Description
Foreliggende oppfinnelse angår fremstilling av cis 1,3-oksatiolan.
Nærmere bestemt angår oppfinnelsen en fremgangsmåte for fremstilling av et terapeutisk aktivt, racemisk cis 1,3-oksatiolan.

Foreliggende oppfinnelse finner anvendelse ved fremstilling av antivirale nukleoside analoger, særlig BCH-189 (2<1>, 3'-dideoksy-3'-tia-cytidin) og mer spesielt ved den selektive syntese av B-isomeren av BCH-189 og relaterte forbindelser så vel som den selektive syntese av enantiomer-anriket BCH-189 og relaterte forbindelser.
Foreliggende søknad er avdelt fra NO-P 923014 som er rettet mot en fremgangsmåte for fremstilling av et enantiomert anriket 1,3-oksatiolan-nukleosid og der denne fremgangsmåte karakteriseres ved at den omfatter anrikning av 1,3-oksatiolan-nukleosid med en optisk isomer i en enantioselektiv, enzymatisk reaksjon.
Nærmere bestemt angår P923014 den selektive syntes av 6-isomeren av BCH-189 (2', 3'-dideoksy-3'-tia-cytidin, og relaterte forbindelser, såvel som den selektive syntese av enantiomeranriket BCH-189 og relaterte forbindelser.
I 1981 begynte dokumenteringen av sykdommen som ble kjent som ervervet immunmangelsyndrom, "Acquired Immune Deficiency Syndrome" eller AIDS, så vel som dennes forløper "AIDS- relatert kompleks" (ARC). I 1983 ble årsaken til sykdommen AIDS henført til en virus kalt human immunodefektvirustype 1 (HIV-1). Vanligvis ville en person som var infisert med virusen til slutt utvikle AIDS, i alle kjente tilfeller av AIDS er det endelig resultat døden.
Sykdommen AIDS er sluttresultatet av en HIV-1-virus som følger sin egne komplekse livscyklus. Virion-livscyklusen begynner med at virionet fester seg selv til vertshuman-T4- lymfocyttimmuncellen via binding av et glycoprotein på overflaten av virionets beskyttende belegg med CD4-glycoproteinet på lymfocyttcellen. Når den først var festet, kastet virionet sitt glycoproteinbelegg, penetrerte inn i membranet i vertscellen og avdekket sitt RNA. Virionenzymet, revers transkriptase, retter prosessen av transkribering av RNA til enkeltstrenget DNA. Det virale RNA brytes ned og en andre DNA-streng dannes. Det nu dobbeltstrengede DNA integreres inn i den humane celles gener og disse gener benyttes for cellereproduksjon.
På dette punkt utfører den humane celle sin reproduktive prosess ved å benytte sin egen RNA-polymerase for å transkribere det integrerte DNA til viralt RNA. Dette virale RNA translateres til glycoproteiner, strukturelle proteiner og virale enzymer som går sammen med det virale RNA intakte. Når vertscellen er ferdig med det reproduktive trinn, fortsetter en ny virioncelle, nu en T4-lymfocytt, virksomheten. Antallet HIV-1-virusceller vokser således mens antallet T4-lymfocytter synker.
Den typisk humanimmunsystemrespons, avlivning av det invaderende virion, vanskeliggjøres på grunn av at en stor del av virionets livscyklus forbringes i en latent tilstand i den immune celle. I tillegg er den virale reverse transkriptase, enzymet som benyttes for dannelse av en ny virioncelle, ikke meget spesifikk, og forårsaker transkrip-sjonsfeil som resulterer i kontinuerlig endrede glycoproteiner på overflaten av det virale beskyttende belegg. Denne mangel på spesifisitet reduseres immunsystemets effektivitet fordi antistoffer som spesifikt produseres mot et glycoprotein kan være ubrukelig mot et annet hvorved man reduserer antallet antistoffer som er tilgjengelige for å bekjempe viruset. Viruset fortsetter å vokse mens immunresponssystemet fortsetter å svekkes. Til slutt hersker HIV generelt fritt over legemets immunsystem og tillater at opportunistiske infeksjoner kan sette inn og medfører, hvis ikke antivirale midler og/eller immunomodulatorer anvendes, død som resultat.
Det er tre kritiske punkter i vimsens livscyklus som er identifisert som mål for antivirale medikamenter:
(1) den første festing av virionet til T4-lymfocytt- eller makrofagasetet,
(2) transkripsjonen av viral RNA til viral DNA, og
(3) sammensetningen av den nye virioncelle under reproduksjon.
