Abstract
Die vorliegende Erfindung ist ausgerichtet auf kristallines Atorvastatin(d.h. [R-(R*,R*)]-2-(4-Fluorphenyl)-ß,d-dihydroxy-5-(1-methylethyl)-3-phenyl-4-[(phenylamino)carbonyl]-1H-pyrrol-1-heptansäure-hemicalciumsalz)-Hydrat der Form I, eine pharmazeutische Zusammensetzung, die kristallines Atorvastatinhydrat der Form I enthält sowie die Verwendung eines Atorvastatinhydrats der Form I.

Hintergrund der Erfindung
Die vorliegende Erfindung betrifft eine neue kristalline Form von Atorvastatin, das unter dem chemischen Namen [R-(R*,R*)]-2-(4-Fluorphenyl)-ß,ö-dihydroxy-5-(1-miethylethyl)-3-phenyl-4-5 [(phenylamino)carbonyl]-1 H-pyrrol-1-heptansäure-hemicalciumsalz bekannt ist, die als pharmazeutisches Mittel verwendbar ist, Verfahren zu deren Herstellung und Isolation, diese Verbindung und einen pharmazeutisch akzeptablen Träger enthaltende pharmazeutische Zusammensetzungen und pharmazeutische Behandlungsverfahren. Die neue kristalline Verbindung der vorliegenden Erfindung ist als Inhibitor des Enzyms 3-Hydroxy-3-methylglutaryl-coenzym-A-io Reduktase (HMG-CoA-Reduktase) verwendbar und sie ist daher ein verwendbares Hypolipidä-mie- und Hypocholesterolämiemittel.
Das US-Patent Nr. 4 681 893, das hier als Bezug aufgenommen ist offenbart bestimmte trans-[2-(3- oder 4-Carboxamido-subtituiertes-Pyrrol-1-yl)alkyl]-4-hydroxy-pyran-2-one einschließlich 15 von trans-(±)-5-(4-Fluorphenyl)-2-(1-methylethyl)-N,4-diphenyl-1-[(2-tetrahydro-4-hydroxy-6-oxo-2H-pyran-2-yl)ethyl]-1H-pyrrol-3-carboxamid.
Das US-Patent Nr. 5 273 995, das hier als Bezug aufgenommen ist, offenbart das Enantiomer mit der R-Form der ringgeöffneten Säure von trans-5-(4-Fluorphenyl)-2-(1-methylethyl)-N,4-20 diphenyl-1 -[(2-tetrahydro-4-hydroxy-6-oxo-2H-pyran-2-yl)ethyl]-1 H-pyrrol-3-carboxamid, d.h. [R-(R*,R*)]-2-(4-Fluorphenyl)-ß,ö-dihydroxy-5-(1-methylethyl)-3-phenyl-4-[(phenylamino)carbonyl]-1 H-pyrrol-1 -heptansäure.
Die US-Patente Nr. 5 003 080, 5 097 045, 5 103 024, 5 124 482, 5 149 837, 5 155 251, 25 5 216 174, 5 245 047, 5 248 793, 5 280 126, 5 397 792 und 5 342 952, die hier als Bezug auf genommen sind, offenbaren verschiedene Verfahren und Schlüsselzwischenprodukte zur Herstellung von Atorvastatin.
Atorvastatin wird als dessen Calciumsalz, d.h. [R-(R*,R*)]-2-(4-Fluorphenyl)-ß,ö-dihydroxy-5-(1-30 methylethyl)-3-phenyl-4-[(phenylamino)carbonyl]-1 H-pyrrol-1 -heptansäure-hemicalciumsalz (2:1) hergestellt. Das Calciumsalz ist günstig, da es eine passende Formulierung von Atorvastatin in beispielsweise Tabletten, Kapseln, Pastillen, Pulvern und dgl. zur oralen Verabreichung ermöglicht. Außerdem besteht Bedarf nach einer Herstellung von Atorvastatin in reiner und kristalliner Form, um Formulierungen zu ermöglichen, die exakte pharmazeutische Anforderun-35 gen und Spezifikationen erfüllen.
