<Desc/Clms Page number 1> Die Erfindung betrifft ein Verfahren zur Herstellung neuer substituierter Pyrrolidine der allgemeinen Formel EMI1.1 worin Ru Cycloalkyl, Cycloalkenyl, Bicycloalkyl oder Tricycloalkyl, A eine Einfachbindung oder eine gesättigte oder ungesättigte Kohlenwasserstoffkette mit 1 bis 3 Kohlenstoffatomen, n 0, 1, 2 oder 3, Rs Wasserstoff, Alkyl mit 1 bis 3 Kohlenstoffatomen, Alkenyl oder Alkinyl und Ri, R2, R3 sowie R6 Wasserstoff, Halogen, Alkyl, Alkoxy, Amino, Acetamido, Sulfamoyl, Alkyl- sulfamoyl, Dialkylsulfamoyl, Alkylsulfonyl oder Alkylsulfinyl bedeuten oder untereinander unter Ausbildung einer Azimidogruppe verknüpft sind, mit den folgenden Massgaben : - wenn Rt für Cycloalkyl, Rs für Wasserstoff oder Alkyl, A für eine Einfachbindung und n für 0 stehen, stellt mindestens einer der Substituenten Ri, R 2, R3 und R6 eine Alkyl- sulfonyl- oder Alkylsulfinylgruppe dar oder bilden zwei dieser Substituenten eine Azimido- gruppe, - wenn Ru für Cycloalkyl, Rs für Methyl, A für Alkylen mit 1 bis 3 Kohlenstoffatomen und n für 1 stehen und die Amidkette an Position 2 des Pyrrolidins gebunden ist, stellt R, Halogen, Sulfamoyl, Alkylsulfamoyl, Dialkylsulfamoyl oder Alkylsulfonyl dar, u. zw. ledig- lich in denjenigen Fällen, in denen R2, R3 und R6 nicht gleichzeitig Wasserstoff bedeuten, sowie deren Additionssalzen mit pharmakologisch verträglichen Säuren. Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass eine substituierte Benzoesäure der allgemeinen Formel EMI1.2 worin Rl, R, R , Rs und R6 wie oben definiert sind, oder ein reaktives Derivat derselben, mit einem primären Amin der allgemeinen Formel EMI1.3 in der n wie oben definiert ist und Hal ein Chlor-, Brom- oder Jodatom bedeutet, und die so erhaltene Verbindung der allgemeinen Formel <Desc/Clms Page number 2> EMI2.1 in der R1, R2, R1, R5, R6, Hal und n wie oben definiert sind, mit einem Amin der allgemeinen Formel H2N - A - R., (V) in der A und R4 wie oben definiert sind, umgesetzt wird. Als reaktive Derivate der Benzoesäure der allgemeinen Formel (II) können beispielsweise ein Säurechlorid, ein Alkylester, reaktive Ester, wie Methoxymethylester oder Cyanmethylester, ein aromatischer Ester, N-Hydroxyimidester, symmetrische Anhydride oder gemischte Anhydride, beispielsweise gebildet aus einem Kohlensäureester oder einem Halogenameisensäureester, ein Säureazid, Hydrazid, Azolid, Säureisothiocyanat, Trichloracetophenon- oder Triphenylphosphin-Derivate eingesetzt werden. Es ist auch möglich, die freie Säure und das freie Amin in Gegenwart eines Kondensierungsmittels, z. B. Siliziumtetrachlorid, Trichlorphenylsilan, Phosphorsäureanhydrid, eines Carbodiimids oder eines Alkoxyacetylens umzusetzen. Das Verfahren zur Herstellung der neuen Verbindungen ist in dem folgenden Reaktionsdiagramm dargestellt. <Desc/Clms Page number 3> EMI3.1 EMI3.2 EMI3.3 <Desc/Clms Page number 4> Weiterhin können sie gegebenenfalls mit Alkylsulfaten oder Alkylhalogeniden zu den quartären Ammoniumsalzen umgesetzt werden. Schliesslich können sie in an sich bekannter Weise, z. B. mit Wasserstoffperoxyd und Mangandioxyd zu den entsprechenden N-Oxyden umgesetzt werden. Zur Herstellung der Tabletten wird die jeweilige