Inhiberingen av virusen på det andre kritiske punkt, virale RNA —► virale DNA-transkripsjonsprosessen, har stått for mesteparten av de terapier som har vært benyttet for behandling av AIDS. Denne transkripsjon må inntre for at virionet skal reprodusere på grunn av at virionets gener er kodet i RNA; vertscellen leser kun DNA. Ved innføring av medikamenter som blokkerer den reverse transkriptase fra å komplettere dannelsen av viral DNA, kan HIV-1-replikering stanses.
Nukleosidanaloger som 3'-azido-3'-deoksythymidin (AZT), 2',3'-dideoksycytidin (DOC), 2',3-dideoksythymidinen (D4T), 2',3'-dideoksyinsoin (DDI) og forskjellige fluorderivater av disse nukleosider er relativt effektive med henblikk på å oppholde HlV-replikeringen på revers transkriptase-trinnet. En annen lovende revers transkriptase-inhibitor er 2',3'- dideoksy-3'-tia-cytidin (BCH-189) som inneholder en oksatiolanring som erstatter sukkerdelen i nukleosidet.
AZT er et vellykket anti-HIV-medikament fordi det saboterer dannelsen av viral DNA i verts-T4-lymfocyttcellen. Når AZT går inn i cellen, aktiverer cellulære kinaser AZT ved fosforylering til AZT-trifosfat. AZT-trifosfat konkurrerer så med naturlige thymidinnukleosider for reseptorsetet av HIV- revers transkriptase-enzymet. Det naturlige nukleosid har to reaktive ender, den første for festing til det tidligere nukleosid og det andre for binding til det neste nukleosid. AZT-molekylet har kun den første reaktive ende, når det er i HIV-enzymsetet terminerer AZT-azidgruppen viral DNA-dannelse fordi azidet ikke kan lage 3',5'-fosfodiesteren med ribosedelen av det følgende nukleosid.
AZT's kliniske fordeler inkluderer øket levetid, redusert frekvens og alvor av opportunistiske infeksjoner og øket perifer CD4-lymfocytt-telling. Immunsorbentanalyser for viral p24, et antigen som benyttes for å spore HIV-1-aktivitet, viser en betydelig reduksjon ved bruken av AZT. Imidlertid må AZT-fordeler veies mot de alvorlig ugunstige reaksjoner av benmargssuppresjon, nausea, myalgi, insomnia, alvorlig hodepine, anemi, perifer neuropati og kramper. Videre opptrer disse ugunstige bivirkninger umiddelbart etter behandlingsbegynnelse, mens et minimum på seks ukers terapi er nødvendig for realisere AZT-fordeler.
Både DDC og D4T er potente inhibitorer for HIV-replikering med aktiviteter som er sammenlignbare (D4T) eller overlegne (DDC) den til AZT. Imidlertid konverteres både DDC og D4T til sine 5-trifosfater mindre effektivt enn deres naturlige analoger og de er motstandsdyktige mot deaminaser og fosforylaser. Klinisk er begge forbindelser toksiske. I dag benyttes DDI i forbindelse med AZT for behandling av AIDS. Imidlertid inkluderer DDI's bivirkninger sporadisk pankreatitt og perifer neuropati. Initialprøver på 3'-fluor-2',3'- dideoksythymidin viser at dens antivirale aktivitet er sammenlignbar med den til AZT.
Nylige prøver på BCH-189 har vist at den har anti-HIV- aktivitet tilsvarende AZT og DDC, men uten den celletoksisitet som forårsaker debiliteringsbivirkningene til AZT og DDC. En tilstrekkelig mengde BCH-189 er nødvendig for å tillate klinisk prøving og behandling ved bruk av medikamentet.
De vanligvis benyttede kjemiske tilnærmelser for syntetisering av nukleosider eller nukleosidanaloger kan klassifiseres i to generelle kategorier: (1) de som modifiserer intakte nukleosider ved å endre karbohydratet, basen eller begge, og (2) de som modifiserer karbohydratene og innarbeider basen, eller dennes syntetiske forløper, på et egnet trinn i syntesen.
Fordi BCH-189 substituerer et svovelatom for et karbonatom i karbohydratringen er den andre tilnærmelse mer gjennomførbar. Den viktigste faktor i denne sistnevnte strategi medfører avgivning av basen fra 13-flaten av karbohydratringen i glycosyleringsreaksjonen for kun 8-isomerene viser brukbar biologisk aktivitet.