Des weiteren muss das Verfahren, nach dem Atorvastatin hergestellt wird, eines sein, das einer Produktion im großen Maßstab zugänglich ist. Außerdem ist es günstig, wenn das Produkt in einer Form vorliegt, die problemlos filtrierbar und leicht zu trocknen ist. Schließlich ist es wirt-40 schaftlich günstig, wenn das Produkt über einen längeren Zeitraum ohne die Notwendigkeit von speziellen Lagerungsbedingungen stabil ist.
Die Verfahren in den obigen US-Patenten offenbaren amorphes Atorvastatin, das ungeeignete Filtrations- und Trocknungseigenschaften zur Herstellung im großen Maßstab aufweist und vor 45 Wärme, Licht, Sauerstoff und Feuchtigkeit geschützt werden muss.
Die WO-A-94/16693 offenbart eine orale pharmazeutische Zusammensetzung zur Behandlung von Hypercholesterolämie oder Hyperlipidämie, die eine vorteilhafte Formulierung zur Stabilisierung des HMG-CoA-Coenzym-A-lnhibitors CI-981 Hemi-Calcium der Formel (IA) mit wirksamen so Mengen von Calciumcarbonat enthält. 55 5 3 AT 008 453 U1

Die US-A-5 316 765 offenbart ein Verfahren zur Verbesserung der Reduktionen von Coenzym 15 Q10 bei Cardiomyopathiepatienten, die einen HMG-CoA-Reduktaseinhibitor erhalten, das das gleichzeitige Verabreichen des HMG-CoA-Reduktaseinhibitors und von Coenzym Q10 mit einer so ausreichenden Konzentration, dass die Serumspiegel von Coenzym Q10 auf mindestens 2,0 pg/ml erhöht werden, umfasst, wobei als HMG-CoA-Reduktaseinhibitor CI-981 mit der folgenden Formel verwendet werden kann: 20

Die EP-A-0409281 offenbart [R-(R*,R*)]-2-(4-Fluorphenyl)-ß,5-dihydroxy-5-(1-methylethyl)-3-pheny l-4-[(phenylamino)carbonyl]-1H-pyrrol-1-heptansäure oder (2R-trans)-5-(4-Fluorphenyl)- 2- (1-methylethyl-N,4-diphenyl-1-[2-(tetrahydro-4-hydroxy-6-oxo-2H-pyran-2-yl)ethyl]-1H-pyrrol- 3- carboxamid, ein Verfahren zu deren Herstellung und pharmazeutisch akzeptable Salze der-50 selben.
Wir ermittelten nun überraschenderweise und unerwarteterweise, dass Atorvastatin in kristalliner Form hergestellt werden kann. Daher erfolgt durch die vorliegende Erfindung die Bereitstellung von Atorvastatin in einer neuen kristallinen Form mit der Bezeichnung Form I. 4 AT 008 453 U1
Atorvastatinhydrat der Form I besteht aus einer kleineren Teilchen- und einer gleichförmigeren Größenverteilung als das frühere amorphe Produkt und es zeigt günstigere Filtrations- und Trocknungseigenschaften. Außerdem ist Atorvastatinhydrat der Form I reiner und stabiler als das amorphe Produkt.
