Description

磁性シリカ粒子を用いた測定方法及び該測定方法用試薬
本発明は、磁性シリカ粒子を用いた測定対象物質測定方法及び該方法に用いられる試薬に関する。更に詳しくは、診断の補助、治療効果確認の補助、タンパク質精製時及び細胞分離時の精製度の確認等に用いる測定方法及び試薬に関する。
従来、生体物質を含有する試料、例えば生体サンプル中のタンパク質等を測定又は精製する方法としてタンパク質が結合し得る粒子表面に生体サンプル中のタンパク質等を結合させ、生体サンプル中の目的タンパク質等以外の不純物を除くために、粒子を洗浄しタンパク質等が結合した粒子を回収して、タンパク質の結合量を測定する方法やタンパク質解離溶液中に解離させて精製する方法が知られている。
また、磁力によって容易に分離、回収が可能であることから、磁性を有する粒子が用いられており、例えば特許文献1には、酸化鉄からなる芯粒子の表面にシリカの被膜が形成されてなる磁性シリカ粒子が記載されている。しかし、この磁性粒子は、磁性体が強磁性であり、回収時の磁場を取り除いても強磁性により磁性体自身が一時的な磁場を示し粒子同士が自己会合し、洗浄性が悪い及び/又は次の操作(例えば、免疫反応)に悪影響を及ぼすという問題がある。
更に、強磁性による磁性体自身の自己会合を解決する目的で、例えば、特許文献2には、磁性体に超常磁性である磁性体を用いた磁性シリカ粒子が開示されている。しかし、この磁性粒子は、粒子径が小さい場合は、磁性体の含有量が低く、磁力で粒子を回収する際に時間がかかり、粒子径が大きい場合は、比表面積が小さいために、結合するタンパク質等の量が少ないという問題がある。

本発明は、測定対象物質、測定対象物質の類似物質、又は、測定対象物質と特異的に結合する物質を担持させる際の結合性に優れ、且つ、磁力による回収性及び磁力を除去した状態での分散性に優れ、短時間の洗浄工程でも免疫測定において優れた感度を出し得る磁性シリカ粒子を用いた測定対象物質測定方法、及び、該測定法用試薬を提供することを目的とする。
本発明者らは、上記目的を達成すべく鋭意検討した結果、本発明に到達した。即ち本発明は、平均粒子径が1~15nmの超常磁性金属酸化物を60~95重量%含有するシリカ粒子の表面に、測定対象物質、測定対象物質の類似物質、又は、測定対象物質と特異的に結合する物質が固定化された磁性シリカ粒子を用いて試料中の測定対象物質を測定することを特徴とする測定対象物質測定方法、及び、平均粒子径が1~15nmの超常磁性金属酸化物を60~95重量%含有するシリカ粒子の表面に、測定対象物質、測定対象物質の類似物質、又は、測定対象物質と特異的に結合する物質が固定化された磁性シリカ粒子を含有することを特徴とする上記測定方法用の試薬である。
本発明の測定方法及び試薬においては、磁性シリカ粒子を磁気を用いて回収することにより簡便なB/F分離を可能とする。また、磁性シリカ粒子が超常磁性金属酸化物を含むものであるため、その磁気特性により集磁後の粒子の再分散性に優れ、その結果、短時間に高感度で測定対象物質を測定(検出)することができる。
本発明における測定対象物質としては、通常この分野で測定されるものであれば特に限定はされず、例えば血清,血液,血漿,尿等の生体体液、リンパ液、血球、各種細胞類等の生体由来の試料中に含まれるタンパク質、脂質タンパク質、核酸、免疫グロブリン、血液凝固関連因子、抗体、酵素、ホルモン、癌マーカー、心疾患マーカー及び各種薬物等が代表的なものとして挙げられる。更に具体的には、例えばアルブミン,ヘモグロビン,ミオグロビン,トランスフェリン,プロテインA,C反応性蛋白質(CRP)等のタンパク質、例えば高比重リポ蛋白質(HDL),低比重リポ蛋白質(LDL),超低比重リポ蛋白質等の脂質蛋白質、例えばデオキシリボ核酸(DNA),リボ核酸(RNA)等の核酸、例えばアルカリ性ホスファターゼ,アミラーゼ,酸性ホスファターゼ,γ-グルタミルトランスフェラーゼ(γ-GTP),リパーゼ,クレアチンキナーゼ(CK),乳酸脱水素酵素(LDH),グルタミン酸オキザロ酢酸トランスアミナーゼ(GOT),グルタミン酸ピルビン酸トランスアミナーゼ(GPT),レニン,プロテインキナーゼ(PK),チロシンキナーゼ等の酵素、例えばIgG,IgM,IgA,IgD,IgE等の免疫グロブリン(或いはこれらの、例えばFc部,Fab部,F(ab)2部等の断片)、例えばフィブリノーゲン,フィブリン分解産物(FDP),プロトロンビン,トロンビン等の血液凝固関連因子、例えば抗ストレプトリジンO抗体,抗ヒトB型肝炎ウイルス表面抗原抗体(HBs抗原),抗ヒトC型肝炎ウイルス抗体,抗リュウマチ因子等の抗体、例えば甲状腺刺激ホルモン(TSH),甲状腺ホルモン(FT3,FT4,T3,T4),副甲状腺ホルモン(PTH),ヒト絨毛性ゴナドトロピン(hCG)エストラジオール(E2)等のホルモン、例えばα-フェトプロテイン(AFP),癌胎児性抗原(CEA),CA19-9,前立腺特異抗原(PSA)等の癌マーカー、例えばトロポニンT(TnT),ヒト脳性ナトリウム利尿ペプチド前駆体N端フラグメント(NT-proBNP)等の心疾患マーカー、例えば抗てんかん薬,抗生物質,テオフィリン等の薬物等が挙げられる。上記したものの中でも、抗体、ホルモン、癌マーカー、心疾患マーカー等が好ましい。
本発明における測定対象物質の類似物質(アナログ)は、測定対象物質と特異的に結合する物質(測定対象物質結合物質)が有する測定対象物質との結合部位と結合し得るもの、言い換えれば、測定対象物質が有する測定対象物質結合物質との結合部位を有するもの、更に言い換えれば、測定対象物質と測定対象物質結合物質との反応時に共存させると該反応と競合し得るものであれば何れでもよい。
本発明における測定対象物質と特異的に結合する物質(測定対象物質結合物質)としては、例えば「抗原」-「抗体」間反応、「糖鎖」-「タンパク質」間反応、「糖鎖」-「レクチン」間反応、「酵素」-「インヒビター」間反応、「タンパク質」-「ペプチド鎖」間反応、「染色体又はヌクレオチド鎖」-「ヌクレオチド鎖」間反応、又は、「ヌクレオチド鎖」-「タンパク質」間反応等の相互反応によって測定対象物質又はその類似物質と結合するもの等が挙げられ、上記各組合せに於いて何れか一方が測定対象物質又はその類似物質である場合、他の一方がこの測定対象物質結合物質である。例えば、測定対象物質又はその類似物質が「抗原」であるときは測定対象物質結合物質は「抗体」であり、測定対象物質又はその類似物質が「抗体」であるときは測定対象物質結合物質は「抗原」である(以下、その他の上記各組合せにおいても同様である)。

上記の如き測定対象物質結合物質としては、「抗原」-「抗体」間反応或いは「糖鎖-タンパク質」間反応によって測定対象物質又はその類似物質と結合するものが好ましい。具体的には、測定対象物質又はその類似物質に対する抗体、測定対象物質又はその類似物質が結合する抗原、或いは、測定対象物質又はその類似物質に結合するタンパク質が好ましく、測定対象物質又はその類似物質に対する抗体、或いは測定対象物質又はその類似物質に結合するタンパク質が更に好ましい。
本発明における磁性シリカ粒子は、シリカのマトリックス中に平均粒子径が1~15nmで超常磁性を有する金属酸化物を分散させたものである。
本発明における超常磁性金属酸化物の平均粒子径は、任意の200個の超常磁性金属酸化物について走査型電子顕微鏡で観察して測定された粒子径の平均値である。
超常磁性金属酸化物の平均粒子径は、後述の超常磁性金属酸化物作製時の金属イオン濃度を調節することにより制御することができる。また、通常の分級等の方法によっても超常磁性金属酸化物の平均粒子径を所望の値にすることができる。
超常磁性とは、外部磁場の存在下で物質の個々の原子磁気モーメントが整列し誘発された一時的な磁場を示し、外部磁場を取り除くと、部分的な整列が損なわれ磁場を示さなくなることをいう。
平均粒子径が1~15nmで超常磁性を示す超常磁性金属酸化物としては、鉄、コバルト、ニッケル及びこれらの合金等の酸化物が挙げられるが、磁界に対する感応性が優れていることから、酸化鉄が特に好ましい。超常磁性金属酸化物は、1種を単独で用いても2種以上を併用してもよい。
平均粒子径が1nm未満の場合は合成が困難であり、平均粒子径が15nmを超える場合は得られた磁性シリカ粒子の磁気特性が強磁性となり、実際の用途面において磁場を取り除いても磁性体自身が一時的な磁場を示し粒子同士が自己会合し、洗浄性が悪い及び/又は免疫反応等に悪影響を及ぼすという問題がある。
酸化鉄としては、公知の種々の酸化鉄を用いることができる。酸化鉄の内、特に化学的な安定性に優れることから、マグネタイト、γ-ヘマタイト、マグネタイト-α-ヘマタイト中間酸化鉄及びγ-ヘマタイト-α-ヘマタイト中間酸化鉄が好ましく、大きな飽和磁化を有し、外部磁場に対する感応性が優れていることから、マグネタイトが更に好ましい。