Det er velkjent i denne teknikk at den stereoselektive innføring av baser til de anomere sentra av karbohydrater kan kontrolleres ved å kapitalisere på nabogruppedeltagelsen til en 2-substituent i karbohydratringen ("Chem. Ber.", 114:1234 (1981)). Imidlertid har BCH-189 og dennes analoger ingen 2-substituent og kan derfor ikke benytte denne prosedyre hvis det ikke i syntesen innarbeides ytterligere trinn for å innføre en funksjonell gruppe som både er rettende og disponerbar. Disse ytterligere trinn vil redusere syntesens totale effektivitet.
Det er også velkjent i denne teknikk at "betydelige mengder av uønskede a-nukleosider alltid dannes under syntesen av 2'-deoksyribosider" ("Chem. Ber.", 114:1234, 1244
(1981)). Videre lærer denne referanse at bruken av enkle Friedel- Crafts-katalysatorer som SnCl4 i nukleosidsynteser gir uønskede emulsjoner ved opparbeiding av reaksjonsblandingen, danner komplekse blandinger av a- og B-isomerene og fører til stabile a-komplekser mellom SnCl4 og de mer basisk silylerte heterocykler som silylert cytosin. Disse kompleksene fører til lengre reaksjonstider, lavere utbytter og gir de uønskede unaturlige N3-nukleosider. Således beskriver den kjente teknikk bruken av trimetylsilyltriflat eller trimetylsilylperklorat som katalysator under koblingen av pyrimidinbaser med en karbohydratring for å oppnå høye utbytter av de biologisk aktive B-isomerer. Imidlertid gir bruken av disse katalysatorer for å syntetisere BCH-189 eller BCH-189- analoger ikke fortrinnsvis B-isomeren; disse reaksjoner resulterer i et omtrentlig 50:50-forhold for isomerene.
Således foreligger det et behov for en effektiv syntetisk rute til BCH-189 og dennes analoger. Det forelå altså et behov for en stereoselektiv syntetisk rute til den biologisk aktive isomer av disse forbindelser, B-BCH-189 og relaterte B-analoger. Videre foreligger det et behov for en stereoselektiv syntetisk rute til enantiomert-anriket 6-BCH-189 fordi den andre enantiomer er inaktiv og derfor representerer en urenhet på 50%.
P923014, foreliggende søknads stamsøknad, er basert på oppdagelsen av en overraskende effektiv, syntetisk rute til rute til BCH-189 og forskjellige analoger av BCH-189 fra rimelige forløpere med muligheten for innføring av funksjonalitet efter behov. Denne syntetiske rute tillater den stereoselektive fremstilling av den biologisk aktive isomer av disse forbindelser, B-BCH-189 og relaterte forbindelser. Videre kan stereokjemien på nukleosid 4-posisjon kontrolleres for å gi enantiomer-anriket B-BCH-189 og dennes analoger.
Uttrykket "BCH-189-analoger" er ment å bety nukleosider som dannes fra pyrimidinbaser som er substituert i 5-posisjon som er koblet til substituerte 1,3-oksatiolaner.
Fremgangsmåten ifølge P923014 inkluderer ozonisering av en allyleter eller -ester med formelen CH2=CH-CH2-OR, der R er en beskyttende gruppe, for eksempel en alkyl-, silyl- eller acylgruppe, for derved å danne et glycoaldehyd med formelen OHC-CH2-OR; tilsetning av tioglycolsyre til glycoaldehydet for å danne laktonet med formelen 2-(R-oksy)- metyl-5-okso-1,3-oksatiolan; omdanning av laktonet til det tilsvarende karboksylat i 5-posisjon av oksatiolanringen; kobling av acetatet med en silylert pyrimidinbase i nærvær av SnCl4 for å danne B-isomeren av en 5'-(R-oksy)-2',3'-dideoksy-3'-tia-nukleosid-analog; og å erstatte den R-beskyttende gruppe med et hydrogen for å danne BCH-189 eller en analog av BCH-189.
Fremgangsmåten kan benyttes for å fremstille BCH-189 eller BCH- 189-analoger som er enantiomer-anriket i 4'-posisjon ved å selektere en egnet R-beskyttende gruppe for å tillate stereoselektiv seleksjon av et enzym. For eksempel kan den R-beskyttende gruppe velges slik at substituenten i 2-posisjon i oksatiolanlaktonet er butyryloksy for å tillate stereoselektiv enzymatisk hydrolyse med svineleveresterase. Det resulterende optisk aktive hydrolyserte lakton kan så omdannes til det tilsvarende diacetat og kobles med en silylerte pyrimidinbase som ovenfor.
Som nevnt innledningsvis angår foreliggende oppfinnelse en fremgangsmåte for fremstilling av et terapeutisk,aktivt, racemis cis 1,3-oksatiolan med formelen:

og denne fremgangsmåte karakteriseres ved at den omfatter omsetning av en forbindelse med formelen
der R* er acyl og R er alkyl, silyl eller acyl,
med en eventuelt beskyttet pyrimidinbase i nærvær av SnCl4.

Fremgangsmåten for fremstilling av BCH- 189 og BCH-189-analoger er angitt i figur 1. En allyleter eller -ester 1 ozoniseres for å gi et aldehyd 2 som omsettes med tioglycolsyre til et lakton 3. Laktonet 3 behandles med et reduksjonsmiddel, for eksempel diisobutylaluminiumhydrid (DIBAL), natrium-bis(2-metoksyetoksy)aluminiumhydrid (som kommersielt kan oppnås som en 3,4 molar oppløsning i toluen, kalt Rd-Al), og NaBH4, fulgt av et karboksylsyreanhydrid for å oppnå karboksylatet 4. Dette karboksylat kobles med en silylert pyrimidinbase i nærvær av en Lewis-syre som kan katalysere stereospesifikk kobling, for eksempel SnCl4 for derved å oppnå B-isomeren av det substituerte nukleosid 5 i et i det vesentlige 100:0-forhold B:cc-isomerer. Det substituerte nukleosid 5 debeskyttes for å gi BCH-189 eller BCH- 189-analogen6.
Denne prosedyre kan skreddersyes for å fremstille BCH-189 eller BCH-189-analoger som er enantiomer-anriket i 4'-posisjon ved å selektere en egnet R-beskyttende gruppe for å tillate stereoselektiv enzymatisk hydrolyse av 3 med et enzym som svineleveresterase, porcinpankreatisk lipase eller subtilisin eller andre enzymer som hydrolyserer 3 på stereoselektiv måte. Den resulterende optisk aktive forbindelse 3 kan omdannes til et enantiomer-anriket karboksylat 4 og kobles med en silylert pyrimidinbase som ovenfor for å gi enantiomer-anriket BCH-189 eller BCH-189-analoger.
Den beskyttende gruppe R i 1 kan selekteres til å gi beskyttelse for den tilsvarende alkohol inntil sluttrinnet i syntesen er utført (debeskyttelse av 5 for å oppnå 6). I tillegg kan den beskyttende gruppe selekteres hvis ønskelig, for å gi et ytterligere erkjennelsessete for et enzym som senere skal benyttes i enantioselektiv hydrolysereaksjon. En alkyl-beskyttende gruppe som her benyttet, betyr trifenylmetyl eller en alkylgruppe som har i det vesentlige de samme beskyttende egenskaper som trifenylmetyl. En silyl-beskyttende gruppe som uttrykket her benyttes, betyr en trisubstituert silylgruppe med formelen:
der Ri, R2 og R3 kan være laverealkyl som metyl, etyl og butyl, og der alkyl har 5 karbonatomer eller mindre; eller fenyl. Videre kan R\ være identisk med R2; videre kan R\, R2 og R3 alle være like. Eksempler på silyl-beskyttende grupper er, men er ikke begrenset til, trimetylsilyl og t-butyldifenylsilyl.
En acylgruppe slik uttrykket her benyttes for å beskrive en acyl-beskyttende gruppe (som i 1) eller for å beskrive et karboksylat (som i 4), er en gruppe med formelen:

En silylert pyrimidinbase slik uttrykket her benyttes, betyr en forbindelse med formelen: der X enten eller en trisubstituert silyloksy- eller en trisubstituert silylaminogruppe, Z er en trisubstituert silylgruppe og Y er som beskrevet ytterligere nedenfor. En trisubstituert silylgruppe som uttrykket her benyttes, betyr en gruppe med formelen: der R\, R2 og R3 kan være laverealkyl som metyl, etyl, butyl og alkyl har 5 karbonatomer eller mindre, eller feriyl. Videre kan R\ være lik R2; Ri, R2 og R3 kan alle være identiske. Eksempler på trisubstituerte silylgrupper er, men er ikke begrenset til, trimetylsilyl og t-butyldifenylsilyl.
Den silylerte pyrimidinbase substitueres så med en ønsket substituent, for eksempel F eller H.
Illustrerende eksempler på syntesen av BCH-189 eller BCH-189- analoger ifølge oppfinnelsen er gitt i figurene 2, 3 og 4 og den følgende beskrivelse.
Figur 2 viser syntesen av BCH-189 ut fra allylalkoholen 7. En NaH-oljesuspensjon (4,5 g, 60%, 110 mmol) ble vasket med 2 x 100 ml THF og det resulterende faststoffet ble suspendert i 300 ml THF. Suspensjonen ble avkjølt til 0°C, allylalkoholen 7 (6,8 ml, 100 mmol) ble tilsatt dråpevis og blandingen ble omrørt i 30 minutter ved 0°C. t-butyl-difenylsilylklorid (25,8 ml, 100,8 mmol) ble tilsatt dråpevis ved 0°C og reaksjonsblandingen ble omrørt i en time ved 0°C. Oppløsningen ble bråkjølt med 100 ml vann og ekstrahert med 2 x 200 ml dietyleter. De kombinerte ekstrakter ble vasket med vann, tørket over MgS04, filtrert, konsentrert og resten ble destillert under vakuum (90-100°C ved 0,5-0,6 mm Hg) hvorved man oppnådde en farveløs væske 8 (28 g, 94 mmol, 94%).

Silylallyleteren 8 (15,5 g, 52,3 mmol) ble oppløst i 400 ml CH2CI2 og ozonisert ved - 78°C. Etter ferdig ozonolyse ble DMS (15 ml, 204 mmol, 3,9 ekv.) tilsatt ved -78°C og blandingen ble oppvarmet til romtemperatur og omrørt over natten. Oppløsningen ble vasket med 2 x 100 ml vann, tørket over MgS04, filtrert, konsentrert og destillert under vakuum (100- 110°C ved 0,5-0,6 mm Hg) hvorved man oppnådde en farveløs væske 9 (15,0 g, 50,3 mmol, 96%).

Silylert glycoaldehyd 9 (15,0 g, 50,3 mmol) ble oppløst i 200 ml toluen og tioglycolalkohol (3,50 ml, 50,3 mmol) ble tilsatt på en gang. Oppløsningen ble kokt under tilbakeløp i 2 timer, mens det resulterende vann ble fjernet med en Dean- Stark-felle. Oppløsningen ble avkjølt til romtemperatur og vasket med mettet NaHC03-oppløsning og det vandige vaskevann ble ekstrahert med 2 x 200 ml dietyleter. De kombinerte ekstrakter ble vasket med 2 x 100 ml vann, tørket over MgS04, filtrert og konsentrert til en farveløs olje 10 (16,5 g, 44,3 mmol, 88%) som langsomt størknet under vakuum. Omkrystallisering fra heksan ga et hvitt faststoff 10 (15,8 g, 84%).

