungsgemäßen Verbindungen sind in der Therapie insbesondere als angstlösende, schmerzstillende,
entzündungshemmende und/oder avf das zentrale Nervensystem einwirkende Mittel anwendbar. Sie alle
wirken analgetisch, wobei die schmerzstillende Wirkung bei den einzelnen Produkten unterschiedlich ist.
Manche dieser Verbindungen zeigen außer der schmerzstillenden Wirkung andere interessante Eigenschaften.
So wirkt das Produkt des Beispiels 6 (CRL 4019), das einen DL-50, oral bei Mäusen von 350 mg/kg aufweist,
auch nach dem Carragenin-Ödem-Test als entzündungshemmende Substanz und das Produkt des Beispiels 7
(CRL-40 218) ist ein das zentrale Nervensystem stimulierendes Mittel, das als angstlösendes MiUe1 -virkt. Auch
die Verbindungen der Beispiele 12, 13 und 14 wirken auf das zentrale Nervensystem.
Die Verbindungen der Formel II können nach an sich bekannten Verfahren durch Anwendung klassischer
Reaktionsmethoden hergestellt werden. In dem folgenden Schema I werden die verschiedenen Reaktionsmechanismen abgebildet, die die Synthese der Sulfide II aus einem gegebenenfalls substituierten Thiophenol
und dann die Herstellung der Sulfinylderivate I aus den genannten Sulfiden gestatten.

Die Reaktionsmechanismen des Schema 1 können je nach den Bedürfnissen der einzelnen Synthese modifiziert
werden. So ist es nicht nötig, zur Herstellung der Amidine, Amidoxime und cyclischen Amidine den aus
dem PhenylthiGalkylnitril erhaltenen Iminoester zu isolieren. Man kann den Iminoester in situ herstellen.
Ferner kann die Reaktion zur Einführung einer Gruppe R (anders als Wasserstoff) in 1-Stellung der cyclischen
Amidingruppe-mit einem anderen Halogenderivat als CIR, beispielsweise mit BrR, durchgeführt werden.
Andere Vorteile und Eigenschaften der Erfindung gehen aus den folgenden Beispielen hervor. Darin sind die
Temperaturen in 0C ausgedrückt, der Ausdruck Äther ohne nähere Kennzeichnung bedeutet Diäthylätherund
z. B. der Ausdruck Isopropyläther bedeutet den Diisopropyläther.
Sämtliche in den Beispielen beschriebenen Verbindungen sind in den nachfolgenden Tabellen enthalten.

Unter Rühren bei 70 bis 800C gibt man 10 ml (0,13 Mol) jS-Chlorpropionitril zu einer Lösung von 17,4 g
(0,12 Mol) p-Chlorthiophenol und 4,8 g Natriumhydroxidpastillen in 50 ml Wasser. Man hält V2 Stunde bei
8O0C. Nach dem Abkühlen extrahiert man mit Äther, wäscht mit Wasser und trocknet. Man erhält 22,5 g
3-(p-Chlorphenylthio)-propionitril. F. = 53 bis 54°C.
b) 2-[2-(p-Chlorphenylthio)-äthyl]-^l'-imidazolinchIorhydrat
Eine Lösung von 22,5 g (0,114 Mol) 3-(p-Chlorphenylthio)-propionitril in 200 ml Äther und 12 ml Äthanol
sättigt man mit trockenem gasförmigem HCl. Man läßt 48 Stunden bei 5°C stehen und zentrifugiert. Man erhält
20 g des Iminoester-chlorhydrats. F. = 108 bis 1100C.
Zu 19 g (0,068 Mol) des genannten Iminoester-chlorhydrats, in 100 ml Äthanol gelöst, gibt man 4,2 g
(0,07 Mol) Äthylendiamin zu und erhitzt 2 Stunden unter Rückfluß. Man dampft unter Vakuum zur Trockne ein,
nimmt mit Wasser auf, und fügt bis pH 1 konzentrierte Salzsäure zu. Man filtriert über Aktivkohle und fällt in
der Kälte mit konzentrierter NaOH. Man filtriert, wäscht mit Wasser und trocknet. Man erhält 14,5 g freie Base.
F. = 68 bis 700C, die man in das Chlorhydrat überfuhrt, F. = 130 bis 1320C.
c) CRL-Nummer 40 065
Man gibt 3,2 ml (0,032 Mol) Wasserstoffperoxidlösung von 110 Volumteilen zu einer Lösung von 8,9 g
2-[2-(p-Chlorphenylthio)-äthyl]-Jd2-imidazolinchlorhydrat in 30 ml Essigsäure. Nach 1 Stunde bei 5O0C und
Stehenlassen über Nacht dampft man im Vakuum zur Trockne ein. Man nimmt mit Aceton auf, filtriert und kristallisiert
aus Isopropanol um. Man erhält die Verbindung CRL 40 065 mit 40%iger Ausbeute. Sie liegt in Form
von kleinen weißen Nadeln vor, die bei 142 bis 144°C schmelzen. Sie ist in Wasserund Äthanol löslich, unlöslich
in Aceton und Äther

