五、發明說明(13) 吾等已構築MVA重組型病毒,其容許人類酪胺酸酶 基因在牛痘病毒早期/晚期啟動子P7.5之控制下的表現。 最近’人類路胺酸酶已被鑑定是為一種容許抗腫瘤溶胞性 τ-淋巴細胞之產生的黑色瘤-專一性腫瘤抗原(Brichard,V. et al· [1993] J. Exp· Med· 178, 489-495)。由於在正常細胞

Bakt· Hyg· I· Abt· Odg· B 167,375-390)。投藥之模式、投 藥之劑量及次數可為熟習此項技藝之人士以一已知之方式 而加以最適化。若適合的話,可方便地採行將該疫苗於一 個漫長時段内投藥數次,俾以得到對抗外來抗原之適當<免 疫反應。 MVA重組型病毒用於生產異種多胜肽之用途 17 575664 五、發明說明(15) 依據本發明之MVA重組型牛痘病毒亦可被使用於在 真核細胞中製備異種多胜肽。這需要有被該重組型牛痘病 毒所感染的細胞。編碼外來多胜肽之基因於該等細胞中被 表現,而被表現之異種多胜肽被分離出。要被用於生產該 等異種多胜肽之方法係為熟習此項技藝之人士一般所知曉 的(ΕΡ-Α·206,920 及 EP-A-205,939)〇 藉由該等 MVA 重組型 病毒之助而被產生之多胜肽,由於該等M VA病毒之特殊 性質,是更適合於供使用作為人類及動物之醫藥。 編碼Τ7 RN Α聚合酶之M VA重組型病毒及其用於表現在一 Τ7 RNA聚合酶啟動子之轉錄控制下的DNA序列之用途 在本發明之進一步較佳具體例中,吾等已構築出MVA 重組型病毒,其容許噬菌體Τ7 RNA聚合酶基因在牛痘病 毒早期/晚期啟動子Ρ7.5之控制下的表現。MVA-T7pol重 組型病毒作為表現系統之有用性已於暫時轉染分析中被測 試,俾以誘發重組型基因在一 T7 RNA聚合酶啟動子之控 制下的表現。利用大腸桿菌氯黴素乙醯基轉移酶(CAT)基 因作為一報告基因(reporter gene),吾等發現MVA-T7pol 會誘發出跟一源自於牛痘病毒之可行複製的WR株之牛痘 /T7pol重組型病毒同樣有效的CAT基因表現。 依據本發明之MVA/T7 RNA聚合酶雜交物系統可因 此被使用作為一用於在沒有生產性牛痘病毒複製下來生產 多胜肽之簡單、有效且安全之哺乳動物表現系統。 此表現系統亦可被使用於產生供用於疫苗接種或基因 治療之重組型病毒粒子,這係藉由以含有全部或某些該等 18 575664

USA 77, 2163-2167 (1980)]或藉由熟習此 項技藝者所知之其他方法。藉由磷酸鈣沈澱法之穿染是為 較佳的。 以下之洋細貫施例係意欲幫助對本發明有一更佳瞭 解。但是,並無意圖給予「本發明被限定於該等實施例中 的標的」之印象。 實施例 ” 1 ·病毒之生長及純化 1 · 1 M VA病毒之生長 23 575664 五、發明說明(21) M VA病毒是一經高度減毒之牛痘病毒,其係源自於牛 痘病毒安哥拉株(CVA)並藉由在原始雞胚纖維母細胞(CEF) 培養物上作長期連續繼代而得。有關MVA株之生產、性 質及用途之歷史的整體性回顧,可參見Mayer等人在 Infection 3,6-14 [1975]中所發表之總結報告。由於在CEF 内之減毒,M VA病毒在此禽類宿主細胞中複製至高效價。 然而,在哺乳類細胞中,M VA生長受到嚴重限制,且由該 病毒所形成之典型菌斑是偵測不到的。因此,MVA病毒是 於CEF細胞上生長。為了製備CEF細胞,自被孵化之雞 蛋中分離出11天大之胚胎,除去端點(extremities),而胚 胎被剁碎並在37°C於一 0.25%胰蛋白酶溶液中予以解離歷 時20分鐘。將所形成之細胞懸浮液過濾,而細胞藉由於室 溫下在一 Sorvall RC-3B離心機内以2000 rpm予以離心歷 時5分鐘而被沉降,將之再懸浮於10倍體積之培養基A (MEM Eagle,例如可得自於 Life Technologies GmbH, Eggenstein,德國)中,再次藉由於室溫下在一 Sorvall RC-3B 離心機内以2000 rpm予以離心歷時5分鐘而將細胞沉降。 細胞沉塊被再重建於含有10%胎牛血清(FCS)、盤尼西林 (100單位/毫升)、鏈黴素(100單位/毫升)以及2 mM麩胺酸 之培養基A中而得到一含有500000個細胞/毫升之細胞懸 浮液。以此方法所得之CEF細胞被塗佈於細胞培養皿上。 