Description
Gegenstand der vorliegenden Erfindung sind neue Verbindungen der in Anspruch 1 angegebenen Formel (III) und ein Verfahren zur Herstellung dieser Verbindungen nach einem Verfahren mit den in Anspruch 2 angegebenen Merkmalen.
Die neuen Verbindungen der Formel (III) eignen sich zur Durchführung des im Patent Nr. 650 265 der Anmelderin beschriebenen neuen und verbesserten Verfahrens zur Herstellung von überwiegend neuen threo-3- Amino-2-hydroxybut-anoylaminoessigsäuren und Estern hiervon, dementsprechend sind die neuen Verbindungen (III) erhältlich durch:

Die Erfindung betrifft neue threo-3-geschützte Amino-2-hydroxy-4-oxobutansäuren und deren Herstellung. Die neuen Verbindungen eignen sich unter anderem zur Herstellung von pharmazeutisch brauchbaren threo-3-Amino-2-hydroxybut-anoylaminoessigsäuren, die zum Teil aus den US-PS Nrn. 55 4 029 547 und 4 052 449 sowie den GB-PS 1 510 477 und 1 540 019 bekannt oder aber neu sind.
Die (2S,3R)-3-Amino-2-hydroxy-4- phenylbutanoylami-noessigsäure (im folgenden auch kurz Bestatin genannt) ist z.B. eine besonders wenig toxische Substanz, die von H. Ume- 60 zawa et al aus Kulturfiltraten von Streptomyces olivoreticuli als Aminopeptidase B-Inhibitor isoliert worden ist (erwähnt in US-PS 4 029 547); diese Substanz zeigt eine zunehmende Wirkung auf Immunoreaktionen, wie verzögerte Hyperästhesie, aktiviert die in-vivo-Abwehrmechanismen und zeigt inhi- 65 bitive Wirkungen auf Krebs (siehe JP-OS Nr. 117 435/1977); es ist zu erwarten, dass diese Verbindung als Pharmazeutikum brauchbar ist.

Im Ausgangsmaterial der Formel (I) kann R, eine Naph-thylgruppe, z.B. 1-Naphthyl oder 2-Naphthyl, oder eine Gruppe der Formel sein, in der R6 und R7 gleich oder verschieden sind.
R6 und R7 können einzeln Wasserstoff, Halogen, wie Chlor, Brom und Fluor, Amino oder geschütztes Amino, und zwar Acylamino, Niederalkoxycarbonylamino, gegebenenfalls substituiertes Benzyloxycarbonylamino, Carbamoyl-amino, Hydroxy, Acyloxy, Niederalkoxy mit 1 bis 6 C-Ato-men, z.B. Methoxy, Éthoxy, Propoxy, Butoxy, Pentyloxy und Hexyloxy, oder Niederalkyl mit 1 bis 6 C-Atomen, z.B. Methyl, Ethyl, Propyl, Butyl, Pentyl und Hexyl, darstellen. Die Niederalkoxy- oder Niederalkylgruppe kann verzweigt sein.
Die Gruppe der Formel umfasst z.B. Phenyl, Chlorphenyl, Dichlorphenyl, Fluor-phenyl, Aminophenyl, Hydroxyphenyl, Methoxyphenyl, Ethoxyphenyl, n-Propoxyphenyl, iso-Propoxyphenyl, n-But-oxyphenyl, iso-Butoxyphenyl, sec-Butoxyphenyl, Methyl-phenyl, n-Propylphenyl, iso-Propylphenyl, n-Butylphenyl, iso-Butylphenyl, sec-Butylphenyl, Biphenyl, Dihydroxy-phenyl, Dimethoxyphenyl und Hydroxymethoxyphenyl. Substituenten an den Benzolkernen können in allen Positionen stehen, d.h. in o-, m- oder p-Position; o- und m-Positio-nen, p- und o-Positionen oder p- und m-Positionen.
Die Schutzgruppe am geschützten Amino R2 ist z.B. ein Acyl, wie Formyl, ein Niederalkylcarbonyl, das Substituenten, wie Halogen, tragen kann, welche die Reaktion nicht stören, z.B. Acetyl, Chloracetyl, Dichloracetyl, Bromacetyl, 2-Chlorpropionyl und 2-Brompropionyl, oder Benzoyl, das Substituenten, wie Niederalkoxy, Niederalkyl oder Halogen am Phenylring tragen, welche die Reaktion nicht stören; Niederalkyloxycarbonyl, das nicht störende Substituenten, wie Halogen, tragen kann, z.B. Methoxycarbonyl, Ethoxy-carbonyl, i-Propyloxycarbonyl, i-Butyloxycarbonyl, t-Butyl-oxycarbonyl, t-Amyloxycarbonyl und 2,2,2-Trichlorethoxy-carbonyl; Cycloalkyloxycarbonyl, z.B. Cyclopentyloxycar-bonyl und Cyclohexyloxycarbonyl; Benzyloxycarbonyl, das nicht störende Substituenten, wie Niederalkoxy, Niederalkyl und Halogen am Phenylring tragen kann, z.B. Ethylcarbamoyl, Propylcarbamoyl und Butylcarbamoyl sowie Phenylcarbamoyl und Phthalyl. Das Alkyl und Phenyl der Phthalyl- oder Carbamoylgruppe können nicht störende Substituenten tragen, d.h. solche, die die Reaktion nicht stören.
Die oben genannten Schutzgruppen können auch zum Schutz von R6 und R7 dienen, wenn diese Amino- oder Hydr-oxygruppen sind.
Zu den für die Herstellung der neuen Verbindungen (III) geeigneten N-geschützten 2-Oxoethylaminen der Formel (I) gehören auch viele neue Verbindungen, die sich analog wie die bekannten Verbindungen aus einem Methylketon der Formel (VIII)