磁性シリカ粒子中の超常磁性金属酸化物の含有量の下限は、通常60重量%、好ましくは65重量%であり、上限は通常95重量%、好ましくは80重量%である。超常磁性金属酸化物の含有量が60重量%未満の場合、得られた磁性シリカ粒子の磁性が十分でないため、実際の用途面における分離操作に時間がかかり、95重量%を超えるものは合成が困難である。
超常磁性金属酸化物の製造方法は、特に限定されないが、Massartにより報告されたものをベースとして水溶性鉄塩及びアンモニアを用いる共沈殿法(R.Massart,IEEE Trans.Magn.1981,17,1247)、又は、水溶性鉄塩の水溶液中の酸化反応を用いた方法により合成することができる。
磁性シリカ粒子の平均粒子径は、好ましくは1~5μm、更に好ましくは1~3μmである。平均粒子径が1μm未満の場合、分離回収の際に時間がかかる傾向にあり、5μmを超えると、表面積が小さくなり、担持する物質(対象物質、測定対象物質の類似物質又は測定対象物質と特異的に結合する物質)の結合量が低く結合効率が低下する傾向にある。
本発明における磁性シリカ粒子の平均粒子径は、任意の200個の磁性シリカ粒子について走査型電子顕微鏡(日本電子株式会社製「JSM-7000F」)で観察して測定された粒子径の平均値である。
磁性シリカ粒子の平均粒子径は、後述の水中油型エマルションを作製する際の混合条件(せん断力等)を調節して水中油型エマルションの粒子径を調整することにより制御することができる。また、磁性シリカ粒子製造時の水洗工程の条件変更又は通常の分級等の方法によっても平均粒子径を所望の値とすることができる。
本発明における磁性シリカ粒子の製造方法としては、例えば平均粒子径が1~15nmの超常磁性金属酸化物粒子、前記超常磁性金属酸化物粒子100重量%に対して30~500重量%の(アルキル)アルコキシシラン及び分散剤を含有する分散液(A1)、又は更に非水溶性有機溶媒を含有する分散液(A2)と、水、水溶性有機溶媒、非イオン性界面活性剤及び(アルキル)アルコキシシランの加水分解用触媒を含有する溶液(B)とを混合して、水中油型エマルションを作製し、(アルキル)アルコキシシランの加水分解反応及び縮合反応を行い、超常磁性金属酸化物がシリカに包含された粒子を製造する方法が挙げられる。
(アルキル)アルコキシシランの加水分解反応及び縮合反応後、遠心分離及び磁石により固液分離し、水又はメタノールで洗浄して乾燥することにより磁性シリカ粒子が得られる。
上記及び以下において、(アルキル)アルコキシシランとは、アルキルアルコキシシラン又はアルコキシシランを意味する。
上記分散剤としては、分子内に1個以上のカルボキシル基を有する有機化合物、1個以上のスルホ基を有する有機化合物及び1個以上のカルボキシル基と1個以上のスルホ基を有する有機化合物等が挙げられる。具体的には、以下に例示する(a-1)~(a-5)の有機化合物等が挙げられ、これらは1種を単独で用いても2種以上を併用してもよい。
(a-1)カルボキシル基を2個以上有する有機化合物:
炭素数2~30の脂肪族ポリカルボン酸(シュウ酸、マロン酸、コハク酸、グルタル酸、アジピン酸、ピメリン酸、スベリン酸、アゼライン酸、セバシン酸、プロパン-1,2,3-トリカルボン酸、クエン酸及びドデカン二酸等の飽和ポリカルボン酸並びにマレイン酸、フマール酸及びイタコン酸等の不飽和ポリカルボン酸等)、炭素数8~30の芳香族ポリカルボン酸(フタル酸、イソフタル酸、テレフタル酸、トリメリット酸及びピロメリット酸等)及び炭素数5~30の脂環式ポリカルボン酸(シクロブテン-1,2-ジカルボン酸、シクロペンテン-1,2-ジカルボン酸、フラン-2,3-ジカルボン酸、ビシクロ[2,2,1]ヘプタ-2-エン-2,3-ジカルボン酸及びビシクロ[2,2,1]ヘプタ-2,5-ジエン-2,3-ジカルボン酸等)等。
(a-2)スルホ基を2個以上有する有機化合物:
炭素数1~30の脂肪族ポリスルホン酸(メチオン酸、1,1-エタンジスルホン酸、1,2-エタンジスルホン酸、1,1-プロパンジスルホン酸、1,3-プロパンジスルホン酸及びポリビニルスルホン酸等)、炭素数6~30の芳香族ポリスルホン酸(m-ベンゼンジスルホン酸、1,4-ナフタレンスルホン酸、1,5-ナフタレンジスルホン酸、1,6-ナフタレンジスルホン酸、2,6-ナフタレンジスルホン酸、2,7-ナフタレンジスルホン酸及びスルホン化ポリスチレン等)、ビス(フルオロスルホニル)イミド及び炭素数2~10のビス(パーフルオロアルカンスルホニル)イミド[ビス(トリフルオロメタンスルホニル)イミド、ビス(ペンタフルオロエタンスルホニル)イミド、ビス(ノナフルオロブタンスルホニル)イミド、ペンタフルオロエタンスルホニルトリフルオロメタンスルホニルイミド、トリフルオロメタンスルホニルヘプタフルオロプロパンスルホニルイミド、ノナフルオロブタンスルホニルトリフルオロメタンスルホニルイミド等]等。
(a-3)カルボキシル基とスルホ基をそれぞれ1個以上有する有機化合物:
炭素数2~30のスルホカルボン酸(スルホ酢酸及びスルホコハク酸等)及び炭素数7~30のスルホ芳香族モノ又はポリカルボン酸(o-、m-又はp-スルホ安息香酸、2,4-ジスルホ安息香酸、3-スルホフタル酸、3,5-ジスルホフタル酸、4-スルホイソフタル酸、2-スルホテレフタル酸、2-メチル-4-スルホ安息香酸、2-メチル-3,5-ジスルホ安息香酸、4-プロピル-3-スルホ安息香酸、4-イソプロピル-3-スルホ安息香酸、2,4,6-トリメチル-3-スルホ安息香酸、2-メチル-5-スルホテレフタル酸、5-メチル-4-スルホイソフタル酸、5-スルホサリチル酸及び3-オキシ-4-スルホ安息香酸等)等。
(a-4)カルボキシル基を1個有する有機化合物:
炭素数1~30の脂肪族飽和モノカルボン酸(ギ酸、酢酸、プロピオン酸、酪酸、イソ酪酸、吉草酸、カプロン酸、エナント酸、カプリル酸、ベラルゴン酸、ラウリン酸、ミリスチン酸、ステアリン酸及びベヘニン酸等)、炭素数3~30の脂肪族不飽和モノカルボン酸(アクリル酸、メタクリル酸、オレイン酸等)、炭素数3~30のヒドロキシ脂肪族モノカルボン酸(グリコール酸、乳酸及び酒石酸等)、炭素数4~30の脂環式モノカルボン酸(シクロプロパンカルボン酸、シクロペンタンカルボン酸及びシクロヘキサンカルボン酸等)、炭素数7~30の芳香族モノカルボン酸(安息香酸、ケイ皮酸及びナフトエ酸等)、炭素数7~20のヒドロキシ芳香族モノカルボン酸(サリチル酸及びマンデル酸等)及び炭素数2~20のパーフルオロカルボン酸(トリフルオロ酢酸、ウンデカフルオロヘキサン酸、トリデカフルオロヘプタン酸、パーフルオロオクタン酸、ペンタデカフルオロオクタン酸、ペプタデカフルオロノナン酸及びノナデカフルオロデカン酸等)等。
(a-5)スルホ基を1個有する有機化合物:
炭素数1~30の脂肪族モノスルホン酸(メタンスルホン酸、エタンスルホン酸、プロパンスルホン酸、ブタンスルホン酸、ヘキサンスルホン酸、デカンスルホン酸、ウンデカンスルホン酸及びドデカンスルホン酸等)、炭素数6~30の芳香族モノスルホン酸(ベンゼンスルホン酸、p-トルエンスルホン酸、o-トルエンスルホン酸、m-トルエンスルホン酸、4-ドデシルベンゼンスルホン酸、4-オクチルベンゼンスルホン酸及びナフタレンスルホン酸等)及び炭素数1~20のパーフルオロアルカンスルホン酸(トリフルオロメタンスルホン酸等)等。
これらの内、(アルキル)アルコキシシランとの相溶性の観点から(a-4)の内の炭素数10~30の有機化合物が好ましい。
分散剤の使用量は、超常磁性金属酸化物の重量を基準として、100~2,000重量%、特に250~1,000重量%であることが好ましい。分散剤が100重量%未満の場合、超常磁性金属酸化物が(アルキル)アルコキシシラン溶液に分散しにくい傾向にあり、2,000重量%を超えると後の工程の水溶液への分散の際にエマルションが形成しにくい傾向にある。
使用する(アルキル)アルコキシシランとしては、下記一般式(1)で表される化合物が挙げられる。

一般式(1)中、R及びRは、アミノ基、カルボキシル基、水酸基、メルカプト基又はグリシジルオキシ基で置換されていてもよい炭素数1~10の1価の炭化水素基を表す。
炭素数1~10の炭化水素基としては、炭素数1~10の脂肪族炭化水素基(メチル基、エチル基、n-又はiso-プロピル基、n-又はiso-ブチル基、n-又はiso-ペンチル基及びビニル基等)、炭素数6~10の芳香族炭化水素基(フェニル基等)及び炭素数7~10の芳香脂肪族基(ベンジル基等)等が挙げられる。
一般式(1)におけるnは1~4の整数を表す。但し、nが1のアルキルアルコキシシランを用いる場合は、nが2~4の(アルキル)アルコキシシランと併用する必要がある。反応後の粒子の強度及び粒子表面のシラノール基の量の観点からnは4であることが好ましい。