Verbindung mit Stärke und Lactose gemäss dem Verfahren der sukzessiven Verdünnungen vermischt. Das Gemisch wird mit Methylcellulose granuliert. Levilit, Magnesiumstearat und Talk werden dem Granulat vor der Weiterverarbeitung durch Kompression zugesetzt. Es ist möglich, die Methylcellulose durch ein beliebiges anderes geeignetes Granuliermittel zu ersetzen, z. B. durch Äthylcellulose, Polyvinylpyrrolidon oder Stärkepaste. Magnesiumstearat kann durch Stearinsäure ersetzt werden. Zur Herstellung injizierbarer Lösungen ist es möglich, die neuen Verbindungen z. B. in den folgenden Säuren zu lösen : Salzsäure oder Levulinsäure, Gluconsäure oder Glucoheptonsäure. Die Lösung wird unter sterilen Bedingungen hergestellt und mit einem Alkalimetallchlorid, z. B. Natriumchlorid, isotonisiert ; anschliessend werden Konservierungsmittel zugesetzt. Es ist auch möglich, die gleiche Lösung ohne Konservierungsmittel herzustellen : Die Ampulle wird dann unter Stickstoff gefüllt und eine halbe Stunde bei 1000C sterilisiert. Die pharmakodynamischen Tests mit den neuen Verbindungen, insbesondere eine Untersuchung ihrer antiemetischen Eigenschaften (der Antagonismus gegenüber Apomorphin bei der subcutanen Verabreichung an Hunde ist fünf- bis zwanzigmal grösser als der von bekannten Verbindungen aus der Reihe der Benzamide) lassen den Schluss zu, dass diese auf das zentrale Nervensystem stark wirken. Ihre niedrige Toxizität und die Abwesenheit von jeglichen unerwünschten Nebenerscheinungen wie Katalepsie, die normalerweise bei ähnlichen Produkten anzutreffen ist, machen diese Verbindungen besonders interessant, insbesondere für die Psychiatrie. Die neuen Pyrrolidine wurden subcutan an männliche Ratten verabreicht. Als Kriterium für den kataleptischen Zustand gilt, dass das Tier 30 s lang mit ausgestreckten Hinterpfoten bewegungslos ist, u. zw. in sorgfältiger Anordnung auf 4 cm hohen Holzklötzen, wodurch das Tier sich in einer ungewohnten und unbequemen Stellung befindet. Die kataleptische Wirkung wurde bestimmt, wenn der Effekt maximal war, d. h. 5 bis 6 h nach Verabreichung des Produktes. Es zeigte sich, dass bei einer Dosierung von 100 mg/kg die Verbindungen 2,5, 6,9, 10, 13,15, 18,19, 20,21 und 23 überhaupt keine kataleptische Wirkung hervorriefen, und dass bei einer Dosierung von 200 mg/kg die Verbindungen 1, 7,12 und 14 bei 10% der Tiere einen kataleptischen Zustand herbeiführen. Die neuen Verbindungen können in Dosierungen von 50 bis 750 mg/Tag in einer Portion oder mehreren verabreicht werden. Die Erfindung wird durch das folgende Beispiel näher erläutert : Beispiel : N-(1-Cyclopropyl-2-pyrrolidinylmethyl)-2-methoxy-5-sulfamoylbenzamid 2, 5-Dichlorpentylamin-hydrochlorid 1010 g (10 Mol) Tetrahydrofurfurylamin werden in einen 10 l-Kolben mit mechanischem Rührer, Rückflusskühler, der an eine mit Schwefelsäure gefüllte Gaswaschflasche angeschlossen ist, und einem Gaseinleitungsrohr gegeben. Es wird abfiltriert, mit Chloroform gewaschen und bei 70 C getrocknet. Man erhält 1512 g (Ausbeute 78, 5%) Produkt mit einem Schmelzpunkt von 160 C. 2-Methoxy-5-sulfamoylbenzoylchlorid 23, 1 g (0, 1 Mol) 2-Methoxy-5-sulfamoylbenzoesäure in 400 ml Dichloräthan und 1 ml Dimethylformamid