in der Ri die obige Bedeutung hat, durch Bromieren zum entsprechenden Brommethylketon, dessen Umsetzung mit Hexa-methylentetramin, Hydrolysieren zum entsprechenden 2-Oxoethylamin und folgendem Schutz von dessen Amino-gruppen durch Umsetzung mit geeignetem Acylierungsmittel synthetisieren lassen.

N-[2-Oxo-2-(4-isopropylphenyl)-ethyl]-benzamid (F 115 bis 117 °C) (3,38 g, 12,0 mM), 2,82 g (33,6 mM) Natriumbi-carbonat und 2,21 g (24,0 mM) Glyoxylsäuremonohydrat wurden in einer Mischung aus 20 ml Methanol, 10 ml Ethanol und 2 ml Wasser gelöst. Die Lösung wurde bei 50 bis 60 °C über Nacht reagieren gelassen, worauf die Umsetzung vollständig war. Die Reaktionslösung wurde unter vermindertem Druck zur Trockenheit eingedampft, der Rückstand in Wasser gelöst und mit Ethylacetat gewaschen. Die wässrige Phase wurde abgetrennt und mit Salzsäure auf pH 1 bis 2 eingestellt. Das abgeschiedene ölige Produkt wurde zweimal mit Ethylacetat extrahiert, die Extrakte vereinigt, mit Wasser gewaschen und über wasserfreiem Natriumsulfat getrocknet. Das Natriumsulfat wurde ausfiltriert und das Filtrat unter vermindertem Druck konzentriert. Herstellung von threo-(2RS)-3-Acetylamino-2-hydroxy -4- oxo-4-(4-isopropoxyphenyl) -butansäure: N-[2-Oxo-2-(4-isopropoxyphenyl)-ethyl]-acetamid (F 92 bis 95 °C) (1,90 g, 8,05 mM), 1,89 g (22,5 mM) Natriumbicarbo-natund 1,33 (14,5 mM) Glyoxylsäuremonohydrat wurden in 30 ml Methanol gelöst. Die Lösung wurde bei 50 bis 60 °C reagieren gelassen und die Umsetzung war nach 61/2 Std. vollständig. Die Reaktionslösung wurde unter vermindertem Druck zur Trockenheit eingedampft und der Rückstand in 30 ml Wasser gelöst. Die unlöslichen Anteile wurden mit Ethylacetat abgetrennt und die wässrige Phase mit verdünnter Salzsäure auf pH 1 bis 2 eingestellt. Das ausgeschiedene ölige Produkt wurde zweimal mit 30 ml Ethylacetat extrahiert. Die Extrakte wurden vereinigt und über wasserfreiem Natriumsulfat getrocknet. Das Natriumsulfat wurde ausfiltriert und das Filtrat unter vermindertem Druck konzentriert. Der Rückstand wurde mit 20 ml Diethyläther verrieben und die ausgefällten Kristalle durch Filtration gesammelt, mit Diethyläther gewaschen und im Vakuum getrocknet. Das Zielprodukt wurde in einer Ausbeute von 1,47 g (58,8%) erhalten; F 157 bis 158 C (Zersetzung).