一般式(1)で表される化合物の具体例としては、テトラメトキシシラン、テトラエトキシシラン、テトライソプロポキシシラン及びテトラブトキシシラン等のアルコキシシラン;メチルトリメトキシシラン及びメチルトリエトキシシラン等のアルキルアルコキシシラン;3-アミノプロピルトリメトキシシラン、3-アミノプロピルトリエトキシシラン、N-2(アミノエチル)3-アミノプロピルトリメトキシシラン及びN-2(アミノエチル)3-アミノプロピルトリエトキシシラン等のアミノ基で置換されたアルキル基を有するアルキルアルコキシシラン;7-カルボキシ-ヘプチルトリエトキシシラン及び5-カルボキシ-ペンチルトリエトキシシラン等のカルボキシル基で置換されたアルキル基を有するアルキルアルコキシシラン;3-ヒドロキシプロピルトリメトキシシラン及び3-ヒドロキシプロピルトリエトキシシラン等の水酸基で置換されたアルキル基を有するアルキルアルコキシシラン;3-メルカプトプロピルトリメトキシシラン及び3-メルカプトプロピルトリエトキシシラン等のメルカプト基で置換されたアルキル基を有するアルキルアルコキシシラン;3-グリシジルオキシプロピルトリメトキシシラン及び3-グリシジルオキシプロピルトリエトキシシラン等のグリシジルオキシ基で置換されたアルキル基を有するアルキルアルコキシシラン等が挙げられる。
(アルキル)アルコキシシランは、1種類を単独で用いても2種以上を併用してもよい。
(アルキル)アルコキシシランの使用量は、超常磁性金属酸化物100重量%に対して、通常30~500重量%、好ましくは40~200重量%である。(アルキル)アルコキシシランが30重量%未満の場合、超常磁性金属酸化物の表面が均一に被覆されにくくなり、500重量%を超えると、超常磁性金属酸化物の含有率が小さくなり、磁力による回収時間が長くなる。
水の使用量は、超常磁性金属酸化物100重量%に対して500~3,000重量%であることが好ましく、特に800~2,000重量%が好ましい。
非水溶性有機溶媒としては、25℃における水への溶解度が0.1g/水100g以下である、炭素数6~16の芳香族炭化水素溶剤(トルエン及びキシレン等)及び炭素数5~16脂肪族炭化水素溶剤(n-ヘプタン、n-ヘキサン、シクロヘキサン、n-オクタン、デカン及びデカヒドロナフタレン等)等が挙げられる。これらは、1種類を単独で用いても2種以上を併用してもよい。
非水溶性有機溶媒の使用量は、超常磁性金属酸化物100重量%に対して、200~1,000重量%、特に250~500重量%が好ましい。有機溶媒の使用量が200重量%未満であると超常磁性金属酸化物の分散性が悪くなる傾向にあり、1,000重量%を超えると、磁性シリカ粒子の粒子径が不均一になる傾向にある。
水溶性有機溶媒としては、25℃における水への溶解度が100g/水100g以上である、炭素数1~3のアルコール(メタノール、エタノール及びn-又はiso-プロパノール等)、炭素数2~9のグリコール(エチレングリコール及びジエチレングリコール等)、アミド(N-メチルピロリドン等)、ケトン(アセトン等)、環状エーテル(テトラヒドロフラン及びテトラヒドロピラン等)、ラクトン(γ-ブチロラクトン等)、スルホキシド(ジメチルスルホキシド等)及びニトリル(アセトニトリル等)等が挙げられる。
これらの内、磁性シリカ粒子の粒子径の均一性の観点から、炭素数1~4のアルコールが好ましい。
水溶性有機溶媒は、1種類を単独で用いても2種以上を併用してもよい。
水溶性有機溶媒の使用量は、水100重量%に対して、100~500重量%であることが好ましい。
非イオン性界面活性剤としては、高級アルコールアルキレンオキサイド(以下、AOと略記)付加物;炭素数8~24の高級アルコール(デシルアルコール、ドデシルアルコール、ヤシ油アルキルアルコール、オクタデシルアルコール及びオレイルアルコール等)のエチレンオキサイド(以下、EOと略記)1~20モル及び/又はプロピレンオキサイド(以下、POと略記)1~20モル付加物(ブロック付加物及び/又はランダム付加物を含む。以下同様)、炭素数6~24のアルキルを有するアルキルフェノールのAO付加物;オクチル又はノニルフェノールのEO1~20モル及び/又はPO1~20モル付加物、ポリプロピレングリコールEO付加物及びポリエチレングリコールPO付加物;プルロニック型界面活性剤等、脂肪酸AO付加物;炭素数8~24の脂肪酸(デカン酸、ラウリン酸、ミリスチン酸、パルミチン酸、ステアリン酸、オレイン酸及びヤシ油脂肪酸等)のEO1~20モル及び/又はPO1~20モル付加物等及び多価アルコール型非イオン性界面活性剤;炭素数3~36の2~8価の多価アルコール(グリセリン、トリメチロールプロパン、ペンタエリスリトール、ソルビット及びソルビタン等)のEO及び/又はPO付加物;前記多価アルコールの脂肪酸エステル及びそのEO付加物[TWEEN(登録商標)20及びTWEEN(登録商標)80等];アルキルグルコシド(N-オクチル-β-D-マルトシド、n-ドデカノイルスクロース及びn-オクチル-β-D-グルコピラノシド等);砂糖の脂肪酸エステル、脂肪酸アルカノールアミド及びこれらのAO付加物(ポリオキシエチレン脂肪酸アルカノールアミド等);等の非イオン性界面活性剤が挙げられる。これらは、1種類を単独で用いても2種以上を併用してもよい。
非イオン性界面活性剤の使用量は、超常磁性金属酸化物100重量%に対して、100~1,000重量%、特に300~500重量%が好ましい。100重量%未満又は1,000重量%を超えると、エマルションが安定せず、生成する粒子の粒度分布が広くなる傾向がある。
非イオン性界面活性剤を含む水溶液の使用量は、超常磁性金属酸化物100重量%に対して、1,000~10,000重量%、特に1,500~4,000重量%が好ましい。1,000重量%未満又は10,000重量%を超えると、エマルションが安定せず、生成する粒子の粒度分布が広くなる傾向がある。
(アルキル)アルコキシシランの加水分解用触媒としては、ルイス酸や塩酸等を用いることができ、具体的には、塩酸等の無機酸、酢酸等の有機酸、アンモニア等の無機塩基化合物、エタノールアミン等のアミン化合物を用いることができる。
加水分解用触媒の使用量は、(アルキル)アルコキシシランの重量に対して、0.01~100重量%、特に0.2~50重量%が好ましい。
分散液(A1)又は分散液(A2)と、溶液(B)との混合方法は、特に限定されず、後述の設備を使用して一括混合することもできるが、磁性シリカ粒子の粒子径の均一性の観点から、溶液(B)を撹拌しながら分散液(A1)又は分散液(A2)を滴下する方法が好ましい。
分散液(A1)又は分散液(A2)と溶液(B)とを混合する際の設備としては、一般に乳化機、分散機として市販されているものであれば特に限定されず、例えば、ホモジナイザー(IKA社製)、ポリトロン(キネマティカ社製)及びTKオートホモミキサー(特殊機化工業社製)等のバッチ式乳化機、エバラマイルダー(荏原製作所社製)、TKフィルミックス、TKパイプラインホモミキサー(特殊機化工業社製)、コロイドミル(神鋼パンテック社製)、クリアミックス(エムテクニック社製)、スラッシャー、トリゴナル湿式微粉砕機(三井三池化工機社製)、キャピトロン(ユーロテック社製)及びファインフローミル(太平洋機工社製)等の連続式乳化機、マイクロフルイダイザー(みずほ工業社製)、ナノマイザー(ナノマイザー社製)及びAPVガウリン(ガウリン社製)等の高圧乳化機、膜乳化機(冷化工業社製)等の膜乳化機、バイブロミキサー(冷化工業社製)等の振動式乳化機、超音波ホモジナイザー(ブランソン社製)等の超音波乳化機等が挙げられ、粒子径の均一化の観点から、APVガウリン、ホモジナイザー、TKオートホモミキサー、エバラマイルダー、TKフィルミックス、TKパイプラインホモミキサー及びクリアミックスが好ましい。
(アルキル)アルコキシシランの加水分解反応及び縮合反応の温度は、25~100℃であることが好ましく、更に好ましくは45~60℃である。また、反応時間は、好ましくは0.5~5時間、更に好ましくは1~2時間である。
本発明における磁性シリカ粒子は、超常磁性金属酸化物がシリカに包含され、粒子表面に存在しないことから、多くの測定対象物質、測定対象物質の類似物質、又は測定対象物質と特異的に結合する物質をその表面に固定化することができる。
本発明における磁性シリカ粒子に、測定対象物質、測定対象物質の類似物質、又は、測定対象物質と特異的に結合する物質(以下、これらを総称して担持用物質と略記する場合がある)を固定化する方法としては、上述の磁性シリカ粒子に担持用物質を物理吸着させる方法が挙げられるが、より効率良く担持用物質を固定化させる観点から、グルタルアルデヒド、アルブミン、カルボジイミド、ストレプトアビジン、ビオチン及び官能基を有するアルキルアルコキシシランからなる群から選ばれる少なくとも1種の有機化合物を磁性シリカ粒子表面に結合させ、それらを介して担持用物質を磁性シリカ粒子に固定化させるのが好ましい。これらの有機化合物の内、特定の担持用物質を結合させる観点から、官能基を有するアルキルアルコキシシランが更に好ましい。
上記アルキルアルコキシシランが有する官能基としては、アミノ基、カルボキシル基、水酸基、メルカプト基及びグリシジルオキシ基等が挙げられ、アルキルアルコキシシラン1分子中に異なる種類の官能基を有していてもよい。
