Ciertos anticuerpos tienen disulfuros intercatenarios reducibles, es decir, puentes de cisteína. Péptidos
En una realización, el agente de unión celular es un péptido lineal o cíclico que comprende 4-20, preferiblemente 6-20, residuos de aminoácidos contiguos. En esta realización, se prefiere que un agente de unión celular esté enlazado con un monómero o compuesto dímero de pirrolobenzodiazepina.
El agente de unión celular comprende un péptido que se une a la integrina αvβ6. El péptido puede ser selectivo para αvβ6 sobre XYS.
En una realización, el agente de unión celular comprende el polipéptido A20FMDV-Cys. El A20FMDV-Cys tiene la secuencia: NAVPNLRGDLQVLAQKVARTC. Como alternativa, una variante de la secuencia de A20FMDV-Cys se puede utilizar en donde uno, dos, tres, cuatro, cinco, seis, siete, ocho, nueve o diez residuos de aminoácidos se sustituyen con otro residuo de aminoácido.
En una realización el anticuerpo es un anticuerpo monoclonal; anticuerpo quimérico; anticuerpo humanizado; anticuerpo totalmente humano; o un anticuerpo de una sola cadena. En una realización, el anticuerpo es un fragmento de uno de estos anticuerpos que tienen actividad biológica. Los ejemplos de tales fragmentos incluyen fragmentos Fab, Fab’, F(ab’)2 y Fv.
En estas realizaciones, cada anticuerpo puede estar enlazado a uno o varios grupos dímero de pirrolobenzodiazepina. Las relaciones preferidas de pirrolobenzodiazepina al agente de unión celular se han dado anteriormente.
El anticuerpo puede ser un anticuerpo de dominio (DAB).
En una realización, el anticuerpo es un anticuerpo monoclonal.
Los anticuerpos para su uso en la presente invención incluyen aquellos anticuerpos descritos en el documento WO 2005/082023. Particularmente preferidos son aquellos anticuerpos para antígenos asociados a tumores. Los ejemplos de los antígenos conocidos en la técnica incluyen aquellos antígenos asociados a tumores establecidos en el documento WO 2005/082023. Véase, por ejemplo, páginas 41-55.
Los conjugados de la invención están diseñados para dirigirse a células tumorales por medio de sus antígenos de superficie celular. Los antígenos son habitualmente antígenos de superficie celular normales que son ya sea sobre-expresados o expresado en un número de veces anormal. Idealmente, el antígeno diana se expresa solamente en células proliferativas (preferiblemente células tumorales), esto se observa raramente en la práctica. Como resultado, los antígenos diana usualmente se seleccionan con base en la expresión diferencial entre tejido proliferativo y sano.

Los antígenos asociados a tumores (TAA) son conocidos en la técnica, y se pueden preparar para su uso en la generación de anticuerpos utilizando métodos e información que son bien conocidos en la técnica. En intentos por descubrir dianas celulares eficaces para el diagnóstico y terapia del cáncer, los investigadores han buscado identificar polipéptidos transmembrana o de otro modo asociados a tumores que se expresan específicamente en la superficie de uno o más tipo(s) particular(es) de células cancerígenas en comparación con una o más célula(s) normal(es) no cancerígena(s). A menudo, tales polipéptidos asociados a tumores se expresan más abundantemente en la superficie de las células cancerígenas en comparación con la superficie de las células no cancerígenas. La identificación de tales polipéptidos del antígenos sobre la superficie celular asociados a tumores ha dado lugar a la capacidad de dirigirse específicamente a células cancerígenas para la destrucción por medio de terapias basadas en anticuerpos.

En una realización preferida, los sustituyentes descritos en el presente documento (que incluyen sustituyentes opcionales) están limitados a aquellos grupos que no son reactivos con un agente de unión celular. El enlace al agente de unión celular en el presente caso se forma a partir de la posición N10 del compuesto de PBD a través de un grupo enlazador (que comprende, por ejemplo, L1, L2 y A) al agente de unión celular. Los grupos funcionales reactivos ubicados en otras partes de la estructura de PBD pueden ser capaces de formar enlaces adicionales al agente de unión celular (esto puede ser denominado como reticulación). Los ejemplos de grupos alquilo lineales o ramificados incluyen iso-propilo (C3), isobutilo (C4), sec-butilo (C4), terc-butilo (C4), iso-pentilo (C5), y neo-pentilo (C5).
Un grupo alquilo puede estar opcionalmente interrumpido por uno o más heteroátomos seleccionados de O, N(H) y S. Tales grupos puede ser denominados como "heteroalquilo".
Heteroalquilo C2-20: La expresión "heteroalquilo C2-12" como se usa en el presente documento, se refiere a una porción monovalente obtenida mediante la eliminación de un átomo de hidrógeno de un átomo de carbono de un compuesto hidrocarburo que tiene de 2 a 12 átomos de carbono, y uno o más heteroátomos seleccionados de O, N(H) y S, preferiblemente O y S.
Los ejemplos de grupos heteroalquilo incluyen, aquellos que comprenden una o más unidades de etilenglicol del tipo -(OCH2CH2)-. El terminal de un grupo heteroalquilo puede ser la forma primaria de un heteroátomo, por ejemplo, -OH, -SH o -NH2. En una realización preferida, el terminal -CH3.
Alquenilo C2-12: La expresión "alquenilo C2-12" como se usa en el presente documento, se refiere a un grupo alquilo que tiene uno o más dobles enlaces carbono-carbono.

Heterociclilo C3-20: La expresión "heterociclilo C3-20" como se usa en el presente documento, se refiere a una porción monovalente obtenida mediante la eliminación de un átomo de hidrógeno de un átomo de anillo de un compuesto heterocíclico, cuya porción tiene de 3 a 20 átomos de anillo, de los cuales de 1 a 10 son heteroátomos de anillo.