werden in einen 1 1-Kolben mit mechanischem Rührer, Tropftrichter und Rückflusskühler gegeben. Man rührt schnell 11 ml (0, 15 Mol) Thionylchlorid ein und erhitzt das Gemisch zum Rückfluss, bis die Feststoffe sich vollständig gelöst haben. Nach dem Abkühlen auf 50 C wird das Reaktionsgemisch filtriert. Das erhaltene Chlorid wird in einem Vakuumexsikkator getrocknet. <Desc/Clms Page number 7> Man erhält 20, 2 g des gewünschten Produktes mit einem Schmelzpunkt von 1670C unter Zersetzung. EMI7.1 : berechnet 14, 22% ;77, 5 g (0, 4 Mol) 2, 5-Dichlorpentylamin-hydrochlorid, gelöst in 500 ml Dichloräthan, werden in einen 2 l-Kolben mit Rührer, Thermometer, Rückflusskühler und Tropftrichter gegeben ; anschlie- ssend werden 112 ml Triäthylamin hinzugesetzt. Schliesslich wird eine Lösung von 100 g (0, 4 Mol) 2-Methoxy-5-sulfamoylbenzoylchlorid in 1 l Methyläthylketon eingegossen, wobei man die Temperatur bei 20 C hält. Nach einstündiger Reaktionszeit wird der Niederschlag aus Triäthylaminhydrochlorid abfiltriert. Das Filtrat wird im Vakuum zur Trockne eingedampft. Der Niederschlag wird in 1 l Wasser gelöst. Es bleiben Kristalle zurück, die abfiltriert und zweimal mit 100 ml Isopropanol gewaschen werden. Sie werden zu dem Rückstand des eingedampften Filtrats gegeben. Der Feststoff wird aus 1600 ml Isopropanol umkristallisiert. Die Kristalle werden filtriert, mit Isopropanol gewaschen und in einem Ofen bei 40 C getrocknet. Man erhält 101 g (68, 4%) des gewünschten Produkts mit einem Schmelzpunkt von 148 C. N- (l-Cyclopropyl-2-pyrrolidinylmethyl)-2-methoxy-5-sulfamoylbenzamid 36, 9 g (0, 1 Mol) N- (2, 5-Dichlorpentyl)-2-methoxy-5-sulfamoylbenzamid und 57 g Cyclopropylamin werden in einen 250 ml-Kolben gegeben. Man erhitzt 6 h am Rückfluss, lässt über Nacht stehen und erhitzt erneut 5 h am Rückfluss. Die erhaltene Lösung wird in 300 ml Wasser und 50 g Eis gegossen. Der farblose Niederschlag wird mit Wasser gewaschen und in einem Ofen bei 60 C getrocknet. Das Produkt schmilzt bei 1630C und wiegt 29, 5 g (83, 6%). Es wird in 2 l Acetonitril gelöst und in Gegenwart von Aktivkohle filtriert. Man lässt das Filtrat zur Kristallisation stehen. Die erhaltenen Kristalle werden abfiltriert, mit Acetonitril gewaschen und bei 40 C getrocknet. Man erhält 67 g (Ausbeute 74, 9%) des gewünschten Benzamids mit einem Schmelzpunkt von 180 C. Analyse : N : berechnet 11, 89 ; gefunden 11, 74% S : berechnet 9, 07 ; gefunden 9, 19%. **WARNUNG** Ende DESC Feld kannt Anfang CLMS uberlappen**.
Claims (1)
- PATENTANSPRÜCHE : 1. Verfahren zur Herstellung neuer substituierter Pyrrolidine der allgemeinen Formel EMI7.2 worin R Cycloalkyl, Cycloalkenyl, Bicycloalkyl oder Tricycloalkyl, A eine Einfachbindung oder eine gesättigte oder ungesättigte Kohlenwasserstoffkette mit 1 bis 3 Kohlenstoffatomen, n 0, 1, 2 oder 3, Rs Wasserstoff, Alkyl mit 1 bis 3 Kohlenstoffatomen, Alkenyl oder Alkinyl und RI, R,., R, sowie R6 Wasserstoff, Halogen, Alkyl, Alkoxy, Amino, Acetamido, Sulfamoyl, Alkyl- sulfamoyl, Dialkylsulfamoyl, Alkylsulfonyl oder Alkylsulfinyl bedeuten oder untereinander unter Ausbildung einer Azimidogruppe verknüpft sind, mit den folgenden Massgaben :<Desc/Clms Page number 8> - wenn