磁性シリカ粒子の表面に官能基を有するアルキルアルコキシシランを結合させる方法としては、磁性シリカ粒子を作製する際の(アルキル)アルコキシシランとして、前述のアミノ基、カルボキシル基、水酸基、メルカプト基又はグリシジルオキシ基で置換されたアルキル基を有するアルキルアルコキシシランを使用する方法、又は、これらの置換基を有しない(アルキル)アルコキシシランを使用して磁性シリカ粒子を作製した後、磁性シリカ粒子をアミノ基、カルボキシル基、水酸基、メルカプト基又はグリシジルオキシ基で置換されたアルキル基を有するアルキルアルコキシシランで処理する方法等が挙げられる。
後者の方法の具体例としては、磁性シリカ粒子をその濃度が1~50重量%になるように溶媒に分散し、この分散液にアミノ基、カルボキシル基、水酸基、メルカプト基又はグリシジルオキシ基で置換されたアルキル基を有するアルキルアルコキシシランの溶液を添加して、1時間以上室温で加水分解反応及び縮合反応を行う方法が挙げられる。
この方法における溶媒は、用いるアルキルアルコキシシランの溶解性に応じて適宜選択され、水に可溶なアミノ基、カルボキシル基、水酸基又はメルカプト基で置換されたアルキル基を有するアルキルアルコキシシランを用いる場合は、水又は水-アルコールの混合溶媒等を用いることが好ましく、水に溶解しにくいグリシジルオキシ基で置換されたアルキル基を有するアルキルアルコキシシランを用いる場合、酢酸ブチル等を用いることが好ましい。
アミノ基、カルボキシル基、水酸基、メルカプト基又はグリシジルオキシ基で置換されたアルキル基を有するアルキルアルコキシシランの使用量は、処理前の磁性シリカ粒子に対して0.1~5重量%であることが好ましい。0.1重量%未満であると、導入される官能基数が充分でない場合があり、5重量%を超えると、粒子表面の官能基量を更に増加させる効果が少なくなる傾向にある。
グルタルアルデヒド、アルブミン、カルボジイミド、ストレプトアビジン又はビオチンを磁性シリカ粒子の表面に結合させる方法は特に限定されないが、例えば、以下のようにして結合させることができる。
アルデヒド基を有するグルタルアルデヒド及びカルボキシル基を有するビオチンは、アミノ基を有するアルキルアルコキシシランが表面に結合した磁性シリカ粒子と反応させることで、磁性シリカ粒子の表面に結合させることができる。また、アミノ基を有するアルブミン及びストレプトアビジン並びにカルボジイミド基を有するカルボジイミドは、カルボキシル基を有するアルキルアルコキシシランが表面に結合した磁性シリカ粒子と反応させることで、磁性シリカ粒子の表面に結合させることができる。
本発明の測定対象物質測定方法は、上記本発明における磁性シリカ粒子を用いて行われる以外はこの分野で通常行われる、文献[例えば、酵素免疫測定法第2版(石川栄治ら編集、医学書院)1982年]記載のサンドイッチ法、競合法及び特開平6-130063号公報記載の測定法に準じて行えばよい。
サンドイッチ法は、例えば、測定対象物質を含む試料と、測定対象物質と特異的に結合する物質(測定対象物質結合物質)を表面に固定化した磁性シリカ粒子と、標識物質により標識された測定対象物質結合物質(標識測定対象物質結合物質)とを接触させて、磁性シリカ粒子上に測定対象物質結合物質と測定対象物質と標識測定対象物質結合物質との複合体(標識複合体)を形成させ、該標識複合体を担持した磁性シリカ粒子をB/F分離して、標識複合体中の標識物質量を測定し、その結果に基づいて試料中の測定対象物質を測定することによりなされる。
具体的には例えば、測定対象物質を含む試料と測定対象物質結合物質を表面に固定化した磁性シリカ粒子とを接触させて、磁性シリカ粒子表面に測定対象物質結合物質と測定対象物質との複合体を形成させ、更に該複合体に、標識測定対象物質結合物質を接触させて、磁性シリカ粒子に担持された測定対象物質結合物質と測定対象物質と標識測定対象物質結合物質との複合体(標識複合体)を形成させ、該標識複合体を担持した磁性シリカ粒子をB/F分離して、標識複合体中の標識物質量を測定し、その結果に基づいて試料中の測定対象物質を測定すればよい。該方法に於いては、測定対象物質と測定対象物質結合物質が固定化された磁性シリカ粒子とを反応させた後、標識測定対象物質結合物質を反応させているが、標識測定対象物質結合物質と測定対象物質とを反応させた後に測定対象物質結合物質が固定化された磁性シリカ粒子とを反応させても、これら3つを同時に反応させても構わない。
上記サンドイッチ法におけるB/F分離とは、上記標識複合体と、標識複合体の形成に関与しなかった標識測定対象物質結合物質との分離を意味し、具体的には、磁性シリカ粒子に担持された測定対象物質結合物質、磁性シリカ粒子に担持された測定対象物質結合物質と測定対象物質との複合体、及び、上記の標識複合体と、他の成分(試料中の測定対象物質以外の成分、標識複合体の形成に関与しなかった標識測定対象物質結合物質等)との分離を意味する。
また、B/F分離工程は標識複合体の形成後には必須の工程であるが、磁性シリカ粒子表面に測定対象物質結合物質と測定対象物質との複合体を形成させた後においても実施することができる。
競合法の一態様としては、測定対象物質を含む試料、標識物質により標識された測定対象物質と特異的に結合する物質(標識測定対象物質結合物質)、及び、測定対象物質又は測定対象物質の類似物質をその表面に固定化している磁性シリカ粒子とを接触させて、磁性シリカ粒子上に測定対象物質又はその類似物質と標識測定対象物質結合物質との複合体(標識複合体)を形成させ、該標識複合体を担持した磁性シリカ粒子をB/F分離して、標識複合体中の標識物質量を測定し、その結果に基づいて試料中の測定対象物質を測定することによりなされる。
具体的には例えば、測定対象物質を含む試料と、標識測定対象物質結合物質と、測定対象物質又はその類似物質を担持した磁性シリカ粒子とを接触させて、標識測定対象物質結合物質に、試料中の測定対象物質と磁性シリカ粒子上の測定対象物質又はその類似物質と競合反応させて、磁性シリカ粒子上に測定対象物質又はその類似物質と標識測定対象物質結合物質との複合体(標識複合体)を形成させ、該標識複合体を担持した磁性シリカ粒子をB/F分離して、標識複合体中の標識物質量を測定し、その結果に基づいて試料中の測定対象物質を測定すればよい。該方法に於いては、測定対象物質、標識測定対象物質結合物質、及び測定対象物質又はその類似物質を担持した磁性シリカ粒子を同時に競合反応させているが、測定対象物質と測定対象物質又はその類似物質を担持した磁性シリカ粒子とを接触させた後に、標識測定対象物質結合物質を加えて競合反応させても、測定対象物質と標識測定対象物質結合物質を接触させた後に、測定対象物質又はその類似物質を担持した磁性シリカ粒子を加えて競合反応させてもよい。
上記競合法におけるB/F分離とは、上記標識複合体と、標識複合体の形成に関与しなかった他の成分(標識測定対象物質結合物質、及び、標識測定対象物質結合物質と測定対象物質の複合体)との分離を意味し、具体的には、測定対象物質又はその類似物質を担持した磁性シリカ粒子、及び、測定対象物質又はその類似物質を担持した磁性シリカ粒子と標識測定対象物質結合物質との複合体(標識複合体)と、他の成分(試料中の測定対象物質以外の成分、標識測定対象物質結合物質、測定対象物質の複合体等)との分離を意味する。
また、競合法の別の態様としては、測定対象物質を含む試料と、標識物質により標識された測定対象物質又はその類似物質(標識測定対象物質又はその類似物質)と、測定対象物質結合物質を担持した磁性シリカ粒子とを接触させて、磁性シリカ粒子上に測定対象物質結合物質と標識測定対象物質又はその類似物質との複合体(標識複合体)を形成させ、該標識複合体を担持した磁性シリカ粒子をB/F分離して、標識複合体中の標識物質量を測定し、その結果に基づいて試料中の測定対象物質を測定することによりなされる。
具体的には例えば、測定対象物質を含む試料と、標識測定対象物質又はその類似物質と、測定対象物質結合物質を担持させた磁性シリカ粒子とを接触させて、磁性シリカ粒子上の測定対象物質結合物質に、試料中の測定対象物質と標識測定対象物質又はその類似物質とを競合反応させて、磁性シリカ粒子上に測定対象物質結合物質と標識測定対象物質又はその類似物質との複合体(標識複合体)とを形成させ、該標識複合体を担持した磁性シリカ粒子をB/F分離して、標識複合体中の標識物質量を測定し、その結果に基づいて試料中の測定対象物質を測定すればよい。該競合法に於いては、測定対象物質、標識測定対象物質又はその類似物質、及び、測定対象物質結合物質を担持させた磁性シリカ粒子を同時に競合反応させているが、測定対象物質と測定対象物質結合物質を担持させた磁性シリカ粒子とを接触させた後に、標識測定対象物質又はその類似物質を加えて競合反応させても、標識測定対象物質又はその類似物質と測定対象物質結合物質を担持させた磁性シリカ粒子とを接触させた後に、測定対象物質を加えて競合反応させてもよい。
