Claims (21)
1,4-ブタンジオール(BDO)経路を有する非天然微生物体であって、該微生物体は、BDOを生成するのに十分な量で発現されるBDO経路酵素をコードする少なくとも1つの外因性核酸を含み、該BDO経路酵素が、
(a)アセトアセチル-補酵素A(CoA)の3-ヒドロキシブチリル-CoAへの変換を触媒する3-ヒドロキシブチリル-CoAデヒドロゲナーゼ;(b)3-ヒドロキシブチリル-CoAのクロトノイル-CoAへの変換を触媒する3-ヒドロキシブチリル-CoAデヒドラターゼ;(c)クロトノイル-CoAのビニルアセチル-CoAへの変換を触媒するビニルアセチル-CoAΔ-イソメラーゼ;及び(d)ビニルアセチル-CoAの4-ヒドロキシブチリル-CoAへの変換を触媒する4-ヒドロキシブチリル-CoAデヒドラターゼを含み、
該微生物体が、(e)4-ヒドロキシブチリル-CoAの1,4-ブタンジオールへの変換を触媒する4-ヒドロキシブチリル-CoAレダクターゼ(アルコール形成);(f)4-ヒドロキシブチリル-CoAの4-ヒドロキシブタナールへの変換を触媒する4-ヒドロキシブチリル-CoAレダクターゼ;及び(g)4-ヒドロキシブタナールの1,4-ブタンジオールへの変換を触媒する1,4-ブタンジオールデヒドロゲナーゼから選択される酵素をコードする少なくとも1つの外因性核酸を更に含む、前記非天然微生物体。

該微生物体が、(e)4-ヒドロキシブチリル-CoAの1,4-ブタンジオールへの変換を触媒する4-ヒドロキシブチリル-CoAレダクターゼ(アルコール形成);(f)4-ヒドロキシブチリル-CoAの4-ヒドロキシブタナールへの変換を触媒する4-ヒドロキシブチリル-CoAレダクターゼ;及び(g)4-ヒドロキシブタナールの1,4-ブタンジオールへの変換を触媒する1,4-ブタンジオールデヒドロゲナーゼから選択される酵素をコードする少なくとも1つの外因性核酸を更に含む、前記非天然微生物体。
前記BDO経路酵素をコードする3以上の外因性核酸を含む、請求項1記載の非天然微生物体。
前記BDO経路酵素をコードする4以上の外因性核酸を含む、請求項1記載の非天然微生物体。
前記BDO経路酵素をコードする5以上の外因性核酸を含む、請求項1記載の非天然微生物体。
前記BDO経路酵素(a)3-ヒドロキシブチリル-CoAデヒドロゲナーゼ、(b)3-ヒドロキシブチリル-CoAデヒドラターゼ、(c)ビニルアセチル-CoAΔ-イソメラーゼ、及び(d)4-ヒドロキシブチリル-CoAデヒドラターゼ;並びに
(e)4-ヒドロキシブチリル-CoAレダクターゼ(アルコール形成)、又は(f)4-ヒドロキシブチリル-CoAレダクターゼ、及び(g)1,4-ブタンジオールデヒドロゲナーゼ
をコードする外因性核酸を含む、請求項1記載の非天然微生物体。
(e)4-ヒドロキシブチリル-CoAレダクターゼ(アルコール形成)、又は(f)4-ヒドロキシブチリル-CoAレダクターゼ、及び(g)1,4-ブタンジオールデヒドロゲナーゼ
をコードする外因性核酸を含む、請求項1記載の非天然微生物体。
前記BDO経路酵素が、4-ヒドロキシブチリル-CoAレダクターゼ(アルコール形成)を含む、請求項1〜5のいずれか1項記載の非天然微生物体。
前記BDO経路酵素が、4-ヒドロキシブチリル-CoAレダクターゼ、及び1,4-ブタンジオールデヒドロゲナーゼを含む、請求項1〜5のいずれか1項記載の非天然微生物体。
前記BDO経路酵素が、4-ヒドロキシブチリル-CoAレダクターゼ(アルコール形成)、4-ヒドロキシブチリル-CoAレダクターゼ、及び1,4-ブタンジオールデヒドロゲナーゼを含む、請求項1〜5のいずれか1項記載の非天然微生物体。
前記少なくとも1つの外因性核酸が異種核酸である、請求項1〜8のいずれか1項記載の非天然微生物体。
実質的に嫌気性の培地に存在する、請求項1〜9のいずれか1項記載の非天然微生物体。
請求項1記載の非天然微生物体を、BDOを生成するための条件下で、且つ十分な時間にわたって培養することを含む、BDOを生成するための方法。
前記微生物体が、前記BDO経路酵素をコードする3以上の外因性核酸を含む、請求項11記載の方法。
前記微生物体が、前記BDO経路酵素をコードする4以上の外因性核酸を含む、請求項11記載の方法。
前記微生物体が、前記BDO経路酵素をコードする5以上の外因性核酸を含む、請求項11記載の方法。
前記微生物体が、前記BDO経路酵素(a)3-ヒドロキシブチリル-CoAデヒドロゲナーゼ、(b)3-ヒドロキシブチリル-CoAデヒドラターゼ、(c)ビニルアセチル-CoAΔ-イソメラーゼ、及び(d)4-ヒドロキシブチリル-CoAデヒドラターゼ;並びに
(e)4-ヒドロキシブチリル-CoAレダクターゼ(アルコール形成)、又は(f)4-ヒドロキシブチリル-CoAレダクターゼ、及び(g)1,4-ブタンジオールデヒドロゲナーゼ
をコードする外因性核酸を含む、請求項11記載の方法。
(e)4-ヒドロキシブチリル-CoAレダクターゼ(アルコール形成)、又は(f)4-ヒドロキシブチリル-CoAレダクターゼ、及び(g)1,4-ブタンジオールデヒドロゲナーゼ
