Description
Die Erfindung betrifft substituierte heterocyclische Benzamide
gemäß Anspruch 1, Verfahren zu ihrer Herstellung sowie diese
enthaltende Arzneimittel.
Die Erfindung betrifft weiterhin Additionssalze der vorgenannten
Benzamide mit pharmakologisch verträglichen Säuren.
Die Verbindungen der Erfindung können hergestellt werden,
indem man jeweils in an sich bekannter Weise eine entsprechend
substituierte Benzoesäure, ausgewählt aus der von 2,3-Dimethoxy-
5-sulfamoyl-, 2-Methoxy-4-amino-5-ethylsulfonyl- und
2-Methoxy-5-methylsufonylbenzoesäure gebildeten Gruppe, oder
ein reaktives Derivat derselben mit einem entsprechenden
heterocyclischen Amin, ausgewählt aus der von
1-Cyclopropylmethyl-2-aminomethylpyrrolidin,
1-(Cyclohexenyl)-methyl-2-aminomethylpyrrolidin,
1-Cyclohexylmethyl-2-aminomethylpyrrolidin,
1-Cycloheptyl-2-aminomethylpyrrolidin und
1-Cyclohexylmethyl-3-aminopyrrolidin
1-(Cyclohexenyl)-methyl-2-aminomethylpyrrolidin,
1-Cyclohexylmethyl-2-aminomethylpyrrolidin,
1-Cycloheptyl-2-aminomethylpyrrolidin und
1-Cyclohexylmethyl-3-aminopyrrolidin
gebildeten Gruppe, oder ein reaktives Derivat derselben in
Gegenwart oder Abwesenheit eines Kondensationsmittels oder an
einer Ionenaustauscherkolonne umsetzt.
Reaktive Derivate der vorgenannten Benzoesäuren sind Säurehalogenide,
Alkylester, reaktive Ester wie Methoxymethylester
oder Cyanmethylester, aromatische Ester, N-Hydroxyimidester,
symmetrische Anhydride oder gemischte Anhydride, beispielsweise
gebildet aus einem Kohlensäureester oder einem Halogenameisensäureester,
Säureazide, Hydrazide, Azolide, Säureisothiocyanate,
Trichloracetophenon- oder Triphenylphosphin-Derivate.
Reaktive Derivate der vorgenannten heterocyclischen Amine sind
z. B. die durch Umsetzung der Amine mit Phosphortrichlorid oder
Phosphorpentachlorid, Phosphoroxychlorid, einem Dialkyl-,
Diaryl- oder Orthophenylenchlorphosphit, einem Alkyl- oder
Aryldichlorphosphit, einem Isothiocyanat des Amins, einem
Sulfamid oder einem substituierten Harnstoff erhaltenen.
Die Amidierungsreaktion kann in situ oder nach Isolierung
eines Zwischenprodukts durchgeführt werden.
Es ist auch möglich, die freie Säure und das freie Amin in
Gegenwart eines Kondensierungsmittels, z. B. Siliciumtetrachlorid,
Trichlorphenylsilan, Phosphorsäureanhydrid, einem
Carbodiimid oder einem Alkoxyacetylen umzusetzen.
Die Amidierungsreaktionen können mit oder ohne Lösungmittel
durchgeführt werden.
Einige Beispiele für erfindungsgemäß einsetzbaren Lösungsmittelsysteme,
die gegenüber der Amidierungsreaktion inert sind,
sind Alkohole, Polyole, Ketone, Benzol, Toluol, Dioxan, Chloroform
und Diethylenglykoldimethylether. Es ist auch möglich,
einen Überschuß des als Ausgangsprodukt eingesetzten Amins als
Lösungsmittel zu verwenden. Es kann vorteilhaft sein, das
Reaktionsgemisch während der Amidierung zu erhitzen, z. B. bis
zum Siedepunkt der obengenannten Lösungsmittel.
Die erfindungsgemäß erhaltenen Verbindungen können gegebenenfalls
mit pharmazeutisch verträglichen anorganischen oder organischen
Säuren zu den Säureadditionssalzen umgesetzt werden,
z. B. mit Salzsäure, Bromwasserstoff, Schwefelsäure, Phosphorsäure,
Oxalsäure, Essigsäure, Weinsäure, Citronensäure oder
Methansulfonsäure.