a) S-Q-Trinuormethylphenylthioi-propioniminoäthyiester-chlorhydrat
Eine Lösung von 15,9 g (0,09 Mol) 3-Trifluormethylthiophenol und 3,8 g (0,095 Mol) NaOH-Pastillen in
100 ml Wasser erhitzt man auf 65 bis 700C und gibt unter Rühren tropfenweise 8,5 g (0,095 Mol)jS-Chlorpropionitril
zu. Man erhitzt' /4 Stunde unter Rückfluß, kühlt ab, extrahiert mit Äther, wäscht mit Wasser und trocknet.
So erhält man 3-(3-Trifluormeihylphenylthio)-propionitril, das man nicht abtrennt. Man filtriert das Magnesiumsulfat,
fügt zum Filtrat 10 ml Äthanol zu und sättigt mit trockenem gasformigem HCl bei 00C. Man läßt
48 Stunden stehen und filtriert. Man erhält 25,6 g des gewünschten Iminoäthylester-chlorhydrats (90%),
F. = 92°C.
b) 2-[2-(3-Trifluonnethylphenylthio)-äthyl]-2l2-imidazolin-chlorhydrat
Man gibt 4,8 g (5,4 ml = 0,08 Mol) Äthylendiamin zu einer Lösung von 25 g (0,08 Mol) 2-(m-Trifluormethylphenylthio)-propioniminoäthylester-chlorhydrat
in 100 ml Äthanol. Man erhitzt 2 Stunden unter Rückfluß, dampft unter Vakuum zur Trockne ein, nimmt mit Wasser, in dem einige Tropfen konzentrierter Salzsäure enthalten
sind, auf. Man filtriert über Aktivkohle, fallt die Base mit konzentrierter Natronlauge aus, filtriert, wäscht
mit Wasser und trocknet. Man erhält so 16,3 g derfreien Base,F. = 71 bis 72°C. Man löst die Base in Äthylacetat,
filtriert über Aktivkohle, gibt zum Filtrat äthz^olische Salzsäure und zentrifugiert. Man erhält 16,4 g (66%).
c) CRL-Nummer 40 094
5,2 ml (0,052 Mol) Wasserstoffperoxid von 110 Volumteilen gibt man bei 2O0C zu einer Lösung von 16,2 g
(0,052 Mol) des vorher erhaltenen Chlorhydrais in 50 ml Essigsäure. Es tritt eine Erwärmung auf, man hält
1 Stunde bei 500C und dampft unter Vakuum ab. Man nimmt den Rückstand mit 100 ml Wasser auf, filtriert über
Aktivkohle und dampft im Vakuum ein. Man erhält 100 ml Äthylacetat und zentrifugiert. Nach Umkristallisieren
aus Aceton erhält man CRL-Nummer 40 094 mit 44%iger Ausbeute in Form von kleinen cremefarbenen
Plättchen. Es ist in Wasser und in Alkoholen löslich, wenig löslich in Aceton in der Kälte, unlöslich in Äther und
Äthylacetat. F. = 109 bis UO0C.

