Neue Eburnanderivate,Verfahren zur Herstellung derselben
und diese enthaltende pharmazeutische Präparate
Die "Erfindung betrifft neue Eburaanderivate, ein
Verfahren zur Herstellung derselben und diese Verbindungen enthaltende pharmazeutische Präparate.
Gegenstand der Erfindung sind neue, razemische oder optisch aktive neue Eburnanderivate der allgemeinen Formel
I
(worin
R eine 1-6 Kohlenstoffatome enthaltende Alkylgruppe ist;
A eine Hydroxygruppe und
Y ein Wasserstoffatom bedeuten, oder
Y und Y zusammen eine Oxogruppe bilden; wobei
das Wasserstoffatom in Stellung 3 und die Gruppe R eine
oc, c<- und/oder ß,ß- bzw. σς,β und/oder ö,oc und die
Gruppe A und Y eine <x,ß und/oder ß,o<
Konfiguration haben)
und Säureadditionssalze davon.
Gegenstand der.Erfindung ist weiterhin ein Verfahren
zur Herstellung von neuen, razemischen oder optisch aktiven Eburnanderivaten der allgemeinen Formel I und deren

Säureadditionssalzen, indem man ein neues, razemisches
oder optisch aktives Eburnanoxim-Derivat der allgemeinen Formel Ha
(Ha)
und/oder
(lib)
ITb (worin die Bedeutung von R und die Konfiguration dieser
Gruppe zum Wasserstoffatom in Stellung 3 wie bei der allgemeinen Formel I angegeben sind) oder ein Säureadditionssalz
davon desoximiert und erwünschtenfalls mit einer
Säure behandelt und/oder erwünschtenfalls das erhaltene razemische oder optisch aktive neue Eburnan-Derivat der
allgemeinen Formel I (in welchem A und T zusammen eine Oxo-Gruppe bilden und"die Bedeutung der Gruppe E und die
Konfiguration dieser Gruppe zum Wasserstoffatom in Stellung
3 wie bei der allgemeinen Formel I angegeben ist) oder ein Säureadditionssalz davon einer partiellen Reduktion
unterwirft und erwünschtenfalls ein 15-Epimerengemisch des erhaltenen neuen Eburnan-Derivats der allgemeinen
Formel I (worin A eine Hydroxygruppe und T ein Wasserstoffatom bedeuten und die Konfiguration des Wasserstoffatoms
in Stellung 3 und der R Gruppe wie oben angegeben ist) in die Epimere aufspaltet bzw. ein erhaltenes 15-Epimer
in das andere 15-Epimer überführt und/oder erwünschtenfalls ein. erhaltenes razemisches Eburnanderivat
der allgemeinen Formel I in die optisch aktiven Antipode aufspaltet und/oder erwünschtenfalls ein erhaltenes razemisches
oder optisch aktives Eburnan-Derivat der allgemeinen Formel I oder gegebenenfalls dessen 15-Epimere mit
R in den obigen allgemeinen Formeln steht für eine geradkettige oder verzweigte, 1-6 Kohlenstoffatome
enthaltende Alkylgruppe (z. B. Methyl, Äthyl, n-Propyl,
Isopropyl, η-Butyl, sec.-Butyl, tert.-Butyl, n-Pentyl,
Isopentyl, n-Hexyl, Isohexyl).
Die neuen razemischen oder optisch aktiven Ausgangsstoffe der allgemeinen Formel Ha und/oder ITb
können aus den entsprechenden Eburnamonin-Derivaten durch Oximbildung hergestellt werden. Diese Reaktion kann
zweckmäßig unter Anwendung eines tertiären-(C^ « Alkyl) nitrits,
in Gegenwart einer starken Base durchgeführt werden.
Die Desoximierung der Verbindungen der allgemeinen
Formel Ha und/oder Hb kann hydrolytisch (z. B.
fflit Hilfe einer verdünnten wäßrigen Salzsäurelösung
und Formaldehyd oder Levulinsäure, oder mit einem Gemisch von Brenztraubensäure und einer Mineralsäure,
oder mit einer wäßrigen Natriumdisulphitlösung) oder auf
oxydativem Wege [ζ. Β. mit Cerium(17) ammoniumnitrat,
Blei(IV)acetat, Tallium(III)nitrat, Perj odsäure, mit einem
Gemisch von Chromtrioxid und Schwefelsäure oder Essigsäure,
mit Ozon oder mit einem Gemisch von Palladium- -triphenylphosphit und Sauerstoff] oder auf reduktivem
Wege [z. B. mit Titan(III)Chlorid, Chrom(II)acetat, Raneynickel
in alkalischem Medium oder Zink in Essigsäure] oder durch Umoximierung mit Aceton durchgefüht werden.