Die Erfindung wird im folgenden anhand von bevorzugten Ausführungsbeispielen
näher erläutert.
419 g (1,6 Mol) 2,3-Dimethoxy-5-sulfamoylbenzoesäure und
1351 g (11,35 Mol) Thionylchlorid werden in eine 2-l-Kolben
mit Rührer, Thermometer und Rückflußkühler gegeben, wobei der
Rückflußkühler an eine mit Soda gefüllte Gaswaschflasche angeschlossen
ist. Man erhitzt das Gemisch eine Stunde am Rückfluß,
wonach man das überschüssige Thionylchlorid im Vakuum
verjagt. Der Rückstand wird in 1000 ml Hexan gelöst, filtriert,
zweimal mit 500 ml Petrolether gewaschen und schließlich in
einem Exsikkator unter Vakuum getrocknet. Man erhält 124 g
(Ausbeute 94,8%) des gewünschten Produktes mit einem Schmelzpunkt
von 153°C.
20 g (0,13 Mol) 1-Cycloproplmethyl-2-aminomethylpyrrolidin
und 150 ml Methylethylketon werden in einen 500-ml-Kolben
mit Rührer, Thermometer und Rückflußkühler gegeben. 36,3 g
(0,13 Mol) 2,3-Dimethoxy-5-sulfamoylbenzoylchlorid werden
ebenfalls zugegeben, und zwar portionsweise, wobei man die
Temperatur zwischen 15 und 20°C hält. Die gebildete dicke
Paste wird 170 ml Wasser verdünnt; man läßt sie anschließend
eine Stunde bei Raumtemperatur reagieren. Das Gemisch
wird anschließend zur Trockene eingedampft. Der Rückstand
wird in 200 ml Wasser gelöst und mit überschüssigem
Ammoniak alkalisch gemacht. Die Base fällt aus und kristallisiert
langsam. Die Kristalle werden abfiltriert, mit Wasser
gewaschen und in einem Ofen bei 50°C getrocknet. Man erhält
50 g (Ausbeute 97%) des gewünschten Produkts. Dieses wird
dreimal aus Butylacetat umkristallisiert, wobei man 26 g
(50,5%) Kristalle erhält. Die Kristalle werden in normaler
Salzsäurelösung gelöst, filtriert, die Lösung wird mit normaler
Sodalösung alkalisch gemacht und erneut filtriert. Die Kristalle
werden mit Wasser bis zur vollständigen Chloridfreiheit gewaschen
und in einem Ofen (50°C) getrocknet. Man erhält 24 g
(46,6%) Kristalle mit einem Schmelzpunkt von 136°C (sie sind
in Wasser unlöslich).

159 g 2-Methoxy-4-amino-5-mercaptobenzoesäure, 355 ml Wasser
und 160 ml Natronlauge werden in einem Kolben mit Rückflußkühler
vorgelegt. Das Gemisch wird bis zur Auflösung der Feststoffe
erwärmt. Anschließend werden 123 g Ethylsulfat zugegeben.
Das Gemisch wird am Rückfluß gekocht, mit 10 ml
30%iger Natronlauge behandelt und schließlich eine Stunde
am Rückfluß erhitzt. Es wird abgekühlt, man setzt 800 ml
Wasser hinzu und filtriert die Lösung. Der Niederschlag,
der sich bei der Zugabe von 100 ml konzentrierter Salzsäure
in Gegenwart von Ether bildet, wird abgesaugt, mit Wasser gewaschen
und getrocknet.
Man erhält 162 g des gewünschten Produkts mit einer Ausbeute
von 88%.
123 g 2-Methoxy-4-amino-5-ethylthiobenzoesäure werden in der
Hitze in 542 ml Essigsäure gelöst. Die erhaltene Lösung wird
auf 35°C abgekühlt. Anschließend setzt man 185 ml Wasserstoffperoxid
(131 Vol.) in kleinen Anteilen hinzu, wobei man die
Temperatur auf 80°C steigert.