Zusammenfassung der Erfindung
Daher ist die vorliegende Erfindung ausgerichtet auf 1. Kristallines Atorvastatin(d.h. [R-(R*,R*)]-2-(4-Fluorphenyl)-ß,ö-dihydroxy-5-(1-methylethyl)-3-phenyl-4-[(phenylamino)carbonyl]-1 H-pyrrol-1 -heptansäure-hemicalciumsalz)-Hydrat der Form I mit einem Pulverröntgenbeugungsmuster, das die im folgenden angegebenen, unter Verwendung von CuKa-Strahlung ermittelten 20-Werte enthält: 9,150, 9,470, 10,266, 10,560, 11,853, 12,195, 17,075, 19,485, 21,626, 21,960, 22,748, 23,335, 23,734, 24,438, 28,915 und 29,234. 2. Kristallines Atorvastatinhydrat der Form I, charakterisiert durch Festkörper-13C-Kernresonanz mit den im folgenden angegebenen chemischen Verschiebungen in ppm (parts per million): 21,3, 25,2, 26,4, 40,2, 41,9, 47,4, 64,9, 68,1, 70,5, 73,1, 113,8, 118,2, 120,9, 123,5, 127,6,129,5,131,1,134,9, 137,0, 159,3,166,7 (breit), 178,4 und 182,8. 3. Kristallines Atorvastatin der Form I gemäß einem der vorhergehenden Punkte, das 1-8 Mole Wasser enthält. 4. Kristallines Atorvastatin der Form I gemäß einem der vorhergehenden Punkte, das 3 Mole Wasser enthält. 5. Pharmazeutische Zusammensetzung, die kristallines Atorvastatinhydrat der Form I gemäß einem der vorhergehenden Punkte im Gemisch mit mindestens einem pharmazeutisch akzeptablen Streckmittel, Verdünnungsmittel oder Träger enthält. 6. Pharmazeutische Zusammensetzung gemäß Punkt 5 in Form von Tabletten, Pillen, dispergierbaren Körnchen, Kachets, Kapseln, Pulvern, Pastillen, Suppositorien oder Retentionsklistieren. 7. Verwendung eines kristallinen Atorvastatinhydrats der Form I gemäß einem der Punkte 1-4 in der Medizin.
Als Inhibitoren von HMG-CoA ist die neue kristalline Form von Atorvastatin ein brauchbares Hypolipidämie- und Hypocholesterolämiemittel.

Detaillierte Beschreibung der Erfindung
Kristallines Atorvastatinhydrat der Form I kann durch sein Pulverröntgenbeugungsmuster und/oder sein Festkörperkernresonanzspektrum (NMR) charakterisiert werden. 5 AT 008 453 U1
Pulverröntgenbeugung
Atorvastatinhydrat der Form I wurde durch sein Pulverröntgenbeugungsmuster charakterisiert. Daher wurde das Röntgenbeugungsmuster von Atorvastatinhydrat der Form I auf einem Sie-mens-D-500-Diffraktometer mit CuKa-Strahlung ermittelt.
Geräte
Siemens-D-500-Diffraktometer-Kristalloflex mit einer IBM-kompatiblen Schnittstelle, Softwaren DIFFRAC AT (SOCABIM 1986, 1992).
CuKa-Strahlung (20 mA, 40 kV, λ = 1,5406 A, Schlitze I und II bei 1°) elektronisch gefiltert durch den Psi-Peltier-gekühlten Silicium[Si(Li)]-Detektor (Schlitze: III bei 1° und IV bei 0,15°).
Methodik
Der Siliciumstandard wird jeden Tag zur Überprüfung der Röntgenröhrenausrichtung laufengelassen. Kontinuierliche Θ/2Θ -gekoppelte Abtastung: 4,00° bis 40,00° in 2Θ, Abtastrate 6°/min: 0,4 s/0,04°-Stufe. Eine Probe wird aus der Phiole laufengelassen und auf Nullhintergrundquarz in Aluminiumhalterung gepresst. Probenbreite 13-15 mm.
Die Proben werden bei Raumtemperatur gelagert und laufengelassen.
Mahlen/Sieben
Mahlen wird zur Minimierung von Intensitätsschwankungen für das hier offenbarte Beugungsdiagramm verwendet. Wenn jedoch ein Mahlen das Beugungsdiagramm deutlich veränderte oder den amorphen Anteil der Probe erhöhte, wurde das Beugungsdiagramm der nichtgemahlenen Probe verwendet. Das Mahlen erfolgte mit einem kleinen Achatmörser und -pistill. Der Mörser wurde während des Mahlens gehalten und auf das Pistill wurde leichter Druck ausgeübt.