上記競合法におけるB/F分離とは、上記標識複合体と、標識複合体の形成に関与しなかった他の成分(標識測定対象物質又はその類似物質)との分離を意味し、具体的には、測定対象物質結合物質を担持させた磁性シリカ粒子、測定対象物質結合物質を担持させた磁性シリカ粒子と測定対象物質との複合体、及び、測定対象物質結合物質を担持させた磁性シリカ粒子と標識測定対象物質又はその類似物質との複合体と、他の成分(試料中の測定対象物質以外の成分、標識複合体の形成に関与しなかった標識測定対象物質又はその類似物質等)との分離を意味する。
また、測定対象物質が酵素の場合には、上記サンドイッチ法及び競合法以外の酵素活性方法を用いる方法、例えば、測定対象物質を含む試料と、測定対象物質結合物質(例えば、抗体等の酵素と結合し得る物質)を担持させた磁性シリカ粒子を表面に固定化した磁性シリカ粒子とを接触させて、磁性シリカ粒子上に酵素と測定対象物質結合物質との複合体を形成させ、複合体を担持した磁性シリカ粒子をB/F分離した後、酵素の種類に応じた基質、又は酵素の種類に応じた基質及び発色剤、要すれば更に共役酵素を添加し、その基質の変化又は発色剤の発色結果に基づいて試料中の酵素量を測定する方法により、測定してもよい。尚、基質、発色剤、共役酵素は、公知のものを用いればよく、例えば酵素がペルオキシダーゼの場合には、過酸化水素とルミノール発光試薬等を用いればよい。これらの使用量も通常この分野で用いられる範囲であればよい。上記方法におけるB/F分離とは、測定対象物質と測定対象物質結合物質を担持させた磁性シリカ粒子との複合体と、その他の成分(試料中の測定対象物質以外の成分等)との分離を意味する。
本発明の測定対象物質測定方法において、試料、磁性シリカ粒子、標識された測定対象物質結合物質、標識測定対象物質又はその類似物質等を接触させる方法としては、通常なされる撹拌、混合等の処理により、磁性シリカ粒子が分散されればよい。反応時間は、測定対象物質、用いられる測定対象物質結合物質、サンドイッチ法、競合法等の違いに応じて適宜設定されればよいが、通常1~10分、好ましくは1~5分である。
本発明の測定対象物質測定方法におけるB/F分離は、例えば、磁性シリカ粒子の磁性を利用し、反応槽の外側等から磁石等により磁性シリカ粒子を集めて、反応液を排出し、洗浄液を加えた後、磁石を取り除き、磁性シリカ粒子を混合して分散させ、洗浄することによりなされる。上記操作を1~3回繰り返してもよい。洗浄液としては、通常この分野で用いられるものであれば特に限定はされない。
測定対象物質結合物質、測定対象物質又はその類似物質等を標識するために用いられる標識物質としては、例えば酵素免疫測定法(EIA)に於いて用いられるアルカリホスファターゼ、β-ガラクトシダーゼ、ペルオキシダーゼ、マイクロペルオキシダーゼ、グルコースオキシダーゼ、グルコース-6-リン酸脱水素酵素、リンゴ酸脱水素酵素、ルシフェラーゼ、チロシナーゼ、酸性ホスファターゼ等の酵素類、例えば放射免疫測定法(RIA)に於いて用いられる99mTc、131I、125I、14C、3H、32P等の放射性同位元素、例えば蛍光免疫測定法(FIA)に於いて用いられるフルオレセイン、ダンシル、フルオレスカミン、クマリン、ナフチルアミン或いはこれらの誘導体、グリーン蛍光タンパク質(GFP)等の蛍光性物質、例えばルシフェリン、イソルミノール、ルミノール、ビス(2,4,6-トリフロロフェニル)オキザレート等の発光性物質、例えばフェノール、ナフトール、アントラセン或いはこれらの誘導体等の紫外部に吸収を有する物質、例えば4-アミノ-2,2,6,6-テトラメチルピペリジン-1-オキシル、3-アミノ-2,2,5,5-テトラメチルピロリジン-1-オキシル、2,6-ジ-t-ブチル-α-(3,5-ジ-t-ブチル-4-オキソ-2,5-シクロヘキサジエン-1-イリデン)-p-トリオキシル等のオキシル基を有する化合物に代表されるスピンラベル化剤としての性質を有する物質等が挙げられる。
これらの内、感度等の観点から、酵素、蛍光性物質が好ましく、更に好ましいのはアルカリホスファターゼ、ペルオキシダーゼ及びグルコースオキシダーゼであり、特に好ましいのはペルオキシダーゼである。
上記した如き標識物質を測定対象物質結合物質、測定対象物質又はその類似物質等に結合させるには、通常この分野で用いられる方法、例えば自体公知のEIA、RIA或いはFIA等に於いて一般に行われている自体公知の標識方法[例えば、医化学実験講座、第8巻、山村雄一監修、第1版、中山書店、1971;図説 蛍光抗体、川生明著、第1版、(株)ソフトサイエンス社、1983;酵素免疫測定法、石川栄治、河合忠、室井潔編、第2版、医学書院、1982等に記載のグルタルアルデヒド法、過ヨウ素酸法、マレイミド法又はピリジルジスルフィド法等]等を利用すればよい。
標識物質の使用量は、用いる標識物質の種類により異なるため一概には言えないが、例えばペルオキシダーゼを標識物質として使用する場合には、測定対象物質結合物質と標識物質とを、例えば通常1:1~20のモル比、好ましくは1:1~10のモル比、更に好ましくは1:1~2のモル比となるように、例えばトリス緩衝液、リン酸緩衝液、ベロナール緩衝液、ホウ酸緩衝液、グッド緩衝液等の通常この分野で用いられている緩衝液中に含有させて用いればよい。尚、当該緩衝液としては、通常この分野で用いられている、例えばトリス緩衝液、リン酸緩衝液、ベロナール緩衝液、ホウ酸緩衝液、グッド緩衝液等が挙げられ、そのpHは、抗原抗体反応を抑制しない範囲であればよく、通常5~9である。また、このような緩衝液中には、目的の抗原抗体反応を阻害しないものであれば、例えばアルブミン、グロブリン、水溶性ゼラチン、ポリエチレングリコール等の安定化剤、界面活性剤、糖類等を含有させておいてもよい。
標識物質又はその活性の測定方法としては、放射免疫測定法(RIA)、酵素免疫測定法(EIA)、蛍光免疫測定法(FIA)及び化学発光免疫測定法(CLIA及びCLEIA)が挙げられ、短時間での免疫測定における感度の観点から好ましいのはEIA、CLIA及びCLEIAであり、更に好ましいのはCLEIAである。
本願発明の測定方法は、例えばサンドイッチ法でAFPを測定する場合、以下のようにして行えばよい。
即ち、例えばAFPを含む試料10~25μLと例えば本発明における磁性シリカ粒子に抗AFP抗体を担持させたものを0.1~1mg/mL含むリン酸緩衝液等の緩衝液(磁性シリカ粒子を含有する試薬)40~50μLとを反応槽に添加し、該反応溶液を攪拌し、磁性シリカ粒子を分散させて、磁性シリカ粒子上に担持されている抗AFP抗体とAFPとを接触、反応させ、複合体を形成させる。次いで、例えば反応槽の外側から磁石等により磁性シリカ粒子を集め、反応液を排出し、生理食塩水等の洗浄液を添加する。その後、磁石を取り除き、該磁性シリカ粒子を分散させて洗浄する。この操作は、1~3回繰り返してもよい。尚、この洗浄操作は、試料又は磁性シリカ粒子を含有する試薬を残したまま、西洋ワサビ由来等ペルオキシダーゼ(以下PODと略記)標識された抗AFP抗体(以下標識試薬と略記)を添加して反応させる場合には、この洗浄操作を省略しても構わない。その後、例えば標識試薬を加え、該反応溶液を攪拌し、磁性シリカ粒子を分散させて、POD標識された抗AFP抗体と上記複合体とを反応結合させる。次いで、上記の洗浄と同様にして生理食塩水等の洗浄液を加えて分散させて洗浄を行う。最後に例えばルミノール及び過酸化水素を加え、化学発光計にて1秒間の化学発光積算量を測定し、該測定値を基に試料中のAFP量を算出する。尚、この場合には予め規定のAFP含有溶液を試料として上記と同様の操作により作られたAFP量と1秒間の化学発光積算量との関係を示す検量線等を用いることで容易に試料中のAFP量を算出し得る。
本発明の試薬は、上記本発明における磁性シリカ粒子を含んでなるものであり、上記測定方法に用いられるものである。具体的には、例えば、本発明における磁性シリカ粒子を含む緩衝液等が挙げられ、該緩衝液としては、免疫測定等に通常用いられる緩衝液が好ましく挙げられ、例えば1,4-ピペラジンジエタンスルホン酸/水酸化ナトリウム緩衝液、MOPS[3-(N-モルホリノ)プロパンスルホン酸]/水酸化ナトリウム緩衝液、トリエタノールアミン/塩酸緩衝液及びPBS(リン酸緩衝液)等が挙げられる。
これらの緩衝液中の緩衝剤の濃度は、1~500mMが好ましく、更に好ましくは5~300mM、特に好ましくは10~200mMである。
上記及び以下において、mMは25℃での濃度(ミリモル/リットル)を表す。
磁性シリカ粒子の量は、特に限定されず、用いられる測定対象物質結合物質又は測定対象物質の類似物質の種類、測定対象物質の種類等により適宜選択できる。
本発明の試薬は、本発明における磁性シリカ粒子が緩衝液に分散された形態であることが好ましい。
本発明の試薬は、本発明における磁性シリカ粒子を含有する試薬以外に、標識物質により標識された測定対象物質と特異的に結合する物質、或いは、標識物質により標識された、測定対象物質又はその類似物質を含有する試薬(以下、標識試薬と略記する場合がある)を含んでいてもよい。該標識物質は、上記本発明の測定方法の項で記載したものと同じものが挙げられ、好ましいものも同じである。