Man setzt die Temperatur auf 40°C herab und hält das Reaktionsgemisch
einige Stunden bei dieser Temperatur. Schließlich
kühlt man auf 10°C.
Der gebildete Niederschlag wird abgesaugt, mit Essigsäure gewaschen
und getrocknet, anschließend in 600 ml Wasser und
100 ml 20%igem Ammoniak gelöst.
Der sich durch Zugabe von 70 ml konzentrierter Salzsäure bildende
Niederschlag wird abgekühlt, abgesaugt, mit Wasser gewaschen
und getrocknet.
Man erhält 61,5 g des wasserhaltigen gewünschten Produkts in
einer Ausbeute von 42% und mit einem Schmelzpunkt von 95-
100°C.
31,3 g (0,31 Mol) Triethylamin, 400 ml Tetrahydrofuran und
80,3 g (0,31 Mol) 2-Methoxy-4-amino-5-ethylsulfonylbenzoesäure
werden in einen 1-l-Kolben mit Rührer, Thermometer,
Rückflußkühler und Tropftrichter gegeben. Es bildet sich ein
gummiartiger Niederschlag, der allmählich zerbröckelt. Nach
30 Minuten bei Raumtemperatur kühlt man auf 0°C und setzt
tropfenweise 33,6 g (0,31 Mol) Chlorameisensäureethylester
hinzu.
Das Gemisch wird eine Stunde unter Rühren bei 0-5°C gehalten.
Anschließend fügt man tropfenweise 62 g (0,40 Mol) 1-Cyclopropylmethyl)-
2-aminomethylpyrrolidin hinzu, wobei man die
Temperatur bei der gleichen Höhe hält. Es bildet sich ein
dicker Niederschlag. Das Reaktionsgemisch wird weitere zwei
Stunden bei Raumtemperatur gerührt; anschließend läßt man
über Nacht stehen. Die erhaltenen Kristalle werden abfiltriert,
zweimal mit 100 ml Tetrahydrofuran gewaschen und in einem Ofen
bei 50°C getrocknet. Man erhält 137 g Produkt, das man in
siedendem Wasser löst. Nach dem Abfiltrieren und Trocknen
erhält man 91 g (74,3%) Kristalle; diese werden aus 600 ml
90%igem Alkohol umkristallisiert. Sie werden abfiltriert,
zweimal mit 50 ml Alkohol gewaschen und in einem Ofen bei
40°C getrocknet. Man erhält 81,5 g (Ausbeute 66,5%) des gewünschten
Produkts mit einem Schmelzpunkt von 181°C.

13 g 2,3-Dimethoxy-5-sulfamoylbenzoesäure (0,05 Mol), 150 ml
Aceton, 35 ml Wasser und 5 g Triethylamin (0,05 Mol) werden in
einen 500-ml-Kolben mit Rührer, Thermometer, Rückflußkühler und
Tropftrichter gegeben. Das Gemisch wird auf 0 bis +5°C abgekühlt.
Anschließend werden tropfenweise 5,5 g Chlorameisensäureethylester
(0,05 Mol) zugegeben. Das Reaktionsmedium wird gerührt,
bis sich der Niederschlag vollständig gelöst hat. Anschließend
wird erneut auf 0°C gekühlt und tropfenweise mit 9,7 g 1-(Cyclohexenyl)-
methyl-2-aminomethylpyrrolidin (0,05 Mol) versetzt.
Nach fünfstündiger Reaktion bei Raumtemperatur läßt man das
Reaktionsgemisch stehen. Die Lösungsmittel werden im Vakuum eingedampft.
Der Rückstand wird in 150 ml Salzsäure (Dichte 1,18)
gelöst. Man dekantiert von einem unlöslichen Öl ab und macht die
wäßrige Lösung mit 13 ml Ammoniak (Dichte 0,91) alkalisch. Der
gebildete Niederschlag wird abfiltriert, mit Wasser gewaschen
und aus 120 ml Isopropanol umkristallisiert. Man erhält 6,1 g
Produkt, das man aus 250 ml Isopropanol umkristallisiert. Die
Ausbeute beträgt 4,5 g (21%), der Schmelzpunkt 169°C.