Tabelle 1 gibt die 20-Werte, die d-Abstände und die relativen Intensitäten aller Linien in der nichtgemahlenen Probe mit einer relativen Intensität von >20 % für kristallines Atorvastatinhydrat der Form I an. Tabelle 1 gibt auch die relativen Intensitäten der gleichen Linien in einem nach zweiminütigem Mahlen gemessenen Beugungsdiagramm an. Die Intensitäten der 2 Minuten gemahlenen Probe sind repräsentativer für das Beugungsmuster ohne bevorzugte Orientierung. Es ist auch anzumerken, dass die Rechner-erzeugten, nichtgerundeten Zahlen in dieser Tabelle angegeben sind.

Festkörperkernresonanz (NMR) 20 Methodik
Alle 13C-Festkörper-NMR-Messungen wurden mit einem 250-MHz-NMR-Spektrometer Bruker AX-250 gemacht. Hochauflösende Spektren wurden unter Verwendung von Hochleistungsprotonenentkopplung und Kreuzpolarisatiuon (CP) mit Magic-Angle-Spinning (MAS) bei etwa 5 kHz 25 erhalten. Der Magic-Angle wurde unter Verwendung des Br-Signals von KBr durch Erfassen der Seitenbanden gemäß der Beschreibung von Frye und Maciel (J.S. Frye und G.E. Maciel, J. Mag. Res., 1982, 48:125) eingestellt. Etwa 300 bis 450 mg Probe, die in einen Kanistermodellrotor gepackt waren, wurden für jedes Experiment verwendet. Die chemischen Verschiebungen wurden auf externes Tetrakis(trimethylsilyl)silan (Methylsignal bei 3,50 ppm) bezogen 30 (J.V. Muntean und L.M. Stock, J. Mag. Res., 1988, 76:54).
Tabelle 2 zeigt das Festkörper-NMR-Spekrum für kristallines Atorvastatinhydrat der Form I.

Kristallines Atorvastatinhydrat der Form I gemäß der vorliegenden Erfindung liegt in Hydratfor-35 men vor. Kristallines Atorvastatinhydrat der Form I enthält etwa 1 bis 8 mol Wasser. Vorzugsweise enthält Atorvastatinhydrat der Form I 3 mol Wasser.
Durch die vorliegende Erfindung erfolgt die Bereitstellung eines Verfahrens zur Herstellung von kristallinem Atorvastatinhydrat der Form I, das das Kristallisieren von Atorvastatin aus einer 40 Lösung in Lösemitteln unter Bedingungen, die kristallines Atorvastatinhydrat der Form I ergeben, umfasst.
Die genauen Bedingungen, unter denen kristallines Atorvastatinhydrat der Form I gebildet wird, können empirisch bestimmt werden und es ist nur möglich, eine Anzahl von Verfahren an-45 zugeben, die sich in der Praxis als geeignet erwiesen.