R, für Cycloalkyl, Rs für Wasserstoff oder Alkyl, A für eine Einfachbindung und n für 0 stehen, stellt mindestens einer der Substituenten R R , R und Ru eine Alkyl- sulfonyl- oder Alkylsulfinylgruppe dar oder bilden zwei dieser Substituenten eine Azimido- gruppe, - wenn R, für Cycloalkyl, R5 für Methyl, A für Alkylen mit 1 bis 3 Kohlenstoffatomen und n für 1 stehen und die Amidkette an Position 2 des Pyrrolidins gebunden ist, stellt R, EMI8.1 sowie deren Additionssalzen mit pharmakologisch verträglichen Säuren, dadurch gekennzeichnet, dass eine substituierte Benzoesäure der allgemeinen Formel EMI8.2 worin R1, R2, R3, R5 und Re wie oben definiert sind, oder ein reaktives Derivat derselben,mit einem primären Amin der allgemeinen Formel EMI8.3 in der n wie oben definiert ist und Hal ein Chlor-, Brom- oder Jodatom bedeutet, und die so erhaltene Verbindung der allgemeinen Formel EMI8.4 in der R1, R2, R3, R5, R6, Hal und n wie oben definiert sind, mit einem Amin der allgemeinen Formel H2N-A-R4, (V) EMI8.5 Alkylsulfamoyl bedeuten, oder ein reaktives Derivat derselben, mit einem primären Amin der allgemeinen Formel (III), worin Hal und n wie in Anspruch 1 definiert sind, die so erhaltene Verbindung der Formel (IV), in der R1, R2, R3, R5, R6, Hal und n wie oben definiert sind, mit einem Amin der allgemeinen Formel (V), in der A wie in Anspruch 1 definiert ist und R, Cycloalkyl darstellt, unter Beachtung der folgenden Massgaben umgesetzt wird :<Desc/Clms Page number 9> EMI9.1 und R, für Alkylsulfonyl oder Alkylsulfinyl, - wenn gleichzeitig A für Alkylen mit 1 bis 3 Kohlenstoffatomen, n für 1 stehen und die Amidkette an Position 2 des Pyrrolidins gebunden ist, wenn R 1 Sulfamoyl, Alkyl- sulfamoyl oder Alkylsulfonyl darstellt, bedeuten R2 und R] nicht gleichzeitig Wasser- stoff.3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass eine substituierte Benzoesäure der allgemeinen Formel (II) und ein primäres Amin der allgemeinen Formel (III) eingesetzt werden, worin R1, R2, R3, R5, R6, Hal und n wie in Anspruch 1 definiert sind und die erhaltene Verbindung der Formel (IV) mit einem Amin der Formel (V), in der A und Ru wie in Anspruch 1 definiert sind, umgesetzt wird, mit den folgenden Massgaben :- wenn Ru für Cycloalkyl, Rs für Wasserstoff oder Alkyl, A für eine Einfachbindung und n für 0 stehen, stellt mindestens einer der Substituenten R R z, R 3 und Re Alkyl- sulfonyl oder Alkylsulfinyl dar oder bilden zwei dieser Substituenten eine Azimidogrup- pe und R e kann nur für den Fall, dass wenigstens einer der Substituenten Ri, R 2 und R3 Halogen, Dialkylsulfamoyl oder Alkyl bedeutet oder dass zwei dieser Substituenten miteinander unter Ausbildung einer Azimidogruppe verknüpft sind, Wasserstoff sein, - wenn R., für Cycloalkyl, Rs für Methyl, A für Alkylen mit 1 bis 3 Kohlenstoffatomen und n für 1 stehen und die Amidkette an Position 2 des Pyrrolidins gebunden ist, kann R e nur dann für Wasserstoff stehen,wenn wenigstens einer der Substituenten Ri, R2 und R3 Alkyl bedeutet oder wenn zwei dieser Substituenten miteinander unter Ausbil- dung einer Azimidogruppe verknüpft sind oder wenn wenigstens einer der Substituenten R2 und Ra Halogen oder Dialkylsulfamoyl darstellt.