2-(t-butyl-difenylsilyloksy)-metyl-5-okso-l,2-oksatiolan 10 (5,0 g, 13,42 mmol) ble oppløst i 150 ml toluen og oppløsningen ble avkjølt til -78°C. Dibal-H-oppløsning (14 ml, 1,0M i heksan, 14 mmol) ble tilsatt dråpevis, mens den indre temperatur ble holdt under -70°C hele tiden. Etter ferdig tilsetning ble blandingen omrørt i 30 minutter ved - 78°C. Eddiksyreanhydrid (5 ml, 53 mmol) ble satt og blandingen ble oppvarmet til romtemperatur og omrørt over natten. 5 ml vann ble satt til blandingen og den resulterende blanding ble omrørt i en time ved romtemperatur. Blandingen ble fortynnet Ått Ula V«1^k
ved romtemperatur. Blandingen ble filtrert, konsentrert og resten flashkromatografert med 20% EtOAc i heksaner hvorved man oppnådde en farveløs væske 11 (3,60 g, 8,64 mmol, 64%) som var en 6:1- blanding av anomerer.

2-(t-butyl-difenylsilyloksy)-metyl-5-acetoksy-1,3-oksatiolan 11 (0,28 g, 0,67 mmol) ble oppløst i 20 ml 1,2-dikloretan og silylert cytosin 12 (0,20 g, 0,78 mmol) ble tilsatt på en gang ved romtemperatur. Blandingen ble omrørt i 10 minutter og det ble tilsatt SnCl4-oppløsning (0,80 ml, 1,0M-oppløsning i CH2C12, 0,80 mmol) dråpevis ved romtemperatur. Ytterligere cytosin 12 (0,10 g, 0,39 mmol) og 0,60 ml SnCl4-oppløsning ble tilsatt på samme måte en time senere. Efter ferdig reaksjon i løpet av 2 timer ble oppløsningen konsentrert og resten ble triturert med 2 ml trietylamin og underkastet flashkromatografi (først med ren EtOAc og deretter med 20% etanol i EtOAc) for å gi et gyldent faststoff 13 (100% 6-konfigurasjon) (0,25 g, 0,54 mmol, 80%).

Silyleteren 13 (0,23 g, 0,49 mmol) ble oppløst i 30 ml THF og dertil ble det satt n-Bu4NF-oppløsning (0,50 ml, 1,0M- oppløsning i THF, 0,50 mmol) dråpevis ved romtemperatur. Blandingen ble omrørt i en time og konsentrert under vakuum. Resten ble tatt opp i etanol:trietylamin (2:1) og underkastet flashkromatografi (først med EtOAc, deretter 20% etanol i EtOAc) hvorved man oppnådde et hvitt faststoff 14 i 100% anomer renhet (BCH-189; 0,11 g, 0,48 mmol, 98%), som ble ytterligere renset fra etanol:CHCl3 :heksaner.

BCH-189 og dennes analoger kan også syntetiseres ved kobling av et silylert uracilderivat med 11. Det silylerte uracilderivat 15 (1,80 g, 7,02 mmol) ble koblet med 11 (1,72 g, 4,13 mmol) i 50 ml 1,2-dikloretan i nærvær av 5,0 ml SnCi4 som beskrevet ovenfor i nærvær av cytosinderivatet 13. Reaksjonen var ferdig efter 5 timer. Flashkromatografi, først med 40% EtOAc i heksan og deretter EtOAc, ga et hvitt skum 16 (1,60 g, 3,43 mmol, 83%).

Uracilderivatet 16 kan omdannes til cytosinderivatet 13. Uracilderivatet 16 (0,20 g, 0,43 mmol) ble oppløst i pyridimdikloretan 2:10 på volumbasis og oppløsningen avkjølt til 0°C. Triflikanhydrid (72 ul, 0,43 mmol) ble tilsatt dråpevis ved 0°C og blandingen ble oppvarmet til romtemperatur og omrørt i en time. Ytterligere triflikanhydrid ble tilsatt og blandingen omrørt i en time. TLC viste ingen mobilitet med EtOAc. Reaksjonsblandingen ble så dekanylert inn i 30 ml av NH3-mettet metanoloppløsning og blandingen ble omrørt i 12 timer ved romtemperatur. Oppløsningen ble konsentrert og resten underkastet flashkromatografi hvorved man oppnådde et gyldent skum 13 (0,18 g, 0,39 mmol, 91%) som er identisk med den forbindelse som ble oppnådd fra cytosinkoblingsreaksj onen.
Figur 3 illustrerer syntesen av 5-metylcytidin- og thymidinderivatene av BCH-189. Acetat 11 (0,93 g, 2,33 mmol) i 50 ml 1,2-dikloretan ble omsatt med det silylerte thyminderivat 17 (1,0 g, 3,70 mmol) og 4,0 ml SnCi4-oppløsning på en måte tilsvarende det som er beskrevet for fremstilling av cytosinderivatet 13.