Daher kann beispielsweise kristallines Atorvastatinhydrat der Form I durch Kristallisation unter kontrollierten Bedingungen hergestellt werden. Insbesondere kann es entweder aus einer wässrigen Lösung des entsprechenden Basesalzes, wie ein Alkalimetallsalz, beispielsweise Lithium, so Kalium, Natrium und dgl.; Ammoniak oder ein Aminsalz; vorzugsweise Natriumsalz durch Zugabe eines Calciumsalzes, wie beispielsweise Calciumacetat und dgl., oder durch Suspendieren von amorphem Atorvastatin in Wasser, hergestellt werden. Beispielsweise ist die Verwendung eines Hydroxyl-Colösemittels, wie beispielsweise ein niedriges Alkanol, beispielsweise Methanol und dgl., bevorzugt. 55 δ ΑΤ 008 453 U1
Wenn das Ausgangsmaterial zur Herstellung des gewünschten kristallinen Atorvastatinhydrat der Form I eine Lösung des entsprechenden Natriumsalzes ist, umfasst eine bevorzugte Herstellung die Behandlung einer Lösung des Natriumsalzes in Wasser, das nicht weniger als etwa 5 % (V/V) Methanol, zweckmäßigerweise etwa 5 % bis 33 % (V/V) Methanol, vorzugsweise 5 etwa 10 % bis 15 % (V/V) Methanol enthält, mit einer wässrigen Lösung von Calciumacetat zweckmäßigerweise bei einer erhöhten Temperatur von bis zu etwa 70 °C, beispielsweise etwa 45-60 °C, vorzugsweise etwa 47-52 °C. Vorzugsweise wird Calciumacetat und im allgemeinen 1 Mol Calciumacetat gegenüber 2 Molen des Natriumsalzes von Atorvastatin verwendet. Unter diesen Bedingungen sollte die Calciumsalzbildung sowie die Kristallisation vorzugsweise bei io einer erhöhten Temperatur, beispielsweise innerhalb der im vorhergehenden genannten Temperaturbereiche, durchgeführt werden. Es zeigte sich, dass es vorteilhaft sein kann, wenn die Ausgangslösung eine kleine Menge von Methyl-tert.-butylether (MTBE), beispielsweise etwa 7 Gew.-% enthält. Es zeigte sich häufig, dass es günstig ist, "Keime" von kristallinem Atorvastatinhydrat der Form I der Kristallisationslösung zuzusetzen, um kristallines Atorvastatinhydrat der 15 Form I konsistent zu erzeugen.
Wenn das Ausgangsmaterial amorphes Atorvastatin oder eine Kombination von amorphem und kristallinem Atorvastatinhydrat der Form I ist, kann das gewünschte kristalline Atorvastatinhydrat der Form I durch Suspendieren des Feststoffs in Wasser, das etwa 40 % (V/V), beispielsweise 20 etwa 0 % bis 20 % (V/V), vorzugsweise etwa 5 % bis 15 % (V/V) Colösemittel, beispielsweise Methanol, Ethanol, 2-Propanol, Aceton und dgl. enthält, bis die Umwandlung in die gewünschte Form vollständig ist, und eine anschließende Filtration erhalten werden. Es zeigte sich häufig, dass es günstig ist, "Keime" von kristallinem Atorvastatinhydrat der Form I zur Suspension zu geben, um eine vollständige Umwandlung in kristallines Atorvastatinhydrat der Form I sicherzu-25 stellen. Alternativ kann ein wasserfeuchter Kuchen, der hauptsächlich aus amorphem Atorvastatin besteht, auf höhere Temperaturen, beispielsweise bis zu etwa 75 °C, vorzugsweise etwa 65-70 °C erhitzt werden, bis eine signifikante Menge von kristallinem Atorvastatinhydrat der Form I vorhanden ist, wobei dann das amorphe/kristalline-Form-l-Gemisch wie im vorhergehenden beschrieben aufgeschlämmt werden kann. 30
Kristallines Atorvastatinhydrat der Form I ist deutlich leichter zu isolieren als amorphes Atorvastatin und es kann aus dem Kristallisationsmedium nach dem Abkühlen abfiltriert und gewaschen und getrocknet werden. Beispielsweise war die Filtration einer 50-ml-Aufschlämmung von kristallinem Atorvastatinhydrat der Form I innerhalb von 10 s beendet. Die Filtration einer ähn-35 lieh große Probe von amorphem Atorvastatin dauerte mehr als 1 h.
Die erfindungsgemäßen Verbindungen können in einer großen Vielzahl oraler und parenteraler Dosierungsformen hergestellt und verabreicht werden. Daher können die Verbindungen der vorliegenden Erfindung durch Injektion, d.h. intravenös, intramuskulär, intrakutan, subkutan, 40 intraduodenal oder intraperitoneal verabreicht werden. Auch können die Verbindungen der vorliegenden Erfindung durch Inhalation, beispielsweise intranasal verabreicht werden. Außerdem können die Verbindungen der vorliegenden Erfindung transdermal verabreicht werden. Einem Fachmann ist klar, dass die im folgenden angegebenen Dosierungsformen als aktive Komponente entweder Verbindungen oder ein entsprechendes pharmazeutisch akzeptables 45 Salz einer Verbindung der vorliegenden Erfindung umfassen können.