Eine Suspension von 10,4 g (0,05 Mol) 2-[2-(Phenylthio)-äthyl]-2f2-imidazolin und 10,6 g (0,1 Mo!) Natriumcarbonat
in 5 ml Wasser erhitzt man auf 7 bis 800C und gibt unter Rühren 5 g (0,053 Mol) Chloressigsäure, in
10 ml Wasser gelöst, zu. Man neutralisiert mit Natriumcarbonat und erhitzt anschließend 10 Minuter, unter
Rühren auf 800C. Die kalte Lösung wird mit konzentrierter HCl angesäuert. Man filtriert und wäscht mit ein
wenig kaltem Wasser. So erhält man in 70%iger Ausbeute die {2-[2-(Phenylthio)-äthyl]-j42-imidazolin-l-yl}-essigsäure,
F. = 62 bis 630C.
Man oxydiert 15,6 g (0,06 Mol) dieser Säure, in 60 ml Essigsäure gelöst, mit 6 ml Wasserstoffperoxidlösung
von 110 Volumteilen. Man dampft unter Vakuum zur Trockne ein, nimmt mit Isopropyläther auf und zentrifugiert.
Man erhält die CRL-Nummer 40 111 mit 60%iger Ausbeute, F. = 110 bis 112°C.

Man tropft eine Lösung von 4 g (0,042 Mol) Chloressigsäure und 5 g (0,05 Mol) Natriumcarbonat bei 70 bis
800C in eine Suspension von 9,6 g (0,04 Mol) 2-[2-(p-Chlorphenylthio)-äthyl]-2l2-imidazolin und 8,5 g
(0,08 Mol) Na2CO3 in 50 mi Wasser. Man erhitzt noch 10 Minuten auf 8O0C. Man kühlt die Lösung ab und
säuert mit konzentrierter HCl an. Man filtriert, wäscht mit kaltem Wasser und trocknet. Man erhält so die
{2-[2-(p-Chlorphenylthio)-äthyl]-2l2-imidazolin-l-yl}-essigsäure, F. = 98 bis 990C.
Man oxydiert 12 g (0,04 Mol) dieser Säure, gelöst in 40 ml Essigsäure, mit4 ml Wasserstoffperoxidlösung von
110 Volumteilen. Man dampft unter Vakuum zur trockne ein, nimmt mit kaltem Wasser auf, filtriert, trocknet
und wäscht mit ein wenig Isopropyläther. Man kristallisiert aus Isopropanol um und erhält so CRL-Nummer
40 112 mit 56%iger Ausbeute, F. = 132 bis 133°C.

erhall so die c-{2-[2-{p-Chloφhenylthΐo)-äthylj-J2-imidazoIin-l-yl}-essigsäuΓe, F. = 92 bis 93°C.
Man oxydiert 9,5 g (0,031 Mol) dieser Säure, in 30 ml Essigsäure gelöst, mit 3,1 ml (0,031 Mol) Wasserstoffperoxid
von 110 VeJy mteüen. Nach 1 Stunde bei 500C dampft man unter Vakuum zur Trockne ein, nimmt den
Rückstand mit verdünnter Natriumbicarbonatlösung auf, filtriert, fällt mit konzentrierter Salzsäure, zentrifugiert
und trocknet. Man kristallisiert aus Athylacetat-Petroläther (50 :50) um und erhält so die Verbindung
CRL-Nummer 40 113 mit einer Ausbeute von 50%. F. = 119 bis 1200C.