rhymidinderivatet 18 (0,20 g, 0,42 mmol) ble oppløst i pyridin:dikloretan 2:10 på 'olumbasis og oppløsningen avkjølt til 0°C. Dertil ble det satt triflikanhydrid (100 ul, 1,60 mmol), dråpevis ved 0°C, og blandingen ble under kontinuerlig omrøring tillatt ippvarming til romtemperatur. Efter oppnådd romtemperatur ble den omrørt i en time. rLC viste ingen mobilitet med EtOAc. Reaksjonsblandingen ble så dekanylert inn i 20 ni NH3-mettet metanoloppløsning og blandingen ble omrørt i 12 timer ved omtemperatur. Oppløsningen ble konsentrert og resten underkastet flashkromatografi ivorved man oppnådde et gyldent skum 19 (0,18 g, 0,38 mmol, 90%).

Figur 4 viser syntesen av enantiomer-anriket BCH-189 og analoger derav. Allylbutyrat 22 (19,0 g, 148 mmol) ble oppløst i CH2C12 (400 ml) og ozonisert ved -78°C. Efter ferdig ozonolyse ble dimetylsulfid (20 ml, 270 mmol, 1,8 ekv.) tilsatt ved -78°C og blandingen oppvarmet til romtemperatur og omrørt over natten. Oppløsningen ble vasket med 2 x 100 ml vann, tørket over MgS04, filtrert, konsentrert og destillert under vakuum (70-80°C ved 0,5-0,6 mm Hg) hvorved man oppnådde en farveløs væske 23 (17,0 g, 131 mmol, 88%).

Butyryloksyacetaldehyd 23 (15,0 g, 115 mmol) ble oppløst i 200 ml toluen og blandet med tioglycolsyre (8,0 ml, 115 mmol). Oppløsningen ble kokt under tilbakeløp i 5 timer mens det resulterende vann ble fjernet med en Dean-Stark-felle. Oppløsningen ble avkjølt til romtemperatur og overført til en 500 ml skilletrakt. Oppløsningen ble så vasket med mettet NaHC03 -oppløsning. Disse vandige vaskevæsker ble ekstrahert med 2 x 200 ml dietyleter for å gjenvinne råproduktet fra det vandige sjikt. Eterekstraktene ble satt til toluensjiktet og den resulterende blanding ble vasket med 2 x 100 ml vann, tørket over MgS04, filtrert, konsentrert og destillert under vakuum (70-80°C ved 0,5-0,6 mm Hg) hvorved man oppnådde en farveløs olje 24 (19 g, 93 mmol, 81%).

