Claims (1)
- 【特許請求の範囲】 (1)植物での外来遺伝子の発現を前記遺伝子の構造コーディング配列を修飾す ることにより改良する方法において、その改良が【配列があります】 および CATAAAからなる群から選択されたポリアデニレーション信号の出現を減少 することを含む方法。 (2)さらに構造コーディング配列内へのATTTA配列の出現を減少という改 良を含む請求項1に記載の方法。 (3)植物において、Bacillus thuringiensisの殺虫効 果のあるタンパク質の発現を強めるために、野生型構造遺伝子配列を修飾するた めの、 a)前記タンパク質を暗号化する配列を残したままで、前記野生型遺伝子に含ま れるポリアデニレーション信号を除去すること、および、 b)前記タンパク質を暗号化する配列を残したままで、前記野生型遺伝子に含ま れるATTTA配列を除去することからなる方法。 (4)さらに前記タンパク質を暗号化する構造遺伝子配列を保持したままで、自 己相補的な配列を除去したり、そのような配列を植物でよく使われるコドンを含 む非自己相補的なDNAと置換することからなる請求項3に記載の方法。 (5)さらにポリアデニレーション信号およびATTTA配列の除去に植物でよ く使われる配列を使用することを含む請求項4に記載の方法。 (6)ポリアデニレーション信号が、【配列があります】およびCATAAAか らなる群から選択される請求項3に記載の方法。 (7)ポリアデニレーション信号が、【配列があります】およびCATAAAか らなる群から選択される請求項4に記載の方法。 (8)ポリアデニレーション信号が【配列があります】およびCATAAAから なる群から選択される請求項5に記載の方法。 (9)植物において、Bacillus thuringiensisの殺虫効 果のあるタンパク質の発現を強めるために、野生型構造遺伝子配列を修飾するた めの方法であって、a)4個より多い連続的なアデニンまたはチミンのヌクレオ チドをもつ前記配列内の領域を同定し、b)前記タンパク質を暗号化する遺伝子 配列を保持したままで、10塩基配列内の2個あるいはそれ以上のポリアデニレ ーション信号をもつ段階a)の領域を、前記信号を除去するために修飾し、そし て、 c)前記タンパク質を暗号化する遺伝子配列を保持したままで、植物の主たるポ リアデニレーション信号、1個より多い重要でないポリアデニレーション信号を 含む連続的な配列および1個より多いATTTA配列を含む連続的な配列を除去 するために、段階a)の領域周辺の15から30塩基の領域を修飾する、 ことからなる方法。 (10)植物の主たるポリアデニレーション信号が、AATAAAおよびAAT AATからなる群から選択される請求項9に記載の方法。 (11)ポリアデニレーション信号が、【配列があります】およびCATAAA からなる群から選択される請求項10に記載の方法。 (12)さらにポリアデニレーション信号およびATTTA配列を除去するのに 、植物でよく使われる配列を使用することを含む請求項11に記載の方法。 (13)Bacillus thuringiensisの殺虫効果をもつタン パク質を暗号化する構造遺伝子で、本質的にポリアデニレーション信号およびA TTTA配列が欠けている遺伝子。 (14)【配列があります】 およびCATAAAからな る群から選択されるポリアデニレーション信号が本質的に欠けている請求項13 に記載の構造遺伝子。 (15)次の配列をもつ、B.t.k.HD−1の殺虫効果をもつタンパク質を 暗号化する請求項13に記載の構造遺伝子:【配列があります】 (16)次の配列をもつ、B.t.k.HD−73の殺虫効果をもつタンパク質 を暗号化する請求項13に記載の構造遺伝子:【配列があります】 (17)次の配列をもつ、B.t.k.HD−1の殺虫効果をもつタンパク質を 暗号化する請求項13に記載の構造遺伝子:【配列があります】 (18)次の配列をもつ、B.t.k.HD−73の殺虫効果をもつタンパク質 を暗号化する請求項13に記載の構造遺伝子:【配列があります】 (19)次の配列をもつ、B.t.k.HD−73の殺虫効果をもつ完全長のタ ンパク質を暗号化する請求項13に記載の構造遺伝子: 【配列があります】 (20)次の配列をもつ、B.t.k.HD−73の殺虫効果をもつ完全長のタ ンパク質を暗号化する請求項13に記載の構造遺伝子: 【配列があります】 (21)次の配列をもつ、B.t.k.HD−73の殺虫効果をもつ完全長のタ ンパク質を暗号化する請求項13に記載の構造遺伝子: 【配列があります】 (22)次の配列をもつ、B.t.t.の殺虫効果をもつタンパク質を暗号化す る請求項13に記載の構造遺伝子:【配列があります】 (23)次の配列をもつ、B.t.entomocidusの殺虫効果をもつタ ンパク質を暗号化する請求項13に記載の構造遺伝子:【配列があります】 (24)次の配列をもつ、P2の殺虫効果をもつタンパク質を暗号化する請求項 13に記載の構造遺伝子:【配列があります】 (25)請求項13に記載の構造遺伝子をもつ植物遺伝子を含む植物形質転換ベ クター。 (26)B.t.k.HD−1のN−末端の610アミノ酸とB.t.k.HD −73のC−末端の567アミノ酸からなる融合タンパク質を暗号化する請求項 13に記載の構造遺伝子配列で、次の配列をもつ遺伝子: 【配列があります】 (27)さらに5個以上の連続的なA+TまたはG+C塩基を含む配列の除去を 含む請求項4に記載の方法。 (28)大多数の植物でよく使われるコドンを含む請求項13に記載の構造遺伝 子配列。 (29)ジャガイモの葉のロールウイルスの外被タンパク質を暗号化する構造遺 伝子で、次の配列をもつ遺伝子:【配列があります】(30)Bacillus thuringiensisの殺虫効果をもつタンパク質を暗号化する構造コ ーディング配列を含む植物のキメラ遺伝子で、この構造コーディング配列の中に あると推定される多くのポリアデニレーション信号を減衰するように修飾されて いる遺伝子。 (31)ポリアデニレーション信号が、【配列があります】およびCATAAA からなる群から選択される請求項30に記載の植物のキメラ遺伝子。 (32)構造コーディング配列が、この構造コーディング配列内の多くのATT TA配列を減衰するようにさらに修飾されている請求項31に記載の植物キメラ 遺伝子。 (33)構造コーディング配列が本質的に、ポリアデニレーション信号とATT TA配列を欠いている請求32に記載の植物キメラ遺伝子。 (34)請求項33に記載の遺伝子を含む形質転換された植物細胞。 (35)大豆、綿、アルファルファ、ナタネ、亜麻、トマト、テンサイ、ヒマワ リ、ジャガイモ、タバコ、トウモロコシ、米および小麦からなる群から選択され る請求項34に記載の形質転換された植物細胞。 (36)請求項34に記載の形質転換された植物細胞を含む植物。 (37)請求項35に記載の植物細胞を含む請求項36に記載の植物。 (38)請求項36に記載の植物により生産された種子。