Nach einer vorteilhaften Ausführungsform des erfindungsgemäßen
Verfahrens wird die Desoximierung durch Hydrolyse durchgeführt. Man verfährt zweckmäßig so, daß
man die razemische oder optisch aktive neue Verbindung der allgemeinen Formel Ha und/oder lib mit der wäßrigen
Lösung einer Mineralsäure (z. B. mit wäßriger Salzsäure oder Schwefelsäure) bei einer Temperatur von 60-120 C,
zweckmäßig bei 95-105 0C erwärmt.
Eine dünnschichtchromatographische Verfolgung der Desoximierungsreaktion hat gezeigt, daß bei der Anwendung
eines Z-Isomers der allgemeinen Formel Ha-zuerst das thermodynamsich
stabilere Ε-Isomer der allgemeinen Formel Hb gebildet wird. In der ersten Stufe findet also eine
Transisomerisierung statt und die Desoximierung erfolgt eigentlich durch das Oximderivat der allgemeinen Formel
Hb.
Die partielle Reduktion der Verbindungen der allgemeinen
Formel I, in welchen A und T zusammen eine Oxogruppe bilden, kann nach beliebigen geeigneten Methoden
durchgeführt werden, welche zur partiellen Reduktion der Oxogruppe in Stellung 15 zu einer alkoholischen
Hydroxygruppe unter Beibehaltung der Oxogruppe in Stellung
14· geeignet sind. Zu diesem Zweck können als Reduktionsmittel Alkalimetallborhydride (z. B. Natriumborhydrid oder
Natriumcyanoborhydrid) eingesetzt werden. Die partielle Reduktion kann vorteilhaft in einem inerten Lösungsmittel
(h. B. 1-6 Kohlenstoffatome enthaltende Alkanole, wie
Methanol oder Gemische von 1-6 Kohlenstoffatome enthaltenden Alkoholen und halogenierten aliphatischen Kohlenwasserstoffen)
durchgeführt werden. Die partielle Reduktion kann vorzugsweise bei niedriger Temperatur - zweckmäßig zwischen
O und 5 0C - vollzogen werden.
Die erfindungsgemäß hergestellten Verbindungen der allgemeinen Formel I, in welchen A eine Hydroxygruppe und
Y ein Wasserstoff atom bedeuten können, weisen c(,ß und/oder
ß,cC Konfiguration auf eine. Ein erhaltenes Gemisch der 15-Epimere kann erwünschtenfalls in an sich bekannter
Weise in die Epimere aufgespaltet werden (z. B. mit Hilfe von Säuiencnromatographie oder Dünnschichtchromatographie).
Ein erhaltenes 15-Epimer kann in an sich bekannter Weise
in das andere 15-Epimer überführt werden.
Die Konfiguration zwischen dem Wasserstoffatom in Stellung 3 und der Gruppe R in den Ausgangsstoffen der
allgemeinen Formel Ha und/oder lib bleibt bei dem erfindungsgemäßen
Verfahren unverändert. Die Konfiguration zwischen dem Wasserstoffatom in Stellung 3 und der Gruppe
R im Endprodukt der allgemeinen Formel I ist mit derselben in den Ausgangsstoffen der allgemeinen Formel Ha und/oder
lib identisch.
Zur Bildung der Säureadditionssalze der Verbindungen der allgemeinen Formel I können die folgenden Sauren
verwendet werden: Mineralsäuren (z. B. Halogenwasserstoffe, wie Salzsäure oder Bromwasserstoff, Schwefelsäure,
Phosphorsäure, Salpetersäure oder Perhalogensäuren, wie Perchlorsäure
usw.), organische Carbonsäuren (ζ. Β. Ameisensäure, Essigsäure, Propionsäure, Glykolsäure, Maleinsäure,

Hydroxymaleinsäure, Fumarsäure, Bernsteinsäure, Weinsäure,
Ascorbinsäure, Citronensäure, Apfelsäure, Salicylsäure, Milchsäure, Zimtsäure, Benzoesäure, Phenylessigsäure,
p-Aminobenzoesäure, p-Hydroxybenzoesäure, p-Aminosalicylsäure
usw.)» Alkylsulphonsäuren (z, B. Methansulphonsäure,
Ethansulphonsäure usw.); cycloaliphatische Sulphonsäuren
(z. B. Cyclohexylsulphonsäure); Arylsulphonsäuren (z. B.
p-Toluolsulphonsäure, Naphthylsulphonsäure, Sulphanylsäure
usw.); Aminosäuren (z. B. Asparaginsäure, Glutaminsäure, N-Acetylasparaginsäure, N-AcetyIglutarsäure
usw.) .
