- ゲノムが、

(b)場合により、宿主非ヒト哺乳動物カッパ定常領域の上流の1つもしくは複数のヒトIg軽鎖カッパV領域および1つもしくは複数のヒトIg軽鎖カッパJ領域、ならびに/または宿主非ヒト哺乳動物ラムダ定常領域の上流の1つもしくは複数のヒトIg軽鎖ラムダV領域および1つもしくは複数のヒトIg軽鎖ラムダJ領域
を含み、
非ヒト哺乳動物定常領域およびヒト可変領域を有するキメラ抗体またはキメラ重鎖および場合によりキメラ軽鎖のレパートリーを生成することができる非ヒト哺乳動物であって、
DNAが挿入される非ヒト哺乳動物ゲノムは、欠失していない内在性のV(D)J領域を含み、
げっ歯動物であり、
ヒト重鎖DNAの挿入を、非ヒト哺乳動物定常領域と、最後の3'非ヒト哺乳動物J領域との間で行う、非ヒト哺乳動物であるか、
あるいは
ゲノムが、
(a)宿主非ヒト哺乳動物カッパ定常領域の上流の複数のヒトIg軽鎖カッパV領域および1つもしくは複数のヒトIg軽鎖カッパJ領域、ならびに/または宿主非ヒト哺乳動物ラムダ定常領域の上流の複数のヒトIg軽鎖ラムダV領域および1つもしくは複数のヒトIg軽鎖ラムダJ領域、さらに、

ヒトカッパまたはラムダ鎖DNAの挿入を、非ヒト哺乳動物定常領域と最後の3'非ヒト哺乳動物J領域との間で行う、非ヒト哺乳動物細胞。 - ゲノムが、ヒト重鎖およびヒト軽鎖由来の、挿入されたヒト可変領域DNAを含み、
(a) 宿主非ヒト哺乳動物定常領域の上流に複数のヒトIgH V領域、1つもしくは複数のヒトD領域および1つもしくは複数のヒトJ領域が存在し、かつ、
キメラ軽鎖が細胞内に産生されるために、ヒト軽鎖DNAが非ヒト哺乳動物軽鎖定常領域の上流にある、
請求項1または請求項2に記載の哺乳動物または細胞。 - 前記哺乳動物がマウスであり、または前記細胞がマウス細胞であり、ヒト重鎖DNAの挿入を、マウス12番染色体の座標114,667,091と座標114,665,190との間のマウスゲノムで行う(座標は、マウスC57BL/6J系統についてのNCBI m37、2007年4月、ENSEMBL Release 55.37hを指す)、請求項1から3のいずれか一項に記載の細胞または非ヒト哺乳動物。
- 細胞または哺乳動物ゲノムが、完全宿主種特異的抗体の発現を阻止するまたは低下させ るように改変され、場合により、非ヒト哺乳動物ゲノムが、非ヒト哺乳動物VDJ領域またはVJ領域の全部または一部の反転により改変される、請求項1から3のいずれか一項に記載の細胞または哺乳動物。
- キメラである抗体または抗体鎖のレパートリーを生成することができる非ヒト哺乳動物に発生することが可能であるか、またはキメラである抗体または抗体鎖のレパートリーを生成することができる非ヒト哺乳動物組織および臓器に寄与することが可能であるES細胞、造血幹細胞またはiPS細胞であって、前記キメラ抗体または抗体鎖が、非ヒト哺乳動物定常領域およびヒト可変領域を有する、請求項2から5のいずれか一項に記載の細胞。
- 抗体産生細胞である、請求項2から5のいずれか一項に記載の細胞。
- ヒト重鎖およびヒト軽鎖に少なくとも由来する挿入したヒト可変領域DNAを含む、請求項1から7のいずれか一項に記載の細胞または哺乳動物。
- 細胞のゲノムが、別の細胞または生物体に由来する定常領域DNAを含まない、請求項1から8のいずれか一項に記載の細胞または哺乳動物。
- ラットのスイッチ配列を含む、請求項1から9のいずれか一項に記載の細胞または哺乳動物。
- 不死化した、請求項2から10のいずれか一項に記載の細胞。
- 抗体産生細胞を提供するための腫瘍細胞との融合か、直接的な細胞の不死化のいずれかによって作製される、不死化した抗体産生細胞である、請求項11に記載の細胞。
- 所望の抗原に特異的な抗体または抗体重鎖もしくは軽鎖を生成するための方法であって、請求項1、3から5および8から10のいずれか一項に記載の非ヒト哺乳動物を、所望の抗原を用いて免疫化するステップと、抗体または抗体鎖を回収するステップあるいは抗体または重鎖もしくは軽鎖を生成する細胞を回収するステップと
を含む方法。 - 前記非ヒト哺乳動物または細胞が、それぞれマウスまたはマウス細胞である、請求項1から3、および、5から13のいずれか一項に記載の非ヒト哺乳動物、細胞または方法。
- 完全にヒト化した抗体または抗体鎖を生成するための方法であって、請求項1、3から5、8から10および14のいずれか一項に記載の、非ヒト哺乳動物を免疫化するステップと、次いで、ヒト定常領域で抗原に特異的な反応性を示す抗体またはキメラ抗体鎖の非ヒト哺乳動物定常領域を、場合により抗体またはキメラ抗体鎖をコードする核酸を操作することによって置き換えるステップとを含む方法。