Zur Herstellung pharmazeutischer Zusammensetzungen aus den Verbindungen der vorliegenden Erfindung können pharmazeutisch akzeptable Träger entweder fest oder flüssig sein. Zubereitungen fester Form umfassen Pulver, Tabletten, Pillen, Kapseln, Kachets, Suppositorien und so dispergierbare Granulatkörnchen. Ein fester Träger können eine oder mehrere Substanzen sein, die auch als Verdünnungsmittel, Geschmacksmittel, löslichmachende Stoffe, Gleitmittel, Suspendiermittel, Bindemittel, Konservierungsmittel, den Tablettenzerfall fördernde Mittel oder Einkapselungsmaterial fungieren können. 55 In Pulvern ist der Träger ein feinverteilter Feststoff, der im Gemisch mit der feinverteilten aktiven 9 AT 008 453 U1
Komponente vorliegt.
In Tabletten ist die aktive Komponente mit dem Träger, der die notwendigen Bindungseigenschaften besitzt, in passenden Anteilen gemischt und in der gewünschten Form und Größe 5 kompaktiert.
Die Pulver und Tabletten enthalten vorzugsweise 2 oder 10 bis etwa 70 % der aktiven Verbindung. Geeignete Träger sind Magnesiumcarbonat, Magnesiumstearat, Talkum, Zucker, Lactose, Pektin, Dextrin, Stärke, Gelatine, Tragant, Methylcellulose, Natriumcarboxymethylcellulose, io ein niedrigschmelzendes Wachs, Kakaobutter und dgl. Der Ausdruck "Zubereitung" soll die Formulierung der aktiven Verbindung mit Einkapselungsmaterial als Träger, das eine Kapsel bildet, in der die aktive Komponente mit oder ohne andere Träger von einem Träger umgeben ist, der daher mit dieser in Verbindung ist, umfassen. In ähnlicher Weise werden Kachets und Pastillen umfasst. Tabletten, Pulver, Kapseln, Pillen, Kachets und Pastillen können als zur 15 oralen Verabreichung geeignete feste Dosierungsformen verwendet werden.
Zur Herstellung von Suppositorien wird ein niedrigschmelzendes Wachs, beispielsweise ein Gemisch von Fettsäureglyceriden oder Kakaobutter, zunächst geschmolzen und die aktive Komponente wird darin, beispielsweise durch Rühren, homogen verteilt. Das geschmolzene 20 homogene Gemisch wird dann in Formen passender Größe gegossen, abkühlen gelassen und dadurch verfestigt.
Zubereitungen flüssiger Formen umfassen Lösungen, Suspensionen, Retentionsklistiere und Emulsionen, beispielsweise Wasser- oder Wasser-Propylenglykol-Lösungen. Zur parenteralen 25 Injektion können flüssige Zubereitungen in Lösung in wässriger Polyethylenglykollösung formuliert werden.
Zur oralen Verwendung geeignete wässrige Lösungen können durch Auflösen der aktiven Komponente in Wasser und Zugeben geeigneter Färbemittel, Geschmacksstoffe, Stabilisatoren 30 und von Dickungsmitteln je nach Wunsch hergestellt werden.
Zur oralen Verwendung geeignete wässrige Suspensionen können durch Dispergieren der feinzerteilten aktiven Komponente in Wasser mit viskosem Material, wie natürlichen oder synthetischen Gummis, Harzen, Methylcellulose, Natriumcarb-oxymethylcellulose, und anderen 35 bekannten Suspendiermitteln hergestellt werden.