a) 2-[2-(Phenylthio)-äthy!]-Jd2-imidazolin-chlorhydrat
Man erwärmt eine Lösung von 13,2 g (0,12 MoI) Thiophenol und 4,3 g (0,12 Mol) NaOH in 100 ml Wasser auf
800C und fügt 10 ml (0,13 Mol) jS-Chlorpropionitril zu, führt noch i0 Minuten bei 800C und läßt abkühlen. Man
extrahiert zweimal mit 100 ml Äther, wäschtmitverdünnter Natronlauge, verdürnter Salzsäure, mit Wasserund
trocknet. Zu dieser Ätherlösung gibt man 8 ml Äthanol und sättigt mit trockenem gasförmigem HCI. Man läßt
48 Stunden lang in der Kälte stehen und dampft unter Vakuum ein.
Der Rückstand (30 g) wird in 90 ml Äthanol gelöst; dann gibt man 10 ml Äthylendiamin zu und erhitzt
2 Stunden unter Rückfluß. Man filtriert das ausgefallene Äthylendiamin-chlorhydrat ab und dampft unter
Vakuum zur Trockne ein. Den Rückstand nimmt man mit verdünnter Salzsäure auf, extrahiert mit Äther, fällt
mit konzentrierter Natronlauge aus, filtriert und trocknet.
Man erhält das Chlorhydrat, indem man die Base (F. = 800C), in Äthylacetat gelöst, mit äthanolischer Salzsäure
behandelt Man erhält so 22 g 2-[2-{PhenyiihiG)-äihyi^2-imidazolin-chIorhydrat mit 75%iger Ausbeute,
F.= 128 bis 1300C.
b) CRL-Nummer 4 019
Man oxydiert 12,1g (0,05 Mol) des so erhaltenen Chlorhydrats, in 50 ml Essigsäure gelöst, mit 5 ml (0,05 Mol)
Wasserstoffperoxid. Man dampft unter Vakuum zur Trockne ein, nimmt mit Aceton auf und filtriert. Man nimmt
den Niederschlag mit der geringsten Menge leicht angesäuerten Wassers auf, filtriert, dampft unter Vakuum ein
und kristallisiert aus Äthanol-Aceton-Gemisch um. Man erhält das 2-[2-(PhenylsuIfinyi)-äthyl]-.42-imidazolinchlorhydrat
mit 55%iger Ausbeute. Es liegt in Form von »weißen, cremefarbigen«, sehr hygroskopischen
Kristallen vor. F. = 115 bis 116°C.

Man erwärmt eine Lösung von 10 g (0,078 Mo!) p-Fluorthiophenoi und 3,2 g (0,08 Mol) Natriumhydroxid in
Pastillen in 100 ml Wasser auf 700C. Unter Rühren gibt man tropfenweise 8,3 g (0,08 Mol) y-Chlorbutyrnitril zu.
Nach 10 Minuten unter Rückfluß und 1 Stunde Ruhen extrahiert man mit Äther, wäscht mit Wasser, trocknet
und dampft unter Vakuum ein. Zum Rückstand gibt man eine Hydroxylaminlösung, die aus 10,5 g (0,15 Mol)
Hydroxylamin-chlorhydrat, 15 g Kaliumbicarbonat in 15 ml Wasser und 60 ml Butanol hergestellt wurde. Man
erhitzt das Gemisch 3 Stunden unter Rühren am Rückfluß. Nach Stehenlassen über Nacht filtiert man die Base
ab und erhält 11 g Produkt, F. = 1040C.
Man erhält das Chlorhydrat, indem man eine Lösung der Base in Äthylacetat mit äthanolischer Salzsäure
ansäuert. F. = 75 bis 76°C.
b) CRL-Nummer40 218
Man oxydiert 11,15 g (0,042MoI) 4-p-Fluorphenylthiobutyramidoxim-chlorhydrat, in 42 ml Essigsäure
gelöst, mit 4,2 ml Wasserstoflperoxidlösung von 110 Volumteilen. Nach anfanglicher Erwärmung läßt man
3 Stunden zusammen stehen und dampft unter Vakuum zur Trockne ein. Man nimmt den Rückstand mit 50 ml
Aceton auf und filtriert. Man erhält die Verbindung CRL-Nummer40 218 mit 55%iger Ausbeute in Form von
kleinen weißen Kristallen. Sie ist in Wasser und Alkoholen löslich, in Aceton, Äther und Benzol unlöslich,
F. = 186 bis 188°C.