標識試薬には、標識物質により標識された測定対象物質と特異的に結合する物質、或いは、標識物質により標識された、測定対象物質又はその類似物質以外に緩衝液等を含むことができる。標識試薬に使用される緩衝液としては、免疫測定に通常用いられる緩衝液が好ましく、例えば、上述の磁性シリカ粒子を含む試薬に使用される緩衝液と同様のものが挙げられ、緩衝液中の緩衝剤の濃度も上述の磁性シリカ粒子を含む試薬に使用される場合と同様である。
本発明の試薬は、本発明における磁性シリカ粒子を含有する試薬、及び、標識試薬以外に、化学発光試薬を含んでいてもよく、該化学発光試薬は、上記の標識物質に基づき選択され、例えば、標識物質がPODである場合、2,3-ジヒドロ-1,4-フタラジンジオン化合物及び化学発光増強剤を必須構成成分としてなる化学発光試薬第1液と、酸化剤及び水を必須構成成分としてなる化学発光試薬第2液とを含んでなる。
2,3-ジヒドロ-1,4-フタラジンジオン化合物としては、例えば、特開平2-291299号公報、特開平10-319015号公報及び特開2000-279196号公報等に記載の公知の2,3-ジヒドロ-1,4-フタラジンジオン化合物及びこれらの混合物等が使用できる。
これらの内、ルミノール、イソルミノール、N-アミノヘキシル-N-エチルイソルミノール(AHEI)、N-アミノブチル-N-エチルイソルミノール(ABEI)及びこれらの金属塩(アルカリ金属塩等)が好ましく、更に好ましいのはルミノール及びその金属塩、特に好ましいのはルミノールのナトリウム塩である。
化学発光試薬における2,3-ジヒドロ-1,4-フタラジンジオン化合物の含有量は、その種類及び適用する測定方法や測定条件等によって適宜設定されるが、化学発光増強効果及び保存安定性等の観点から、0.5~80mMが好ましく、更に好ましくは1.8~40mM、特に好ましくは3.5~21mMである。
化学発光増強剤としては、例えば、特表昭59-500252号公報、特開昭59-171839号公報及び特開平2-291299号公報等に記載の公知の化学発光増強剤及びこれらの混合物等が使用できる。
これらの内、化学発光増強効果等の観点から、フェノールが好ましく、更に好ましいのはP-ヨードフェノール、4-(シアノメチルチオ)フェノール及び4-シアノメチルチオ-2-クロロフェノール、特に好ましいのは4-(シアノメチルチオ)フェノールである。
化学発光試薬における化学発光増強剤の含有量は、その種類及び適用する測定方法や測定条件等によって適宜設定されるが、化学発光増強効果及び保存安定性等の観点から、0.1~15mMが好ましく、更に好ましくは0.3~7.0mM、特に好ましくは0.6~3.4mMである。
化学発光試薬第1液には、2,3-ジヒドロ-1,4-フタラジンジオン化合物及び化学発光増強剤以外に、緩衝液及び/又はキレート剤等を含むことができる。
緩衝液としては、例えば、特開平10-319015号公報及び特開2003-279489号公報等に記載の公知の緩衝液等が使用できる。
これらの内、化学発光増強効果及び保存安定性等の観点から、3-[4-(2-ヒドロキシエチル)-1-ピペラジニル]プロパンスルホン酸/水酸化ナトリウム緩衝液、2-ヒドロキシ-3-[4-(2-ヒドロキシエチル)-1-ピペラジニル]プロパンスルホン酸・1水和物/水酸化ナトリウム緩衝液及びピペラジニル-1,4-ビス(2-ヒドロキシ-3-プロパンスルホン酸)・2水和物/水酸化ナトリウム緩衝液が好ましく、更に好ましいのは3-[4-(2-ヒドロキシエチル)-1-ピペラジニル]プロパンスルホン酸/水酸化ナトリウム緩衝液及び2-ヒドロキシ-3-[4-(2-ヒドロキシエチル)-1-ピペラジニル]プロパンスルホン酸・1水和物/水酸化ナトリウム緩衝液、特に好ましいのは3-[4-(2-ヒドロキシエチル)-1-ピペラジニル]プロパンスルホン酸/水酸化ナトリウム緩衝液である。
これらの緩衝液における緩衝剤の含有量は、化学発光増強効果及び保存安定性等の観点から、1~500mMが好ましく、更に好ましくは5~300mM、特に好ましくは10~200mMである。
キレート剤としては、例えば、特開平9-75099号公報及び特開2003-279489号公報等に記載の公知のキレート剤等が使用できる。
これらの内、化学発光増強効果及び保存安定性等の観点から、4配位キレート剤が好ましく、更に好ましいのはエチレンジアミン四酢酸(EDTA)及びその塩(エチレンジアミン四酢酸二ナトリウム、エチレンジアミン四酢酸三ナトリウム、エチレンジアミン四酢酸四ナトリウム、エチレンジアミン四酢酸二カリウム及びエチレンジアミン四酢酸三カリウム等)並びにトランス-1,2ジアミノシクロヘキサン-N,N,N’,N’-四酢酸(CyDTA)、特に好ましいのはエチレンジアミン四酢酸(EDTA)及びその塩である。
キレート剤を含有する場合、その含有量は化学発光増強効果及び保存安定性の観点から、2,3-ジヒドロ-1,4-フタラジンジオン化合物の重量に基づいて、0.001~4重量%が好ましく、更に好ましくは0.01~2重量%、特に好ましくは0.05~1重量%である。
化学発光試薬第1液は、液体であることが好ましく、また、酵素の蛍光強度の観点からはアルカリ性であることが好ましい。第1液のpHは、7~11が好ましく、更に好ましくは8~10である。尚、pHは、JIS K0400-12-10:2000に準拠して測定される(測定温度25℃)。
化学発光試薬第1液は、2,3-ジヒドロ-1,4-フタラジンジオン化合物、化学発光増強剤並びに必要により緩衝液及び/又はキレート剤を均一混合することにより容易に得ることができる。
化学発光試薬第2液が含有する酸化剤としては、例えば、特開平8-261943号公報及び特開2000-279196号公報等に記載の公知の酸化剤等[無機の過酸化物(過酸化水素、過ホウ酸ナトリウム及び過ホウ酸カリウム等)、有機過酸化物(過酸化ジアルキル及び過酸化アシル等)、ペルオクソ酸化合物(ペルオクソ硫酸及びペルオクソリン酸等)等]の水溶液が挙げられる。
これらの内、保存安定性等の観点から、過酸化水素水溶液、過ホウ酸ナトリウム水溶液及び過ホウ酸カリウム水溶液が好ましく、更に好ましいのは過酸化水素水溶液である。
化学発光試薬第2液における酸化剤の濃度は、その種類及び適用する測定方法や測定条件等によって適宜設定されるが、化学発光増強効果等の観点から、0.5~40mMが好ましく、更に好ましくは1~20mM、特に好ましくは2.5~10mMである。
化学発光試薬第2液が含有する水としては、蒸留水、逆浸透水及び脱イオン水等が挙げられる。これらの内、化学発光増強効果及び保存安定性等の観点から、蒸留水及び脱イオン水が好ましく、更に好ましいのは脱イオン水である。
化学発光試薬第2液は、酸化剤及び水以外にキレート剤等を含むことができる。
キレート剤としては、上述の第1液に含むことができるキレート剤として例示したものと同様のものが挙げられ、好ましいものも同様である。
キレート剤を含有する場合、その含有量は、化学発光増強効果及び保存安定性の観点から、酸化剤の重量に基づいて、0.2~100重量%が好ましく、更に好ましくは0.5~20重量%、特に好ましくは1~10重量%である。
化学発光試薬第2液は、酸化剤、水及び必要によりキレート剤を均一混合することにより容易に得られる。
以下、実施例により本発明を更に説明するが、本発明はこれらに限定されるものではない。以下、特に定めない限り、%は重量%、部は重量部を示す。
製造例1
反応容器に塩化鉄(III)6水和物2.7部、塩化鉄(II)4水和物1.0部及び水375部を仕込んで溶解させて50℃に昇温し、攪拌下温度を50~55℃に保持しながら、25%アンモニア水3.8部と水100部を混合した溶液を1時間かけて滴下し、滴下後1時間攪拌し、オレイン酸10.5部加え、2時間攪拌を継続した。室温に冷却後、デカンテーションにより固液分離して得られたオレイン酸が吸着したマグネタイト粒子を水50部で洗浄する操作を3回行った。得られたオレイン酸が吸着したマグネタイト粒子を容器に仕込み、デカン5.7部及びテトラエトキシシラン2.2部を加えて混合し、分散液(A2-1)を調製した。
反応容器に25%アンモニア水溶液39.0部、イソプロパノール55.4部、ソルビタンモノオレエート2.9部(三洋化成工業社製「イオネットS-80」)及びポリオキシエチレン(付加モル数20モル)アルキル(C16~18)エーテル(三洋化成工業社製「エマルミン200」)2.0部を加えてクリアミックス(エムテクニック社製)を用いて混合し、50℃に昇温後、クリアミックスの回転数6,000rpmで攪拌しながら、上記分散液(A2-1)を1時間かけて滴下後、50℃で1時間反応させた。反応後、2,000rpmで5分間遠心分離して微粒子の存在する上清を除いた。得られた固相に水50部を加えて粒子を分散させて1,000rpmで10分間遠心分離後、微粒子の存在する上清を除く操作を10回行った。続いて、得られた固相に水50部を加えて粒子を分散させて500rpmで5分間遠心分離することにより、大きな粒子径の粒子を沈降させて目的とする粒子径を有する粒子を含有する上清(1)を回収した。残った固相に水50部を加えて500rpmで5分間遠心分離後、上清(2)を回収する操作を2回行い、固相中に存在する目的とする粒子径を有する粒子を回収した。次に、上清(1)及び(2)について、磁石を用いて粒子を集磁し、集磁された粒子を80℃で8時間乾燥させて磁性シリカ粒子(S1)を得た。