Ebenfalls umfasst werden Zubereitungen fester Form, die kurz vor der Verwendung in Zubereitungen flüssiger Form zur oralen Verabreichung umgewandelt werden sollen. Diese flüssigen Formen umfassen Lösungen, Suspensionen und Emulsionen. Diese Zubereitungen können 40 zusätzlich zu der aktiven Komponente Färbemittel, Geschmacksstoffe, Stabilisatoren, Puffermittel, künstliche und natürliche Süßungsmittel, Dispergiermittel, Dickungsmittel, löslichmachende Mittel und dgl. enthalten.
Die pharmazeutische Zubereitung liegt vorzugsweise in Einheitsdosisform vor. In dieser Form 45 ist die Zubereitung in Einheitsdosen, die passende Mengen der aktiven Komponente enthalten, unterteilt. Die Einheitsdosisform kann eine abgepackte Zubereitung sein, wobei die Packung diskrete Menge der Zubereitung enthält, wie abgepackte Tabletten, Kapseln und Pulver in Phiolen oder Ampullen. Auch kann die Einheitsdosisform eine Kapsel, Tablette, ein Kachet oder eine Pastille selbst sein oder es kann die entsprechende Anzahl von diesen in abgepackter so Form sein.
Die Menge der aktiven Komponente in einer Einheitsdosiszubereitung kann von 0,5 mg bis 100 mg, vorzugsweise 2,5 mg bis 80 mg gemäß der speziellen Anwendung und der Stärke der aktiven Komponente variiert oder eingestellt werden. Die Zusammensetzung kann je nach 55 Wunsch auch andere kompatible therapeutische Mittel enthalten.
Claims (7)
10 AT 008 453 U1 Bei der therapeutischen Verwendung als Hypolipidämie- und/oder Hypocholesterolämiemittel wird das beim erfindungsgemäßen pharmazeutischen Verfahren verwendete kristalline Ator-vastatinhydrat der Form I mit einer Anfangsdosis von täglich etwa 2,5 mg bis etwa 80 mg verabreicht. Ein Tagesdosisbereich von etwa 2,5 mg bis etwa 20 mg ist bevorzugt. Die Dosisgaben 5 können jedoch in Abhängigkeit von den Bedürfnissen des Patienten, der Schwere der zu behandelnden Störung und der verwendeten Verbindung variiert werden. Die Bestimmung der passenden Dosierung für eine spezielle Situation ist einschlägig bekannt. Im allgemeinen wird eine Behandlung mit kleineren Dosisgaben, die kleiner als die optimale Dosis der Verbindung sind, begonnen. Danach wird die Dosierung in kleinen Inkrementen erhöht, bis die unter den io Umständen optimale Wirkung erreicht ist. Passenderweise kann die Gesamttagesdosis unterteilt und in Portionen während des Tages je nach Wunsch verabreicht werden. Die im folgenden angegebenen nichtbeschränkenden Beispiele erläutern die bevorzugten Verfahren der Erfinder zur Herstellung der erfindungsgemäßen Verbindungen. 15 Beispiel 1 [R-(R*,R*)]-2-(4-Fluorphenyl)-ß,ö-dihydroxy-5-(1-methylethyl)-3-phenyl-4-[(phenylamino)carbo-nyl]-1H-pyrrol-1-heptansäure-hemicalciumsalz (Atorvastatin der Form I) 20 Verfahren A Ein Gemisch von (2R-trans)-5-(4-Fluorphenyl)-2-(1-methylethyl)-N,4-diphenyl-1-[2-(tetrahydro-4-hydroxy-6-oxo-2H-pyran-2-yl)ethyl]-1 H-pyrrol-3-carboxamid (Atorvastatinlacton) (US-Patent 25 Nr. 5 273 995) (75 kg), Methy-tert.