erhitzt noch 10 Minuten unter Rückfluß und extrahiert nach dem Abkühlen mit Äther, wäscht mit Wasser und
trocknet. Der so erhaltene Ester wird in 50 ml Butanol gelöst und 4 Stunden zusammen mit einer Hydroxylaminlösung,
die aus 7 g (0,1 Mol) Hydroxylaminchlorhydrat, 10 g Kaüumbicarbonat und 15 ml Wasser hergestellt
wurde, unter Rückfluß erwärmt. Man säuert dann mit konzentrierter Salzsäure an und dampft unter
Vakuum zur Trockne ein. Man nimmt den Rückstand mit Wasser auf, extrahiert mit Äther, verwirft die Ätherphase,
filtriert die wäßrige Phase über Aktivkohle und fällt die Base mit Natriumcarbonat aus. Man filtriert,
wäscht mit Wasser und trocknet. Man erhält 10,1 g Produkt vom F. = 86°C.
Man erhält das Chlorhydrat (F. = 136 bis 138°C) durch Ansäuern einer Lösung der Base in Äthylacetat mit
äthanolischer Salzsäure.

b) CRL-Nummer 40 220 U
Man oxydiert 11,5 g (0,046 Mol) 3-p-FluorphenyIthio-propionamidoxim-chlorhydrat, in 46 ml Essigsäure
gelöst, mit 4,6 ml Wasserstoffperoxid von 110 Voiumteilen. Nach anfänglicher Erwärmung (etwa 400C) lät man
2 Stunden ruhen, dampft unter Vakuum zur Trockne ein, nimmt mit Aceton auf, filtiert und kristallisiert aus
Methanol um. Man erhält die Verbindung CRL-Nummer 40 220 mit 58%iger Ausbeute. Das Produkt liegt in
Form von kleinen weißen Kristallen vor, die bei 146 bis 148°C unter Zersetzung schmelzen.

a) 4-(p-Chlorphenylthio)-butyramidoxim-chlorhydrat
Unter Rühren erwärmt man ein Gemisch von 29 g (0,2 Mol) p-Chlorthiophenol und 8 g Natriumhydroxid in
200 ml Wasser auf 800C. Man gibt tropfenweise 22 g (0,21 Mol) y-Chlorbutyronitril zu und erhitzt V4 Stunde W,
unter Rückfluß. Man kühlt ab, extrahiert mit Methylenchlorid, wäscht mit Wasser, trocknet und dampft unter |
Vakuum zur Trockne ein. Das zurückbleibende Öl (42 g, 0,2 Mol) wird, in 180 ml Butanol gelöst, zu einer §
Lösung von 21 g (0,3 Mol) 1 lydroxylmnmoniumchlorid und 30 g Kaliumbicarbonat in 40 ml Wasser gegeben.
Man erhitzt das Gemisch 5 Stunden unter Rücknuß und dampft das Butanol unter Vakuum ab. Man gibt 1 I
(i5 Wasser zu, zentrifugiert, wäscht mit Wasser, dann zweimal mit 50 ml Isopropyläther, trocknet und kristallisiert
aus Isopropanol um.
Man erhält 34,5 g der Base (F. = 99°C), die man durch Zugabe von ätherischer Salzsäure zur Lösung der Base
in Äthylacetat in das Chlorhydrat umwandelt. Man erhält 38 g (68%) Chlorhydrat, F. = 88 bis 89°C.
b) CRL-Nummer 40 266
20,5 g (0,075 MoI) des vorstehend erhaltenen Chlorhydrats, in 75 ml Essigsäure gelöst, werden mit 6,9 ml
"Wasserstoffperoxid von 120 Volumteilen während 1 Stunde bei 500C oxydiert. Man läßt über Nacht stehen,
dampft unter Vakuum zurTrockne ein, nimmt mit Aceton auf, zentrifugiert und kristallisiert aus Propanol um.
Man erhält die Verbindung CRL 40 266 mit58%iger Ausbeute in Form eines weißen Pulvers. F. = 178 bis 18O°C.