25,9 g 2-Methoxy-4-amino-5-ethylsulfonylbenzoesäure, 40 ml
Wasser, 200 ml Aceton und 13,9 ml Triethylamin (Dichte 0,726)
werden in einen 500-ml-Kolben mit Rührer, Thermometer und
Tropftrichter gegeben. Die Lösung wird auf etwa 0 bis 5°C
abgekühlt und tropfenweise mit 10,9 g Chlorameisensäureethylester
versetzt. Das Gemisch wird 40 Minuten bei etwa 0°C gerührt;
anschließend werden 19,6 g 1-Cyclohexylmethyl-2-aminomethylpyrrolidin
tropfenweise zugegeben. Das Gemisch wird zwei
Stunden bei Raumtemperatur gerührt und anschließend stehengelassen.
Das Aceton wird im Vakuum eingedampft. Der Rückstand
wird in 100 ml Wasser und 25 ml Essigsäure gelöst und in Gegenwart
von Aktivkohle filtriert. Das Filtrat wird mit 100 ml
40%iger Natronlauge alkalisch gemacht. Es bildet sich ein
Niederschlag, der abfiltriert, mit viel Wasser gewaschen und
feucht in 230 ml siedendem Aceton gelöst wird. Die heiße Lösung
wird in Gegenwart von Aktivkohle filtriert; man läßt das Filtrat
auskristallisieren. Das Produkt wird abgesaugt, mit wenig
Aceton gewaschen und in einem Ofen bei 50°C getrocknet.
Man erhält 25 g (57%) des Produkts mit einem Schmelzpunkt
von 155°C.
52 g 1-Cycloheptyl-2-aminomethylpyrrolidin und 240 ml Wasser
werden in einen 500-ml-Kolben mit Rührer, Rückflußkühler und
Thermometer gegeben. Man kühlt auf 0° und setzt portionsweise
62,1 g 2-Methoxy-5-methylsulfonylbenzoylchlorid (0,250 Mol)
hinzu, wobei man die Temperatur zwischen 0 und +5°C hält. Die
Temperatur wird gesteigert; bei etwa 20°C setzt eine exotherme
Reaktion ein und die Temperatur steigt auf 30°C. Nach einstündigem
Rühren hat sich alles gelöst. Die Reaktion wird weitere
30 Minuten fortgesetzt; anschließend wird die Lösung in Gegenwart
von Aktivkohle filtriert. Das Filtrat wird mit 20 ml
Natronlauge, verdünnt mit 80 ml Wasser, alkalisch gemacht. Der
Niederschlag ist anfänglich gummiartig und kristallisiert
später. Das Produkt wird abgesaugt, mit Wasser gewaschen und
in einem Ofen bei 50°C getrocknet.
Man erhält 90,5 g Produkt mit einem Schmelzpunkt von 112-114°C.
Die Base wird erneut zweimal umkristallisiert, und zwar aus
225 ml bzw. 200 ml Methanol mit einem Gehalt von 30% Wasser.
Die farblosen Kristalle werden abgesaugt und in einem Ofen bei
40°C getrocknet.
Man erhält 51 g Produkt mit einem Schmelzpunkt von 118°C und
in einer Ausbeute von 53%.
36,4 g 1-Cyclohexylmethyl-3-aminopyrrolidin und 200 ml Wasser
werden in einen 500-ml-Kolben mit Rührer und Thermometer gegeben.
Die Lösung wird auf +5°C abgekühlt; anschließend werden
47,4 g 2-Methoxy-5-methylsulfonylbenzoylchlorid portionsweise
zugegeben. Die Suspension wird erneut zwei Stunden lang
bei 20°C gerührt, anschließend eine Stunde bei 30°C. Das
Reaktionsgemisch wird stehengelassen und anschließend mit 30 ml
Salzsäure (Dichte 1,18) stark angesäuert. Eine geringe unlösliche
Substanz wird abfiltriert. Die Base wird durch 60 ml
Natronlauge ausgefällt. Die Base ist anfänglich ölig und
kristallisiert später. Die farblosen Kristalle werden abfiltriert,
mit Wasser gewaschen und in einem Ofen bei 50°C
getrocknet. Man erhält 57,2 g Produkt mit einem Schmelzpunkt
von 152°C.