-butylether (MTBE) (308 kg), Methanol (190 I) wird mit einer wässrigen Lösung von Natriumhydroxid (5,72 kg in 950 I) bei 48-58 °C während 40-60 min umgesetzt, wobei das ringgeöffnete Natriumsalz gebildet wird. Nach dem Abkühlen auf 25-35 °C wird die organische Schicht verworfen und die wässrige Schicht erneut mit MTBE (230 kg) extrahiert. Die organische Schicht wird verworfen und die mit MTBE gesättigte wässri-30 ge Lösung des Natriumsalzes wird auf 47-52 °C erhitzt. Zu dieser Lösung wird über mindestens 30 min eine Lösung von in Wasser (410 I) gelöstem Calciumacetathemihydrat (11,94 kg) gegeben. Das Gemisch wird mit einer Aufschlämmung von kristallinem Atorvastatin der Form I (1,1 kg in 11 I Wasser und 5 I Methanol) kurz nach der Zugabe der Calciumacetatlösung beimpft. Das Gemisch wird dann mindestens 10 min auf 51-57 °C erhitzt und dann auf 15-40 °C 35 gekühlt. Das Gemisch wird filtriert, mit einer Lösung von Wasser (300 I) und Methanol (150 I) und anschließend Wasser (450 I) gewaschen. Der Feststoff wird unter Vakuum bei 60-70 °C 3-4 Tage lang getrocknet, wobei kristallines Atorvastatin der Form I (72,2 kg) erhalten wird. Verfahren B 40 Amorphes Atorvastatin (9 g) und kristallines Atorvastatin der Form I (1 g) werden bei etwa 40 °C in einem Gemisch von Wasser (170 ml) und Methanol (30 ml) insgesamt 17 h lang gerührt. Das Gemisch wird filtriert mit Wasser gespült und bei 70 °C unter vermindertem Druck getrocknet, wobei kristallines Atorvastatin, der Form I (9,7 g) erhalten wird. 45 Ansprüche: 1. Kristallines Atorvastatin(d.h. [R-(R*,R*)]-2-(4-Fluorphenyl)-ß,ö-dihydroxy-5-(1-methylethyl)-50 3-phenyl-4-[(phenylamino)carbonyl]-1 H-pyrrol-1-heptansäure-hemicalciumsalz)-Hydrat der Form I mit einem Pulverröntgenbeugungsmuster, das die im folgenden angegebenen, unter Verwendung von CuKa-Strahlung ermittelten 20-Werte enthält: 9,150, 9,470, 10,266, 10,560, 11,853, 12,195, 17,075, 19,485, 21,626, 21,960, 22,748, 23,335, 23,734, 24,438, 28,915 und 29,234. 55 AT008 453U1
2. Kristallines Atorvastatinhydrat der Form I, charakterisiert durch Festkörper-13C-Kernresonanz mit den im folgenden angegebenen chemischen Verschiebungen in ppm (parts per million): 21,3, 25,2, 26,4, 40,2, 41,9, 47,4, 64,9, 68,1, 70,5, 73,1, 113,8, 118,2, 120,9,123,5, 127,6, 129/5, 131,1, 134,9, 137,0, 159,3, 166,7 (breit), 178,4 und 182,8.
3. Kristallines Atorvastatin der Form I gemäß einem der vorhergehenden Ansprüche, das 1-8 Mole Wasser enthält.
4. Kristallines Atorvastatin der Form I gemäß einem der vorhergehenden Ansprüche, das 3 Mole Wasser enthält.
5. Pharmazeutische Zusammensetzung, die kristallines Atorvastatinhydrat der Form I gemäß einem der vorhergehenden Ansprüche im Gemisch mit mindestens einem pharmazeutisch akzeptablen Streckmittel, Verdünnungsmittel oder Träger enthält.
6. Pharmazeutische Zusammensetzung gemäß Anspruch 5 in Form von Tabletten, Pillen, dispergierbaren Körnchen, Kachets, Kapseln, Pulvern, Pastillen, Suppositorien oder Retentionsklistieren.
7. Verwendung eines kristallinen Atorvastatinhydrats der Form I gemäß einem der Ansprüche 1-4 in der Medizin. Hiezu 2 Blatt Zeichnungen