Die Salzbildung kann in einem inerten organischen Lösungsmittel (z. B. 1-6 Kohlenstoffatome enthaltende aliphatische
Alkohole) durchgeführt werden. Man kann z. B.
so verfahren, daß man die razemische oder optisch aktive
Verbindung der allgemeinen Formel I in einem obigen Lösungsmittel löst und den pH-Wert der Lösung durch Zugabe
der entsprechenden Säure oder einer Lösung der Säure in einem obigen Lösungsmittel schwach sauer einstellt.(z. B.
pH 5-6)· Das ausgeschiedene Säureadditionssalz kann aus
dem Reaktionsgemisch nach geeigneten Methoden (z. B. Filtrieren) isoliert werden.
Eine erhaltene razemische Verbindung der allgemeinen Formel I kann in an sich bekannter Weise in die
optisch aktiven Antipode aufgespaltet werden. Die optisch aktiven Verbindungen der allgemeinen Formel I können jedoch
auch durch Anwendung von optisch aktiven Ausgangsstoffen der allgemeinen Formel Ha und/oder lib hergestellt
werden.
Die razemisehen Verbindungen der allgemeinen Formel
I werden zweckmäßig durch Anwendung von razemischen Ausgangsstoffen der allgemeinen Formel Ha und/oder Hb,
und die optisch aktiven Verbindungen der allgemeinen Formel I werden zweckmäßig unter Anwendung von optisch
aktiven Ausgangsstoffen der allgemeinen Formel Ha und/oder lib hergestellt.
Die nach dem erfindungsgemäßen Verfahren erhaltenen razemischen oder optisch aktiven Verbindungen der allgemeinen
Formel I und deren Säureadditionssalze können gegebenenfalls gereinigt werden (z. B. durch Umkristallisierung)
. Das zur Umkristallisierung verwendete Lösungsmittel wird auf Grund der Lösbarkeit und Kristallisierungseigenschaften der zu reinigenden Verbindimg gewählt.
Die erfindungsgemäßen Verbindungen der allgemeinen Formel I besitzen wertvolle pharmazeutische Eigenschaften.
Einige Vertreter dieser Verbindungsgruppe haben antiallergische Wirkung, wobei andere eine antidepressive,
magensekretionshemmende und antikonvulsive Wirkung ausüben. Gegenstand der Erfindung sind weiterhin pharmazeutische
Präparate, welche als Wirkstoff eine Verbindung der allgemeinen Formel I oder ein Säureadditionssalz davon
und inerte, nicht-toxische, zur parenteralen oder enteralen Verabreichung geeignete, übliche, feste oder flüssige
Träger und/oder Hilfsstoffe enthalten.
Als Träger können z. B. Wasser, Gelatin, Laktose, Milchzucker, Stärke, Peptin, Magnesiumstearat, Stearinsäure,
pflanzliche öle (z. B. Erdnußöl, Olivenöl usw.) Verwendung finden. Die pharmazeutischen Präparate können in
in der Pharmazie üblichen Formen hergestellt werden. Die festen Präparate können z. B. als eckige oder runde
Tabletten, Kapseln (wie Eartgelatinkapseln) , Pilulen, Suppositorien und die flüssigen Präparate als ölige oder
wäßrige Lösungen, Suspensionen, Emulsionen, Sirupe, Weichgelatinkapseln, injizierbare Ölige oder wäßrige Lösungen
oder Suspensionen usw. vorliegen. Die Menge des festen Trägers kann zwischen breiten Grenzen variiert werden (z.B.
zwischen 25 mg und 1 g) . Die Präparate können gegebenenfalls
übliche pharmazeutische Hilfsstoffe (z. B. Konservierungs-,
Stabilisierungs-, Netz-, Emulgierungsmittel, Salze zur Einstellung des osmotischen Druckes, Puffer,
Geschmacksmittel, Aromastoffe usw.) enthalten. Die Präparate können gegebenenfalls auch weitere pharmazeutisch
wertvolle bekannte Verbindungen enthalten. Die Dosierungseinheiten
entsprechen der gewünschten Verabreichungsart.