製造例2
乳化の際のクリアミックスの回転数を6,000rpmから7,500rpmに代えた以外は、製造例1と同様にして磁性シリカ粒子(S2)を得た。
製造例3
テトラエトキシシランの仕込量を2.2部から0.5部に代えた以外は、製造例1と同様にして磁性シリカ粒子(S3)を得た。
製造例4
乳化の際のクリアミックスの回転数を6,000rpmから7,500rpmに代えた以外は、製造例3と同様にして磁性シリカ粒子(S4)を得た。
製造例5
マグネタイト粒子作製時における25%アンモニア水3.8部と混合する水の量を100部から33.8部に代えた以外は、製造例1と同様にして磁性シリカ粒子(S5)を得た。
製造例6
乳化の際のクリアミックスの回転数を6,000rpmから7,500rpmに代えた以外は、製造例5と同様にして磁性シリカ粒子(S6)を得た。
製造例7
テトラエトキシシランの仕込量を2.2部から0.5部に代えた以外は、製造例5と同様にして磁性シリカ粒子(S7)を得た。
製造例8
乳化の際のクリアミックスの回転数を6,000rpmから7,500rpmに代えた以外は、製造例7と同様にして磁性シリカ粒子(S8)を得た。
製造例9
マグネタイト粒子作製時における25%アンモニア水3.8部と混合する水の量を100部から55部に代え、テトラエトキシシランの仕込量を2.2部から1.2部に代えた以外は、製造例2と同様にして磁性シリカ粒子(S9)を得た。
比較製造例1
テトラエトキシシランの仕込量を0.5部から8.0部に代えた以外は、製造例4と同様にして磁性シリカ粒子(H1)を得た。
比較製造例2
テトラエトキシシランの仕込量を1.2部から12.5部に代えた以外は、製造例9と同様にして磁性シリカ粒子(H2)を得た。
比較製造例3
テトラエトキシシランの仕込量を1.2部から8.0部に代え、乳化の際のクリアミックスの回転数を7,500rpmから6,000rpmに代えた以外は、製造例9と同様にして磁性シリカ粒子(H3)を得た。
比較製造例4
100部の硫酸第一鉄を1,000部の水に溶解し、攪拌下、水500部に水酸化ナトリウム28.8部を溶解した水溶液を1時間かけて滴下後、攪拌しながら、85℃まで昇温して空気を懸濁液に吹き込み8時間酸化し、遠心分離することにより得られたマグネタイト粒子2.2部をデカン5.7部及びテトラエトキシシラン2.2部と混合して分散液(R-1)を得た。分散液(A2-1)を分散液(R-1)に代えた以外は、製造例2と同様にして磁性シリカ粒子(H4)を得た。
比較製造例5
テトラエトキシシランの仕込量を2.2部から12.5部に代えた以外は、比較製造例4と同様にして磁性シリカ粒子(H5)を得た。
比較製造例6
平均粒子径50nmのマグネタイト粒子(シグマ アルドリッチ ジャパン社製)2.2部をデカン5.7部及びテトラエトキシシラン2.2部と混合して分散液(R-2)を得た。分散液(A2-1)を分散液(R-2)に代える以外は、製造例1と同様にして磁性シリカ粒子(H6)を得た。
実施例1
以下の操作により、磁性シリカ粒子を含有する試薬(抗AFP抗体結合磁性シリカ粒子試薬)、標識試薬(POD標識抗AFP抗体試薬)、化学発光試薬第1液及び化学発光試薬第2液から構成される本発明の試薬を得た。
磁性シリカ粒子を含有する試薬の作製:
1重量%γ-アミノプロピルトリエトキシシラン含有アセトン溶液40mLの入った蓋付きポリエチレン瓶に製造例1で作製した磁性シリカ粒子(S1)40mgを加え、25℃で1時間反応させ、ネオジウム磁石で磁性シリカ粒子を集磁後、液をアスピレーターで吸引除去した。次いで脱イオン水40mLを加えて蓋をし、ポリスチレン瓶をゆっくりと2回倒置攪拌した後、ネオジウム磁石で磁性シリカ粒子を集磁後、液をアスピレーターで吸引除去して磁性シリカ粒子を洗浄した。この洗浄操作を3回行った。次いで、この洗浄後の磁性シリカ粒子を2重量%グルタルアルデヒド含有水溶液40mLの入った蓋付きポリエチレン瓶に加え、25℃で1時間反応させた。そして、脱イオン水40mLを加えて蓋をし、ポリスチレン瓶をゆっくりと2回倒置攪拌したのち、ネオジウム磁石で磁性シリカ粒子を集磁後、液をアスピレーターで吸引除去して磁性シリカ粒子を洗浄した。この洗浄操作を3回行った。更にこの洗浄後の磁性シリカ粒子を抗AFPモノクローナル抗体(ダコジャパン社より購入)を20μg/mLの濃度で含む0.02Mリン酸緩衝液(pH8.7)40mLの入った蓋付きポリエチレン瓶に加え、25℃で1時間反応させた。反応後、ネオジウム磁石で磁性シリカ粒子を集磁後、抗AFP抗体含有リン酸緩衝液を除去し、抗AFP抗体結合磁性シリカ粒子を作製した。これを1%の牛血清アルブミン含有の0.02Mリン酸緩衝液(pH7.0)で、抗AFP抗体結合磁性シリカ粒子濃度として0.5mg/mLの濃度に希釈し、磁性シリカ粒子を含有する試薬を調製し、冷蔵(2~10℃)で保存した。
標識試薬の作製:
抗AFPポリクローナル抗体(ダコジャパン社より購入)、西洋ワサビ由来POD(東洋紡社製)を用い、文献(エス・ヨシタケ、エム・イマガワ、イー・イシカワ、エトール;ジェイ.バイオケム,Vol.92,1982,1413-1424)に記載の方法でPOD標識抗AFP抗体を調製した。これを1%の牛血清アルブミン含有の0.02Mリン酸緩衝液(pH7.0)で、POD標識抗AFP抗体濃度として200nMの濃度に希釈し、標識試薬を調製し、冷蔵(2~10℃)で保存した。
化学発光試薬第1液の調製:
ルミノールのナトリウム塩[シグマ アルドリッチ ジャパン社製]0.7g及び4-(シアノメチルチオ)フェノール0.1gを1,000mLメスフラスコに仕込んだ。3-[4-(2-ヒドロキシエチル)-1-ピペラジニル]プロパンスルホン酸/水酸化ナトリウム緩衝液(10mM、pH8.6)を溶液の容量が1,000mLになるように仕込み、25℃で均一混合して化学発光試薬第1液を調製した。測定に用いるまで冷蔵(2~10℃)保存した。
化学発光試薬第2液の調製:
過酸化水素[和光純薬工業社製、試薬特級、濃度30重量%]6.6gを1,000mLメスフラスコに仕込んだ。脱イオン水を溶液の容量が1,000mLになるように仕込み、25℃で均一混合して化学発光試薬第2液を調製した。測定に用いるまで冷蔵(2~10℃)保存した。
実施例2~9及び比較例1~6
磁性シリカ粒子(S1)を磁性シリカ粒子(S2)~(S9)又は(H1)~(H6)に代える以外は実施例1と同様にして、本発明の又は比較用の磁性シリカ粒子を含有する試薬、標識試薬、化学発光試薬第1液及び化学発光試薬第2液から構成される本発明の試薬を得た。
得られた試薬を用いて、以下の方法により短時間での免疫測定における洗浄性及び感度を評価した結果を表1に示す。また、使用した磁性シリカ粒子(S1)~(S9)及び(H1)~(H6)について、以下に示す方法で超常磁性金属酸化物の平均粒子径、磁性シリカ粒子の平均粒子径、超常磁性金属酸化物の含有量を測定し、集磁性、再分散性及び再凝集性を評価した結果を併せて表1に示す。
<本発明の試薬を用いた短時間での免疫測定における洗浄性及び感度の評価方法>
磁性シリカ粒子を含有する試薬0.025mLと1重量%の牛血清アルブミンを含有したリン酸緩衝液で調製したAFP濃度が2ng/mLの標準AFP液0.025mLを試験管に入れて混合し、試験管中で37℃で3分間反応させ、抗AFP抗体結合磁性シリカ粒子/AFP複合体を形成させた。反応後、試験管の外側からネオジウム磁石で磁性シリカ粒子を10秒間集め、試験管中の液をアスピレーターで除き、ネオジウム磁石を側面から十分に離し、生理食塩水0.5mLを加えて磁性シリカ粒子を分散させて集磁後、アスピレーターで液を除く洗浄操作を3回行った。
続いて、標識試薬0.025mLを試験管に注入し、試験管中で37℃で3分間反応させ、抗AFP抗体結合磁性シリカ粒子/AFP/POD標識抗AFP抗体複合体を形成させた。反応後、試験管の外側からネオジウム磁石で磁性シリカ粒子を10秒間集め、試験管中の液をアスピレーターで除き、ネオジウム磁石を側面から十分に離し、生理食塩水0.5mLを加えて磁性シリカ粒子を分散させて集磁後、アスピレーターで液を除く洗浄操作を2回行った。
最後に、化学発光試薬第1液0.07mLと化学発光試薬第2液0.07mLとを同時に加え、37℃で43秒間発光反応させ、化学発光試薬を添加後43~45秒の平均発光量を発光検出器[Lumat LB9507(ベルトールドジャパン社製)]で測定した。尚、AFP濃度が2ng/mLの標準AFP液の代わりにAFP濃度が0ng/mLの標準AFP液を使用して上記と同様の操作を行いバックグラウンドとして用いた。