視所需的細胞密度而定,它們被留在培養基A中,並在37 °C下於一 C02培養箱中生長歷時1-2天,並被直接地或是 於再一次的細胞繼代後被用於感染。原始培養物的製備之 24 575664 五、發明說明(22) 一詳細描述可見於R.I. Natl· Acad. Sci. USA 89,10847-10851; Sutter,G. Wyatt,L·,Foley,P·, Bennink,J. and Moss,B. MVA重組型病毒之構築及特性判定 2.1載體質體之構築 為了容許MVA重組型病毒之產生,新質體載體被建 構。外來基因至M VA基因組内之插入被精確地對準至位 在MVA基因組内之天然發生的刪除位置II之位址。位在 MVA基因組之Hindlll N片段中之一個2500 bp删除位置 的側翼之 MVA DNA 序列(Altenburger,W·,Sutter,C.P· and Altenburger,J· [1989],J. Arch. Virol. 105, 15-27)以 PCR 予 以擴增並被選殖至質體pUC18之多重選殖位址内。有關左 端600 bp DNA側翼之引子係為5’-CAG CAG GGT ACC CTC ATC GTA CAG GAC GTT CTC-3’及 5,-CAG CAG CCC GGG TAT TCG ATG ATT ATT TTT AAC AAA ATA ACA-3’(有關於限制酶尤及Smal之位址被劃底線表 27 575664 五、發明說明(25) 示)。有關右端550 bp DNA側翼之引子係為5’-CAG CAG CTG CAG GAA TCA TCC ATT CCA CTG AAT AGC-3’及 5,-CAG CAG GCA TGC CGA CGA ACA AGG AAC TGT AGC AGA-3 ’(有關於限制酶PWI及办ΛΙ之位址被劃底線表 示)。在被插入至pUC 18内之此等側翼MVA DNA之間, 處在牛痘病毒晚期啟動子P11之控制下之大腸桿菌McZ基 因(藉由限制消化而從pill LZ被製得,Sutter,G. and Moss, Β· [1992] PNAS USA 89, 10847-10851),使用 BamHI 位址, 而被插入以產生MVA插入載體pUCII LZ (第一圖)。接下 來,一個包含牛痘病毒早期-晚期啟動子P7.5之289 bp片 段連同一用於選殖之Smal位址(藉由使用五及 之限制消化而從質體載體pSCll 被製得[Chakrabartietal· 1985, Molecular and Cellular Biology 5, 3403-3409])被插入 至pUC II LZ之Smal位址内以生成MVA載體pUC II LZ P7.5 (第二圖)。為了構築一容許經由報告酵素/3-半乳糖苷 酶之暫時合成來作MVA重組型病毒之分離的載體質體, 一源自於大腸桿菌ΖμΖ開放解讀框架(open reading frame) 之3’端之330 bp DNA片段以PCR予以擴增(引子為 5,-CAG CAG GTC GAC CCC GAC CGC CTT ACT GCC GCC-3,及 5,-GGG GGG CTG CAG AATG GTA GCG ACC GGC GCT CAG-3’),並將之選殖入 pUC II LZ P7.5 之心/1 及PWI位址内而得到MVA載體pUC II Lzdel Ρ7·5 (第三 圖)。利用Smal位址,此載體質體可被用於將編碼一處在 牛痘病毒啟動子P7.5之轉錄控制下的外來基因之DNA序 28 575664 五、發明說明(26) 列插入至M VA基因組中。 在所欲之重組型病毒已藉由篩選Θ -半乳糖苷酶活性 之表現而被分離出後,該重組型病毒之進一步繁殖導致被 再處理之Pll-LacZ表現卡匣藉由同源性重組的自我刪除。 2.2重組型病毒MVA T7pol之構築及特性判定 一個包含處在牛痘病毒早期/晚期啟動子Ρ7.5之控制 下的噬菌體Τ7 RNA聚合酶之全部基因的3.1 kbp DNA片 段藉由EcoRI而自質體pTF7-3被切出(Furest,T.R·,Niles, E.G., Studier, F.W. and Moss, B.? 1986, P.N.A.S. Meyer zum Buschenfelde, K. nad Boon, T. (1994) Eur. J.