-CH20) J=3,2 og 12,2 Hz; 4,29 (1H, dd, -CH20) J=5,7 og 12,2 Hz; 3,72 (1H, d, -CH2S) J=16,2 Hz; 3,64 (1H, d, -CH2S); 2,34 (2H, t, -CH2CO) J=7,2Hz; 1,66 (2H, seks,-CH2); 0,95 (3H, t, CH3) J=7,2 Hz. IR (topp): 2980, 2960,2900,1780,1740,1460,1410,1390, 1350, 1300, 1290, 1260, 1220, 1170, 1110, 1080, 1070, 1000, 950, 910, 830, 820, 800, 760. 90 ml svineleveresteraseoppløsning ble tilsatt til 700 ml av pH 7-bufferoppløsning ved romtemperatur og blandingen omrørt heftig i 5 minutter. Butyratet 24 (2,8 g, 13,7 mmol) ble satt, alt på en gang, til esterase/buffer-oppløsningen og blandingen ble omrørt heftig ved romtemperatur i 2 timer. Reaksjonsblandingen ble helt i en skilletrakt. Reaksjonskolben ble vasket med 10 ml eter og vaskevæsken ble kombinert med reaksjonsblandingen i trakten. Den kombinerte blandingen ble ekstrahert med 3 x 100 ml heksan. De tre heksanekstrakter ble kombinert, tørket over MgS04, filtrert og konsentrert og man oppnådde det optisk aktive butyrat 24 (1,12 g, 5,48 mmol, 40%). Enantiomert overskudd ble bestemt ved et NMR-forsøk ved bruk av et tris[3-heptafluorpropyl-hydroksymetylen)-(+)- kamforat]europium(IH)derivat som kjemisk skiftreagens; denne prosedyre viste ca. 40% anrikning for en enantiomer. Det gjenværende vandige sjikt fra reaksjonen ble underkastet en kontinuerlig ekstrahering med CH2CI2 i 20 timer. Det organiske sjikt ble fjernet fra ekstraheringsapparaturen, tørket over MgSC>4, filtrert og konsentrert til tørr tilstand og man oppnådde 1,24 g av en olje som ved NMR-analyse ble vist å bestå av hovedsakelig 2-hydroksymetyl-okso- 1,3-oksatiolan 25 med små mengder butyrsyre og butyrat 24.
Laktonet 25 (0,85 g, 4,16 mmol) ble oppløst i 30 ml toluen og oppløsningen avkjølt til - 78°C. Dibal-H-oppløsning (9 ml, 1,0M på heksaner, 9 mmol) ble tilsatt dråpevis, mens den indre temperatur ble holdt under -70°C under reaksjonen. Efter at tilsetningen var ferdig, ble blandingen omrørt i 0,5 timer ved -78°C. Eddiksyreanhydrid (5 ml, 53 mmol) ble satt til blandingen under kontinuerlig omrøring, ble så tillatt å vende tilbake til romtemperatur over natten. 5 ml vann ble satt til reaksjonsblandingen og den resulterende blanding ble omrørt i en time. 40 mg MgSC«4 ble så tilsatt og blandingen ble omrørt heftig i en time ved romtemperatur. Blandingen ble filtrert, konsentrert og resten flashkromatografert med 20% EtOAc i heksaner hvorved man oppnådde en farveløs væske 26 (0,41 g, 1,86 mmol, 45%) som var en blanding av anomerer på C4-posisjonen.
2-acetoksymetyl-5-acetoksy-l,3-oksatiolanet 26 (0,40 g, 1,82 mmol) ble oppløst i 40 ml 1,2-dikloretan og dertil ble det silylerte cytosin 12 (0,70 g, 2,74 mmol) satt, alt på en gang, ved romtemperatur. Blandingen ble omrørt i 10 minutter og dertil ble det satt en SnCi4-oppløsning (3,0 ml, 1,0M- oppløsning i CH2CI2, 3,0 mmol), dråpevis, ved
romtemperatur. Ytterligere 1,0 ml SnCi4-oppløsning ble tilsatt efter en time. Reaksjonen ble fulgt av TLC. Etter ferdig kobling ble oppløsningen konsentrert, resten triturert med 2 ml trietylamin og underkastet flashkromatografi, først med ren EtOAc og deretter med 20% etanol i EtOAc for å oppnå et gyldent faststoff 27 (0,42 g, 1,55 mmol, 86%).
<*>H NMR: 7,73 (1H, d, H6) J=7,5 Hz; 6,33 (1H, t, H40 J=4,8 Hz; 5,80 (1H, d, H5) J=7,5 Hz; 4,52 (1H, dd, 1H5-) J=5,7 og 12,3 Hz; 4,37 (1H, dd, 1H50 J=3,3 og 12,3 Hz; 3,54 (1H, dd, H20 J=5,4 og 12,0 Hz; 3,10 (1H, dd, 1H3); 2,11 (3H, s, CH3).
5-acetatet av BCH-189 27 (140 mg, 0,52 mmol) ble oppløst i 10 ml vannfri metanol og dertil ble det satt natriummetoksyd (110 mg, 2,0 mmol) i en porsjon. Blandingen ble omrørt ved romtemperatur inntil hydrolysen var ferdig. Hydrolysen tok ca. 1 time og reaksjonen ble fulgt av TLC. Efter ferdig reaksjon ble blandingen konsentrert og resten tatt opp i 2 ml etanol. Etanoloppløsningen ble underkastet kolonnekromatografi ved bruk av først etylacetat, så 20% etanol i EtOAc og man oppnådde et hvitt skum (110 mg, 92%) som viste et NMR- spektrum identisk det til det autentiske BCH-189, 14.