Die pharmazeutischen Präparate werden nach an sich bekannten Methoden der pharmazeutischen Industrie hergestellt
(z. B. Sieben, Vermischung, Granulieren, Verpressung
oder Auflösung der Komponenten) . Die Präparate können üblichen Maßnahmen der pharmazeutischen Industrie
unterworfen werden (z. B. Sterilisierung).
Weitere Einzelheiten des erfindungsgemäßen Verfahrens
sind den nachstehenden Beispielen zu entnehmen, ohne den Schutzumfang auf diese Beispiele einzuschränken.
Herstellung; von (-)-14,15-Dioxo-eburaan(3 «* %16*· )
2,0 g (6,2 Millimol) (+) -W-Oxo-^-hydroxyimino-
-eburnan(3 «Λ,ΐδιχ:) werden in 12 ml einer I5 # wäßrigen
Salζsäurelösung auf einem Wasserbad 6Ο-9Ο Minuten lang erwärmt.
Das Reaktionsgemisch wird abgekühlt und unter Eiskühlung durch Zugabe einer konzentrierten wäßrigen Ammoniaklösung
auf pH 9 alkalisiert. Die ausgeschiedene organische Substanz wird dreimal mit je 30 ml Dichlormethan extrahiert.
Die vereinigten organischen Extrakte werden über festem wasserfreiem Magnesiumsulphat getrocknet, filtriert
und unter vermindertem Druck eingeengt. Das zurückgeblie-

bene Öl wird aus 4 ml Äthanol umkristallisiert. Bs werden
1,50 g der im Titel genannten Verbindung erhalten. Ausbeute: 78,5 %-
Nach Dünnschichtchromatographie ist der R--Wert
des Endproduktes größer als derselbe des Ausgangsstoffes (KG-G, Benzol:Methanol=14:3). P: 161-162 0C
(Äthanol).

Herstellung von (-) -14-0xo-15 ^.-hydroxy-eburnan-
4,00 g (13 Millimol) des nach Beispiel 1 hergestellten (-)-14;15-oxo-eburaans werden in 40 ml wasserfreiem
Methanol unter Erwärmen gelost, worauf der Lösung unter Eiskühlung bei 0 0C in mehreren Portionen 240 mg
(6,5 Millimole) Natriumborhydrid zugegeben werden.
Die Reaktion wird durch Dünnschichtchromatographie
verfolgt. Der Rf Wert des Ausgangsstoffes ist größer
als derselbe des Endproduktes (KG-G, Dichlormethan:Methanol=
= 20:1).
Nach Beendigung der Reduktion wird das über-.flüssige
Reduktionsmittel unter Eiskühlung mit Eisessig zersetzt und das Lösungsmittel unter vermindertem Druck
entfernt. Der Rückstand wird in 15 ml Dichlormethan gelöst,
mit 10 ml einer wäßrigen 10 #-igen Natriumcarbonat lösung
alkalisch gemacht und extrahiert. Die abgetrennte wäßrige Phase wird zweimal mit je 10 ml Dichlormethan
extrahiert. Die organischen Phasen werden vereinigt, über wasserfreiem Magnesiumsulphat getrocknet, filtriert und unter
vermindertem Druck eingeengt. Das zurückgebliebene öl (4»b S) wird m 8 ml Methanol gelöst und der pH Wert der
Lösung durch Zugabe von salzsaurem Methanol auf 2 eingestellt. Es werden 3j83 g der im Titel genannten Verbindung
in Form des kristallinen Hydrochlorids erhalten. Ausbeute: 85,5 #. F: 255 0C (Zersetzung).