a) 4-(p-Fluorphenylthio)-butyroiminoäthylester-chlorhydrat
Eine Lösung von 14,5 g (0,075 Mol) 4-(p-Fluorphenylthio)-butyronitril und 8 ml Äthanol in 75 ml Äther
sättigt man bei 0°C mit trockenem gasförmigem HCl. Man läßt 24 Stunden in der Kälte stehen, zentrifugiert und
wäscht mit Äther. Man erhält 17 g Iminoäthylester (Ausbeute 82%), F. = 103 bis 1040C).
b) 4-(p-Fluorphenylthio)-butyramidin-chlorhydrat
In der Kälte (etwa ö°C) werden 17 g (0,061 Mol) des vorstehend erhaltenen Iminoester-chlorhydrats, in
100 ml Äthanol gelöst, mit trockenem gasförmigem NH3 gesättigt. Man läßt 24 Stunden in der NH3-Atmo-Sphäre,
dampft dea Alkohol im Vakuum ab, nimmt mit 100 ml Äther auf und zentrifugiert. Man erhält 15 g
Sulfid (Ausbeute 96%), F. - 71°C.
c) CRL-Nummer 40 267
Während 1 Stunde oxydiert man bei 500C 15 g (0,06 Mol) 4-(p-Fluorphenylthio)-butyramidin-chlorhydrat, in
50 mi Essigsäure geiösi, mit 5,6 nil Wassersioffperoxidiörüng von 120 Valurnteilen. Man läßt über Nacht
stehen, dampft unter Vakuum ab, nimmt mit Aceton auf, zentrifugiert und kristallisiert aus Isopropanol um.
Man erhält die Verbindung CRL 40 267 mit 76%iger Ausbeute in Form von kleinen weißen Kristallen,
F. = 138°C.
Beispiel 11 4-(Phenylsulfinyl-butyramidoxim-chlorhydrat

a) 4-(Phenylthio)-butyramidoxim-chlorhydrat
Man gibt tropfenweise 22 g (0,21 MoI) y-Chlorbutyronitril zu einer bei 800C gerührten Lösung von 22 g
(0,2 Mol) Thiophenol und 8 g (0,2 MoI) NaOH in 200 ml Wasser. Man erhitzt V4 Stunde unter Rückfluß, kühlt
ab, extrahiert mit Mcthylenchlorid, wäscht mit Wasser, trocknet und dampft ab.
Das zurückbleibende Öl, das aus 35,4 g (0,2 Mol) 4-Phenylthiobutyronitril besteht, wird in 180 ml Butanol
gelöst und zu einer Lösung von 0,3 Mol Hydroxylamin in 40 ml Wasser gegeben. Man erhitzt 4 Stunden unter
Rückfluß, dampft unter Vakuum ein, gibt 11 Wasser zu, zentrifugiert, wäscht mit Wasser, dann mit Isopropyläther.
Mari erhält die freie Base (F. = 9p bis 91°C). Diese Base wird in Äthylacetat gelöst und durch Zugabe von
äthanbüscher Salzsäure in das Chlörhydrat überführt. Man erhält 22 g (Ausbeute 44%), F. = 77 bis 78°C.
b) CRL-Nummer 40 268
18,5 g (0,075 Mol) des voranstehend beschriebenen Chlorhydrats, gelöst in 75 ml Essigsäure, werden mit
7 ml Wasserstoffperoxid von 120 Volumteilen oxydiert. Man dampft die Essigsäure im Vakuum ab, nimmt
mit 100 ml Aceton aufund zentrifugiert. Man kristallisiert aus dem Äthanol-Aceton-Gemisch um und erhält die
Verbindung CRL-Nummer 40 268 mit 38%iger Ausbeute. Das Produkt liegt in Form eines weißen Pulvers
vor, F. = 152°C.
Beispiel 12