洗浄性については、AFP濃度が0ng/mLの標準溶液を用いて免疫測定を行った場合の平均発光量から以下の基準で判定した。
○:10,000cps未満
△:10,000cps以上かつ30,000cps未満
×:30,000cps以上
感度については、AFP濃度が0ng/mLと2ng/mLの標準溶液を用いた免疫測定を行った場合の発光量の差から以下の基準で判定した。
○:25,000cps以上
△:10,000cps以上かつ25,000cps未満
×:10,000cps未満
<超常磁性金属酸化物の平均粒子径の測定方法>
任意の200個の超常磁性金属酸化物について、走査型電子顕微鏡で観察して粒子径を測定し、その平均値を平均粒子径とした。
<磁性シリカ粒子の平均粒子径の測定方法>
任意の200個の磁性シリカ粒子について、走査型電子顕微鏡で観察して粒子径を測定し、その平均値を平均粒子径とした。
<磁性シリカ粒子中の超常磁性金属酸化物の含有量の測定方法>
任意の20個の磁性シリカ粒子について、走査型電子顕微鏡で観察し、エネルギー分散型X線分光装置により超常磁性金属酸化物の含有量を測定してその平均値を含有量とした。
<磁性シリカ粒子の集磁性の評価方法>
1.0mgの磁性粒子シリカ粒子を2mLのイオン交換水に分散させ、口内径×胴径×全高=φ10.3mm×φ12.0mm×35mmのガラス容器に入れ、1cm×1cm×1cmのネオジウム磁石を側面につけ、初期吸光度が20%となるまでの時間を測定し、以下の基準で判定した。
○:15秒未満
△:15秒以上かつ30秒未満
×:30秒以上
<磁性シリカ粒子の再分散性の評価方法>
1.0mgの磁性粒子シリカ粒子を2mLのイオン交換水に分散させ、口内径×胴径×全高=φ10.3mm×φ12.0mm×35mmのガラス容器に入れ、1cm×1cm×1cmのネオジウム磁石を側面につけて、磁性シリカ粒子を完全に集磁し、上清を除去し、ネオジウム磁石を側面から十分に離し、2mLのイオン交換水を磁性シリカ粒子に噴きつけながら加え、3回のピペッティングにより混合した後、10秒以内に顕微鏡で観察し、視野内の全粒子数に対する凝集した粒子数の割合を算出して以下の基準で再分散性を判定した。
○:凝集粒子が5%未満
△:凝集粒子が5%以上かつ20%未満
×:凝集粒子が20%以上
<磁性シリカ粒子の再凝集性の評価方法>
3回のピペッティングによる混合後、顕微鏡で観察するまでの時間を10秒以内から5分に代える以外は上記再分散性の評価方法と同様にして、視野内の全粒子数に対する凝集した粒子数の割合を算出して以下の基準で判定した。
○:凝集粒子が5%未満
△:凝集粒子が5%以上かつ20%未満
×:凝集粒子が20%以上

本発明の磁性シリカ粒子を用いた測定対象物質測定方法は、磁性シリカ粒子の磁気特性及び集磁後の粒子の再分散性が優れることから、簡便且つ短時間に高感度で測定対象物質を測定することができるため、放射免疫測定法、酵素免疫測定法、蛍光免疫測定法及び化学発光免疫測定法等の臨床検査に幅広く適用できる。また、本発明の試薬は、上記測定方法に用いるのに適したものであり、同様に、放射免疫測定法、酵素免疫測定法、蛍光免疫測定法及び化学発光免疫測定法等の臨床検査薬として用いることができる。
 
 

Claims (18)

  1. 平均粒子径が1~15nmの超常磁性金属酸化物を60~95重量%含有するシリカ粒子の表面に、測定対象物質、測定対象物質の類似物質、又は、測定対象物質と特異的に結合する物質が固定化された磁性シリカ粒子を用いて試料中の測定対象物質を測定することを特徴とする測定対象物質測定方法。
  2. 前記磁性シリカ粒子が、測定対象物質と特異的に結合する物質(測定対象物質結合物質)をその表面に固定化しているものであって、前記測定対象物質を含む試料と、前記磁性シリカ粒子と、標識物質により標識された測定対象物質結合物質(標識測定対象物質結合物質)とを接触させて、前記磁性シリカ粒子上に前記測定対象物質結合物質と前記測定対象物質と前記標識測定対象物質結合物質との複合体(標識複合体)を形成させ、前記標識複合体を担持した磁性シリカ粒子をB/F分離して、前記標識複合体中の標識物質量を測定し、その結果に基づいて前記試料中の測定対象物質を測定する、請求項1記載の測定方法。
  3. 前記磁性シリカ粒子が、測定対象物質又は測定対象物質の類似物質をその表面に固定化しているものであって、前記測定対象物質を含む試料と、標識物質により標識された測定対象物質と特異的に結合する物質(標識測定対象物質結合物質)と、前記磁性シリカ粒子とを接触させて、前記磁性シリカ粒子上に前記測定対象物質又は測定対象物質の類似物質と前記標識測定対象物質結合物質との複合体(標識複合体)を形成させ、前記標識複合体を担持した磁性シリカ粒子をB/F分離して、前記標識複合体中の標識物質量を測定し、その結果に基づいて試料中の測定対象物質を測定する、請求項1記載の測定方法。
  4. 前記磁性シリカ粒子が、測定対象物質と特異的に結合する物質(測定対象物質結合物質)をその表面に固定化しているものであって、前記測定対象物質を含む試料と、標識物質により標識された測定対象物質又は測定対象物質の類似物質(標識測定対象物質又はその類似物質)と前記磁性シリカ粒子とを接触させて、前記磁性シリカ粒子上に前記測定対象物質結合物質と前記標識測定対象物質又はその類似物質との複合体(標識複合体)を形成させ、前記標識複合体を担持した磁性シリカ粒子をB/F分離して、前記標識複合体中の標識物質量を測定し、その結果に基づいて試料中の測定対象物質を測定する、請求項1記載の測定方法。
  5. 前記B/F分離が、磁性シリカ粒子の磁性を利用してなされるものである、請求項2~4の何れか記載の測定方法。
  6. 前記磁性シリカ粒子の平均粒子径が1~5μmである、請求項1~5の何れか記載の測定方法。
  7. 前記超常磁性金属酸化物が酸化鉄である請求項1~6の何れか記載の測定方法。
  8. 前記酸化鉄がマグネタイト、γ-ヘマタイト、マグネタイト-α-ヘマタイト中間酸化鉄及びγ-ヘマタイト-α-ヘマタイト中間酸化鉄からなる群から選ばれる少なくとも1種の酸化鉄である請求項7記載の測定方法。
  9. 前記測定対象物質、前記測定対象物質の類似物質、又は、前記測定対象物質と特異的に結合する物質が固定化される前の前記磁性シリカ粒子の表面にグルタルアルデヒド、アルブミン、カルボジイミド、ストレプトアビジン、ビオチン及び官能基を有するアルキルアルコキシシランからなる群から選ばれる少なくとも1種の有機化合物を結合させてなる、請求項1~8の何れか記載の測定方法。
  10. 前記アルキルアルコキシシランが有する官能基が、アミノ基、カルボキシル基、水酸基、メルカプト基及びグリシジルオキシ基からなる群から選ばれる少なくとも1種の官能基である、請求項9記載の測定方法。
  11. 前記測定対象物質と特異的に結合する物質が、測定対象物質又は測定対象物質の類似物質に対する抗体、測定対象物質又は測定対象物質の類似物質が結合する抗原、或いは、測定対象物質又は測定対象物質の類似物質に結合するタンパク質である、請求項1~10の何れか記載の測定方法。
  12. 平均粒子径が1~15nmの超常磁性金属酸化物を60~95重量%含有するシリカ粒子の表面に、測定対象物質、測定対象物質の類似物質、又は、測定対象物質と特異的に結合する物質が固定化された磁性シリカ粒子を含有することを特徴とする、請求項1~11の何れか記載の測定方法用試薬。
  13. 前記磁性シリカ粒子の平均粒子径が1~5μmである請求項12記載の試薬。
  14. 前記超常磁性金属酸化物が酸化鉄である請求項12又は13記載の試薬。
  15. 前記酸化鉄がマグネタイト、γ-ヘマタイト、マグネタイト-α-ヘマタイト中間酸化鉄及びγ-ヘマタイト-α-ヘマタイト中間酸化鉄からなる群から選ばれる少なくとも1種の酸化鉄である請求項14記載の試薬。
  16. 測定対象物質、測定対象物質の類似物質、又は、測定対象物質と特異的に結合する物質が固定化される前の前記磁性シリカ粒子の表面にグルタルアルデヒド、アルブミン、カルボジイミド、ストレプトアビジン、ビオチン及び官能基を有するアルキルアルコキシシランからなる群から選ばれる少なくとも1種の有機化合物を結合させてなる請求項12~15の何れか記載の試薬。
  17. 前記アルキルアルコキシシランが有する官能基が、アミノ基、カルボキシル基、水酸基、メルカプト基及びグリシジルオキシ基からなる群から選ばれる少なくとも1種の官能基である請求項16記載の試薬。
  18. 前記測定対象物質と特異的に結合する物質が、測定対象物質又は測定対象物質の類似物質に対する抗体、測定対象物質又は測定対象物質の類似物質が結合する抗原、或いは、測定対象物質又は測定対象物質の類似物質に結合するタンパク質である請求項12~17の何れか記載の試薬。