Beispiel 17
Herstellung von threo-(2RS)-3-Benzoylamino-2-hydroxy -4- oxo-4-phenyl-butansäure:
N-(2-Oxo-2-phenylethyl)-benzamid (16,7 g, 0,07 Mol) und 13,0 g(0,155 Mol) Natriumbicarbonat wurden in einer Mischung aus 3,75 g (0,13 Mol) einer 25%igen wässrigen Glyoxylsäurelösung, 100 ml Wasser und 250 ml Methanol gelöst. Die Lösung wurde über Nacht bei 50 bis 60 °C reagieren gelassen.
Nach Ausfiltrieren von unlöslichen Anteilen wurde das 30 Filtrat der Reaktionslösung unter vermindertem Druck zur Abdestillation von Methanol konzentriert. Die konzentrierte Lösung wurde mit verdünnter Salzsäure zur Einstellung des pH-Wertes auf 1 bis 2 versetzt. Die abgeschiedenen Kristalle wurden ausfiltriert, mit Wasser gewaschen und im Vakuum 35 über Phosphorpentoxid getrocknet. Das kristalline Rohprodukt (17,6 g) wurde aus Ethylacetat umkristallisiert und lieferte das reine Zielprodukt in einer Ausbeute von 13,5 g (61,8%), F 174 bis 176 °C (Zersetzung).

Herstellung von threo-(2RS)-2-Hydroxy-4-oxo-4-phenyl so -3- phthalimino-butansäure:
N-(2-Oxo-2-phenylethyl)-phthalimid (10,0 g, 0,0377 Mol) und 9,0 g (0,107 Mol) Natriumbicarbonat wurden in einer Mischung aus 20,1 g (0,068 Mol) einer 25%igen wässrigen Glyoxylsäurelösung und 20 ml Ethanol gelöst. Die Lösung 55 wurde 24 Std. bei 50 bis 60 C reagieren gelassen.
Die Reaktionslösung wurde unter vermindertem Druck zur Abdestillation des Ethanols konzentriert. Die konzentrierte Lösung wurde in einer Mischung aus 100 ml Ethylacetat und 50 ml einer 5%igen wässrigen Natriumbicarbonatlö-60 sung aufgenommen und mit dieser geschüttelt. Die wässrige Phase wurde abgetrennt und mit verdünnter Salzsäure auf pH 1 bis 2 eingestellt. Die abgeschiedenen Kristalle wurden ausfiltriert, mit Wasser gewaschen und im Vakuum über Phosphorpentoxid getrocknet. Es wurde das Zielprodukt in einer 65 Ausbeute von 1,97 g (15,4%), F 168 bis 170 C (Zersetzung) erhalten.

Verfahren zur Herstellung einer threo-3-geschützten Amino-2-hydroxy-4-oxobutansäure oder deren Ester der Formel (III) gemäss Anspruch 1, gekennzeichnet durch Umsetzung eines N-geschützten 2-Oxoethylamins der Formel (I)25Es ist auch bekannt, dass verschiedene 3-Amino-2-hydr-oxybutanoylaminoessigsäuren ausser Bestatin eine gleiche oder stärkere Inhibierungsaktivität für Aminopeptidase B wie Bestatin haben; Synthesen für solche Verbindungen sind in der GB-PS 1 510 477 beschrieben. Das dort beschriebene Verfahren liefert zwar beispielsweise (2S,3R)-3-Amino-2-hydr-oxybutansäure als Zwischenverbindung für Bestatin, hat aber verschiedene, weiter unten beschriebene Nachteile.Obwohl sich dieses bekannte Verfahren im Labormassstab ohne Schwierigkeiten durchführen lässt, ist es für die grosstechnische Produktion ungeeignet, da es grosse Mengen des teuren (R)-Phenylalanins, das keine natürliche Aminosäure ist, den Einsatz von leicht entzündlichem Lithiumaluminiumhydrid für die Reduktion des Pyrazolides und die Verwendung von hochgiftigem Cyanid in der Synthese des Cyan-hydrins erfordert.