R eine 1-6 Kohlenstoffatome enthaltende Alkylgruppe ist; A eine Hydroxygruppe undT eins Wasserstoffatom bedeuten, oder A und Y zusammen eine Oxogruppe bilden; wobei das Vasserstoffatom in Stellung 3 und die Gruppe R eine oc ,cc - und/oder ß,ß- bzw. -/,ß und/oder β ,oc und die Gruppen A und Y eine oc,ß und/oder β , χ Konfiguration haben)
und Säureadditionssalze' davon. - 2. Pharmazeutische Präparate, dadurch gekennzeichnet , daß diese als Wirkstoff ein razemisches oder optisch aktives Eburnan-Derivat der allgemeinen Formel I(D(worinß eine 1-6 Kohlenstoffatome enthaltende Alkylgruppe ist; A eine Hydroxygruppe und
T ein Wasserstoffatom bedeuten, oder
A und Y zusammen eine Oxogruppe bilden; wobei
das Wasserstoffatom in Stellung 3 und die Gruppe R eineor ,*<·- und/oder ß,ß- bzw. <x,ß und/oder 2,W1C und die Gruppen A und Y eine Λ,β und/oder β,Α Konfiguration haben)
oder ein pharmazeutisch geeignetes Säureadditionssalz davon und geeignete, inerte, übliche pharmazeutische Träger und/oder Hilfsstoffe enthalten. - 3. Verfahren zur Her stellring von razemischen und optisch aktiven Eburnan-Derivaten der allgemeinen Formel I(D(worinR eine 1-6 Kohlenstoffatome enthaltende Alkylgruppe ist; A eine Hydroxygruppe und
Y ein Wasserstoffatom bedeuten, oder A und Y zusammen eine Oxogruppe bilden; wobei das Wasserstoffatom in Stellung 3 und die Gruppe R eine o<, ·Λ - und/oder ß,ß- bzw. ·λ,β und/oder ß,.A und die Gruppen A und Y eine Λ,β und/oder ß,oC Konfiguration haben)und Säureadditionssalzen davon,dadurch gekennzeichnet , daß man ein neues razemisches oder optisch aktives Aburnanoxim-Derivat der allgemeinen Formel Ha(Ilä)und/oder(lib)(worin die Bedeutung von R und die Konfiguration dieser Gruppe zum Wasserstoffatom in Stellung 3 wie "bei der allgemeinen Formel I angegeben sind) oder ein Säureadditionssalz davon desoximiert und erwünschtenfalls mit einer Säure behandelt und/oder erwünschtenfalls das erhaltene razemische oder optisch aktive neue Eburnan-Derivat der allgemeinen Formel I (in welchem A und Y zusammen eine Oxo-Gruppe bilden und die Bedeutung der Gruppe R und die Konfiguration dieser Gruppe zum Wasserstoffatom in Stellung 3 wie bei der allgemeinen Formel I angegeben ist) oder ein Säureadditionssalz davon einer partiellen Reduktion unterwirft und erwünschtenfalls ein 15-Epimerengemisch des erhaltenen neuen Eburnan-Derivats der allgemeinen Formel I (worin A eine Hydroxylgruppe und Y ein Wasserstoffatom bedeuten und die Konfiguration des Wasserstoffatoms in Stellung 3 und der R Gruppe wie oben angegeben ist) in die Epimere aufspaltet bzw. ein erhaltenes 15-Epimer in das andere 15-Epimer überführt und/oder erwünschtenfalls ein erhaltenes razemisches Eburaanderivat der allgemeinen Formel I in die optisch aktiven Antipode aufspaltet und/oder erwünschtenfalls ein erhaltenes razemisches oder optisch aktives Eburnan-Derivat derallgemeinen Formel I oder gegebenenfalls dessen 15-Epimere mit einer Säure behandelt. - 4. Verfahren nach Anspruch 3, dadurch gekennzeichnet , daß man die Desoximierung hydrolytisch, vorteilhaft mit einer wäßrigen Mineralsäurelösung, durchführt.
- 5. Verfahren nach Anspruch 2, dadurch gekennzeichnet , daß man die partielle Reduktion mit einem Alkalimetallborhydrid durchführt.
- 6. Verfahren zur Herstellung von pharmazeutischen Präparaten, dadurch gekennzeichnet , daß man eine razemische oder optisch aktive Verbindung der allgemeinen Formel I(DA" T(worinR eine 1-6 Kohlenstoffatome enthaltende Alkylgruppe ist; A eine Hydroxygruppe und '
T ein Wasserstoffatom bedeuten, oder Y und Y zusammen eine Oxogruppe bilden; wobei das Vasserstoffatom in Stellung 3 und die Gruppe R eine oC,<*.- und/oder ß,ß- bzw. of,β und/oder β,-α und die Gruppen A und Y eine ^, β und/oder ß,;\ Konfiguration haben), oder ein pharmazeutisch geeignetes Säureadditionssalζ davon mit geeigneten, inerten, üblichen pharmazeutischen Trägern und/oder Hilfsstoffen vermischt.