Das Produkt wird aus 600 ml Methanol umkristallisiert, wobei
heiß filtriert wird. Man läßt es über Nacht in einem Kühlschrank
auskristallisieren. Die Kristalle werden abfiltriert,
mit wenig Methanol gewaschen und in einem Ofen bei 60°C
getrocknet.
Man erhält 51 g des Benzamids mit einem Schmelzpunkt von
157°C in einer Ausbeute von 68%.

Zur Herstellung der Tabletten wird die jeweilige Verbindung
mit Stärke und Lactose gemäß dem Verfahren der sukzessiven
Verdünnungen vermischt. Das Gemisch wird mit Methylcellulose
granuliert. Levilit, Magnesiumstearat und Talk werden dem
Granulat vor der Weiterverarbeitung durch Kompression zugesetzt.
Es ist möglich, die Methylcellulose durch ein beliebiges
anderes geeignetes Granulierungsmittel zu ersetzen, z. B.
durch Ethylcellulose, Polyvinylpyrrolidon oder Stärkepaste.
Magnesiumstearat kann durch Stearinsäure ersetzt werden.
Zur Herstellung injizierbarer Lösungen ist es möglich, die
Verbindungen der Erfindung z. B. in den folgenden Säuren zu
lösen: Salzsäure oder Levulinsäure, Gluconsäure oder Glucoheptonsäure.
Die Lösung wird unter sterilen Bedingungen hergestellt und mit
einem Alkalimetallchlorid, z. B. Natriumchlorid, isotonisiert;
auch möglich, die gleiche Lösung ohne Konservierungsmittel
herzustellen: Die Ampulle wird dann unter Stickstoff gefüllt
und eine halbe Stunde bei 100°C sterilisiert.
Die pharmacodynamischen Tests mit den Verbindungen der Erfindung,
insbesondere eine Untersuchung ihrer antiemetischen
Eigenschaften (der Antagonismus gegenüber Apomorphin bei der
subcutanen Verabreichung an Hunde ist 5- bis 20mal größer als
der von bekannten Verbindungen aus der Reihe der Benzamide)
lassen den Schluß zu, daß diese auf das zentrale Nervensystem
stark wirken.
Ihre niedrige Toxizität und die Abwesenheit von jeglichen unerwünschten
Nebenerscheinungen wie Katalepsie, die normalerweise
bei ähnlichen Produkten anzutreffen ist, machen diese
Verbindungen besonders interessant, insbesondere für die
Psychiatrie.
Die akute Toxizität der Verbindungen der Erfindung wurde an
der Maus untersucht. Die DL₅₀-Werte sind in der folgenden
Tabelle zusammengefaßt. Die Tabelle enthält weiterhin Ergebnisse
von Versuchen zur Ermittlung der antiemetischen Wirkung
der Verbindungen der Erfindung an Hunden gemäß der Methode von
Chen und Ensor. Hierzu wurden die Verbindungen subkutan 30 min
vor dem Apomorphin verabreicht; letzteres wurde subkutan in
einer Dosierung von 100 µg/kg verabreicht.
Die Verbindungen der Erfindung zeigen weiterhin keine
kataleptische Wirkung.
Die Benzamide wurden subcutan an männliche Ratten verabreicht.
Als Kriterium für den kataleptischen Zustand gilt,
daß das Tier 30 Sekunden lang mit ausgestreckten Hinterpfoten
bewegungslos ist, und zwar in sorgfältiger Anordnung
auf vier cm hohen Holzklötzen, wodurch das Tier sich
in einer ungewohnten und unbequemen Stellung befindet. Die
kataleptische Wirkung wurde bestimmt, wenn der Effekt
maximal war, d. h. 5 bis 6 Stunden nach Verabreichung des
Produktes.
