Claims (9)
- 호흡쇄 경로로서 고에너지 효율의 호흡쇄 경로 및 저에너지 효율의 호흡쇄 경로를 갖는 미생물의 모(母)균주로 부터 창제된, (a) 고에너지 효율의 호흡쇄 경로가 증가되거나 (b) 저에너지 효율의 호흡쇄 경로가 결손되거나 상기 (a) 및 (b)의 특성을 모두 갖는 돌연변이 균주 또는 유전자 재조합 균주인 미생물을 배지에서 배양하여 배지중에 목적 물질이 생성되어 축적되도록 한 후, 목적 물질을 채취함을 특징으로 하는, 미생물을 이용한 목적 물질의 제조 방법.
- 제1항에 있어서, 호흡쇄에 관련된 효소를 암호화하는 유전자의 복사체 수를 증가시키거나 당해 유전자의 발현 조절 서열을 변형시켜 고에너지 효율의 호흡쇄 경로를 증강시킴을 특징으로 하는, 목적 물질의 제조 방법.
- 제1항 또는 제2항에 있어서, 호흡쇄에 관련된 효소를 암호화하는 유전자를 파괴시켜 저에너지 효율의 호흡쇄 경로를 결손시킴을 특징으로 하는, 목적 물질의 제조 방법.
- 제1항 내지 제3항중의 어느 한항에 있어서, 고에너지 효율의 호흡쇄 효소가 SoxM형 옥시다제, bc1 복합체, NDH-1, 또는 이들중 2 또는 3종을 포함하는 것을 특징으로 하는, 목적 물질의 제조 방법.
- 제1항 내지 제4항중의 어느 한항에 있어서, 저에너지 효율의 호흡쇄 효소가 사이토크롬 bd형 옥시다제, NDH-II, 또는 이들 모두를 포함하는 것을 특징으로 하는, 목적 물질의 제조 방법.
- 제1항 내지 제5항중의 어느 한항에 있어서, 미생물에서 SoxM형 옥시다제의 활성을 증강시키고, NDH-II를 결손시킴을 특징으로 하는, 목적 물질의 제조 방법.
- 제1항 내지 제6항중의 어느 한항에 있어서, SoxM형 옥시다제가 사이토크롬 bo형 옥시다제인 것을 특징으로 하는, 목적 물질의 제조 방법.
- 제1항 내지 제7항중의 어느 한항에 있어서, 미생물이 에스체리키아속 세균 및 코리네형 세균으로 이루어진 그룹중에서 선택되는 것을 특징으로 하는, 목적 물질의 제조 방법.
- 제1항 내지 제8항중의 어느 한항에 있어서, 목적 물질이 L-아미노산 및 핵산으로 이루어진 그룹중에서 선택되는 것을 특징으로 하는, 목적 물질의 제조 방법.