a) 4-Phenylthio-butyrohydroxamsäure
Man mischt 8,1 ml (0,078 Mol) Thiophenol, 15,6 g (0,08 Mol) Äthyl-y-brombutyrat in 50 ml Äthanol, gibt
3,2 g Natriumhydroxid in 40 ml Wasser hinzu, läßt nach anfänglicher Erwärmung 1 Stunde ruhen, dampft den
Alkohol ab, extrahiert mit Äther, wäscht mit Wasser, trocknet und dampft den Äther unter \y;uum ab. Man gibt
den so erhaltenen Ester in eine Kydroxyiarninlösung, die in der Kälte mit 7 g (0,1 Mol)! lydroxylarnrnoniurnchlorid
und 10,8 g (0,2 Mol) Natriumaiethylat in 100 ml Methanol hergestellt wurde. Nach 48stündigem
Kontakt dampft man unter Vakuum zur Tcockne ein. Man nimmt den Rückstand mit 200 ml Wasser auf, filtriert
über Aktivkohle, fallt das Filtrat mit konzentrierter Salzsäure aus, zentrifugiert und trocknet. Man kristallisiert
aus Isopropanol um und erhält 8 g (66%) vom F. = 78 bis 79°C.
b) CRL-Nummer 40 216
Man oxydiert 7,6 g (0,036 Mol) 4-Phenylthio-butyrohydroxamsäure, in 36 ml Essigsäure gelöst, mit 3,6 ml
(0,036 Mol) Wasserstoffperoxidlösung von 110 Volumteilen. Man dampft unter Vakuum zur Trockne ein und
zieht das Wasser ab, indem man mehrere Male mit wasserfreiem Äthanol aufnimmt und abdampft. Man kristallisiert
aus Äthylacetat um und erhält so die Verbindung CRL 40 216 mit 38%iger Ausbeute. Dieses Produkt liegt
in Form von kleinen beigen Kristallen vor, F. = 61 bis 62°C.

a) 4-(p-Fluorphenylthio)-butyrohydroxamsäure
Bei 00C gibt man 10 g (0,078 Mol) p-Fluorthiophenol zu einer Lösung von 4,3 g (0,08 Mol) Natriummethylat
in 25 ml Methanol, rührt und gibt tropfenweise bei 200C eine Lösung von 15,6 g (0,08 Mol) Äthyl-y-brombutyrat
in 20 ml Methanol zu. Die Temperatur des Gemisches errei^i? am Ende der Zugabe 400C. Man erwärmt 2 Stunden
unter Rückfluß, dampft unter Vakuum zur Trockne ein, nimmt mit Äther auf, wäscht mit verdünnterNatronlauge
und mit Wasser, trocknet und dampft unter Vakuum ein.
Man stellt eine Lösung von 7 g (0,1 Mol) Hydroxyiammoniumchlorid in 100 ml Methanol her, gibt bei 100C
eine kalte Lösung von 10,8 g (0,2 Mol) Natriummethylatin40 ml Methanol zu, filtriert und fügt 18 g (0,075 Mol)
Äthyl-4-(p-Fluorphenylthio)-butyrat zu. Man läßt 24 Stunden im Kontakt stehen, dampft unter Vakuum zw
trockne ein, nimmt mit 250 ml Wasser auf, filtriert über Aktivkohle unu Fällt mit konzentrierter HCl. Man filtrierl,
wäscht mit Wasser, trocknet und kristallisiert aus Äthylacetat um. Man erhält 12,8 g (74%), F. = 96°C.
b) CRL-Nummer 40 217
Man oxydiert 10,5 g (0,046 Mol) 4-(p-Fluorphenylthio)-butyrhydroxamsäure, in 45 ml Essigsäure gelöst, mit
4,6 ml Wasserstoffperoxidlösung von 110 Volumteilen. Nach 1 Stunde bei 500C läßt man über Nacht stehen und
dampft unter Vakuum zur Trockne ein. Man nimmt mit Äthylacetat auf und zentrifugiert. So erhält man die
Verbindung CRL 40 217 mit 62% Ausbeute in Form von kleinen beigen Kristallen. F. = 109 bis 1100C.