Es zeigte sich, daß bei einer Dosierung von 100 mg/kg die
Verbindungen der Erfindung überhaupt keine kataleptische
Wirkung hervorriefen, und daß bei einer Dosierung von 200 mg/kg
die Verbindngen, wenn überhaupt, nur schwach kataleptisch
wirken, so daß bei therapeutischen Dosierungen keine
kataleptischen Nebenwirkungen zu erwarten sind.
Claims (3)
1. Heterocyclische Benzamide, ausgewählt aus der von
N-(1-Cyclopropylmethyl-2-pyrrolidinylmethyl)-2,3-dimethoxy-
5-sulfamoylbenzamid,
N-(1-Cyclopropylmethyl-2-pyrrolidinylmethyl)-2-methoxy- 4-amino-5-ethylsulfonylbenzamid,
N-[(1-Cyclohexenyl)-methyl-2-pyrrolidinylmethyl]-2,3-dimethoxy- 5-sulfamoyl-benzamid,
N-(1-Cyclohexylmethyl-2-pyrrolidinylmethyl)-2-methoxy- 4-amino-5-ethylsulfonylbenzamid,
N-(1-Cycloheptyl-2-pyrrolidinylmethyl)-2-methoxy-5-methylsulfonylben-zamid und
N-(1-Cyclohexylmethyl-3-pyrrolidinyl)-2-methoxy-5-methylsulfonylbenz-amid
gebildeten Gruppe,
sowie deren Additionssalze mit pharmakologisch verträglichen Säuren.
N-(1-Cyclopropylmethyl-2-pyrrolidinylmethyl)-2-methoxy- 4-amino-5-ethylsulfonylbenzamid,
N-[(1-Cyclohexenyl)-methyl-2-pyrrolidinylmethyl]-2,3-dimethoxy- 5-sulfamoyl-benzamid,
N-(1-Cyclohexylmethyl-2-pyrrolidinylmethyl)-2-methoxy- 4-amino-5-ethylsulfonylbenzamid,
N-(1-Cycloheptyl-2-pyrrolidinylmethyl)-2-methoxy-5-methylsulfonylben-zamid und
N-(1-Cyclohexylmethyl-3-pyrrolidinyl)-2-methoxy-5-methylsulfonylbenz-amid
gebildeten Gruppe,
sowie deren Additionssalze mit pharmakologisch verträglichen Säuren.
2. Verfahren zur Herstellung der heterocyclischen Benzamide
nach Anspruch 1, dadurch gekennzeichnet, daß man jeweils
in an sich bekannter Weise eine entsprechend substituierte
Benzoesäure, ausgewählt aus der von 2,3-Dimethoxy-5-sulfamoyl-,
2-Methoxy-4-amino-5-ethylsulfonyl- und 2-Methoxy-5-
methylsulfonylbenzoesäure gebildeten Gruppe, oder ein
reaktives Derivat derselben mit einem entsprechenden
heterocyclischen Amin, ausgewählt aus der von
1-Cyclopropylmethyl-2-aminomethylpyrrolidin,
1-(Cyclohexenyl)-methyl-2-aminomethylpyrrolidin,
1-Cyclohexylmethyl-2-aminomethylpyrrolidin,
1-Cycloheptyl-2-aminomethylpyrrolidin und
1-Cyclohexylmethyl-3-aminopyrrolidingebildeten Gruppe, oder ein reaktives Derivat derselben in Gegenwart oder Abwesenheit eines Kondensationsmittels oder an einer Ionenaustauscherkolonne umsetzt.
1-(Cyclohexenyl)-methyl-2-aminomethylpyrrolidin,
1-Cyclohexylmethyl-2-aminomethylpyrrolidin,
1-Cycloheptyl-2-aminomethylpyrrolidin und
1-Cyclohexylmethyl-3-aminopyrrolidingebildeten Gruppe, oder ein reaktives Derivat derselben in Gegenwart oder Abwesenheit eines Kondensationsmittels oder an einer Ionenaustauscherkolonne umsetzt.
3. Arzneimittel, enthaltend ein heterocyclisches Benzamid
nach Anspruch 1 sowie übliche pharmazeutische Träger
und/oder Verdünnungsmittel.