Man gibt 10 g (0,078 MoI) p-Fluorthiophenol zu einer Lösung von 4,5 g (0,0083 Mol) Natriummethylat in
25 ml Methanol und dann tropfenweise eine Lösung von 14,5 g (0,08 Mol) Äthyl-3-brompropionat in 20 ml
Methanol hinzu. Man erwärmt 2 Stunden unter Rückfluß, zieht den Alkohol unter Vakuum ab, nimmt mit Äther
auf, wäscht mit Wasser, trocknet und dampft den Äther ab. Man behandelt 24 Stunden mit einer Lösung von
Hydroxylamin in Methanol, die aus 7,7 g (0,11 Mol) Hydroxylammoniumchlorid und 12 g (0,22 MoI) Natriummethylat
in 150 ml Methanol hergestellt wurde, bei 200C. Man dampft das Methanol unter Vakuum ein, gibt
200 ml Wasser zu, extrahiert mit Äther und fällt die Hydroxamsäure mit konzentriertem HCl aus. Man zentrifugiert,
wäscht mit Wasser und trocknet und erhält 10 g (62%). F. = 93°C.
b) CRL-Nummer40 219
Man oxydiert 9,85 g (0,046 Mol) 3-(p-Fluorphenylthio)-propiohydroxamsäure, in 46 ml Essigsäure gelöst,
durch Zugabe von 4,6 ml Wasserstoffperoxidlösung von 110 Volumteilen. Man läßt 3 Stunden in Kontakt,
dampft unter Vakuum zur Trockne ein, nimmt mit Äthylacetat auf, filtriert und kristallisiert aus Äthanol um.
Man erhält die Verbindung CRL 40 219 mit 52%iger Ausbeute. Dieses Produkt liegt in Form von kleinen
weißen Kristallen vor. F. = 133°C.

a) Äthyl-4-(p-chlorphenylthio)-butyrat
Bei 00C gibt man 14,5 g (0.1 Mol) p-ChlorthiophenoI zu einer Lösung von 5,4 g (0,1 Mol) Natriummethylat in
50 mi Methanol, dann, unter Rühren, bei 2O0C tropfenweise 19,5 g (0,1 Mol) Äthyl-y-brombutyrat in 40 ml
Methanol gelöst, hinzu. Man erhitzt 2 Stunden unter Rückfluß, zieht den Alkohol unter Vakuum ab, extrahiert so
mit Äther, wäscht mit Wasser, trocknet und dampft den Äther ab. Man erhält 24 g (93%) eines weißen Öls.
b) 4-(p-Chlorphenylthio)-butyrhydroxamsäure
Man stellt eine Hydroxylaminlösung mit Hilfe von 10,5 g (0,15 Mol) Hydroxylammoniumchlorid in 150 ml
Methanol gelöst, her, zu der man eine Lösung von 13,5 g (0,25 Mol) Natriummethylat in 100 ml Methanol
zugibt. Man filtriert das Natriumchlorid ab und gibt zum Filtrat 24 g (0,092 Mol) Äthyl-4-(p-chlorphenylthio)-butyrat.
Man läßt 24 Stunden im Kontakt, zieht den Alkohol im Vakuum ab, nimmt mit 250 ml Wasser auf,
filtriert und fällt die Säure mit konzentrierter Salzsäure aus. Man zentrifugiert, wäscht mit Wasser, trocknet und
kristallisiert aus Eenzol um. Man erhält 22 g (96%) des Produkts vom F. = 76 bis 77°C.
c) CRL-Nummer 40 265
In eine Lösung von 19,6 g (0,08 Mol) des vorstehend beschriebenen Sulfids in 80 ml Essigsäure gibt man 8 ml
V/ässersiofiperoxidiösung von 110 Voiurnteilen. Nach Isiündigern Erwärmen auf 500C und Stehenlassen über
Nacht dampft man unter Vakuum ein, gibt 100 ml Äthylacetat hinzu, zentrifugiert und kristallisiert aus Äthanol
um. Man erMlt so die Verbindung CRL 40 265 mit 80%iger Ausbeute in Form von feinen weißen Nadeln.
F. = 126 bis 127°C.
